Osteomalacia in a hemodialysis patient receiving an active vitamin D sterol.

BACKGROUND: A 17-year-old Filipino male hemodialysis patient presented for renal transplant evaluation. He had significant skeletal abnormalities characterized by bone pain, an inability to walk, and secondary hyperparathyroidism despite therapy with an active vitamin D sterol (paricalcitol). INVESTIGATIONS: The patient underwent a physical examination, and his serum levels of calcium, phosphorus, alkaline phosphatase, parathyroid hormone, aluminum and 25-hydroxycholecalciferol (25OH-vitamin D) were determined. X-rays of hips and lower extremities, MRI, and bone histomorphometry after double

Inactivation of sterol Delta5,6-desaturase attenuates virulence in Candida albicans.

Two clinical Candida albicans isolates that exhibited high-level resistance to azoles and modest decreases in susceptibility to amphotericin B were cultured from unrelated patients. Both isolates harbored homozygous nonsense mutations in ERG3, which encodes an enzyme, sterol Delta5,6-desaturase, involved in ergosterol synthesis. Extraction and analysis of the sterols from both isolates confirmed the absence of sterol Delta5,6-desaturase activity. Although the loss of sterol Delta5,6-desaturase activity is known to confer resistance to azoles, this mechanism of resistance has rarely been seen i

Human oocyte maturation in vitro is stimulated by meiosis-activating sterol.

In-vitro studies in mouse oocytes have shown that the C-29 endogenously occurring sterol FF-MAS (follicular fluid meiosis-activating sterol) is a potent inducer of meiotic maturation leading to increased fertilization rates. We have used synthetic FF-MAS to induce meiotic maturation in immature human oocytes aspirated from polycystic ovarian syndrome patients. The patients were asked to give written consent to donate half of their aspirated oocytes to investigate the influence of culture conditions on maturation kinetics. The oocytes were aspirated from follicles 8-12 mm in diameter under ultr

The role of wax and sterol ester fatty acids in chronic blepharitis.

The authors analyzed the long-chain fatty acids derived from the wax and sterol ester fractions of meibomian secretions from patients with chronic blepharitis and normal individuals. Meibomian secretions were partitioned into separate lipid classes by thin-layer chromatography (TLC). Wax and sterol esters were eluted and transesterified. The liberated fatty acid methyl esters (FAME) were analyzed by gas liquid chromatography. Equivalent chain lengths (ECL) were determined for the 58 peaks found. Thirty-three peaks were positively identified by standards. Peaks were quantified by area normaliza

The role of wax and sterol esters of meibomian secretions in chronic blepharitis.

We analyzed the fatty wax esters and sterol esters found in the expressed lipid secretions of six patients from each of six clinical groups of chronic blepharitis, plus eight normal controls. Using gas liquid chromatography (GLC), 12 peaks corresponding to equivalent chain lengths (ECL) of 33.6, 35.4, 36.1, 37.3, 38.2, 39.2, 40.1, 41.2, 42.1, 43.2, 44.9 and 45.7 were found in the fatty wax esters and five peaks corresponding to ECL 19.1, 20.0, 21.1, 22.0 and 23.2 were found in the sterol esters. The clinical groups showed significant differences in several of these components. Sterol and wax e

Characterization of microsomal methyl sterol demethylase in two Morris hepatomas.

Previously, we reported that the rate of metabolism of methyl sterol intermediates of cholesterol biosynthesis by broken-cell preparations of Morriss hepatoma 7777 is very slow, whereas the intact tumors are known to synthesize cholesterol quite efficiently. Active preparations have now been obtained by substitution of pyrophosphate for phosphate buffer. Although substitution of pyrophosphate buffer markedly enhances microsomal methyl sterol demethylation rates 3- to 4-fold in hepatoma 7777, other microsomal enzymes and electron carriers in either liver or a more slowly growing hepatoma appear

Targeting Cancer Signaling Pathways With Plant Sterols: Emerging Roles of Stigmasterol, Campesterol, and β-Sitosterol.

Plant sterols are naturally occurring tetracyclic triterpenes that serve as essential structural components of plant cell membranes. Structurally similar to cholesterol, these bioactive compounds have gained considerable attention for their diverse therapeutic properties, particularly their anticancer potential. Among them, stigmasterol, campesterol, and β-sitosterol are the most abundant and well-studied phytosterols known for their multifaceted roles in tumor suppression and apoptosis induction. They modulate several oncogenic signaling pathways, including PI3K/AKT/mTOR, JAK/STAT, NF-κB, and

Plasma Non-cholesterol Sterols as Markers of Cholesterol Synthesis and Intestinal Absorption: A Critical Review.

Plasma concentrations of phytosterols and non-cholesterol sterol precursors of cholesterol synthesis have been used as markers of intestinal cholesterol absorption and synthesis in inherited and secondary dyslipidemias and in population-based investigations to evaluate the risk for cardiovascular disease, respectively. The method aims at replacing initial research procedures such as the use of stable isotopes associated with fecal steroid balance, which are limited by the high cost and tedious procedures. However, we show in this review that numerous results obtained with serum sterol measurem

ABCG5/G8 gene is associated with hypercholesterolemias without mutation in candidate genes and noncholesterol sterols.

CONTEXT: Approximately 20% to 40% of clinically defined familial hypercholesterolemia (FH) cases do not show a causative mutation in candidate genes (mutation-negative FH), and some of them may have a polygenic origin. OBJECTIVE: The aim of this work was to study the prevalence of ABCG5/G8 genetic variants in mutation-negative FH, as defects in these genes relate to intestinal hyperabsorption of cholesterol and thus ABCG5/G8 variants could explain in part the mechanism of hypercholesterolemia. DESIGN, SETTING, AND PATIENTS: We sequenced the ABCG5/G8 genes in 214 mutation-negative FH and 97 c

Recombinant anthrax protective antigen: Observation of aggregation phenomena by TEM reveals specific effects of sterols.

