Alarm pheromone activates posterior medial amygdala-to-bed nucleus of the stria terminalis projections in male rats.

Animals communicate using olfactory signals called pheromones. Alarm pheromones are evolutionarily conserved and alert conspecifics to danger. In rats, a mixture of hexanal and 4-methylpentanal was identified as the main component that evokes anxiety responses by activating the anterior bed nucleus of the stria terminalis (aBNST). In addition, the posteroventral medial amygdala (pvMeA) was proposed to transmit the alarm pheromone signal to the aBNST. Here, we assessed whether the binary mixture activates direct pvMeA-to-aBNST projections or indirect projections via other MeA subdivisions in ma

Reduced oxytocin signaling in the dBNST drives the transition from acute pain to persistent anxiety.

Transient sensory experiences can trigger sustained emotional disturbances, yet the underlying neural mechanisms remain unclear. Here, we show that acute pain induces persistent anxiety in male mice, independent of ongoing nociceptive input, through reduced oxytocin signaling in the dorsal bed nucleus of the stria terminalis (dBNST). Reactivating oxytocin receptors (Oxtrs) in the dBNST markedly alleviated anxiety-like behaviors following pain resolution. Mechanistically, chemogenetic inhibition of somatostatin-expressing (SST) neurons in the dBNST (dBNSTSST neurons) abolished oxytocin's anxiol

Neuroticism/Negative Emotionality Is Associated with Increased Reactivity to Uncertain Threat in the Bed Nucleus of the Stria Terminalis, Not the Amygdala.

Neuroticism/negative emotionality (N/NE)-the tendency to experience anxiety, fear, and other negative emotions-is a fundamental dimension of temperament with profound consequences for health, wealth, and well-being. Elevated N/NE is associated with a panoply of adverse outcomes, from reduced socioeconomic attainment to psychiatric illness. Animal research suggests that N/NE reflects heightened reactivity to uncertain threat in the bed nucleus of the stria terminalis (BST) and central nucleus of the amygdala (Ce), but the relevance of these discoveries to humans has remained unclear. Here we us

Social anxiety is associated with BNST response to unpredictability.

BACKGROUND: Anxiety disorders are highly prevalent and cause substantial suffering and impairment. Whereas the amygdala has well-established contributions to anxiety, evidence from rodent and nonhuman primate models suggests that the bed nucleus of the stria terminalis (BNST) may play a critical, and possibly distinct, role in human anxiety disorders. The BNST mediates hypervigilance and anticipatory anxiety in response to an unpredictable or ambiguous threat, core symptoms of social anxiety, yet little is known about the BNST's role in social anxiety. METHODS: Functional magnetic resonance i

Activity alterations in the bed nucleus of the stria terminalis and amygdala during threat anticipation in generalized anxiety disorder.

Sustained anticipatory anxiety is central to Generalized Anxiety Disorder (GAD). During anticipatory anxiety, phasic threat responding appears to be mediated by the amygdala, while sustained threat responding seems related to the bed nucleus of the stria terminalis (BNST). Although sustained anticipatory anxiety in GAD patients was proposed to be associated with BNST activity alterations, firm evidence is lacking. We aimed to explore temporal characteristics of BNST and amygdala activity during threat anticipation in GAD patients. Nineteen GAD patients and nineteen healthy controls (HC) underw

Neuronal activity in the bed nucleus of the stria terminalis in a rat model for obsessive-compulsive disorder.

In search of a new potential target for deep brain stimulation in patients with obsessive-compulsive disorder (OCD), we evaluated the single-cell activity of neurons in the bed nucleus of the stria terminalis (BST) in urethane-anesthetized rats in an animal model for OCD, the schedule-induced polydipsia (SIP) model, and compared this to the BST activity in control rats and to a third group of rats which were introduced in the model but did not develop the SIP, and thus were considered resistant. We compared the firing rate and firing pattern of BST neurons between these groups, between hemisph

Forebrain hypoplasia following acute prenatal ethanol exposure: quantitative analysis of effects on specific forebrain nuclei.

The effects of acute prenatal exposure to ethanol on the volumes and neuronal populations of selected forebrain nuclei of postnatal animals have been examined in a mouse model. Pregnant mice were exposed to ethanol (25% ethanol, either two doses at 0.015 ml/g separated by 4 hrs, or a single dose at 0.03 ml/g) on the 8th gestational day and the cytoarchitecture of the basal forebrain, paleo- and neocortex examined quantitatively at P15. Significant reductions in the volume and total number of neurons of ventromedial and central forebrain structures, such as the olfactory bulb, septal nuclei, di

Heterogeneous expression of transketolase in rat brain.

