Growth and ultrastructure of Streptomyces venezuelae during chloramphenicol production.

Streptomyces venezuelae (3022a) was grown in flask cultures and fermentors, using three media having differential effects on chloramphenicol production. Micromorphology, ultrastructure and chloramphenicol concentrations were studied during the growth cycle in each medium. Chloramphenicol production was greatest in the glycerol-serine-lactate (GSL) medium, less in the glycerol-nutrient broth-yeast extract (GNY) medium and very low in glucose-mineral salts (GA) medium. In GSL and GA, much growth was in the form of microcolonies, especially in flask cultures, while short hyphal fragments predomin

Titer improvement of milbemycins via coordinating metabolic competition and transcriptional co-activation controlled by Streptomyces antibiotic regulatory protein family regulator in Streptomyces bingchenggensis.

Streptomyces bingchenggensis is a promising producer of milbemycins (MILs), the macrolide pesticide used widely in agriculture. The relationship between different biosynthetic gene clusters (BGCs) and the MIL BGC remains unclear, which hinders the precise metabolic engineering of S. bingchenggensis for titer improvement. To address this issue, this study discovered the regulatory function of a previously unidentified regulator KelR on a type-II polyketide BGC, MIL BGC, and two other BGCs, and caused titer improvement. First, a type II polyketide synthase (PKS) gene cluster kel with a bidirecti

Rare distribution of butenolide-type signaling molecules among Streptomyces strains and functional importance as inducing factors for secondary metabolite production in Streptomyces rochei 7434AN4.

Streptomyces rochei 7434AN4 produces two structurally unrelated polyketide antibiotics, lankacidin (LC) and lankamycin (LM), and their biosynthesis is tightly controlled by 2,3-disubstituted butenolide-type signaling molecules SRB1 and SRB2. We here investigated the distribution of 2,3-disubstituted butenolides (SRB-type butenolides) among randomly selected 122 Streptomyces strains using two approaches; (1) feeding of their culture extracts into an srrX-deficient strain KA20 of S. rochei, and (2) co-fermentation with strain KA20. All the randomly selected donor strains, except for Streptomyces

Streptomyces as a Prominent Resource of Future Anti-MRSA Drugs.

Methicillin-resistant Staphylococcus aureus (MRSA) pose a significant health threat as they tend to cause severe infections in vulnerable populations and are difficult to treat due to a limited range of effective antibiotics and also their ability to form biofilm. These organisms were once limited to hospital acquired infections but are now widely present in the community and even in animals. Furthermore, these organisms are constantly evolving to develop resistance to more antibiotics. This results in a need for new clinically useful antibiotics and one potential source are the Streptomyces w

Investigation on antimicrobial agents of the terrestrial Streptomyces sp. BCC71188.

The terrestrial actinomycete strain BCC71188 was identified as Streptomyces by its morphology (having spiral chain spore on the aerial mycelium), chemotaxonomy (containing LL-diaminopimelic acid in the cell wall), and 16S rRNA gene sequence analysis [showing high similarity values compared with Streptomyces samsunensis M1463T (99.85 %) and Streptomyces malaysiensis NBRC 16446T (99.40 %)]. The crude extract exhibited antimalarial against Plasmodium falciparum (IC50 0.19 μg/ml), anti-TB against Mycobacterial tuberculosis (MIC 6.25 μg/ml), and antibacterial against Bacillus cereus (MIC 1.56 μg/ml

The cellobiose sensor CebR is the gatekeeper of Streptomyces scabies pathogenicity.

UNLABELLED: A relatively small number of species in the large genus Streptomyces are pathogenic; the best characterized of these is Streptomyces scabies. The pathogenicity of S. scabies strains is dependent on the production of the nitrated diketopiperazine thaxtomin A, which is a potent plant cellulose synthesis inhibitor. Much is known about the genetic loci associated with plant virulence; however, the molecular mechanisms by which S. scabies triggers expression of thaxtomin biosynthetic genes, beyond the pathway-specific activator TxtR, are not well understood. In this study, we demonstrat

Hopanoids are not essential for growth of Streptomyces scabies 87-22.

Hopanoids are triterpenoic, pentacyclic compounds that are structurally similar to sterols, which are required for normal cell function in eukaryotes. Hopanoids are thought to be an important component of bacterial cell membranes because they control membrane fluidity and diminish passive diffusion of ions, and a few taxons modulate their hopanoid content in response to environmental stimuli. However, to our knowledge, mutational studies to assess the importance of hopanoids in bacterial physiology have never been performed. Genome sequencing of the potato scab pathogen, Streptomyces scabies 8

A New Strain of Streptomyces Causing Common Scab in Potato.

Common scab is a serious disease of potatoes (Solanum tuberosum) and other root and tuber crops, affecting the quality and market value of these crops. The disease is caused by gram-positive soil bacteria in the genus Streptomyces. A new common scab-causing streptomycete was isolated from scabby potatoes originating in southeastern Idaho. Research has supported a model of horizontal transfer of pathogenicity determinants among streptomycetes, and the new strain has hallmarks of the recently characterized Streptomyces pathogenicity island (PAI); it has genes encoding the synthetase for the path

Recommendation for the conservation of the name Streptomyces scabies. Request for an Opinion.

The primary streptomycete inciting common scab of potato was first legitimately described by Thaxter in 1892 as 'Oospora scabies', preserving the spelling of an epithet in use since 1846. The name Streptomyces scabies, dating to 1948, was revived in 1989, but changed to Streptomyces scabiei in 1997 to follow grammatical convention. Considering the long-established use and general recognition of 'scabies', it is proposed that the original epithet be conserved.

