Anti fi larial potential of the fruits and leaves extracts of Pongamia pinnata on cattle filarial parasite Setaria cervi.

The effect of aqueous and alcohol extracts of the fruits and leaves of Pongamia pinnata (Linn.) Merr. (Syn. Pongamia glabra Vent, leguminosae) on the spontaneous movements of both the whole worm and the nerve-muscle preparation of Setaria cervi and on the survival of micro fi lariae in vitro was studied. The aqueous and alcohol extracts of fruits and the alcohol extract of leaves caused an inhibition of spontaneous movements of the whole worm and the nerve-muscle preparation of S. cervi. The initial stimulatory effect was not observed with the aqueous extract of fruits on the nerve-muscle prep

Molecular identification and genetic variability of equine and bovine ocular setariasis in india: molecular profiling by mitochondrial genes.

BACKGROUND: Ocular setariasis is an ectopic infection caused by a parasite under the genus Setaria. Adult worms belong to the Setariidae family and typically reside in the peritoneal cavity of ungulates. However, immature forms of these species may aberrantly migrate to the eyes of cattle, buffalo, goats, horses and several other hosts, leading to corneal opacity and blindness. Here, we have distinguished the Setaria digitata collected from both equine and buffalo hosts based on the morphology, molecular profiling of mitochondrial cytochrome c oxidase subunit 1 (Cox1), cytochrome c oxidase sub

The annual, temporal and spatial pattern of Setaria tundra outbreaks in Finnish reindeer: a mechanistic transmission model approach.

BACKGROUND: In northern Finland (Lapland), reindeer are reared as semi-domesticated animals. The region has a short summer season of 2-3 months, yet reindeer are infected with the mosquito-borne filarioid parasite Setaria tundra. The infection causes peritonitis and perihepatitis, which cause significant economic losses due to reduced body weight of infected animals. The objective of this study was to: (i) describe the spatial and temporal pattern of outbreaks in three different areas across Finnish Lapland; and (ii) construct a temperature-driven mechanistic transmission model to quantify the

Filarial glutathione S-transferase: its induction by xenobiotics and potential as drug target.

Glutathione-S-transferase (GST) a Phase-II drug detoxification enzyme, was detected in Setaria cervi, a bovine filarial parasite. In vitro effect of diethylcarbamazine, butylated hydroxyanisole and phenobarbitone on the GST of adult female S. cervi was assayed by the addition of these compounds in the maintenance medium. The specific activity of GST towards 1-chloro-2,4-dinitrobenzene was increased progressively 1.2-1.97, 1.3-2.4 and 1.2-2.7 times at 10-100 microM of diethylcarbamazine, butylated hydroxyanisole and phenobarbitone, respectively, after 5 h at 37 degrees C. Substrate specificity

Subconjunctival setariasis due to Setaria equina infection; a case report and a literature review.

A rare case of human subconjunctival setariasis due to Setaria equina infection is reported herein. A 15-years old girl was referred with a 24h history of edema and redness in her left eye. On slit lamp examination, a thread-like cylindrical worm was moving in the subconjunctival area. The worm was extracted, stained and measured 110mm in length 510μm in width. The isolated worm was identified as adult female S. equina based on morphometric criteria. Identification of the species of the worm was confirmed using molecular methods. For this purpose, the 12S rRNA gene was PCR-amplified and the pu

Comparison of microfilaria concentration method for Setaria digitata infection in cattle and for Dirofilaria immitis infection in dogs.

Several peripheral blood microfilaria concentration methods that use Acetone (Acetone test), 2% formalin (modified Knott method), 5% Tween 20 solution, distilled water, 1% or 0.1% SDS were compared for their efficacy in detecting Setaria digitata microfilaria in cattle. The Acetone test was found to be more efficacious than the modified Knott method or the 5% Tween 20 solution test for detecting the S. digitata microfilaria in bovine blood. However, besides the Acetone test, the modified Knott method was also found to be suitable for Dirofilaria immitis microfilaria detection in dogs. SDS and

Setaria digitata microfilaraemia in Mastomys coucha: an animal model for chemotherapeutic and immunobiological studies.

Intraperitoneal implantation of adult gravid females of the bovine filarial parasite, Setaria digitata in Mastomys coucha was found to induce microfilaraemia lasting for about 125 days. The microfilariae (mf) could be detected as early as 4 days post-implantation (p.i.) and peak levels of about 30 mf in 20 microliters of blood were observed by 21 days. A significant positive correlation was found between mf density and the body weight of recipients pre-implantation. The implanted adult worms were generally viable only for less than 1 week. Implantation resulted in a significant decrease in tot

Molecular characterization and phylogenetic analysis of Setaria digitata of Sri Lanka based on CO1 and 12S rDNA genes.