Transketolase (TK; EC 2.2.1.1) is a key pentose phosphate shunt enzyme that plays an important role in the production of reducing equivalents and pentose sugars. TK activity declines in the brains of patients with Alzheimer's disease or Wernicke-Korsakoff syndrome, as well as in thiamine-deficient rats. Understanding the role of TK in the pathophysiology of these neurodegenerative conditions requires knowledge of its regional, cellular, and subcellular distribution within the brain. The current study employed in situ hybridization and immunocytochemistry to examine the distribution of TK mRNA

A regulatory role of the medial septum in the chloroquine-induced acute itch through local GABAergic system and GABAergic pathway to the anterior cingulate cortex.

Itch, a common somatic sensation, serves as a crucial protective system. Recent studies have unraveled the neural mechanisms of itch at peripheral, spinal cord as well as cerebral levels. However, a comprehensive understanding of the central mechanism governing itch transmission and regulation remains elusive. Here, we report the role of the medial septum (MS), an integral component of the basal forebrain, in modulating the acute itch processing. The increases in c-Fos+ neurons and calcium signals within the MS during acute itch processing were observed. Pharmacogenetic activation manipulation

Anterior Limb of Internal Capsule and Bed Nucleus of Stria Terminalis Stimulation for Gilles de la Tourette Syndrome with Obsessive-Compulsive Disorder in Adolescence: A Case of Success.

Gilles de la Tourette syndrome (GTS) is a neurobehavioral disorder comprising motor and vocal tics. In most cases it is associated with other disorders such as obsessive-compulsive disorder (OCD). In refractory cases deep brain stimulation (DBS) is a valid treatment option. This paper describes the case of a 15-year-old adolescent with an extremely refractory GTS with associated OCD. The patient developed catatonia associated with OCD, which partially remitted after electroconvulsive therapy. At the peak of the disease the Yale Global Tic Severity Scale (YGTSS) was 100 and the patient required

Protracted withdrawal from alcohol and drugs of abuse impairs long-term potentiation of intrinsic excitability in the juxtacapsular bed nucleus of the stria terminalis.

The juxtacapsular bed nucleus of the stria terminalis (jcBNST) is activated in response to basolateral amygdala (BLA) inputs through the stria terminalis and projects back to the anterior BLA and to the central nucleus of the amygdala. Here we show a form of long-term potentiation of the intrinsic excitability (LTP-IE) of jcBNST neurons in response to high-frequency stimulation of the stria terminalis. This LTP-IE, which was characterized by a decrease in the firing threshold and increased temporal fidelity of firing, was impaired during protracted withdrawal from self-administration of alcoho

Effects of vasopressin and related peptides on neurons of the rat lateral septum and ventral hippocampus.

The effects of vasopressin (VP), VP fragments and propressophysin glycopeptide on neuronal activities in the septum-hippocampus complex of rats were studied in vitro and in vivo. The frequency of the hippocampus theta rhythm in Brattleboro rats homozygous for diabetes insipidus was significantly slower than that of heterozygous litter mates and normal rats. Intracerebroventricular micro-injection of des-glycine-amide vasopressin corrected for several hours the frequency deficit of the theta rhythm in the homozygous Brattleboro rats and the centrally administered VP slowed down theta rhythm in

Control of the neuronal rhythmic bursts in the septal pacemaker of theta-rhythm: effects of anaesthetic and anticholinergic drugs.

The drugs, described as blocking the high-frequency (pentobarbital) or low-frequency (scopolamine, atropine) theta rhythm of the hippocampal electroencephalogram, were tested upon the rhythmically bursting septal cells. Three groups of chronic, unanaesthetized rabbits were used for the experiments: with intact septum; with septohippocampal disconnection; with complete basal undercutting of the septum, depriving it of ascending brainstem influences (MFB lesion). While the frequency and other parameters of theta bursts did not differ in the first two groups (5.2-5.5 Hz), in MFB-lesioned septum t

[Acetylcholinesterase activity in the brains of rats in a model of alcoholic embryopathy].

