A new mathematical model of bacterial interactions in two-species oral biofilms.

Periodontitis are bacterial inflammatory diseases, where the bacterial biofilms present on the tooth-supporting tissues switch from a healthy state towards a pathogenic state. Among bacterial species involved in the disease, Porphyromonas gingivalis has been shown to induce dysbiosis, and to induce virulence of otherwise healthy bacteria like Streptococcus gordonii. During biofilm development, primary colonizers such as S. gordonii first attach to the surface and allow the subsequent adhesion of periodontal pathogens such as P. gingivalis. Interactions between those two bacteria have been exte

Binding of the streptococcal surface glycoproteins GspB and Hsa to human salivary proteins.

GspB and Hsa are homologous surface glycoproteins of Streptococcus gordonii that bind sialic acid moieties on platelet membrane glycoprotein Ibalpha. Since this species is an important member of the oral flora, we examined the direct binding of these adhesins to human salivary proteins. Both GspB and Hsa bound low-molecular-weight salivary mucin MG2 and salivary agglutinin. Hsa also bound several other salivary proteins, including secretory immunoglobulin A. Screening of six oral streptococcal isolates revealed that at least two of the strains expressed GspB homologues. These results indicate

Photodynamic antimicrobial effect of safranine O on an ex vivo periodontal biofilm.

BACKGROUND AND OBJECTIVE: The increasing resistance of oral pathogens against antibiotic measures urgently requires new therapeutic strategies. In this context, antimicrobial photodynamic therapy (aPDT) may play a crucial part in the future. The aim of the present study was to compare the antibacterial efficiency of aPDT using the photosensitizer safranine O with that of chlorhexidine (0.2% CHX) on an ex vivo biofilm. METHODS: First the antibacterial activity of both measures against planktonic cultures of Streptococcus gordonii ATCC 33399, Streptococcus mutans ATCC 25175, Fusobacterium nucle

Building a better gingival seal in dental implants: hydrothermal ion incorporation into titanium abutment surfaces.

Despite the recognised biocompatibility of grade V titanium (Ti-6Al-4 V), dental implant abutments have limited affinity for connective tissue, compromising the gingival seal and increasing infection risk. Improving the abutment-tissue seal remains a key challenge. In this study, anodised titanium V surfaces were modified via hydrothermal treatment to incorporate divalent metal ions (Mg2+, Zn2+, Mn2+, Cu2+ and Sr2+). Untreated anodised surfaces (YA) served as controls. The effects of these ion functionalised surfaces were evaluated using Human Gingival Fibroblasts (HGFs) and THP-1 macrophages.

Molecular analysis of bacterial flora associated with chronically inflamed maxillary sinuses.

Chronic maxillary sinusitis is a chronic inflammatory condition in which the role of microbial infection remains undefined. Bacteria have been isolated from chronically inflamed sinuses; however, their role in the chronicity of inflammation is unknown. The objective of this study was to determine whether bacteria are present in clinical samples from chronic maxillary sinusitis and to assess the diversity of the flora present. Washes and/or tissue samples from endoscopic sinus surgery on 11 patients with chronic maxillary sinusitis were subjected to PCR amplification of bacterial 16S rDNA using

Bioactivity of WLBU2 peptide antibiotic in combination with bioerodible polymer.

WLBU2 is a peptide antibiotic designed for broad antimicrobial activity, including bacteria associated with periodontal disease. Although periodontitis is associated with various systemic conditions, ranging from cardiovascular disease to preterm birth, local therapy is needed to treat the source of infection. Biodegradable polymers are often used to control locally the amount and rate of delivery of drugs. In the present study, a bioerodible association polymer comprising cellulose acetate phthalate (CAP) and Pluronic F-127 (PF-127) was explored for its interaction with WLBU2. The intrinsic a

The Effects of Antimicrobial Peptide Nal-P-113 on Inhibiting Periodontal Pathogens and Improving Periodontal Status.

Periodontal disease consists of chronic gingival inflammation characterized by both degradation of the periodontal connective tissue and alveolar bone loss. Drug therapy is used as an auxiliary treatment method in severe chronic periodontitis, aggressive periodontitis, and periodontitis-associated systemic disease. Nal-P-113, a modified antimicrobial peptide, specifically replaces the histidine residues of P-113 with the bulky amino acid β-naphthylalanine, and our previous studies have verified that this novel peptide is not toxic to the human body within a certain concentration range. The obj

Structure of the C-terminal domain of AspA (antigen I/II-family) protein from Streptococcus pyogenes.

