From Teeth to Body: The Complex Role of Streptococcus mutans in Systemic Diseases.

Streptococcus mutans, the principal pathogen associated with dental caries, impacts individuals across all age groups and geographic regions. Beyond its role in compromising oral health, a growing body of research has established a link between S. mutans and various systemic diseases, including immunoglobulin A nephropathy (IgAN), nonalcoholic steatohepatitis (NASH), infective endocarditis (IE), ulcerative colitis (UC), cerebral hemorrhage, and tumors. The pathogenic mechanisms associated with S. mutans frequently involve collagen-binding proteins (CBPs) and protein antigens (PA) present on th

Effects of Sucrose and Farnesol on Biofilm Formation by Streptococcus mutans and Candida albicans.

Candida albicans (C. albicans) and Streptococcus mutans (S. mutans) are frequently detected in the plaque biofilms of children with early childhood caries. This study investigated the effects of sucrose and farnesol on biofilm formation by the oral pathogens S. mutans and C. albicans, including their synergistic interactions. Biofilm formation dynamics were monitored using the Cell Index (CI). The CI for S. mutans increased in the brain-heart infusion medium, peaking at 10 h; however, the addition of sucrose reduced the CI. For C. albicans yeast cells, the CI increased at sucrose concentration

Anti-biofilm and bacteriostatic effects of three flavonoid compounds on Streptococcus mutans.

Streptococcus mutans (S. mutans) is the main cariogenic pathogen associated with dental caries. Orientin-2''-O-β-L-galactoside, orientin and vitexin are natural flavonoids compound. In this study, the antibacterial ability of these flavonoids and their mechanisms in inhibiting S. mutans biofilm formation were investigated. Inhibition zone and 2-fold-dilution tests showed that these flavonoids exerted inhibitory effects on S. mutans. Phenol sulfuric acid method and lactate dehydrogenase (LDH) test revealed that they could reduce EPS formation and stimulate S. mutans to release LDH. Moreover, cr

Microbial Complexes and Caries in 17-Year-Olds with and without Streptococcus mutans.

Streptococcus mutans is a key bacterial species in the caries process, which affects >90% of the population worldwide. However, other acidogenic and aciduric/acidophilic species may contribute to disease development. In Sweden, a country with low prevalences of caries and S. mutans, a significant portion of caries-affected adolescents lack detectable levels of S. mutans. The objectives of the present study were 1) to characterize the tooth biofilm and saliva microbiota of adolescents with caries disease, with or without detectable S. mutans, from tooth biofilm and saliva samples and 2) to asse

A longitudinal study of Streptococcus mutans colonization in infants after tooth eruption.

We previously reported that, before tooth eruption, over one-half of infants aged 6 mos were already infected with Streptococcus mutans. The aim of this investigation was to determine the colonization of S. mutans after tooth eruption in the same cohort of 111 infants (35 pre-term, 76 full-term). Our results showed that S. mutans colonization increased with increasing age, so that by 24 mos of age, 84% harbored the bacteria (p < 0.01). The mean and median ages of S. mutans colonization in dentate infants were 15.7 mos and 16.0 mos, respectively. Factors associated with S. mutans colonization w

Genetic characterization of a Streptococcus mutans LraI family operon and role in virulence.

Proteins belonging to the LraI (for "lipoprotein receptor antigen") family function as adhesins in several streptococci, as a virulence factor for endocarditis in at least one of these species, and potentially as metal transporters in many bacteria. We have identified and characterized the chromosomal locus containing the LraI family gene (designated sloC) from Streptococcus mutans, an agent of dental caries and endocarditis in humans. Northern blot analysis indicated that sloC is cotranscribed with three other genes. As with other LraI operons, the sloA and sloB genes apparently encode compon

Inhibition of Streptococcus mutans adsorption to hydroxyapatite by low-molecular-weight chitosans.

