Recovering host DNA from their invertebrate parasites: a minimally invasive genetic sampling tool for the critically endangered flapper skate.

Obtaining host DNA from the blood meals of their invertebrate parasites (invertebrate-derived DNA, iDNA) has proven to be a powerful tool in terrestrial ecology for detecting vertebrates to answer questions relating to host community diversity. However, the use of iDNA in the marine environment is limited. Here, we assessed the efficacy of sampling parasites from the critically endangered flapper skate (Dipturus intermedius) to determine whether iDNA could (i) identify the host species and (ii) obtain host haplotype sequences. In total, 34 copepods (Trebius caudatus specimens, n = 30; Caligus

Morphology and morphometry of blood cells of two species of freshwater stingrays (Chondrichthyes: Potamotrygoninae) from the lower Solimões river, Amazon.

Cytomorphological evaluation of peripheral blood provides information about the health status of animals, in addition to being indicators of environmental contamination. However, reference data regarding the size and shape of elasmobranch blood cells is limited. This work aimed to determine the morphological and morphometric of Potamotrygon motoro and Potamotrygon orbignyi blood cells. The stingrays were captured in white water environments in the lower Solimões River region, state of Amazonas, Brazil. Blood was obtained through puncture of the branchial artery using syringes with 10% EDTA. Mo

INTRAOCULAR PRESSURES OF AQUARIUM-HOUSED COWNOSE RAYS (RHINOPTERA BONASUS) WITH NORMAL AND ABNORMAL OPHTHALMIC EXAMS.

Cownose rays (Rhinoptera bonasus) are common elasmobranchs in zoos and aquaria; however, there is a lack of published information regarding ocular findings in this species. Intraocular pressure (IOP) was measured in a total of 52 cownose rays (Rhinoptera bonasus) from two unrelated aquaria (n = 22 from A1, n = 30 from A2) using a TonoVet rebound tonometer on two settings (dog = D, and unidentified species = P) as part of a full ophthalmologic examination. Adult (n = 38) and juvenile (n = 14) rays were sampled out of water briefly in sternal recumbency. Intraocular pressure (mean ± SD [range])

Microplastic footprints in sharks and rays: First assessment of microplastic pollution in two cartilaginous fishes, hardnose shark and whitespotted whipray.

Microplastic (MP) pollution is an emerging environmental problem worldwide and has caused widespread concern both in terrestrial and aquatic ecosystems due to their potential impacts on the human health, and health of aquatic organisms and the environment. Little is known about the exposure of top marine predators to MP contamination (debris 0.1μm - <5mm, also called MPs). For the first time, MPs have been characterized in carnivore demersal elasmobranch specimens of hardnose shark Carcharhinus macloti, and the whitespotted whipray Maculabatis gerrardi. The specimens were from the Persian Gulf

PHARMACOKINETICS OF A SINGLE DOSE OF INTRAMUSCULAR AND ORAL MELOXICAM IN YELLOW STINGRAYS (UROBATIS JAMAICENSIS).

Elasmobranchs are popular display animals in public aquaria and zoos, but medical management gaps remain in the understanding of the pharmacokinetics of analgesics and pain management in these species. Meloxicam is a nonsteroidal anti-inflammatory drug that has been evaluated intravenously and intramuscularly in teleosts, but has yet to be studied in any elasmobranch species. The pharmacokinetics of meloxicam were determined in 17 yellow stingrays (Urobatis jamaicensis). All stingrays were determined to be healthy from complete physical examinations and baseline bloodwork performed prior to st

Management of wounds in a loggerhead sea turtle (Caretta caretta) caused by traumatic bycatch injury from the spines of a spotted eagle ray (Aetobatus narinari).

A subadult female loggerhead sea turtle (Caretta caretta) was caught in a trawl net off the west coast of Florida with a spotted eagle ray (Aetobatus narinari) spine lodged in the left stifle. Surgical removal of the spine was performed and antibiotic treatment was initiated. Four weeks later, endoscopy revealed a second spine entering an intestinal lumen. The fistulous tract of the left prefemoral fossa was surgically excised and the intestinal perforation was repaired. Dehiscence occurred and a vacuum-assisted closure (VAC) system was used on the wound for approximately 18 days to help reduc

The first confirmed cases of full albinism in rajid species.

Three albino skate specimens (Rajidae) were captured from the North Sea and English Channel between 2008 and 2011. Using DNA barcoding (COI gene) and morphometric analyses, species were identified as a spotted ray Raja montagui, a blonde ray Raja brachyura and a thornback ray Raja clavata. This finding represents the first record of full albinism (a lack of skin and retinal pigmentation) in rajid species.

Four new species of Acanthobothrium van Beneden, 1849 (Cestoda: Onchoproteocephalidea) from the spotted skate, Raja straeleni Poll, off the Western Cape, South Africa.

The examination of eight spotted skates, Raja straeleni Poll, resulted in the discovery of four new species of Acanthobothrium van Beneden, 1849, namely A. microhabentes sp. n., A. microtenuis sp. n., A. crassus sp. n., and A. dolichocollum sp. n., located off the Western Cape of South Africa. With a total of over 200 valid species of Acanthobothrium recognised worldwide, the use of an integrative approach becomes imperative in the interest of simplifying interspecific comparisons between congeners. In accordance with this, the four new species were incorporated into the category classificatio

Calcified cartilage or bone? Collagens in the tessellated endoskeletons of cartilaginous fish (sharks and rays).

