Genetic Polymorphism and Natural Selection of Apical Membrane Antigen-1 in Plasmodium falciparum Isolates from Vietnam.

Apical membrane antigen-1 of Plasmodium falciparum (PfAMA-1) is a leading malaria vaccine candidate antigen. However, the genetic diversity of pfama-1 and associated antigenic variation in global P. falciparum field isolates are major hurdles to the design of an efficacious vaccine formulated with this antigen. Here, we analyzed the genetic structure and the natural selection of pfama-1 in the P. falciparum population of Vietnam. A total of 37 distinct haplotypes were found in 131 P. falciparum Vietnamese isolates. Most amino acid changes detected in Vietnamese pfama-1 were localized in the ec

Genetic and Psychosocial Predictors of Aggression: Variable Selection and Model Building With Component-Wise Gradient Boosting.

Rationale: Given datasets with a large or diverse set of predictors of aggression, machine learning (ML) provides efficient tools for identifying the most salient variables and building a parsimonious statistical model. ML techniques permit efficient exploration of data, have not been widely used in aggression research, and may have utility for those seeking prediction of aggressive behavior. Objectives: The present study examined predictors of aggression and constructed an optimized model using ML techniques. Predictors were derived from a dataset that included demographic, psychometric and g

Selection of drug resistance-mediating Plasmodium falciparum genetic polymorphisms by seasonal malaria chemoprevention in Burkina Faso.

Seasonal malaria chemoprevention (SMC), with regular use of amodiaquine plus sulfadoxine-pyrimethamine (AQ/SP) during the transmission season, is now a standard malaria control measure in the Sahel subregion of Africa. Another strategy under study is SMC with dihydroartemisinin plus piperaquine (DP). Plasmodium falciparum single nucleotide polymorphisms (SNPs) in P. falciparum crt (pfcrt), pfmdr1, pfdhfr, and pfdhps are associated with decreased response to aminoquinoline and antifolate antimalarials and are selected by use of these drugs. To characterize selection by SMC of key polymorphisms,

Whole-Genome Resequencing Reveals Genetic Variation and Selection Signals in Fusarium acuminatum Causing Astragalus Root Rot.

Astragalus root rot is a soil-borne disease primarily caused by Fusarium spp., which severely hampers the sustainable development of the Astragalus industry. F. acuminatum is a predominant pathogen causing this disease. To elucidate the genetic variation and adaptive evolutionary characteristics of F. acuminatum from different geographical origins, this study conducted whole-genome resequencing analysis on 28 isolates of F. acuminatum collected from four major Astragalus production regions. Approximately 124.9 Gb of high-quality sequencing data were obtained, and a large number of single-nucle

Genetic diversity and natural selection of Plasmodium falciparum Pf41 vaccine candidate in clinical isolates from Senegal.

The merozoite surface antigen Pf41 was previously identified among the top 10 of new malaria vaccine candidates. Pf41 possesses red blood cell binding regions and conserved domains. We used population genetics approaches to determine the genetic diversity and to identify regions under balancing selection for the potential inclusion of Pf41 as candidate in a multicomponent vaccine. We screened 116 clinical isolates collected from different administrative regions in Senegal for P. falciparum positivity, Pf41 amplification and sequencing. We analyzed Pf41 sequences for polymorphism, natural selec

Demographic History, Genetic Load, and the Efficacy of Selection in the Globally Invasive Mosquito Aedes aegypti.

Aedes aegypti is the main vector species of yellow fever, dengue, Zika, and chikungunya. The species is originally from Africa but has experienced a spectacular expansion in its geographic range to a large swath of the world, and the demographic effects of which have remained largely understudied. In this report, we examine whole-genome sequences from six countries in Africa, North America, and South America to investigate the demographic history of the spread of A. aegypti into the Americas and its impact on genomic diversity and deleterious genetic load. In the Americas, we observe patterns

Development of ptxD/Phi as a new dominant selection system for genetic manipulation in Cryptococcus neoformans.

Cryptococcus neoformans is a globally distributed pathogenic fungus posing a significant threat to immunocompromised individuals, particularly those with HIV/AIDS. Effective genetic manipulation tools are essential for understanding its biology and developing new therapies. However, current genetic tools, including the variation of versatile selectable markers, are limited. This study develops and validates the phosphite dehydrogenase gene (ptxD)/phosphite (Phi) selection system as a non-antibiotic selectable marker for genetic manipulation in C. neoformans. A codon-optimized ptxD gene from Ps

Genetic selection for reduced health treatment costs in Holstein cows: implications from a long-term study.

The objective of this study was to estimate genetic parameters of health treatment cost of Holstein cows from producer-recorded health treatments in 8 herds over an 8-yr period of time. Genetic parameters of health treatment cost were estimated in first (n = 2,214), second (n = 1,487) and third (n = 800) parities of US Holstein cows. The health treatments were uniformly defined and consistently recorded by 8 high-performance dairy herds in Minnesota from 2008 to 2015. A fixed treatment cost was assigned to 14 types of health treatments, and the cost included the mean veterinary expense obtaine

Genetic selection for crescent formation yields mouse strain with rapidly progressive glomerulonephritis and small vessel vasculitis.

We have established a recombinant inbred strain of mouse named spontaneous crescentic glomerulonephritis-forming mouse/Kinjoh or SCG/Kj. Mice of this strain spontaneously develop rapidly progressive glomerulonephritis. This strain of mice was derived from (BXSB/Mp x MRL/Mp-lpr/lpr)F1 hybrid mice by brother x sister mating coupled with repeated histopathologic selection for breeding of mice whose parents had the highest frequency of crescent formation in the kidneys. In this strain of mice, nephritis appears earlier and is more rapidly progressive than in any other murine model of systemic lupu

Beyond Precision: A Multidimensional Framework for Selecting Genetic Medicine Platforms.

Gene therapy is undergoing continued clinical translation and technological development. This progress has been marked by regulatory approvals and broadened therapeutic indications across genetic, metabolic, and oncologic diseases and disorders. The field has evolved over decades from early viral-mediated gene addition to approaches capable of targeted editing, regulation, or replacement of genetic information. These systems include base and prime editors, epigenetic modulators, CRISPR-Cas, RNA therapeutics and programmable integration platforms. When paired with increasingly sophisticated vir

A Genetically-Engineered Thyroid Gland Built for Selective Triiodothyronine Secretion.

Thyroid hormones (thyroxine, T4, and triiodothyronine, T3) are indispensable for sustaining vertebrate life, and their deficiency gives rise to a wide range of symptoms characteristic of hypothyroidism, affecting 5-10% of the world's population. The precursor for thyroid hormone synthesis is thyroglobulin (Tg), a large iodoglycoprotein consisting of upstream regions I-II-III (responsible for synthesis of most T4) and the C-terminal CholinEsterase-Like (ChEL) domain (responsible for synthesis of most T3, which can also be generated extrathyroidally by T4 deiodination). Using CRISPR/Cas9-mediate

Genetic background of selected hyperuricemia causing gout with pediatric onset.

Elevated serum uric acid levels are the essential pathophysiology of gout. Although gout rarely develops in childhood, chronic persistent hyperuricemia can induce precipitation and deposition of sodium urate crystals, leading to the development of gout. Hyperuricemia is caused by increased uric acid production and/or decreased uric acid excretion capacity of the kidneys and/or intestinal tract. Increased production of uric acid, the final metabolite of purine, is associated with an increase of phosphoribosyl pyrophosphate, the key compound in the purine synthesis pathways, as observed in hypox
