Morphological and molecular identification of Sarcocystis falcatula from the emerald toucanet (Aulacorhynchus albivitta) in Colombia.

Sarcocystis spp. are cyst-forming coccidia characterized by a two-host predator-prey life cycle. Sarcocysts are formed in muscles or nervous system of the intermediate host, while sporocysts develop in the small intestine of the definitive host. The intermediate hosts of Sarcocystis falcatula are wild birds. Colombia is one of the countries with the greatest biodiversity of birds, however, there are few studies related to this parasite in wild birds. This study presents the morphological and molecular detection of Sarcocystis falcatula collected from the emerald toucanet (Aulacorhynchus albivi

The Genetic Identification of Numerous Apicomplexan Sarcocystis Species in Intestines of Common Buzzard (Buteo buteo).

The common Buzzard (Buteo buteo) was previously shown to transmit two Sarcocystis species (S. glareoli and S. microti) forming cysts in the brains of rodents. Due to a lack of research, the richness of Sarcocystis species spread by these birds of prey is expected to be much higher. A total of 30 samples of the small intestine of the Common Buzzard were collected in Lithuania and subjected to Sarcocystis species identification based on nested PCR of 28S rRNA and ITS1, following the sequencing of amplified DNA fragments. Six known Sarcocystis spp., S. cornixi, S. glareoli, S. halieti, S. kutkien

White-tailed eagle (Haliaeetus albicilla) as the definitive host of Sarcocystis lutrae in the Czech Republic.

The white-tailed eagle, Haliaeetus albicilla, has been involved in the life cycle of several Sarcocystis species as the intermediate and definitive host. To date, it has been supposed that the eagle might play the role as the definitive host for S. Lutrae, and, herein, we tried to elucidate it based on morphometric and molecular analyses. One out of two eagles harbored oocysts (17.0-17.4 × 11.3-11.9 μm) and sporocysts (11.3-12.3 × 8.3-9.3 μm) in the intestinal mucosa, whose sequences at 18S rRNA, 28S rRNA, ITS1, and cox1 showed similar identity (97.64-100%) to published sequences of S. lutrae

Detection of Sarcocystis spp. and Toxoplasma gondii in swine and detection of DNA of these protozoa in tissues and sausages.

The seroprevalence of Sarcocystis spp. and Toxoplasma gondii was researched in swine raised in Santa Maria, RS, Brazil. Serum samples from 84 pigs from 31 farms were tested using indirect immunofluorescence assay (IFA) for both agents. Additionally, 53 samples of pork sausages and tissues destined for human consumption, including: salami, sausage, black pudding, heart, tongue, brain, and rib muscle, were submitted to PCR to detect DNA for each agent. The frequency of anti-Sarcocystis spp. antibodies was 36.9% (31/84), with titers ranging from 32 to 1024, and 25% (21/84) for anti-T. gondii anti

Molecular identification of Sarcocystis halieti in the muscles of two species of birds of prey from Spain.

BACKGROUND: Members of the genus Sarcocystis are protozoan parasites characterized by a prey-predator two-host life-cycle. Sarcocysts are formed in the muscles or central nervous system of the intermediate host (IH), while sporocysts develop in the small intestine of the definitive host (DH). Various birds of prey have been confirmed to be DH for Sarcocystis spp. Three Sarcocystis species, S. wobeseri, S. halieti and S. falcatula, have been identified in the muscles of birds of prey, of which the latter are known to be pathogenic and can cause encephalitis in various birds. The aim of this stu

Serosurvey of Toxoplasma gondii, Neospora caninum and Sarcocystis neurona in raptors and risk factor analysis.

Raptors are carnivorous birds with great hunting ability. Toxoplasma gondii, Neospora caninum and Sarcocystis spp. are intracellular Apicomplexan protozoans which infect a wide range of intermediate hosts, including birds. The aims of this study were to evaluate the serological reactivity of captive raptors serum to T. gondii, N. caninum and S. neurona antigens and identify possible risk factors associated with the infection. From August 2014 to September 2015, blood samples from 72 raptors were collected and serum samples were tested by immunofluorescence antibody test (IFAT). Antigen slides

First molecular detection and characterization of Sarcocystis species in slaughtered cattle in North-West Tunisia.