The primary skeletal tissue in elasmobranchs -sharks, rays and relatives- is cartilage, forming both embryonic and adult endoskeletons. Only the skeletal surface calcifies, exhibiting mineralized tiles (tesserae) sandwiched between a cartilage core and overlying fibrous perichondrium. These two tissues are based on different collagens (Coll II and I, respectively), fueling a long-standing debate as to whether tesserae are more like calcified cartilage or bone (Coll 1-based) in their matrix composition. We demonstrate that stingray (Urobatis halleri) tesserae are bipartite, having an upper Coll

GnRH receptors and peptides: skating backward.

Gonadotropin-releasing hormone (GnRH) and its receptor are essential for reproduction in vertebrates. Although there are three major types of GnRH peptides and two major types of receptors in vertebrates, the pattern of distribution is unusual. Evidence is presented from genome mining that type I GnRHRs are not restricted to mammals, but can be found in the lobe-finned and cartilaginous fishes. This implies that this tail-less GnRH receptor emerged early in vertebrate evolution, followed by several independent losses in different lineages. Also, we have identified representatives from the thre

Genetic and morphological characterization of Mawsonascaris vulvolacinata n. sp. (Nematoda: Anisakidae) and associated histopathology in a wild caught cowtail stingray, Pastinachus ater.

There are limited reports of infectious agents affecting Australian cowtail stingrays. In the present study, a new species of ascaridoid nematode belonging to the genus Mawsonascaris is described. The most distinct characteristic features were observed in females (the presence of a polar spine in the eggs and a flap-like projection in the vulval area). An identification key for Mawsonascaris spp. is provided. Additionally, internal transcribed spacers (ITS) sequences were obtained for the new species. Alignment of the ITS sequence of the specimens in the present study with those deposited in G

Maternal investment evolves with larger body size and higher diversification rate in sharks and rays.

Across vertebrates, live bearing evolved at least 150 times from ancestral egg laying into diverse forms and degrees of prepartum maternal investment.1,2 A key question is how reproductive diversity arose and whether reproductive diversification underlies species diversification.3,4,5,6,7,8,9,10,11 To test this, we evaluate the most basal jawed vertebrates: the sharks, rays, and chimaeras, which have one of the greatest ranges of reproductive and ecological diversity among vertebrates.2,12 We reconstruct the sequence of reproductive mode evolution across a phylogeny of 610 chondrichthyans.13 W

Mitogenomic evidence of population differentiation of thorny skate, Amblyraja radiata, in the North Atlantic.

Management of thorny skate (Amblyraja radiata) in the Northwest Atlantic has posed a conservation dilemma for several decades due to the species' lack of response to strong conservation efforts in the US Gulf of Maine and the Canadian Scotian Shelf, confusion over the relationship between two reproductive size morphs of differing life histories that are sympatric in the Northwest Atlantic, and conflicting data on regional population connectivity throughout the species' broader range. To better assess potential A. radiata regional population differentiation and genetic links to life-history var

An integrated system to assess marine extinctions.

More than 20 global marine extinctions and over 700 local extinctions have reportedly occurred during the past 500 years. However, available methods to determine how many of these species can be confidently declared true disappearances tend to be data-demanding, time-consuming, and not applicable to all taxonomic groups or scales of marine extinctions (global [G] and local [L]). We developed an integrated system to assess marine extinctions (ISAME) that can be applied to any taxonomic group at any geographic scale. We applied the ISAME method to 10 case studies to illustrate the possible ways

A new genus with two new species of lecanicephalidean tapeworms (Cestoda) from the mangrove whipray, Urogymnus granulatus (Myliobatiformes: Dasyatidae),from the Solomon Islands and northern Australia.

A new lecanicephalidean genus is erected for cestodes previously recognised as "New Genus 12" (Polypocephalidae) in a phylogenetic analysis of the interrelationship of members of this order. Examination of the cestode fauna of the mangrove whipray, Urogymnus granulatus (Macleay) (Myliobatiformes: Dasyatidae) from the Solomon Islands and northern Australia revealed the existence of specimens representing two new species, consistent in morphology with "New Genus 12." Corollapex gen. n. is unique among the 24 valid lecanicephalidean genera in its possession of an apical organ in the form of an ex

Testicular Morphology and Spermatogenesis in Potamotrygon motoro: Insights Into Reproduction of Freshwater Stingrays.

This study investigated the spermatogenesis of the freshwater stingray Potamotrygon motoro using microscopic analyses. The testes of this species were described as being composed of germinal zones, a degenerated zone, and the epigonal organ, consisting of connective tissue and lymphomyeloid cells. A cystic pattern of spermatogenesis was observed, in which each cyst is formed and maintained by Sertoli cells that undergo morphological and positional changes throughout the process. After the release of spermatozoa into the duct, the peripheral cysts formed a degenerating layer. Spermiogenesis, th
