Spermidine promotes osteogenic differentiation of BMSCs via the m6A methylation-autophagy axis under inflammatory stress.

Disordered osteogenic potential in bone marrow mesenchymal stem cells (BMSCs) is a hallmark of inflammatory jaw diseases. Recent studies suggest that inflammatory stimulating disrupts energy metabolism during BMSC osteogenesis. However, the impact of key metabolites on BMSC ossification remains unclear. In this study, a comprehensive targeted metabolomics analysis was performed during BMSC osteogenic differentiation, with and without inflammatory stimulation induced by lipopolysaccharide (LPS). We identified a significant downregulation of spermidine (SPD), along with methylated purines and py

Spermidine mitigates blue-light-induced corneal injury by alleviating oxidative stress and restoring autophagy impairment.

This study investigated the protective efficacy of spermidine on corneal epithelium exposed to photodamage from artificial blue light (BL). After six weeks of BL exposure, the rabbit cornea exhibited epithelial injury and delayed wound healing accompanied by downregulation of spermidine, as identified through metabolomic analysis. Pathways related to autophagy and the antioxidant response were implicated in this process. In vitro, spermidine mitigated BL-induced reductions in viability and proliferation of human corneal epithelial cells, decreased cell death, and enhanced colony formation. Spe

Molecular dynamics simulation analysis of a modelled spermidine synthase from Yersinia pseudotuberculosis docked with cyclohexylamine.

The gram-negative bacterium Yersinia pestis is the causative agent of plague 1 and has been responsible for major pandemics in the past. Therefore, it is of interest to document the molecular docking and simulation analysis of spermidine synthase from Yersinia pseudotuberculosis with cyclohexylamine. The sequence and structure analysis showed an abundance of Leu, Val, Gly, Glu and Ala, the least presence of Trp and Cys, higher negatively charged residues and a GRAVY score of -0.125, suggesting the stability of the protein. The cyclohexylamine conformer 4-fluorocyclohexan-1-amine (CID 21027526)

Maternal supplementation spermidine during gestation improves placental angiogenesis and reproductive performance of high prolific sows.

Spermidine (SPD) is a widely recognized polyamine compound found in mammalian cells and plays a key role in various cellular processes. We propose that SPD may enhance placental vascular development in pregnant sows, leading to increased birth weight of piglets. Six hundred and nine sows at 60 days of gestation were randomly assigned into a basal diet (CON group), basal diet supplemented 10 mg/kg of SPD (SPD1 group), and basal diet supplemented 20 mg/kg of SPD (SPD2 group), respectively. Compared with the CON, SPD1 significantly increased the average number of healthy piglets per litter and th

Chemoproteomics-based profiling reveals potential antimalarial mechanism of Celastrol by disrupting spermidine and protein synthesis.

BACKGROUND: Malaria remains a global health burden, and the emergence and increasing spread of drug resistance to current antimalarials poses a major challenge to malaria control. There is an urgent need to find new drugs or strategies to alleviate this predicament. Celastrol (Cel) is an extensively studied natural bioactive compound that has shown potentially promising antimalarial activity, but its antimalarial mechanism remains largely elusive. METHODS: We first established the Plasmodium berghei ANKA-infected C57BL/6 mouse model and systematically evaluated the antimalarial effects of Cel

Spermidine Associated with Gut Microbiota Protects Against MRSA Bloodstream Infection by Promoting Macrophage M2 Polarization.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major human pathogen that causes various diseases. Extensive researches highlight the significant role of gut microbiota and its metabolites, particularly spermidine, in infectious diseases. However, the immunomodulatory mechanisms of spermidine in MRSA-induced bloodstream infection remain unclear. Here, we confirmed the protective effects of spermidine in bloodstream infection in mice. Spermidine reduced the bacterial load and expression of inflammatory factors by shifting the macrophage phenotype to an anti-inflammatory phenotype, ultim

Spermidine rescues proximal tubular cells from oxidative stress and necrosis after ischemic acute kidney injury.