The pathogenic bacteria Streptococcus pyogenes can cause an array of diseases in humans, including moderate infections such as pharyngitis (strep throat) as well as life threatening conditions such as necrotizing fasciitis and puerperal fever. The antigen I/II family proteins are cell wall anchored adhesin proteins found on the surfaces of most oral streptococci and are involved in host colonization and biofilm formation. In the present study we have determined the crystal structure of the C2-3-domain of the antigen I/II type protein AspA from S. pyogenes M type 28. The structure was solved to

Absence of capsule reveals glycan-mediated binding and recognition of salivary mucin MUC7 by Streptococcus pneumoniae.

Salivary proteins modulate bacterial colonization in the oral cavity and interact with systemic pathogens that pass through the oropharynx. An interesting example is the opportunistic respiratory pathogen Streptococcus pneumoniae that normally resides in the nasopharynx, but belongs to the greater Mitis group of streptococci, most of which colonize the oral cavity. Streptococcus pneumoniae also expresses a serine-rich repeat (SRR) adhesin, PsrP, which is a homologue to oral Mitis group SRR adhesins, such as Hsa of Streptococcus gordonii and SrpA of Streptococcus sanguinis. As the latter bind t

A Rare Case of Localised Isolated Penile Fournier's Gangrene and a Short Review of the Literature.

Penile Fournier's gangrene (FG) is very rare clinical entity, which is also known as penile necrotizing fasciitis or wet gangrene of the penis. It is associated with increased morbidity and mortality and in the majority of the described cases it affects not only penis but also the adjacent organs and tissues (e.g., bladder, muscles, rectum, testis, and scrotum). We report a rare case of a previously healthy 68-year-old male, who presented with acute isolated penile Fournier's gangrene. Pus culture was identified with pathogens Enterococcus faecalis, Streptococcus gordonii, and Prevotella melan

Characterization of Streptococcus suis genes encoding proteins homologous to sortase of gram-positive bacteria.

Many surface proteins which are covalently linked to the cell wall of gram-positive bacteria have a consensus C-terminal motif, Leu-Pro-X-Thr-Gly (LPXTG). This sequence is cleaved, and the processed protein is attached to an amino group of a cross-bridge in the peptidoglycan by a specific enzyme called sortase. Using the type strain of Streptococcus suis, NCTC 10234, we found five genes encoding proteins that were homologous to sortases of other bacteria and determined the nucleotide sequences of the genetic regions. One gene, designated srtA, was linked to gyrA, as were the sortase and sortas

Probiotic Outer Membrane-Encapsulated Antimicrobial Delivery System against Cariogenic Tristreptococcal Species Biofilms and Oral Caries.

An in vitro model intended to mimic the cariogenic conditions found in the oral cavity is the tristreptococcal species biofilms. Extracellular polysaccharides are the main pathogenic element in these biofilms. Owing to the obstruction of dental surface extracellular polysaccharides, conventional antibiotics have a limited residence time in cariogenic biofilms, leading to poor inhibition of dental plaque. To overcome this problem, we drew inspiration from cell membrane-coated nanoparticles (CMCNPs) and synthesized LA/TCS@PLGA by encapsulating triclosan (TCS)-loaded poly(lactic-co-glycolic acid)

Liposome nanoparticles with photothermal effect for the treatment of bacterial infectious periodontitis.

A novel strategy is imperative to address the limitations of conventional mechanical and pharmacological periodontal treatments, which are constrained by the inherent difficulty in completely eradicating plaque biofilm and the tendency for bacterial resistance to develop. Photothermal therapy (PTT) emerges as an innovative therapeutic approach for managing periodontitis, offering the distinct advantage of circumventing drug resistance. Furthermore, Near-Infrared-II (NIR-II) guided PTT enhances therapeutic efficacy and provides a viable method for eliminating bacteria in deep periodontal pocket

Organization of the capsule biosynthesis gene locus of the oral streptococcus Streptococcus anginosus.

The capsular polysaccharide (CPS) of the important oral streptococcus Streptococcus anginosus, which causes endocarditis, and the genes for its synthesis have not been clarified. In this study, we investigated the gene locus required for CPS synthesis in S. anginosus. Southern hybridization using the cpsE gene of the well-characterized bacterium S. agalactiae revealed that there is a similar gene in the genome of S. anginosus. By using the colony hybridization technique and inverse PCR, we isolated the CPS synthesis (cps) genes of S. anginosus. This gene cluster consisted of genes containing t

Oral mitis group streptococci reduce infectivity of influenza A virus via acidification and H2O2 production.

Members of the mitis group streptococci are the most abundant inhabitants of the oral cavity and dental plaque. Influenza A virus (IAV), the causative agent of influenza, infects the upper respiratory tract, and co-infection with Streptococcus pneumoniae is a major cause of morbidity during influenza epidemics. S. pneumoniae is a member of mitis group streptococci and shares many features with oral mitis group streptococci. In this study, we investigated the effect of viable Streptococcus oralis, a representative member of oral mitis group, on the infectivity of H1N1 IAV. The infectivity of IA
