Taurine (2-aminoethanesulfonic acid) is the most abundant free amino acid in mammalian cells. It plays a significant role in cell development, nutrition, and survival, such as in the regulation of ion transport and osmoregulation. Cysteine sulfinate decarboxylase (CSD) is the rate-limiting biosynthetic enzyme of taurine. Recently, the synthesis of taurine has been observed in the central nervous system, kidney, liver, and muscle. However, the synthesis of taurine in the salivary glands has still not been described in detail. We have detected CSD expression in the major salivary glands of adult

Cysteine-bound persulfides (Cys-SSH) in proteins are sulfur carrier intermediates in the synthesis of essential cofactors such as iron-sulfur clusters, molybdenum cofactor, vitamin (thiamine), and thionucleosides (thiolated tRNA). Protein-bound persulfides are also used for signaling purposes as a carrier of the "H2S" signal. Several methods have been developed to detect and quantify cysteine-bound persulfides in protein and monitor their exchange. The main challenge in developing these techniques is to discriminate persulfidated cysteine from cysteine and other cysteine modifications. It is a

Nitrile hydratase (NHase, EC 4.2.1.84) is a metalloenzyme which catalyses the conversion of nitriles to amides. The high efficiency and broad substrate range of NHase have led to the successful application of this enzyme as a biocatalyst in the industrial syntheses of acrylamide and nicotinamide and in the bioremediation of nitrile waste. Crystal structures of both cobalt(III)- and iron(III)-dependent NHases reveal an unusual metal binding motif made up from six sequential amino acids and comprising two amide nitrogens from the peptide backbone and three cysteine-derived sulfur ligands, each a

Allicin, a bioactive sulfur compound from garlic known for its antimicrobial and immunomodulatory properties, was evaluated in this study for its effects on growth, antioxidant activity, gut microbiota, and antibiotic resistance genes (ARGs) in Trachidermus fasciatus. Fish were administered allicin at concentrations of 100 mg/kg, 200 mg/kg, and 300 mg/kg. The 200 mg/kg allicin group had significantly higher WGR, LGR, and SGR than the control group. Hepatic SOD and LZM activities were also higher in the 200 mg/kg group. Metagenomics showed that allicin altered the gut microbiota composition, de

Domestic cats and dogs are carnivores that have evolved differentially in the nutrition and metabolism of amino acids. This article highlights both proteinogenic and nonproteinogenic amino acids. Dogs inadequately synthesize citrulline (the precursor of arginine) from glutamine, glutamate, and proline in the small intestine. Although most breeds of dogs have potential for adequately converting cysteine into taurine in the liver, a small proportion (1.3%-2.5%) of the Newfoundland dogs fed commercially available balanced diets exhibit a deficiency of taurine possibly due to gene mutations. Cyclic peptides hold great promise in drug discovery due to their conformational stability, proteolytic resistance, and ability to disrupt protein-protein interactions (PPIs). However, their therapeutic potential is often constrained by reversible binding, while covalent labeling strategies such as Sulfur(VI) Fluoride Exchange (SuFEx) have largely been limited to linear scaffolds. Here, we report a late-stage Pd(II)-catalyzed ortho-C(sp2)-H alkenylation of tyrosine that achieves directing-group-free late-stage peptide macrocyclization. This strategy preserves the phenolic side chain, which can

Peptidases are regulated by latency and inhibitors, as well as compatibilization and cofactors. Ulilysin from Methanosarcina acetivorans, also called lysargiNase, is an archaeal metallopeptidase (MP) that is biosynthesized as a zymogen with a 60-residue N-terminal prosegment (PS). In the presence of calcium, it self-activates to yield the mature enzyme, which specifically cleaves before basic residues and thus complements trypsin in proteomics workflows. Here, we obtained a low-resolution crystal structure of proulilysin, in which 28 protomers arranged as 14 dimers form a continuous double hel