The aim of the present research is to determine the phylogenetic position of Setaria digitata of Sri Lanka in the evolutionary tree of filarial worms. DNA sequences of portions of the mitochondrial genes cytochrome c oxidase subunit 1 (CO1) and small subunit ribosomal RNA (12S rDNA) were analysed. Intra-specific variation was observed in CO1 but not in 12S rDNA. Phylogenetic trees inferred from these two genes resembled one another in recognizing monophyly of Setaria. S. digitata and Setaria labiatopapillosa appear to be sister species.

Discovery of potent inhibitors targeting Glutathione S-transferase of Wuchereria bancrofti: a step toward the development of effective anti-filariasis drugs.

Lymphatic filariasis (LF) is one of the major health problems for the human kind in developing countries including India. LF is caused by three major nematodes namely Wuchereria bancrofti, Brugia malayi, and Brugia timori. The recent statistics of World Health Organization (WHO) showed that 51 million people were affected and 863 million people from 47 countries around worldwide remain threatened by LF. Among them, 90% of the filarial infection was caused by the nematode W. bancrofti. Approved drugs were available for the treatment of LF but many of them developed drug resistance and no longer

Cross-reaction of antigen preparations from adult and larval stages of the parasite Setaria equina with sera from infected humans with Wuchereria bancrofti.

Crude antigenic preparations from Setaria equina were used in ELISA and Western blotting to examine cross-reaction with human sera from areas endemic for bancroftian filariasis. Sera from normal subjects from non-endemic areas were included as negative controls. Cross-reaction was found between S. equina antigens and antibodies in the sera of Wuchereria bancrofti-infected patients, with the highest levels observed between sera of chronic infected patients and Setaria spp. crude female worm surface antigen (CFSWA). In the absence of active transmission of Setaria spp. infection, CFWSA is useful

Adult 175 kDa collagenase antigen of Setaria cervi in immunoprophylaxis against Brugia malayi in jirds.

A 175 kDa antigen fraction with collagenase activity was isolated and purified from somatic extracts of adult Setaria cervi females using column chromatography involving consecutive steps of DEAE-Sepharose CL6B and Sephadex G-100. The optimum pH for 175 kDa collagenase was found to be pH 7.0. Sensitivities to a variety of inhibitors and activators indicated that the 175 kDa coIlagenolytic enzyme was metalloserine in nature. The enzyme hydrolysed a variety of protein substrates such as haemoglobin, casein, azocasein (general substrates) and collagen, FALGPA (furanoyl-acryloyl-leu-gly-pro-ala),

Development of a rapid, nonradioactive, oligonucleotide-based assay for the detection of Setaria digitata.

Setaria species are filarial parasites which inhabit the peritoneal cavity of cattle and other ungulates. The parasite is generally considered to be nonpathogenic in its natural hosts, but the transmission of the infective larvae through mosquito vectors to its abnormal hosts (goats, sheep, or horses) can result in a serious and often fatal neuropathological disorder commonly referred to as cerebrospinal nematodiasis. We have previously described the cloning and characterization of a repetitive DNA sequence of Setaria digitata that could be used as a diagnostic probe to detect the parasite in

Study of parasitic fauna of three species of deer occurring in southeastern Mexico.

Three species of deer can be found in southeastern Mexico, the white-tailed deer (Odocoileus virginanus), the red brocket deer (Mazama temama), and the Yucatan brocket deer (O. pandora). Here, we applied a combination of non-invasive sampling, necropsy examinations, and molecular diagnostics to assess the endoparasite infections fauna of in wild and captive populations of these three deer species. We analyzed a total of 140 samples and found eleven taxa parasitizing these hosts, including two trematodes (Paramphistomum sp., an unidentified trematode), one anoplocephalid cestode, six nematodes

Presence of ecto-protein tyrosine phosphatase activity is vital for survival of Setaria cervi, a bovine filarial parasite.

The ecto protein tyrosine phosphatases (PTP) are known to play a crucial role in the pathogenesis and survival of the intracellular parasites. However, their presence and role in filarial parasites is still unknown. We found a significant amount of tyrosine phosphatase activity in the surface antigen fraction extracted from Setaria cervi (S. cervi), a bovine filarial parasite. An antibody designed against the conserved catalytic core of human protein tyrosine phosphatases, PTP1B cross reacted with a 63 kDa band in the surface antigen. We detected a significant amount of PTP activity in the int

Cytosolic and microsomal glutathione-S-transferases from bovine filarial worms Setaria cervi.

The work presented here deals with the status of glutathione-S-transferase (GST; E.C. 2.5.1.18), the major enzyme of the phase II detoxification pathway, in bovine filarial worms Setaria cervi. GST activity was determined in various subcellular fractions of bovine filarial worms S. cervi (Bubalus bubalis Linn.) and was found to be mainly associated with cytosolic and microsomal fractions. The respective specific activities of the enzyme from cytosolic and microsomal fractions of S. cervi females were determined to be 0.122 +/- 0.024 and 0.010 +/- 0.0052 micromol/min/mg protein, respectively.