The role of Streptococcus mutans in the initiation of dental caries has been recognized and attributed, at least in part, to its ability to colonize the tooth surface. Therefore, factors which prevent S. mutans attachment to hydroxyapatite (HA) are of considerable interest for the prophylaxis of this infectious disease. Chitosan, a chitin derivative by N-deacetylation, is an interesting candidate in this respect, since it stimulates the ordered regeneration of oral soft tissues, prevents the deleterious action of organic acid, and exhibits bactericidal action against several pathogens. Sequence analysis of the wall-associated protein precursor of Streptococcus mutans antigen A.

The nucleotide sequence has been determined for the Streptococcus mutans wall-associated protein A (wapA) gene from serotype c strains Ingbritt and GS5. The nucleotide sequence for each wapA gene was virtually identical, although the gene from strain GS5 contained a 24 base pair deletion. A 29 amino acid signal peptide was specified by each wapA gene with a mature protein of 424 amino acids (Mr, 45,276) for strain Ingbritt and 416 amino acids (Mr, 44,846) for strain GS5. In the C-terminal region of the wall-associated protein A, considerable sequence similarity was found with the membrane anch

Geraniol attenuates Streptococcus mutans biofilm and virulence as an anti-caries agent.

Streptococcus mutans-induced dental caries is largely mediated by robust biofilm formation, making it notoriously difficult to treat with conventional antimicrobials. To overcome the limited efficacy and penetration challenges associated with current conventional treatments, this study investigated the in vitro and in silico effects of geraniol on S. mutans biofilm and the expression of genes associated with biofilm formation. Antibacterial activity was evaluated using disk diffusion and minimum inhibitory concentration (MIC) assays. Membrane integrity, anti-biofilm activity, and structural al

Oral hygiene agents at work: effects on Streptococcus mutans and caries risk.

Dental caries remains one of the most prevalent chronic polymicrobial diseases worldwide, driven by acidogenic and aciduric bacteria, most notably Streptococcus mutans, that thrive within oral biofilms. Conventional strategies for caries prevention rely on mechanical plaque removal combined with agents that inhibit bacterial growth, disrupt biofilm formation, or enhance enamel remineralization. Here, we synthesize current evidence regarding a range of key agents that are incorporated into modern oral hygiene products. In addition to describing the mechanisms and efficacy of these agents, we de

Recent advances in membrane vesicles of Streptococcus mutans: biogenesis, regulations, and functions.

BACKGROUND: Streptococcus mutans (S. mutans) is a major cariogenic pathogen in the oral cavity that actively releases membrane vesicles (MVs) encapsulating proteins, lipids, and nucleic acids. These highly heterogeneous MVs not only help regulate homeostasis of the local oral microecology but may also affect systemic health. A comprehensive review of the physiological characteristics of S. mutans MVs is therefore of considerable research value. OBJECTIVE: By summarizing the latest advances in research on S. mutans MVs, this review provides a broad overview of the relevant studies to elucidate

Isofloridoside: a novel inhibitor of Streptococcus mutans biofilm formation and glucosyltransferase activity.

OBJECTIVE: Isofloridoside, a galactose-containing heteroside derived from marine red algae, has potential applications as a sweetener because it can activate the sweetness receptors T1R2/T1R3. The purpose of this study was to investigate the effects of isofloridoside on the growth and sucrose-dependent biofilm formation of the cariogenic bacterium Streptococcus mutans, to evaluate its potential as a caries-preventing agent. RESULTS: The results showed that S. mutans did not grow when isofloridoside was the sole carbon source. Isofloridoside inhibited sucrose-dependent biofilm formation by S.

V-ATPase contributes to the cariogenicity of Candida albicans- Streptococcus mutans biofilm.

The interaction between Candida albicans and Streptococcus mutans plays an important role in the progression of dental caries. The vacuolar proton pump (V-ATPase) is a vital enzyme regulating the growth and virulence of C. albicans, which is a potential target for caries prevention. However, the effect of V-ATPase on the cariogenicity of C. albicans-S. mutans biofilm remains to be explored. In this study, the detection rate of C. albicans in caries-active (group CA) (22.03%) was significantly higher than that in caries-free (group CF) children (8.00%), and the expression of V-ATPase related ge