Sarcocystis spp. is one of the most prevalent foodborne parasites infecting both animals and humans. Consumption of raw or undercooked infected meat is a risk factor of human intestinal sarcocystosis. The present study aimed to estimate the prevalence of Sarcocystis species infecting slaughtered Tunisian cattle in North-West Tunisia (Béja governorate). DNA was extracted from 150 beef meat samples and a PCR-restriction fragment length polymorphism was used for identification. The overall infection prevalence of Sarcocystis spp. was 38% (57/150). Two species were identified, namely S. hominis (2

Diagnosis and treatment of Sarcocystis neurona-induced myositis in a free-ranging California sea lion.

CASE DESCRIPTION: An underweight, lethargic adult female California sea lion (Zalophus californianus) became stranded along the California shore and was captured and transported to a rehabilitation hospital for assessment and care. CLINICAL FINDINGS: Initial physical assessment revealed the sea lion was lethargic and in poor body condition. Active myositis was diagnosed on the basis of concurrent elevations in activities of alanine aminotransferase and creatine kinase detected during serum biochemical analysis. Infection with Sarcocystis neurona was diagnosed after serologic titers increased

Rhinitis and disseminated disease in a ferret (Mustela putorius furo) naturally infected with Sarcocystis neurona.

Naturally occurring Sarcocystis neurona infection in a ferret (Mustela putorius furo) with rhinitis and disseminated disease are described for the first time. The ferret exhibited severe rhinitis with intra-lesional S. neurona merozoites and schizonts. Diagnosis was confirmed immunohistochemically by staining with S. neurona-specific antibodies, and by phylogenetic analyses of conserved and variable portions of nuclear ribosomal DNA. On the basis of intense schizogony in the nasal mucosa, we propose the possibility of an olfactory nerve pathway route of infection for S. neurona meningoencephal

Abortion and other signs of disease in cows experimentally infected with Sarcocystis fusiformis from dogs.

Six five- to eight-year-old multiparous cows were given oral inoculations of sporocysts of Sarcocystis fusiformis five to six and one-half months after field breeding. All cows became ill. Clinical signs of infection included muscle tremors, hypersalivation, reduced consumption of feed, weight loss, excessive shedding of hair, elevated temperature, reduced packed cell volumes and total red blood cell counts, and abortion. One cow aborted on day 41 after inoculation, and another aborted on day 69. A moribund cow was sacrificed, and a normal fetus was found in utero. Two cows did not produce cal

SARCOCYSTIS CANADENSIS N. SP. (PROTOZOA: SARCOCYSTIDAE) FROM LIMB MUSCLES OF THE CANADA LYNX (LYNX CANADENSIS).

Most species of endotherms and some ectotherms harbor muscle cysts (sarcocysts) of 1 or more species of apicomplexan parasites in the genus Sarcocystis. We report a novel species discovered in the muscles of a Canada lynx (Lynx canadensis). We found microscopic sarcocysts in limb muscles of 1 of 3 endangered Canada lynx from Minnesota. The sarcocyst wall was relatively thin (<2 µm) and had short, stubby protrusions with apical thickening. We characterized these further by sequencing the 18S, 28S, and ITS1 portions of nuclear ribosomal RNA genes. The sequences of each marker perfectly matched t

Differential expression of host invasion-associated genes by Sarcocystis calchasi in intermediate versus definitive hosts.

Sarcocystis calchasi is a pathogenic apicomplexan parasite affecting avian species of several orders. To complete its heteroxenous life cycle, S. calchasi infects a wide range of avian intermediate hosts and accipitriform raptors serve as definitive hosts. The mechanism of invasion into host cells is largely understood in other apicomplexan parasites, particularly Toxoplasma gondii, which also belongs to the family of Sarcocystidae. However, Sarcocystis species exhibit several distinguishing features in their life cycles and in their secretory organelles. The composition of secretory pathogene

Molecular detection of Sarcocystis neurona in cerebrospinal fluid from 210 horses with suspected neurologic disease.