Negatively stained transmission electron microscope images are presented that depict the aggregation of recombinant anthrax protective antigen (rPA83 monomer and the PA63 prepore oligomer) under varying in vitro biochemical conditions. Heat treatment (50°C) of rPA83 produced clumped fibrils, but following heating the PA63 prepore formed disordered aggregates. Freeze-thaw treatment of the PA63 prepore generated linear flexuous aggregates of the heptameric oligomers. Aqueous suspensions of cholesterol microcrystals were shown to bind small rPA83 aggregates at the edges of the planar bilayers. Wi

A comprehensive method for extraction and quantitative analysis of sterols and secosteroids from human plasma.

We describe the development of a method for the extraction and analysis of 62 sterols, oxysterols, and secosteroids from human plasma using a combination of HPLC-MS and GC-MS. Deuterated standards are added to 200 μl of human plasma. Bulk lipids are extracted with methanol:dichloromethane, the sample is hydrolyzed using a novel procedure, and sterols and secosteroids are isolated using solid-phase extraction (SPE). Compounds are resolved on C₁₈ core-shell HPLC columns and by GC. Sterols and oxysterols are measured using triple quadrupole mass spectrometers, and lathosterol is measured using GC

Fatty acids, sterols, phenols and antioxidant activity of Phoenix theophrasti fruits growing in Crete, Greece.

In the present study we report the chemical composition of the Cretan Phoenix theophrasti Gr. fruits, in comparison with dates, from its close relative Phoenix dactylifera L. for their nutritional value and their potential exploitation as a source of bioactive components such as phytosterols, lipids and polyphenols. The non polar dichloromethane (CH2Cl2) extract of the fruits was analysed by several techniques (TLC, CC, GC and GC-MS) and was found to consist mainly from fatty acids, hydrocarbons and phytosterols. Palmitic acid was the most abundant fatty acid, 12.49% of total saponifiables, wh

[The demonstration of a methodology for identifying and analysing oxygenated sterols present in radicular cysts].

Our work has two main aims: to identify oxygenated sterols that accompany cholesterol in dental cyst and to develop effective methods for "profile" analyses of these sterols. Attention as focused on a family of products derived from cholesterol, characterized by the presence of one or more oxygenated functions. More than fifty of these oxysterols are known and find most of time in different parts of the body. In the procedure, lipids are isolated from dental cyst, the fraction is trimethylsilylated and analysed by capillary gas chromatography. Sterols are identified by comparison with referenc

Serum and hepatic cholestanol, squalene and noncholesterol sterols in man: a study on liver transplantation.

Serum noncholesterol sterols indicate overall cholesterol metabolism in a variety of experimental and clinical conditions. In patients with advanced primary biliary cirrhosis serum cholestanol, a 5 alpha-derivative of cholesterol, is markedly increased, and cholesterol precursors, which are indicators of cholesterol synthesis, are clearly reduced, as is the ratio of plant sterols (campesterol/sitosterol). Therefore these variables were studied in the livers and sera of 23 patients undergoing liver transplantation (16 patients with chronic liver disease, 4 with acute liver failure and 3 receivi

Incorporation of precursors into sterols and proteins in liver biopsies from patients with liver disorders.

Incorporation of 14C-acetate into 3-beta-OH sterols and of 3H-leucine into proteins was examined in liver biopsies from patients with liver disorders. Biopsies obtained from patients in whom no liver disease was found served as controls. Reduced 14C-acetate incorporation into sterols was found in biopsies from patients with chronic active hepatitis and primary biliary cirrhosis. Stimulated incorporation of 3H-leucine into proteins was demonstrated in patients with alcoholic liver cirrhosis and in patients with ulcerative colitis associated with liver disease. No correlation could be establishe

Coupling between sterol and sphingolipid structure in ordered membrane domains.

A hallmark of eukaryotic membranes is the pairing of lineage-specific sterols with characteristic sphingolipid species. Mammalian cell membranes are enriched in both cholesterol and long-chain sphingolipids like sphingomyelin, whereas fungi synthesize ergosterol and very long-chain sphingolipids with sugar-containing head groups. It has been proposed that these two lipid classes co-evolved to support membrane structure and organization. Here we investigated how sterol structure and sphingolipid chain length together control membrane order and phase behavior. In the yeast Saccharomyces cerevisi

Maternal cariprazine exposure effects on lactating offspring sterol biosynthesis.

In the developing brain cholesterol is synthesized by both neurons and glia, and sterol biosynthesis peaks in early postnatal life. Genetic disruptions of sterol biosynthesis genes lead to complex intellectual and developmental disabilities. In addition, multiple commonly prescribed medications can impede sterol homeostasis. Of these, cariprazine (CAR) is one of the strongest prescription medications with sterol biosynthesis inhibiting side effects. CAR inhibits the final steps in cholesterol biosynthesis mediated by the enzyme dehydrocholesterol reductase 7 (DHCR7). This inhibition leads to a

Reversible tuning of membrane sterol levels by cyclodextrin in a dialysis setting.

Large unilamellar vesicles are popular membrane models for studying the impact of lipids and bilayer properties on the structure and function of transmembrane proteins. However, the functional reconstitution of transmembrane proteins in liposomes can be challenging, especially if the hydrophobic thickness of the protein does not match the thickness of the lipid bilayer. Such hydrophobic mismatch causes protein aggregation and low yields during the reconstitution procedure, which are exacerbated in sterol-rich membranes featuring low membrane compressibility. Here, we explore new approaches to