Effect of amino acids on thaxtomin A biosynthesis by Streptomyces scabies.

The regulatory effect of amino acids on the production of thaxtomin A, a phytotoxin produced by Streptomyces scabies, was investigated. Tryptophan had an important inhibitory effect on the toxin biosynthesis in all five strains of S. scabies tested. Two other aromatic amino acids (tyrosine and phenylalanine) also inhibited thaxtomin A biosynthesis, while aliphatic amino acids did not cause an important decline in thaxtomin A production. Methylation of tryptophan prevented or reduced the inhibitory effect on thaxtomin A biosynthesis. In spite of the inhibitory action of tryptophan and phenylala

Pathogenicity of Streptomyces scabies Mutants Altered in Thaxtomin A Production.

ABSTRACT To investigate the role of thaxtomin A in the pathogenicity of Streptomyces scabies, mutants altered in thaxtomin A production were obtained by N-methyl-N'-nitro-N-nitrosoguanidine mutagenesis. Mutants of S. scabies EF-35 could be differentiated according to levels of thaxtomin production. Mutants M1, M8, and M19 produced 2 to 20 times less thaxtomin A in oat bran medium than did EF-35. M1 and M19 were deficient in tryptophan catabolism. Thaxtomin production was reduced by about 300 times in mutant M16, which was a glutamic acid auxotroph. No thaxtomin A was detected in M13 culture su

Production of hybrid glycopeptide antibiotics in vitro and in Streptomyces toyocaensis.

BACKGROUND: The glycopeptide antibiotics vancomycin and teicoplanin are currently the last line of defence against some microorganisms that are resistant to many drugs. The emergence of vancomycin-resistant and teicoplanin-resistant enterococci underscores the need for more potent antibiotics. The glycosylation patterns of glycopeptides and chemical modifications of the glycosyl moieties have been shown to greatly influence their antibiotic activity, and certain combinations have resulted in highly active new compounds. To explore further the production of more potent glycopeptide antibiotics,

A novel variant of the catalytic triad in the Streptomyces scabies esterase.

The crystal structure of a novel esterase from Streptomyces scabies, a causal agent of the potato scab disease, was solved at 2.1 A resolution. The tertiary fold of the enzyme is substantially different from that of the alpha/beta hydrolase family and unique among all known hydrolases. The active site contains a dyad of Ser 14 and His 283, closely resembling two of the three components of typical Ser-His-Asp(Glu) triads from other serine hydrolases. Proper orientation of the active site imidazol is maintained by a hydrogen bond between the N delta-H group and a main chain oxygen. Thus, the enz

Regulation and secretion of an extracellular esterase from Streptomyces scabies.

Production of a heat-stable, extracellular esterase by Streptomyces scabies is regulated by zinc ions. The esterase-encoding gene (est) from S. scabies was cloned and expressed in Streptomyces lividans. In S. lividans, expression of the est gene is also regulated by Zn2+, and the esterase is efficiently secreted in this organism. The sequence of the est gene suggests that a 39-amino acid signal peptide is removed during secretion of this protein. Deletion analysis has indicated that the hydrophobic domain of the signal peptide is required for secretion. Gel retardation assays and DNaseI footpr

Catabolism of benzoate and monohydroxylated benzoates by Amycolatopsis and Streptomyces spp.

Eight actinomycetes of the genera Amycolatopsis and Streptomyces were tested for the degradation of aromatic compounds by growth in a liquid medium containing benzoate, monohydroxylated benzoates, or quinate as the principal carbon source. Benzoate was converted to catechol. The key intermediate in the degradation of salicylate was either catechol or gentisate, while m-hydroxybenzoate was metabolized via gentisate or protocatechuate. p-Hydroxybenzoate and quinate were converted to protocatechuate. Catechol, gentisate, and protocatechuate were cleaved by catechol 1,2-dioxygenase, gentisate 1,2-

Effect of some inhibitors on kanamycin formation by Streptomyces kanamyceticus.

The formation of kanamycin is markedly inhibited by mercuric chloride, sodium iodoacetate, 2,4-dinitrophenol, sodium arsenite and sodium azide particularly when these are added at the start of fermentation. Less inhibition of kanamycin synthesis is observed in case of sodium 5,5-diethylbarbiturate, malonic acid, sodium arsenate and sodium fluoride. Inhibition of kanamycin synthesis is associated with growth inhibition in case of 2,4-dinitrophenol, sodium arsenite and sodium azide. Bacitracin and D-cycloserine have a stimulatory effect on kanamycin synthesis with slight inhibition of cellular g

A novel actinobacterium strain BI20T, isolated from the rhizosphere of Dryopteris marginalis, was subjected to a polyphasic taxonomic study. Analysis of the 16S rRNA gene sequence showed that strain BI20T belonged to the genus Streptomyces and shared the highest sequence similarities to Streptomyces cinnamoneus JCM 4633T (98.75%), Streptomyces hiroshimensis JCM 4586T (98.74%) and Streptomyces pseudoechinosporeus NBRC 12518T (98.68%). The Oxytetracycline Producer Streptomyces lydicamycinicus bja295 and Its Genomic Context.

Oxytetracycline (OTC) production is classically attributed to Streptomyces rimosus, and the distribution of its biosynthetic gene cluster (BGC) across the genus is poorly understood. We describe Streptomyces lydicamycinicus bja295 as an OTC producer, the first demonstrated for this species, and the distribution of the BGC across the genus. OTC was identified by mass spectrometry, and the titre reached 200 mg/L on ISP2, the most productive medium tested.