An ante-mortem diagnosis of equine protozoal myeloencephalitis (EPM) is presently based on clinical presentation, immunodiagnostics performed on serum and cerebrospinal fluid (CSF), and ruling out other neurological disorders. Molecular techniques introduce a novel and promising approach for the detection of protozoal agents in CSF. Hypothesizing that real-time PCR (rtPCR) can be a useful complement to EPM diagnostics, 210 CSF samples from horses suspected of neurological disease with EPM included as a differential diagnosis were tested using rtPCR to detect Sarcocystis neurona DNA and immunod

Atypical fatal sarcocystosis associated with Sarcocystis neurona in a White-nosed coati (Nasua narica molaris).

The protozoan parasite Sarcocystis neurona is an important cause of disease in horses (equine protozoal myeloencephalitis, EPM) and marine mammals. Isolated reports of clinical EPM-like disease have been documented in a zebra, raccoon, domestic cat, domestic dog, ferret, skunk, mink, lynx, red panda and fisher. The predominant disease is encephalomyelitis associated with schizonts in neural tissues. Here, we report highly disseminated sarcocystosis, in many tissues of a captive White-nosed coati (Nasua narica molaris). The 14year old, neutered male coati was euthanized due to progressive weakn

Prevalance, Morphology, and Molecular Characterization of Sarcocystis heydorni Sarcocysts from Cattle (Bos Taurus) in China.

Cattle are intermediate hosts for 2 zoonotic species of Sarcocystis, Sarcocystis hominis and Sarcocystis heydorni. Here we report S. heydorni from cattle for the first time in China. Sarcocysts of S. heydorni were found in muscle from 173 of 1,630 (10.6%) cattle in abattoirs (9.7% in skeletal muscles, 3.4% esophagus, 2.5% diaphragm, and 0.1% tongue; heart muscle was negative). By means of light microscopy, S. heydorni sarcocysts were thin-walled (<1 μm). Using transmission electron microscopy, the sarcocyst wall had short (0.3-0.5 × 0.5-0.9 μm) stubby protrusions, the tips of which contained e

Selective inhibition of Sarcocystis neurona calcium-dependent protein kinase 1 for equine protozoal myeloencephalitis therapy.

Sarcocystis neurona is the most frequent cause of equine protozoal myeloencephalitis, a debilitating neurological disease of horses that can be difficult to treat. We identified SnCDPK1, the S. neurona homologue of calcium-dependent protein kinase 1 (CDPK1), a validated drug target in Toxoplasma gondii. SnCDPK1 shares the glycine "gatekeeper" residue of the well-characterized T. gondii enzyme, which allows the latter to be targeted by bumped kinase inhibitors. This study presents detailed molecular and phenotypic evidence that SnCDPK1 can be targeted for rational drug development. Recombinant

[Sarcocystis cruzi (Protozoa: Apicomplexa: Sarcocystiidae) infection in european bison (Bison bonasus) from Białowieza Forest, Poland].

Samples of oesophagus, diaphragm and heart muscles were taken from one European bison from Białowieza Forest during seasonal European bison elimination in 2008. Five gram of each muscle was examined by staining small samples in 0.2% aqueous solution of methylene blue. After that they were placed between compressor glasses and examined under a dissecting microscope in order to detect Sarcocystis sp. infection. All sarcocysts observed were counted. The 5-g sample of the heart muscle yielded a total of 756 sarcocysts while that of the diaphragm contained 107 of them and that of the oesophagus--89

Protozoan infections (Toxoplasma gondii, Neospora caninum and Sarcocystis spp.) in sheep and goats: recent advances.

The protozoan parasite Toxoplasma gondii is a serious cause of fetal mortality in sheep and goats. Oocysts, the parasite stage responsible for initiating infection, are produced following a primary infection in cats. A primary infection in pregnant sheep and goats can establish a placental and fetal infection which may result in fetal death and resorption, abortion or stillbirth. Diagnosis is aided by the clinical picture, the presence of characteristic small white necrotic foci in placental cotyledons, the possible presence of a mummified fetus and on fetal serology and histopathology.
