Shewanella putrefaciens 200 is a nonfermentative bacterium that is capable of dehalogenating tetrachloromethane to chloroform and other, unidentified products under anaerobic conditions. Since S. putrefaciens 200 can respire anaerobically by using a variety of terminal electron acceptors, including NO3-, NO2-, and Fe(III), it provides a unique opportunity to study the competitive effects of different electron acceptors on dehalogenation in a single organism. The results of batch studies showed that dehalogenation of CT by S. putrefaciens 200 was inhibited by O2, 10 mM NO3-, and 3 mM NO2-, but

The anaerobic decolorization of azo dyes by Shewanella species relies on an intricate electron transfer network, yet the full complement of genes involved remains incompletely defined. In this study, we used mini-Tn5 transposon mutagenesis to identify genes required for the decolorization of the azo dye acid yellow 36 in Shewanella putrefaciens CN32. Two genes were uncovered: menD, encoding a key enzyme in menaquinone biosynthesis, and lutB, involved in lactate metabolism. Deletion of menD caused a nearly complete loss of decolorization capacity without affecting growth. Supplementation with v

Shewanella algae is an emerging marine bacterium capable of causing serious infections in immunocompromised individuals. We report the first documented case in Lithuania of bullous cellulitis caused by S. algae, complicated by necrotizing soft-tissue infection and ultimately requiring leg amputation. To determine whether similar cases had been described earlier in Lithuania, a structured literature review was conducted. We searched PubMed/MEDLINE, Embase, Scopus, Web of Science, and Google Scholar by combining the terms "Shewanella algae," "Shewanella," "Baltic Sea," "wound infection," "necrot

Microbial electrosynthesis (MES) is a rapidly growing technology at the forefront of sustainable chemistry, leveraging the ability of microorganisms to catalyze electrochemical reactions to synthesize valuable compounds from renewable energy sources. The reduction of CO2 is a major target application for MES, but research in this area has been stifled, especially with the use of direct electron transfer (DET)-based microbial systems. The major fundamental hurdle that needs to be overcome is the low efficiency of CO2 reduction largely attributed to minimal microbial access to CO2 owing to its l

Shewanella algae is a rod-shaped, Gram-negative bacterium that is considered an emerging human pathogen. Traditionally associated with warmer climates, S. algae has now been isolated from patients worldwide, and reports of infection are increasing. In a regional hospital on the outskirts of Lisbon, Portugal, four cases have been detected in the past 10 years. Two of the patients were migrants from African countries with daily contact with water; the other two patients were Portuguese, and no epidemiological risk factors were found among them. These are the first cases reported in Portugal. Ris

Interfacial reactions between iron (Fe) (hydr)oxide surfaces and the activity of bacteria during dissimilatory Fe reduction affect extracellular electron transfer. The presence of organic matter (OM) and exposed facets of Fe (hydr)oxides influence this process. However, the underlying interfacial mechanism of facet-dependent hematite and its toxicity toward microbes during bioreduction in the presence of OM remains unknown. Herein, humic acid (HA), as typical OM, was selected to investigate its effect on the bioreduction of hematite {100} and {001}. When HA concentration was increased from 0 t

Dehalococcoides is able to completely dehalogenate tetrachloroethene (PCE) and trichloroethene (TCE) to ethene (ETH). However, the dechlorination efficiency of Dehalococcoides is low and result in the accumulation of toxic intermediates. In this study, Shewanella oneidensis MR-1 (S. oneidensis MR-1) was added to the Dehalococcoides-containing culture and the complete TCE to ETH dechlorination was shortened from 24 days to 16 days. Dehalococcoides-targeted 16S rRNA gene and two model reductive dehalogenase (RDase) genes (tceA and vcrA), responsible for dechlorinating TCE to vinyl chloride (VC)

UNLABELLED: Hydrogen sulfide (H2S), well known for its toxic properties, has recently become a research focus in bacteria, in part because it has been found to prevent oxidative stress caused by treatment with some antibiotics. H2S has the ability to scavenge reactive oxygen species (ROS), thus preventing oxidative stress, but it is also toxic, leading to conflicting reports of its effects in different organisms. Here, with Shewanella oneidensis as a model, we report that the effects of H2S on the response to oxidative stress are time dependent. When added simultaneously with H2O2, H2S promote

Bioelectrochemical reactions using whole-cell biocatalysts are promising carbon-neutral approaches because of their easy operation, low cost, and sustainability. Bidirectional (outward or inward) electron transfer via exoelectrogens plays the main role in driving bioelectrochemical reactions. However, the low electron transfer efficiency seriously inhibits bioelectrochemical reaction kinetics. Here, a three dimensional and artificial nanoparticles-constituent inverse opal-indium tin oxide (IO-ITO) electrode is fabricated and employed to connect with exoelectrogens (Shewanella loihica PV-4). Quasielastic neutron scattering (QENS) is an ideal technique for studying water transport and relaxation dynamics at pico- to nanosecond timescales and at length scales relevant to cellular dimensions. Studies of high pressure dynamic effects in live organisms are needed to understand Earth's deep biosphere and biotechnology applications. Here we applied QENS to study water transport in Shewanella oneidensis at ambient (0.1 MPa) and high (200 MPa) pressure using H/D isotopic contrast experiments for normal and perdeuterated bacteria and buffer solutions to distinguish intracellular and transme

