Central projections of the nervus terminalis in lampreys, lungfishes, and bichirs.

Central projections of the nervus terminalis were investigated in a cyclostome (Lampetra planeri), in a sarcopterygian (Protopterus dolloi), and in an actinopterygian fish (Polypterus palmas), following the injection of horseradish peroxidase into the olfactory epithelium. Despite differences in forebrain morphology (inversion versus eversion of the hemispheres), projections of the terminal nerve are similar in the species investigated. The nervus terminalis courses through the subpallium (septum) and mainly innervates periventricular nuclei in the telencephalon and diencephalon. In lampreys,

The development of a simple scaffold of axon tracts in the brain of the embryonic zebrafish, Brachydanio rerio.

We have examined neuronal differentiation and the formation of axon tracts in the embryonic forebrain and midbrain of the zebrafish, between 1 and 2 days postfertilisation. Axons were visualised with three techniques; immunocytochemistry (using HNK-1 and antiacetylated tubulin antibodies) and horseradish peroxidase (HRP) labelling in whole-mounted brains, and transmission electron microscopy. Differentiation was monitored by histochemical staining for acetylcholinesterase (AChE). These independent methods demonstrated that a simple grid of tracts and commissures forms the initial axon scaffold

Effect of methotrexate on neuroepithelium in the rat fetal brain.

Pregnant rats were treated with 30 mg/kg of methotrexate (MTX) on gestation day 13, and fetal brains were examined histopathologically from 6 to 48 hr after the treatment. In the telencephalon of the control group, there were few pyknotic neuroepithelial cells throughout the experimental period. Six hr after MTX treatment, several pyknotic neuroepithelial cells scattered throughout the telencephalic wall. At 12-36 hr, pyknotic neuroepithelial cells increased significantly and were diffusely distributed throughout the telencephalic wall. Neuroepithelial cells were eliminated and showed sparse c

Differential brain distribution of gonadotropin-releasing hormone receptors in the goldfish.

The present study describes the differential distributions in the brain of the two goldfish gonadotropin-releasing hormone (GnRH) receptors, using both immunohistochemistry and in situ hybridization approaches. The goldfish GnRH GfA and GfB receptors are variant forms of the same receptor subtype, although with distinct differences in ligand binding characteristics, and differential distributions in the pituitary and body tissues [Proc. Natl. Acad. Sci. USA 96 (1999) 2526]. The goldfish GnRH GfA receptor was found to be widespread throughout the brain, with neurons showing immunoreactivity in

Immunocytochemical localization of mammalian GnRH (gonadotropin-releasing hormone) and chicken GnRH-II in the brain of the European silver eel (Anguilla anguilla L.).

Using specific antibodies for the two molecular forms of gonadotropin-releasing hormone (GnRH) present in the European eel, Anguilla anguilla, (mammalian GnRH, mGnRH, and chicken GnRH II, cGnRH-II), we employed immunocytochemistry to determine the distribution of these two peptides in the brain and in the pituitary. The results indicate that mGnRH and cGnRH-II are localized in different neurons: mGnRH-immunoreactive (ir) perikaria were observed in the olfactory bulbs, the junction between olfactory bulbs and telencephalon (nucleus olfactoretinalis), the telencephalon, the preoptic region and t

Hif1α-dependent hypoxia signaling contributes to the survival of deep-layer neurons and cortex formation in a mouse model.

Hypoxia-inducible factor 1 α (Hif1α) plays a crucial role in brain development. To study the function of Hif1α in early brain development, we generated neuroepithelial cell-specific Hif1α-knockout mice. Hif1α-knockout mice died soon after birth; these mice exhibited an abnormal head shape, indicating the presence of brain defects. Morphological analysis revealed that Hif1α ablation reduced the overall size of the brain, especially affecting the telencephalon. Neuronal apoptosis predominantly occurred in deep-layer neurons, consequently the alignment of cortical layers was severely disorganized

Ectopic expression of telencephalin in brains with holoprosencephaly.

Telencephalin (TLN), a telencephalon-specific glycoprotein, is exclusively expressed in neurons of the mammalian telencephalon. In the normally developing human brain, TLN immunoreactivity appeared and increased from 35 gestational weeks (GW) in the temporal cortex, and reached adult level at 5 months of postnatal age, being strong in the molecular layer, and weak in the external and internal granular layers. TLN expression corresponded with the development of neuronal dendrites and synapses. In brains with holoprosencephaly TLN immunoreactivity was already strong from as early as 28 GW. Stain

Two differing salmon GnRH precursor mRNAs are co-expressed in the brain of sockeye salmon.

The localization of two salmon-type gonadotropin-releasing hormone (sGnRH) precursors, pro-sGnRH-I (short type) and pro-sGnRH-II (long type), was investigated by using in situ hybridization techniques in the brain of the landlocked sockeye salmon, Oncorhynchus nerka. We used 30-mer oligonucleotide probes complementary to pro-sGnRH-I and pro-sGnRH-II cDNA. No significant differences were observed in the localization of sGnRH neurons expressing pro-sGnRH-I and pro-sGnRH-II mRNAs; both were expressed in the olfactory nerve, the olfactory bulbs, the regions between the olfactory bulb and telenceph

FMRFamide-like immunoreactivity in brain and pituitary of Xenotaca eisenii: (Ciprinidontoformes, Teleostei).

