Gastric ulcer localization by direct in vivo labeling of sucralfate. Work in progress.

We developed and evaluated a new procedure for imaging gastric ulcer disease with technetium 99m-labeled sucralfate. The new method employs direct in vivo labeling of sucralfate instead of in vitro labeling using human serum albumin, as previously reported in the literature. Tests using hydrochloric acid and a rabbit ulcer model showed the efficacy of the direct in vivo labeling technique and the ability of the tagged material to bind to ulcers, respectively. In 26 studies using humans with sucralfate labeled directly in vivo, 15 gave true-negative results and 11 gave true-positive results. Of

Sucralfate in the relief of gastrointestinal symptoms associated with nonsteroidal anti-inflammatory drugs.

Sucralfate is an unabsorbed antiulcer drug that binds to gastrointestinal tissue and protects it from acid and pepsin. Twenty-two arthritic patients taking nonsteroidal anti-inflammatory drugs (NSAIDs) were given sucralfate concomitantly for two weeks in an attempt to lessen gastrointestinal side effects. Changes from baseline in abdominal discomfort were assessed after 2, 7, 10 (or 11), and 14 days of treatment. Sucralfate administration was accompanied by the disappearance of heartburn, epigastric pain, epigastric distress, or epigastric burning in 42 of 59 occurrences, and by statistically

Sucralfate Prevents Pin Site Infections of External Fixators in Open Tibia Fractures.

Pin site infections are the main complication of external fixators. The most common pathogens are Staphylococcus epidermidis and Staphylococcus aureus. The incidence of pin site infections ranges from 2% to 30%. Until now, no satisfactory prevention strategy exists. Therefore, we performed this study to assess the effect of a sucralfate gel 25% applied around the pins of external fixation systems in trauma patients with open tibia fractures. We prospectively studied two groups of patients with open tibia fractures treated with external fixators. In group A patients, pin site care was performed

Sucralfate paste enema: a new method of topical treatment for haemorrhagic radiation proctitis.

AIM: Rectal sucralfate has been reported to deliver clinical improvement in haemorrhagic radiation proctitis, but patients with active proctitis find the enema suspension difficult to retain thus reducing compliance and effectiveness. We describe a novel method of rectal administration via a low-volume sucralfate paste and report its results in a series of 23 patients. METHOD: Patients with rectal bleeding occurring more than 6 months after radiotherapy were shown how to prepare and self-administer sucralfate paste enemas (SPEs) twice daily for 6 weeks. The SPE was prepared using two sucralfa

Topical treatment of chronic venous ulcers with sucralfate: a placebo controlled randomized study.

Venous leg ulcers are an important medical issue due to their high incidence in the elderly and the lack of a standard curative approach. Apart from surgical therapy, different medical treatments to effect ulcer wound repair and regeneration are currently being investigated. Sucralfate is a cytoprotective agent employed to prevent or treat several gastrointestinal diseases such as gastroesophageal reflux, gastritis, peptic ulcer, stress ulcer and dyspepsia. In this study we evaluated the efficacy, safety and tolerability of topical sucralfate (SUC-LIS 95) on the healing of chronic venous leg u

Sucralfate impedes intestinal epithelial sheet migration in a Caco-2 cell culture model.

Sucralfate, which binds to the matrix of the ulcer bed, is theoretically advantageous for duodenal ulcer therapy, but has not fulfilled its promise clinically. We examined the effects of sucralfate and related compounds in a human intestinal epithelial (Caco-2) cell culture model of restitution on sheet migration across and adhesion to collagen I. Migration was quantitated across a collagen I matrix treated with sucralfate or related compounds and correlated with cell adhesion. Caco-2 motility was significantly and dose-responsively inhibited by sucralfate at therapeutic luminal concentrations

Prevention of upper gastrointestinal bleeding in long-term ventilated patients. Sucralfate versus ranitidine.

Thirty-two long-term ventilated patients were randomly selected for a study of the efficacy of sucralfate (1 g six times per day via gastric tube) versus ranitidine (six 50-mg to six 100-mg doses per day intravenously) for the prevention of upper gastrointestinal bleeding. The patients of the two treatment groups (each 16) were comparable with respect to diseases precipitating acute respiratory failure and risk factors of bleeding, e.g., renal failure, thrombopenia, coagulopathy, and anticoagulant treatment. Mean duration of mechanical ventilation was 7.4 in sucralfate- and 7.7 days in ranitid

Effects of different antisecretory drugs on gastric potential difference in the rat: comparison with sucralfate.

