AIM OF THE STUDY: Sensory gating is a human higher cognitive function that serves to suppress excessive sensory information and prevent brain overactivity. To elucidate this function, a paired-pulse stimulation paradigm has been used while recording electroencephalography (EEG), and evaluated as an amplitude ratio of responses to a second stimulus (S2) over responses to the first stimulus (S1). The present study investigated the effects of the inter-stimulus interval (ISI) and inter-trial interval (ITI) on somatosensory gating using somatosensory-evoked potentials (SEPs).

Impaired cerebral inhibition is commonly observed in neurodevelopmental disorders and may represent a vulnerability factor for their development. The hippocampus plays a key role in inhibition among adults and undergoes significant and rapid changes during early brain development. Therefore, the structure represents an important candidate region for early identification of pathology that is relevant to inhibitory dysfunction. To determine whether hippocampal function corresponds to inhibition in the early postnatal period, the present study evaluated relationships between hippocampal activity

Neurobiological evidence indicates that sensory processing is dynamically shaped by the physiological state of the organism. In chemical communication, stimulus effectiveness depends on ligand-receptor interactions together with environmental transport, sensory access, and endocrine, autonomic, vascular and neural conditions that regulate stimulus availability and system responsiveness. This review develops physiological gating as a receiver-state model of chemosensory effectiveness. Physiological gating describes coordinated regulation across three interacting levels: stimulus access and rece

BACKGROUND: Impairment in computerized dynamic posturography scores has been documented in Huntington disease patients. Tetrabenazine is approved to treat chorea in Huntington disease, but its effect on posturography scores, and balance in general, is unknown.
MATERIALS AND METHODS: We designed a study to test computerized dynamic posturography performance while taking tetrabenazine and after stopping tetrabenazine for at least three days.
RESULTS: 10 Huntington disease patients were studied both ON and OFF tetrabenazine. The composite score was statistically different between ON and OFF con

Schizophrenic patients show loss of the normal inhibition of the acoustic startle response (ASR) when the startle stimulus is preceded by a weak prepulse stimulus. In rats experimentally induced to have locally supersensitive dopamine receptors within forebrain dopamine terminal regions, we find a similar deficit in "prepulse inhibition" of the ASR. Our results suggest that there are intriguing parallels between the aberrant sensory gating of the ASR demonstrated by schizophrenics and that demonstrated by rats during stimulation of supersensitive brain dopamine receptors. Thus, prepulse inhibi

Animals' response to a stimulus in one sensory modality is usually influenced by other modalities.1 One important type of multisensory integration is the cross-modal modulation, in which one sensory modality modulates (typically inhibits) another. Identification of the mechanisms underlying cross-modal modulations is crucial for understanding how sensory inputs shape animals' perception and for understanding sensory processing disorders.2,3,4 However, the synaptic and circuit mechanisms that underlie cross-modal modulation are poorly understood. This is due to the difficulty of separating cros

BACKGROUND/AIM: Sleep deprivation disrupts prepulse inhibition of acoustic startle reflex and can be used to mimic psychosis in ex- perimental animals. On the other hand, it is also a model for other disorders of sensory processing, including migraine. This study aims to assess the effects of sodium valproate, a drug that is used in a variety of neuropsychiatric disorders, on normal and disrupted sensorimotor gating in rats.
MATERIALS AND METHODS: Sixty-two Wistar albino rats were randomly distributed into 8 groups. Subchronic and intraperitoneal sodium valproate were administrated to the sle

Evidence of normalized auditory P50 suppression with acute nicotine in schizophrenia has supported the contention that elevated smoking rates in this disorder may be an attempt to correct a nicotinic receptor pathophysiology that may underly impaired sensory gating in these patients. There is very little information regarding the neurochemical or genetic pathways through which nicotine regulates P50 amplitude and its suppression in human studies. In a randomized, double-blind, placebo-controlled design with 24 non-smokers, this study examined the influence of TaqIA dopamine D2 receptor gene po

BACKGROUND: Perioperative benzodiazepines increase the risk of sensory hypersensitivity and agitation during recovery, but the neural mechanisms remain unclear. Auditory gating filters redundant information to prevent network overload. We hypothesise that benzodiazepines impair auditory gating during recovery, leading to heightened auditory responsiveness.
METHODS: Simultaneous electroencephalography, electromyography, and behavioural analyses assessed the effects of remimazolam, a novel benzodiazepine, on arousal states. Multi-region microelectrodes and Neuropixels probes recorded cortical a

Although current knowledge attributes movement disorders to a dysfunction of the basal ganglia-motor cortex circuits, abnormalities in the peripheral afferent inputs or in their central processing may interfere with motor program execution. We review the abnormalities of sensorimotor integration described in the various types of movement disorders. Several observations, including those of parkinsonian patients' excessive reliance on ongoing visual information during movement tasks, suggest that proprioception is defective in Parkinson's disease (PD). The disturbance of proprioceptive regulatio