We assessed whether allicin, through its antihypertensive and antioxidant effects, relieves vascular remodeling, endothelial function, and oxidative stress (OS), thereby improving experimental pulmonary arterial hypertension (PAH). Allicin (16 mg/kg) was administered to rats with PAH (monocrotaline 60 mg/kg). Allicin encouraged body weight gain and survival rate, and medial wall thickness and the right ventricle (RV) hypertrophy were prevented. Also, angiotensin II concentrations in the lung (0.37 ± 0.01 vs. 0.47 ± 0.06 pmoles/mL, allicin and control, respectively) and plasma (0.57 ± 0.05 vs.

Garlic (Allium sativum L.) is a well-known spice widely utilised for its medicinal properties. There is an extensive record of the many beneficial health effects of garlic which can be traced back to as early as the ancient Egyptian era. One of the most studied properties of garlic is its ability to cure certain ailments caused by infections. In the 1940s, the antimicrobial activities exhibited by garlic were first reported to be due to allicin, a volatile compound extracted from raw garlic. Since then, allicin has been widely investigated for its putative inhibitory activities against a wide

Candidiasis is a term describing infections by yeasts from the genus Candida, and the type of infection encompassed by candidiasis ranges from superficial to systemic. Treatment of such infections often requires antifungals such as the azoles, but increased use of these drugs has led to selection of yeasts with increased resistance to these drugs. In this study, we used allicin, an allyl sulfur derivative of garlic, to demonstrate both its intrinsic antifungal activity and its synergy with the azoles, in the treatment of these yeasts in vitro. In this study, the MIC(50) and MIC(90) of allicin

A modular, three-component, and sustainable one-pot/one-step protocol has been developed for the efficient and regioselective synthesis of libraries of γ-keto sulfones and γ-keto phosphine oxides through concurrent CC and CX bond formation (X = S or P). The acidic eutectic mixture ChCl/p-TSA·H2O (1:2) (ChCl = choline chloride; p-TSA = p-toluenesulfonic acid) serves as both promoter and reaction medium. This transformation involves a cascade process comprising three consecutive steps: i) hydration of terminal alkynes to methyl ketones; ii) Claisen-Schmidt condensation with aldehydes, and iii)

BACKGROUND: Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is one of the major targets of NO in cells, especially in neurodegenerative diseases. S-Nitrosylation of GAPDH is accompanied by its translocation into the nucleus with subsequent apoptosis. The product of GAPDH modification by NO is considered to be S-nitrosylated GAPDH (GAPDH-SNO). However, this has not been confirmed by direct methods.
METHODS: Products of GAPDH modification in the presence of the NO donor diethylamine NONOate were analyzed by MALDI- and ESI- mass spectrometry methods.
RESULTS: The adduct between GAPDH and dimed

Taurine is the most abundant free amino acid in the body and is present at high concentrations during development and in the early milk. It is synthesized from cysteine via oxidation of cysteine to cysteinesulfinate by the enzyme cysteine dioxygenase (CDO), followed by the decarboxylation of cysteinesulfinate to hypotaurine, catalyzed by cysteine sulfinic acid decarboxylase (CSAD). To determine whether the taurine biosynthetic pathway is present in mammary gland and whether it is differentially expressed during pregnancy and lactation, and also to further explore the possible regulation of hep

Ascites syndrome (AS) is a deadly condition in fast-growing chickens, preceded by pulmonary arterial hypertension (PAH), where the angiotensin II type 1 receptor (ATR1) plays a role. We investigated whether allicin (ALLI), a garlic derivative, could (a) interact with broiler ATR1, (b) affect ascites-related traits [haematocrit content (Hct%), blood oxygen saturation (SaO2), and the right-to-total ventricular weight ratio (RV:TV)], (c) modify ATR1 expression in the lung, heart, and liver, alongside ascites mortality and growth performance in Ross 308 broilers raised at high altitude and under c
