Talaromycosis is an opportunistic fungal infection caused by the thermally dimorphic fungus Talaromyces marneffei. During infection, T. marneffei survives within host phagocytic cells, where it encounters iron restriction and host-derived stress conditions. Therefore, regulation of iron homeostasis is essential for fungal adaptation and pathogenicity. HapX is a conserved bZIP transcription factor involved in iron-responsive regulation in several fungal species, but its role in T. marneffei has not been investigated. In this study, we examined the function of HapX in iron-dependent growth, stre

The bioassay-guided isolation of an endophytic fungus Talaromyces funiculosus TF37 yielded a total of eleven compounds (1-11), including three undescribed compounds, a bicyclic tetralone named tanicutone D (1) and two prenylated indole alkaloids named mangrovamides L (4) and M (5). The undescribed structures were elucidated by comprehensive spectroscopic analyses of HR-ESI-MS, 1D and 2D NMR, as well as ECD calculations. All isolates were evaluated by the antimicrobial assay against Staphylococcus aureus, Saccharomyces cerevisiae, Candida albicans, and MRSA. Compounds 1-3 were found to have sig

Five previously unreported N-methyl-4-hydroxy-3,5-disubstituted-2-pyridinone analogues, funiculosins B-F (1-5), along with the known compound funiculosin (6) were isolated by bioassay-guided fractionation from Talaromyces cecidicola J-4. Their structures were established by extensive spectroscopic analysis. The absolute configurations were tentatively assigned based on biogenetic considerations and electronic circular dichroism calculations. The availability of these compounds enabled a subsequent small antifungal structure-activity relationship study that revealed tetrahydroxycyclopentyl subs

PURPOSE: Talaromycosis is an invasive fungal infection caused by the pathogenic fungus Talaromyces marneffei (T. marneffei), prevalent in Southeast Asia and Southern China. This disease is rare in non-endemic areas and primarily affects the respiratory system with atypical manifestations. Therefore, the condition is often misdiagnosed as other respiratory infections. This article presents two cases of talaromycosis in non-endemic areas, aiming to provide diagnostic and therapeutic references for this rare fungal infection.

T.marneffei, encountered mostly in Southeast Asia, leads to a systemic infection, especially in immunocompromised individuals such as HIV-infected patients with low CD4 level. A 32-year-old male patient, residing in Hong Kong for the last two years, admitted with fever, cough, weakness, and weight loss. Physical examination revealed bilateral cervical and axillary multiple lymph nodes and hepatosplenomegaly. Screening of the pancytopenic patient revealed HIV infection. Histopathological examination of the cervical lymph node revealed plasmoblastic lymphoma. Blood and urine cultures remained st

Pathogenic eukaryotes including fungi release extracellular vesicles (EVs) which are composed of a variety of bioactive components, including peptides, nucleic acids, polysaccharides, and membrane lipids. EVs contain virulence-associated molecules suggesting a crucial role of these structures in disease pathogenesis. EVs derived from the pathogenic yeast phase of Talaromyces (Penicillium) marneffei, a causative agent of systemic opportunistic mycoses "talaromycosis," were studied for their immunogenic components and immunomodulatory properties. Some important virulence factors in EVs including

BACKGROUND: Little is known about the clinical characteristics of talaromycosis with hyper-immunoglobulin E syndrome (HIES).
METHODS: We conducted a multicenter retrospective study, which included 7 hospitals from 2016 to 2022. Five consecutive cases of human immunodeficiency virus (HIV)-negative patients with systemic Talaromyces marneffei infections due to STAT3-HIES were identified. A systematic literature review of original articles published in English identified an additional 7 cases. Clinical characteristics and laboratory parameters were collected.
RESULTS: Forty-two percent (5/12) o