Acetylcholinesterase (ACE) activity was studied by the Karnovsky-Ruts method from the 5th to the 30th day in the brain of young rats born to chronically alcoholized animals receiving ethanol for 3 to 5 months prior to conception as well as during pregnancy and breast feeding. Alcohol was found to induce deviations in the formation of the enzyme in the adjacents nucleus and Broca's diagonal strip on days 5 and 20 of the postnatal development, respectively. It is assumed that the above deviations are involved in manifestations of the alcoholic syndrome of the fetus.

Distribution of mRNAs encoding transforming growth factors-beta1, -2, and -3 in the intact rat brain and after experimentally induced focal ischemia.

Transforming growth factors-beta1 (TGF-beta1), -2, and -3 form a small group of related proteins involved in the regulation of proliferation, differentiation, and survival of various cell types. Recently, TGF-betas were also demonstrated to be neuroprotective. In the present study, we investigated their distribution in the rat brain as well as their expression following middle cerebral artery occlusion. Probes were produced for all types of TGF-betas, and in situ hybridization was performed. We demonstrated high TGF-beta1 expression in cerebral cortex, hippocampus, central amygdaloid nucleus,

Neuropeptide Y: distribution of immunoreactivity and quantitative analysis in diencephalic structures and cerebral cortex of dwarf hamsters under different photoperiods.

The distribution of neuropeptide Y-like immunoreactivity (NPY-LI) was investigated by immunohistochemistry and radioimmunoassay (RIA) in the brain of the Djungarian hamster (Phodopus sungorus) held under either long or short photoperiods. In the diencephalic and telencephalic structures studied, distinct patterns of NPY-LI were basically consistent in male and female animals of both groups. NPY levels detected by RIA from tissue samples taken at six time points throughout the 24-hour cycle were in the range of 15-60 pmol/mg protein in the diencephalon or below 5 pmol/mg protein in cerebral cor

The organization of neural inputs to the medial preoptic nucleus of the rat.

There is general agreement that the medial preoptic nucleus (MPN) receives projections from widespread regions of the brain, although there are significant discrepancies in the literature with regard to certain specific inputs. Therefore, we have reexamined the inputs to this nucleus with both retrograde and anterograde axonal transport techniques. First, injections of the retrograde tracers true blue, SITS, or wheat germ agglutinin were made into the region of the MPN and the distribution of retrogradely labeled cells was charted. Then, autoradiographic material was used to confirm the result

Basic organization of projections from the oval and fusiform nuclei of the bed nuclei of the stria terminalis in adult rat brain.

The organization of axonal projections from the oval and fusiform nuclei of the bed nuclei of the stria terminalis (BST) was characterized with the Phaseolus vulgaris-leucoagglutinin (PHAL) anterograde tracing method in adult male rats. Within the BST, the oval nucleus (BSTov) projects very densely to the fusiform nucleus (BSTfu) and also innervates the caudal anterolateral area, anterodorsal area, rhomboid nucleus, and subcommissural zone. Outside the BST, its heaviest inputs are to the caudal substantia innominata and adjacent central amygdalar nucleus, retrorubral area, and lateral parabrac

A detailed mapping of histamine H1-receptors in guinea-pig central nervous system established by autoradiography with [125I]iodobolpyramine.

[125I]Iodobolpyramine, a potent and selective histamine H1-receptor antagonist derived from mepyramine, was used to generate light microscopic autoradiograms on sections of guinea-pig brain and spinal cord. Histamine H1-receptors were labelled with high sensitivity over a low background as determined using mianserin or other H1-receptor antagonists as competing agents. An atlas of H1-receptors was established using five sagittal sections and 39 frontal sections, the latter serially prepared at 50 micron intervals. Labelled areas were identified by comparison with corresponding, classically sta

Distribution, morphology and habenular projections of adenosine deaminase-containing neurons in the septal area of rat.

Adenosine deaminase (ADA) was localized within several types of neurons within the septum and in septal efferent projections to the habenula by immunohistochemical, biochemical, retrograde tracing and lesion methods. Numerous ADA-immunoreactive (ADA-IR) neurons were observed in the septofimbrial nucleus, the triangular septal nucleus and the bed nucleus of the anterior commissure, while considerably fewer numbers were seen in the lateral septal area. Based on their size, shape and dendritic features, 4 morphologically distinct types of ADA-IR neurons were recognized in these septal structures.