Kidney ischemia and reperfusion injury (IRI) is associated with a high mortality rate, which is attributed to tubular oxidative stress and necrosis; however, an effective approach to limit IRI remains elusive. Spermidine, a naturally occurring polyamine, protects yeast cells against aging through the inhibition of oxidative stress and necrosis. In the present study, spermidine supplementation markedly attenuated increases in plasma creatinine concentration and tubular injury score after IRI. In addition, exogenous spermidine potently inhibited oxidative stress, especially lipid peroxidation af

Injectable hydrogel-mediated spermidine delivery triggers tumor pyroptosis via oxidative metabolism for enhanced cancer therapy.

Intracellular polyamine metabolic homeostasis is closely linked to therapeutic outcomes in cancer. However, their intrinsic antitumor mechanisms have not been fully clarified. Herein, we demonstrate that the catabolism of spermine (Spm) and spermidine (Spd) mediated by polyamine oxidase directly generates two potent cytotoxic metabolites, namely acrolein and hydrogen peroxide. These metabolites act synergistically to trigger a marked elevation in intracellular reactive oxygen species (ROS), which in turn activate the caspase-1/GSDMD and caspase-3/GSDME signaling pathways, ultimately driving tu

Spermidine Secreted by Apoptotic Cells Enhances Chemotherapy Resistance by Modulating β-Catenin Activity in Osteosarcoma.

PURPOSE: Chemoresistance remains a key hurdle in osteosarcoma therapy. This study aims to delineate the role and underlying mechanisms of spermidine (SPD) in osteosarcoma chemoresistance. EXPERIMENTAL DESIGN: Using osteosarcoma cell lines and xenografts, we combined flow cytometry, Western blotting, proteomic mass spectrometry, and RNA sequencing to characterize SPD-driven changes in cellular pathways and resistance signatures. We tested whether pharmacologic inhibition of SPD biosynthesis, alone or in combination with standard chemotherapy, improves therapeutic response in vivo. RESULTS: Fo

Dendrobium officinale polysaccharide-spermidine composite hydrogels: Tunable rheological properties and improved in vitro colonic fermentation.

Dendrobium officinale polysaccharide (DOP) and spermidine (Spd) exhibit synergistic anti-aging effects in Caenorhabditis elegans, suggesting potential functional complementarity. Here, a DOP-Spd composite hydrogel (DS) was constructed to integrate structural tunability with fermentation-related functionality. Three weight ratios (18:2, 18:4, and 18:6, DOP:Spd) successfully formed hydrogels. Increasing Spd content enhanced gel strength and thermal stability, as reflected by higher hardness (149.62-353.75 g), chewiness (41.04-119.07), and DTG peak temperature (331.7-334.0 °C). All DS hydrogels e

Secreted spermidine synthase reveals a paracrine role for PGC1α-induced growth suppression in prostate cancer.

Prostate cancer is the fifth cause of death by cancer worldwide, second in incidence in the male population. The definition of the molecular basis of its development and the oncogenic signals driving lethality continue to be important objectives in prostate cancer research. Prior work from others and us has demonstrated that loss of PGC1α expression results in a metabolic, signaling and transcriptional reprogramming that supports the development of metastatic disease. However, we do not fully understand the spectrum of tumor suppressive effects regulated by this co-regulator. Here we show that

Spermidine exerts cardiometabolic protection in estrogen-deprived rats via mitigating cardiac mitochondrial dysfunction and apoptosis.

Postmenopausal conditions are linked to elevated risks of various complications, particularly cardiovascular diseases (CVDs). While estrogen supplementation is a standard intervention, it increase adverse effects of CVDs and breast cancer prevalence in postmenopausal women. Spermidine, a naturally occurring polyamine, has recently emerged attention for its protective effects in heart pathologies. However, the cardioprotective effects of spermidine against estrogen deprivation-related cardiac dysfunctions remain unclear. We hypothesized that spermidine exerted cardiometabolic protection by miti

Spermidine-encapsulated chondroitin sulfate methacryloyl hydrogel delivery system for remodeling bone homeostasis in rheumatoid arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent synovial inflammation, progressive cartilage degradation, and osteoclast-mediated bone erosion. While current systemic therapies alleviate inflammation, they often fail to prevent structural joint damage or promote local tissue regeneration. Herein, we developed a chondroitin sulfate methacryloyl (ChSMA) hydrogel-based delivery system encapsulating spermidine (SPD), a naturally occurring polyamine with emerging anti-inflammatory, senescence-attenuating, and chondroprotective properties, to achieve localized

CaMKIIα mediates spermidine-induced memory enhancement in rats: A potential involvement of PKA/CREB pathway.