Talaromyces marneffei tends to induce systemic infection in immunocompromised individuals, which is one of the causes of the high mortality. The underlying molecular mechanisms of T.marneffei-induced abnormal liver function are still poorly understood. In this study, we found that T.marneffei-infected patients could develop abnormal liver function, evidenced by reduced albumin and increased levels of aspartate aminotransferase (AST) and AST/alanine aminotransferase (ALT). T. marneffei-infected mice exhibited similar characteristics. In vitro investigations showed that T.marneffei induced the d

Quinones represent an important group of highly structurally diverse, mainly polyketide-derived secondary metabolites widely distributed among filamentous fungi. Many quinones have been reported to have important biological functions such as inhibition of bacteria or repression of the immune response in insects. Other quinones, such as ubiquinones are known to be essential molecules in cellular respiration, and many quinones are known to protect their producing organisms from exposure to sunlight. Most recently, quinones have also attracted a lot of industrial interest since their electron-don

BACKGROUND: Talaromyces marneffei (TM) is a life-threatening opportunistic fungal pathogen, particularly in individuals with advanced HIV/AIDS. Early and accurate diagnosis remains challenging due to the limited sensitivity and specificity of conventional microbiological and antigen-based tests. Mp1p, a secreted TM-specific virulence protein, has emerged as a promising target for serological diagnosis. This study aimed to evaluate the diagnostic performance of Mp1p antigen and antibody detection using ELISA in sera from HIV infected patients with confirmed TM infection. METHODS: A total of 36

In the last few years, next-generation sequencing (NGS) has emerged as a technology for laboratory diagnosis of many culture-negative infections and slow-growing microorganisms. In this study, we describe the use of metagenomic NGS (mNGS) for rapid diagnosis of T. marneffei infection in a 37-year-old renal transplant recipient who presented with chronic pneumonia syndrome. Bronchoalveolar lavage for mNGS was positive for T. marneffei sequence reads. Prolonged incubation of the bronchoalveolar lavage revealed T. marneffei colonies after 6 days of incubation. Analysis of 23 cases of T. marneffei

Siderophores are compounds with low molecular weight with a high affinity and specificity for ferric iron, which is produced by bacteria and fungi. Fungal siderophores have been characterized and their feasibility for clinical applications has been investigated. Fungi may be limited in slow growth and low siderophore production; however, they have advantages of high diversity and affinity. Hence, the purpose of this study was to generate a genetically modified strain in Talaromyces marneffei that enhanced siderophore production and to identify the characteristics of siderophore to guide its me

Talaromyces marneffei (T. marneffei), formerly Penicillium marneffei, is a dimorphic fungus prevalent in Southeast Asia that can cause severe systemic infection, especially in immunocompromised patients. There are few reports about the use of posaconazole in T. marneffei infection. Here, we present a case of pulmonary T. marneffei infection in a renal transplant recipient. The patient responded rapidly to oral posaconazole administration but experienced serum creatinine fluctuation because of the interaction between posaconazole and immunosuppressants. Seven months after adjusting the dose of

Waste-derived fertilizers can contribute to circular nutrient management when nutrient availability is coupled with reduced leaching. This study evaluated a 1:1 sewage sludge ash (SSA)-fish meal (FM) formulation applied to soil at 1% (w/w), with or without 0.1% (w/w) macerated Talaromyces adpressus preparation. Soil column leaching assays demonstrated that fungal bioaugmentation significantly reduced cumulative nutrient losses, particularly for phosphorus (P), potassium (K), and sodium (Na) indicating enhanced retention. Soil columns (n = 3 per treatment) were leached for four weeks wherein ev

Verticillium wilt, caused by Verticillium dahliae Kleb., is a destructive vascular disease that is difficult to control using chemical fungicides and remains a major challenge in plant disease management. Microbial biocontrol represents a sustainable alternative, and the identification of highly antagonistic strains against V. dahliae is of considerable importance. In this study, a strain of Talaromyces purpureogenus TS, which demonstrated strong antagonistic and mycoparasitic activity against V. dahliae, was isolated from soil. Its cell-free supernatant inhibited the growth of V. dahliae by d