Auditory P50 suppression, which is assessed using a paired auditory stimuli (S1 and S2) paradigm to record the P50 mid-latency evoked potential, is assumed to reflect sensory gating. Recently, P50 suppression deficits were observed in patients with anxiety disorders, including panic disorder, post-traumatic stress disorder and obsessive-compulsive disorder, as we previously reported. The processes of fear conditioning are thought to play a role in the pathophysiology of anxiety disorders. In addition, we found that the P50 sensory gating mechanism might be physiologically associated with fear

Prepulse inhibition (PPI) is an operational measure of sensorimotor gating that is reduced in several neuropsychiatric disorders that are characterized by deficits in inhibition or gating of intrusive or irrelevant stimuli. Clinically, panic disorder (PD) patients have been described as having difficulties in inhibition of responding to sensory and cognitive events. Because such difficulties may be due to failures in early stages of information processing, we examined PPI in patients with PD. Acoustic startle reactivity, habituation, and PPI (30-, 60-, 120-, 240-, and 2,000-ms interstimulus in

Tic related disorders affect 4-20% of the population, mostly idiopathic, can be grouped in a wide spectrum of severity, where the most severe end is Tourette Syndrome (TS). Tics are arrhythmic hyperkinesias to whom execution the subject is forced by a "premonitory urge" that can be classified as sensory tic, just-right experience or urge without obsession. If an intact volitional inhibition allows patients to temporarily suppress tics, a lack or deficit in automatic inhibition is involved in the genesis of the disorder. Studies have assessed the presence of intrinsic microscopic and macroscopi

Purpose The goal of this study was to assess whether peripheral auditory sensitivity in frequency regions above 8 kHz is related to central inhibitory function, as measured through a sensory gating paradigm, in normal-hearing adults with tinnitus (TINN) and without tinnitus (NTINN). The contribution of gating processes and peripheral sensitivity in extended high frequencies to tinnitus severity was evaluated via a hierarchical multiple regression method. Method Cortical auditory evoked potentials (CAEPs) were recorded in response to pairs of tones in normal-hearing adults without tinnitus, NTI

INTRODUCTION: Impaired inhibitory gating is a sensory processing symptom of Parkinson's disease (PD) that may be associated with bradykinesia and motor inhibition. Acute stress impairs inhibitory gating in healthy adults; however, it is unclear how stress impacts inhibitory gating in people with PD.
METHODS: Using a Socially Evaluated Cold Pressor (SECP) to induce acute physical stress, inhibitory gating was assessed through electroencephalography (EEG) in eight individuals diagnosed with PD and 11 age- and gender-matched healthy older adults (HOAs) by measuring the p50 ratio during an audito

End-organs such as the bladder rely on a delicate balance between internal urgency and voluntary restraint. However, the specific spinal circuits coordinating these functions remain poorly defined. Here, we identify a genetically defined population of lumbosacral spinal interneurons marked by Trpc4 expression that gate bladder sensory input and regulate micturition reflexes. Single-cell transcriptomics and in situ physiology reveal molecularly distinct Trpc4 + subtypes. Functional manipulation reveals that Trpc4 + neurons are essential for coordinating bladder-sphincter activity and gating vis

During locomotion, reflex responses to sensory stimulation are usually modulated and may even be reversed. This is thought to be the result of phased inhibition, but the neurons responsible are usually not known. When the hatchling Xenopus tadpole swims, responses to cutaneous stimulation are modulated. This occurs because sensory pathway interneurons receive rhythmic glycinergic inhibition broadly in phase with the motor discharge on the same side of the trunk. We now describe a new whole-cell recording preparation of the Xenopus tadpole CNS. This has been used with neurobiotin injection to d

BACKGROUND: Functional neurological disorder (FND) is increasingly understood as a multinetwork disorder. Deficits in subcortical sensorimotor gating, indexed by reduced prepulse inhibition (PPI) of the startle reflex, are reported in FND and may reflect impaired filtering and contextual weighting of sensory input. However, the neural correlates linking impaired gating to intrinsic network dysfunction remain unclear.
METHODS: We examined the relationship between PPI and resting-state functional connectivity in 38 patients with functional movement disorder (FMD) and 39 healthy controls (HC). P

Thalamocortical (TC) projections provide the major pathway for ascending sensory information to the mammalian neocortex. Arrays of these projections form synaptic inputs on thalamorecipient neurons, thus contributing to the formation of receptive fields (RFs) in sensory cortices. Experience-dependent plasticity of RFs persists throughout an organism's life span but in adults requires activation of cholinergic inputs to the cortex. In contrast, synaptic plasticity at TC projections is limited to the early postnatal period. This disconnect led to the widespread belief that TC synapses are the pr