The proton pump inhibitors omeprazole and lansoprazole and the histamine H2 receptor antagonists ranitidine and nizatidine were investigated for their effects on gastric transmucosal potential difference (PD) in the rat, in comparison with the gastroprotective compound sucralfate. Omeprazole (1-3 mg kg-1, i.v.) and lansoprazole (1-3 mg kg-1, i.v.) did not modify basal PD, but significantly reduced (by approx. 50-60%) the drop in PD caused by intragastric administration of acetylsalicylic acid (ASA, 60 mg kg-1). Ranitidine (3-100 mg kg-1, i.v.) and nizatidine (10-30 mg kg-1, i.v.) behaved simil

Effect of long-term sucralfate ingestion on antral and fundic epithelial proliferation in the rat.

Sucralfate accelerates the healing of chronic gastric ulcers, but its mechanism is not well understood. We studied the effect of long-term administration of sucralfate on gastric epithelial proliferation in the rat by means of tritiated thymidine autoradiography. Rats were treated perorally with 500 mg/kg sucralfate once a day. After 28 days, rats were injected with tritiated thymidine 1 microCi/g body weight and sacrificed 1 hr later. Autoradiographs from antral and fundic mucosae were prepared and a number of proliferative measurements were made. Long-term sucralfate administration produced

Sucralfate and sodium sucrose sulphate in the treatment of superficial corneal disease in keratoconjunctivitis sicca.

We have studied the effect of the aluminum complex of sucrose sulphate (Sucralfate suspension) and the sodium salt of sucrose sulphate (sodium sucrose sulphate solution) on patients with keratoconjunctivitis sicca. Eyes treated with either of these two drugs showed a decrease in painfulness and blurring of vision. On examination the surface area of the corneal lesions, stained with fluorescein, diminished during treatment. As for the difference in effect between the two eyedrops, the solution was better tolerated.

Oesophageal reflux disease. Diagnosis, pathophysiology and treatment with special reference to the role of sucralfate.

The direct cause of oesophageal reflux disease is readily apparent, as oesophageal reflux of gastric contents has a deleterious effect on the oesophageal mucosa. Reflux is a direct result of a multiplicity of aetiological factors which interact in many ways. The symptoms of the disorder can masquerade as other conditions such as cardiovascular, respiratory and abdominal problems. The diagnosis is suggested from the symptoms and confirmed by endoscopy with biopsy. Among additional diagnostic test, oesophageal pH monitoring using a portable system is a simple procedure providing qualitative and

High Economic Burden Yet Low Clinical Value: Why Is so Much Sucralfate Prescribed?

Sucralfate is a commonly prescribed medication in use for decades. Despite the availability of more effective medications for acid-dyspeptic conditions, including proton pump inhibitors and histamine H2-receptor antagonists, sucralfate continues to be utilized for upper gastrointestinal symptoms. We reviewed the limited evidence base supporting the approval and ongoing usage of sucralfate and performed a cost analysis using the ClinCalc database. The financial impact of sucralfate was comparable to famotidine, lansoprazole and ranitidine. We encourage further studies to accurately gauge the tr

Triple therapy with sucralfate, tetracycline, and metronidazole for Helicobacter pylori-associated duodenal ulcers.

UNLABELLED: Triple therapy with bismuth, metronidazole, and tetracycline or amoxicillin is effective for the treatment of Helicobacter pylori, but side effects are common. Sucralfate inhibits H. pylori hemagglutinin, protease, and lipase and thus might affect colonization of the bacterium in the stomach. OBJECTIVE: We compared the efficacy and side effects of triple therapy with sucralfate versus triple therapy with bismuth plus omeprazole in the treatment of H. pylori-associated duodenal ulcer (DU). METHODS: One hundred and fifty DU patients were recruited in this study; 71 cases were rando

Right colonic hemangioma diagnosed by scintigraphy with 99-mTc sucralfate. A case report.

99-mTc-SF gastrointestinal scintigraphy used in recent years in the assessment of site and activity for ulcerative lesions, was shown to be very important for the diagnosis of a rare case of right colonic hemangioma. A young female patient suffering for some time from functional intestinal disorders, following negative radiologic and endoscopic findings, underwent 99-mTc-SF scintigraphy, which showed massive uptake of radioactivity in the ascending and right flexure of colon. A repeated colonoscopy with biopsy allowed the histologic diagnosis of capillary hemangioma.

Effective Use of Topical Sucralfate in the Conservative Management of Expanded Gastrostomy Tract Reduction.

Sucralfate is a common medication used to treat duodenal ulcers, gastric ulcers, and gastritis. The off-label use of topical sucralfate has been described in the literature to induce wound healing in epithelial injury. Yet, current literature lacks clinical depictions in the application of sucralfate to treat a common gastrostomy tube complication, that of a dilated gastrostomy site. We present a case report of a medically complex pediatric patient where topical sucralfate was applied to reduce the size of a large gastrostomy stomal defect. Sucralfate was used to reduce healing time and allow
