Caudal regression syndrome (sacral agenesis), which impairs development of the caudal region of the body, occurs with a frequency of about 2 live births per 100 000 newborns although this incidence rises to 1 in 350 infants born to mothers with gestational diabetes. The lower back and limbs can be affected as well as the genitourinary and gastrointestinal tracts. The axial skeleton is formed during embryogenesis through the process of somitogenesis in which the paraxial mesoderm periodically segments into bilateral tissue blocks, called somites. Somites are the precursors of vertebrae and asso

Class II non-muscle myosins are implicated in diverse biological processes such as cytokinesis, cellularization, cell shape changes and gastrulation. Two distinct non-muscle myosin heavy chain genes have been reported in all vertebrates: non-muscle myosin heavy chain-A (NMHC-A) and -B (NMHC-B). We report here the isolation of the Xenopus homolog of NMHC-A and present a comparative analysis of the developmental and spatial expression patterns of NMHC-A and the previously isolated NMHC-B to address the role of NMHCs in Xenopus development. A 7.5 kb NMHC-A mRNA is present, maternally in unfertili

The mesenchymal origin muscular tissues entailing some difficulties in the knowledge of certain muscles embryogeny, two methods are therefore applied: the comparison of the ontogenesis data with those of comparative anatomy and the use as a guide of the functional motor unit with which the muscle combines. In resorting to these methods necessary precautions are precisely defined. The origins of the concerned muscles are, at first, situated among those of the whole musculature subjected to will, which are of two types "somitic" and "branchial". The realities lying under these two misleading ter

The notochord functions primarily as a supporting tissue to maintain the anteroposterior axis of primitive chordates, a function that is replaced entirely by the vertebral column in many vertebrates. The notochord still appears during vertebrate embryogenesis and plays a crucial role in the developmental pattern formation of surrounding structures, such as the somites and neural tube, providing the basis for the vertebrate body plan. The indispensable role of the notochord has often been referred to as the developmental burden and used to explain the evolutionary conservation of notochord; how

The vertebral column exhibits segmentation and regionalization along the antero-posterior axis. During embryogenesis, the rhythmic production of the precursors of the vertebrae, the somites, imposes a segmented aspect to the spine, whereas the spine's regional differentiation is controlled by Hox genes. Here we show that in the paraxial mesoderm, Hoxb genes are first activated in a temporal collinear fashion in precursors located in the epiblast lateral to the primitive streak. Our data suggest that collinear activation of Hoxb genes regulates the flux of cells from the epiblast to the streak

Somitogenesis is a term that encompasses somite formation, patterning and differentiation and it is a process that is fundamental to the formation of the axial skeleton in vertebrates. Notch signalling is a mechanism used to specify cell fate in many different contexts, with signalling occurring between cells in contact. Notch signalling is fundamental to the formation and patterning of somites and importantly a ligand of Notch, is mutated in the abnormal vertebral segmentation syndrome spondylocostal dysostosis. Here we discuss what is known about the expression and function of this ligand, D

Three consecutive pairs of head cavities (premandibular, mandibular, and hyoid) found in elasmobranchs have been considered as remnants of preotic 'head' somites-serial homologues of the myotomic compartments of trunk somites that give rise to the extraoccular musculature. Here, we study a more derived vertebrate, and show that cavitation is more complex in the head of Crocodylus niloticus, than just the occurrence of three pairs of cavities. Apart from the premandibular cavities, paired satellite microcavities, and unpaired extrapremandibular microcavities are recognized in the prechordal reg

Two series of experiments were carried out on chick embryos, stages 24 to 28 pairs of somites. (1) Denuded mesenchyme obtained from leg presumptive area previously treated with nitrogen mustard, was grafted into the right flank of embryos, stages 15 to 24 pairs of somites. (2) Conversely, denuded mesenchyme was implanted into the flank of nitrogen mustard-treated embryos. In most of the embryos, a supernumerary abnormal limb appeared; 58% of the legs produced in the first set of experiments had distal parts, whereas only 15% of the legs in the second set of experiments showed toes. Approximately 10% of cases of Alzheimer's disease are familial and associated with autosomal dominant inheritance of mutations in genes encoding the amyloid precursor protein, presenilin 1 (PS1) and presenilin 2 (PS2). Mutations in PS1 are linked to about 25% of cases of early-onset familial Alzheimer's disease. PS1, which is endoproteolytically processed in vivo, is a multipass transmembrane protein and is a functional homologue of SEL-12, a Caenorhabditis elegans protein that facilitates signalling mediated by the Notch/LIN-12 family of receptors. To examine potential roles for PS1 in facili