In this work we investigated the specific spoilage organism (SSO) of large yellow croaker (Pseudosciaena crocea) stored at 4°C and role of quorum sensing (QS) system of SSO isolated from the spoiled fish. According to microbial count and 16S rRNA gene of the isolated pure strains, Shewanella, mainly Shewanella baltica and Shewanella putrefaciens, was predominant genera at the end of shelf-life of P. crocea. Among Shewanella isolates, S.baltica02 was demonstrated as SSO in spoilage potential characteristics by inoculation into sterile fish juice using sensory and chemical analyses. Autoinducer

Shewanella is a Gram-negative bacillus found in marine environments throughout the world. Its role in human pathogenesis has mostly been described in patients with seawater exposures and immunocompromising conditions. We present a case of a previously healthy young woman who presented with fever and weakness 4 days after routine elective plastic surgery. Violaceous bullae were found from her bilateral breasts tracking down to her right thigh, corresponding to surgical sites. Despite linezolid and meropenem administration, the patient had hemodynamic instability, requiring intubation and vasopr

Shewanella xiamenensis is an emerging pathogen causing intra-abdominal infection and intestinal colonization. Epidemiologic clues suggest its role as a potential food-borne zoonotic agent. To date, four genome sequences of S. xiamenensis have been made publicly available. All of them were isolated from water samples. In this study, we characterized the genome of a S. xiamenensis strain isolated from a giant grouper in Taiwan. The genome of S. xiamenensis ZYW1 is 4,827,717 bp in length and encodes 4,239 open reading frames. Its genomic sequence shares high homology with other S. xiamenensis str

Fic enzymes mediate diverse post-translational modifications, including adenosine monophosphate (AMP) transfer and removal, referred to as AMPylation and deAMPylation, respectively. We identified the prokaryotic translation elongation factor Tu (EF-Tu) as an AMPylation target of the Fic enzyme SoFic. SoFic can constitutively reverse EF-Tu modification via deAMPylation whereas AMPylation depends on SoFic homodimerization. The complex crystal structure between SoFic and EF-Tu confirms a conserved target binding mode across evolutionarily distant Fic enzymes. AMPylation disrupts EF-Tu's regulator

Vanadium, essential for steel production and energy storage, is increasingly found in groundwater due to extensive mining and industrial activities. Its high mobility and reactivity pose significant environmental risks. This study developed an Shewanella oneidensis MR-1- Fe2O3-biochar composite to enhance vanadium bioremediation. The composite exhibited strong vanadium resistance, achieving 92.5 ± 1.48% removal of pentavalent vanadium [V(V)] at 100 mg/l with an optimal biochar/Fe₂O₃ ratio of 10:1. Its efficiency was further assessed under varying pH, organic carbon levels, and V(V) concentrati

Electroactive bacteria (EAB) and metal oxides are capable of synergistically removing chloramphenicol (CAP). However, the effects of redox-active metal-organic frameworks (MOFs) on CAP degradation with EAB are not yet known. This study investigated the synergism of iron-based MOFs (Fe-MIL-101) and Shewanella oneidensis MR-1 on CAP degradation. 0.5 g/L Fe-MIL-101 with more possible active sites led to a three-fold higher CAP removal rate in the synergistic system with MR-1 (initial bacterial concentration of 0.2 at OD600), and showed a superior catalytic effect than exogenously added Fe(III)/Fe

AhpC is a bacterial representative of 2-Cys peroxiredoxins (Prxs) with broad substrate specificity and functional plasticity. However, details underpinning these two important attributes of AhpC remain unclear. Here, we studied the functions and mechanisms of regulation of AhpC in the facultative Gram-negative anaerobic bacterium Shewanella oneidensis, in which AhpC's physiological roles can be conveniently assessed through its suppression of a plating defect due to the genetic loss of a major catalase. We show that successful suppression can be achieved only when AhpC is produced in a dose- a

We describe the first Italian case of Shewanella algae septicemia in an immunocompetent patient with chronic leg ulcers. The patient had been exposed to seawater before the onset of symptoms. Despite the absence of severe underlying diseases, the primary soft tissue infection of the leg was complicated by hematogenous dissemination.

To better understand gene expression in the intestine after Shewanella algae infection and provide insights into its immune roles in the tongue sole, Cynoglossus semilaevis, sequencing-based high-throughput RNA analysis (RNA-Seq) for the intestines between the control group and 12 h post-injection group was performed. After assembly, there was an average of 23,957,159 raw sequencing reads, and 23,943,491 clean reads were obtained after filtering out low-quality reads. Then, 383 differentially expressed genes (DEGs) in the intestines in response to S. algae infection were identified. Subsequent