Memory consolidation is associated with the regulation of protein kinases, which impact synaptic functions and promote synaptogenesis. The administration of spermidine (SPD) has been shown to modulate major protein kinases associated with memory improvement, including the Ca2+-dependent protein kinase (PKC) and cAMP-dependent protein kinase (PKA), key players in the cAMP response element-binding protein (CREB) activation. Nevertheless, the initial mechanism underlying SPD-mediated memory consolidation remains unknown, as we hypothesize a potential involvement of the memory consolidation precur

Voluntary activity reverses spermidine-induced myocardial fibrosis and lipid accumulation in the obese male mouse.

Obesity due to high calorie intake induces cardiac hypertrophy and dysfunction, thus contributing to cardiovascular morbidity and mortality. Recent studies in aging suggest that oral supplementation with the natural polyamine spermidine has a cardioprotective effect. Here, the hypothesis was tested that spermidine or voluntary activity alone or in combination protect the heart from adverse effects induced by obesity. Therefore, C57Bl/6 mice (n = 8-10 per group) were subjected to control or high fat diet (HFD) and were left untreated, or either received spermidine via drinking water or were vol

Spermidine supplementation and voluntary activity differentially affect obesity-related structural changes in the mouse lung.

Obesity is associated with lung function impairment and respiratory diseases; however, the underlying pathophysiological mechanisms are still elusive, and therapeutic options are limited. This study examined the effects of prolonged excess fat intake on lung mechanics and microstructure and tested spermidine supplementation and physical activity as intervention strategies. C57BL/6N mice fed control diet (10% fat) or high-fat diet (HFD; 60% fat) were left untreated or were supplemented with 3 mM spermidine, had access to running wheels for voluntary activity, or a combination of both. After 30

Spermidine reduces lipid accumulation and necrotic core formation in atherosclerotic plaques via induction of autophagy.

BACKGROUND AND AIMS: Spermidine is an endogenous biological polyamine that exhibits broad longevity-extending activities via the induction of autophagy. Because basal autophagy is atheroprotective during early atherosclerosis but dysfunctional in advanced plaques, the aim of the present study was to assess the potential beneficial effects of autophagy induction by spermidine on atherosclerotic plaque progression and composition. METHODS: Apolipoprotein E-deficient (ApoE(-/-)) mice were fed a Western-type diet for 20 weeks with or without 5 mM spermidine in the drinking water. RESULTS: (Immun

Induction of spermidine/spermine N1-acetyltransferase (SSAT) by aspirin in Caco-2 colon cancer cells.

Epidemiological, experimental and clinical results suggest that aspirin and other NSAIDs (non-steroidal anti-inflammatory drugs) inhibit the development of colon cancer. It has been shown that the NSAID sulindac induces apoptosis and suppresses carcinogenesis, in part, by a mechanism leading to the transcriptional activation of the gene encoding SSAT (spermidine/spermine N1-acetyltransferase), a rate-limiting enzyme in polyamine catabolism. In the present study, we show that a variety of NSAIDs, including aspirin, sulindac, ibuprofen and indomethacin, can induce SSAT gene expression in Caco-2

Novel chimeric spermidine synthase-saccharopine dehydrogenase gene (SPE3-LYS9) in the human pathogen Cryptococcus neoformans.

The Cryptococcus neoformans LYS9 gene (encoding saccharopine dehydrogenase) was cloned and found to be part of an evolutionarily conserved chimera with SPE3 (encoding spermidine synthase). spe3-lys9, spe3-LYS9, and SPE3-lys9 mutants were constructed, and these were auxotrophic for lysine and spermidine, spermidine, and lysine, respectively. Thus, SPE3-LYS9 encodes functional spermidine synthase and saccharopine dehydrogenase gene products. In contrast to Saccharomyces cerevisiae spe3 mutants, the polyamine auxotrophy of C. neoformans spe3-LYS9 mutants was not satisfied by spermine. In vitro ph