Pax3-deficient Splotch mice display neural tube defects and an array of neural crest related abnormalities including defects in the cardiac outflow tract, dorsal root ganglia and pigmentation. Pax3 is expressed in neural crest cells that emerge from the dorsal neural tube. Pax3 is also expressed in the somites, through which neural crest cells migrate, where it is required for hypaxial muscle development. Homozygous mutant Splotch embryos die by embryonic day 14. We have utilized the proximal 1.6 kb Pax3 promoter and upstream regulatory elements to engineer transgenic mice reproducing endogeno

Somites, metameric structures, give rise to the vertebral column, ribs, skeletal muscles and subcutaneous tissues. In mouse embryos, a pair of somites is formed every 2h by segmentation of the anterior parts of the presomitic mesoderm. This periodic event is regulated by a biological clock called the segmentation clock, which involves cyclic expression of the basic helix-loop-helix gene Hes7. Hes7 oscillation is regulated by negative feedback with a delayed timing. This process has been mathematically simulated by differential-delay equations, which predict that negative feedback with shorter

Mayer-Rokitansky-Kuster-Hauser (MRKH) is a malformation complex comprising absent vagina and absent or rudimentary uterus. MRKH syndrome may be attributed to an initial affection of the intermediate mesoderm consequently leading (by the end of the 4(th) week of fetal life) to an alteration of the blastema of the cervicothoracicsomites and the pronephricducts. These latter subsequently induce the differentiation of the mesonephric and then the Wolffian and Mullerian ducts. There are very sparse such cases reported. We present a case of type II MRKH or Mullerian renal cervical somite association

The distribution and migration of the cardiac neural crest was studied in chick embryos from stages 11 to 17 that were immunochemically stained in whole-mount and sectioned specimens with a monoclonal antibody, HNK-1. The following results were obtained: 1) The first phase of the migration in the cardiac crest follows the dorsolateral pathway beneath the ectoderm. 2) In the first site of arrest, the cardiac crest forms a longitudinal mass of neural-crest cells, called in the present study, the circumpharyngeal crest; this mass is located dorsolateral to the dorsal edge of the pericardium (peri

We analyzed the International Mouse Phenotyping Consortium (IMPC) release 19 set of 8,539 phenotyped whole-gene knockouts to identify 204 genes that alter vertebral anatomy and development. These genes are broadly grouped into six categories based on their phenotype: "vertebral number" (22 genes); "vertebral processes" (35 genes); "spine shape" (16 genes); "tail morphology" (73 genes); "vertebral form" (62 genes); and "somitogenesis" (24 genes), with minimal overlap between groups. Gene expression analysis of somite trios across six developmental stages show that 182 of these genes are express

A central question in developmental biology has been the initiation of cell-specific gene expression and its temporal relationship to morphogenesis. We have coupled embryo microdissection with the exquisite sensitivity of the polymerase chain reaction to define the onset of cell-specific gene expression during pancreatic organogenesis. Using the precise assignment of gestational age by the number of somites in each embryo, we determined the onset of transcription of major genes of the endocrine and exocrine pancreas during mouse development to within 2-3 hr. Somatostatin mRNA was detected at t

The migration pathways and spatial distribution of neural crest cells largely depend on the embryonic architecture. At the preotic level in the chick embryo, cephalic crest adhere to even-numbered rhombomeres proximally, and populate each pharyngeal arch distally, thus prefiguring the morphology of the branchiomeric nerves. This distribution pattern is possible because of the absence of somites in the head. In the postotic region, however, somites and pharyngeal arches coexist at the same axial level. The caudalmost cephalic crest cell population, the circumpharyngeal crest cells, are derived

Removal of premigratory neural crest over somites 1 through 3 in chick embryos has been shown previously to result in a significant incidence of persistent truncus arteriosus. In the present study, single somite-length pieces of premigratory neural crest were removed unilaterally at slightly different embryonic ages. These lesions resulted in a number of different heart defects. Two defects, ventricular septal defect and double outlet right ventricle, were significantly correlated with the location of neural crest removed. The stage at which the ablation was performed was important in determin

In the present study, we re-evaluated the presence of a row of denticles on the posterior margin of the genital double-somite as one of the morphological diagnostic characters used to distinguish Paracalanus indicus Wolfenden, 1906, from P. tropicus Andronov, 1977. The results demonstrated that these denticles are present in both dorsal and lateral margins in these two species. Consequently, the previously reported absence of denticles from the dorsal surface of P. indicus can no longer be regarded as a reliable diagnostic character to separate Paracalanus indicus from P. tropicus. Head-trunk differences are well established in the most primitive vertebrates, and are clear from early developmental stages of all modern forms. The boundary between the two regions is not constant in all vertebrate classes in terms of the number of occipital somites. The occipital region is in some respects a transitional zone, giving rise to trunk-like somitic derivatives in the head. It is also highly specialised, providing a unique population of neural crest cells that are essential for formation of the aorticopulmonary septum (which divides the outflow tract of the heart) in mammals and