FMRFamide-immunoreactive neurons were determined immunocytochemically by means of the PAP-technique for unlabeled antibodies. FMRFamide-containing perikarya were found in the frontoventral telencephalon, in the valvula and corpus cerebelli, and in the rostral and caudal medulla oblongata. FMRFamide immunoreactive cells were localized in the anterio lobe of the pituitary. FMRFamide fibres occurred in the bulbus olfactorius, in the anterior, medial and lateral parts of the telencephalon, in the commissura anterior, beneath the nucleus praeopticus, beneath the nucleus dorsomedialis, ventrolis and

Novel Insights into Emx2 and Dmrta2 Cooperation during Cortex Development and Evidence for Dmrta2 Function in the Choroid Plexus.

Early dorsal telencephalon development is coordinated by an interplay of transcription factors that exhibit a graded expression pattern in neural progenitors. How they function together to orchestrate cortical development remains largely unknown. The Emx2 and Dmrta2 genes encode transcription factors that are expressed in a similar caudomedialhigh/rostrolaterallow gradient in the ventricular zone of the developing dorsal telencephalon with, in the medial pallium, Dmrta2 but not Emx2 expressed in the developing choroid plexus (ChP). Their constitutive loss has been shown to impart similar corti

Fluoxetine affects weight gain and expression of feeding peptides in the female goldfish brain.

Serotonin has been implicated in the regulation of feeding and growth in vertebrates. However, the mechanisms through which serotonin mediates its anorectic effects are only partially understood. In this study we measured food intake and difference in weight gain in sexually regressed female goldfish intraperitionally injected with fluoxetine, a selective serotonin reuptake inhibitor (SSRI). The experiment was conducted in July, a period in which female goldfish show maximum body growth rates. After an acclimation period of one week, goldfish were injected every 3 d with 5 microg/g body weight

The brain of the basking shark (Cetorhinus maximus).

The brain of a basking shark is described for the first time. Allometrical analysis of brain-body weight relationships indicates that Cetorhinus maximus shows by far the lowest degree of cerebralization of any shark investigated to date, possibly reflecting its plankton-feeding mode of life. Furthermore, some external morphological features of the brain appear to be species-specific. The proportions of brain parts indicate a primitive vertebrate brain organization. The size of the telencephalon, 34% of the total brain, equals that in some other sharks, whereas the cerebellum, 30% of the total

Correlations between major brain regions in Chiroptera.

Unlike small homogeneous nuclei, progression indices of large heterogeneous brain regions show no negative correlations and give no evidence of compensatory effects, presumably because the evolutionary progressions and regressions of multiple neighboring functions mask each other. The diencephalon and various parts of the telencephalon show large positive correlations, apparently reflecting the general increase in size of the brain, relatively to body weight, from presumably primitive to advanced forms. The neocortex is strongly correlated with the striatum, a putative precursor, and with the

Effects of Pregnant Fatigue on the Development of Offspring in Rats.

AIM: To explore the correlation between pregnant fatigue and intrauterine physical and neural development of offspring in rats. METHODS: Sprague-Dawley pregnant rats were randomly divided into a normal control group, a mild fatigue group (stand in water for 6 hours/day), and a severe fatigue group (stand in water for 15 h/day). The levels of lactic acid, 5-Hydroxytryptamine and Interleukin-6 in cardiac serum of rats were used to evaluate the fatigue. The expression of S100β in the telencephalon, Insulin-like growth factor-1 (IGF-1) in the liver and Cyclooxygenage-2 (COX-2) in the small intest

Spred-2 expression is associated with neural repair of injured adult zebrafish brain.

Sprouty-related protein-2 (Spred-2) is a negative regulator of extracellular signal-regulated kinases (ERK) pathway, which is important for cell proliferation, neuronal differentiation, plasticity and survival. Nevertheless, its general molecular characteristics such as gene expression patterns and potential role in neural repair in the brain remain unknown. Thus, this study aimed to characterise the expression of spred-2 in the zebrafish brain. Digoxigenin-in situ hybridization showed spred-2 mRNA-expressing cells were mainly seen in the proliferative zones such as the olfactory bulb, telence

Modeling human cortical development in vitro using induced pluripotent stem cells.

Human induced pluripotent stem cells (hiPSCs) are emerging as a tool for understanding human brain development at cellular, molecular, and genomic levels. Here we show that hiPSCs grown in suspension in the presence of rostral neuralizing factors can generate 3D structures containing polarized radial glia, intermediate progenitors, and a spectrum of layer-specific cortical neurons reminiscent of their organization in vivo. The hiPSC-derived multilayered structures express a gene expression profile typical of the embryonic telencephalon but not that of other CNS regions. Their transcriptome is

Induction of agenesis of the corpus callosum by the destruction of anlage of the olfactory bulb using fetal laser surgery exo utero in mice.

It has long been discussed why some congenital anomalies were often involved with abnormalities in other organs, for example, brain anomalies accompanied by limb anomalies or cleft palate. The mechanism of combined abnormalities has been mysterious, and usually explained as pleiotropism. A combination between agenesis of the olfactory bulb and agenesis of the corpus callosum has been reported. In the present experiments, it has been suggested that non-attachment of the olfactory nerve to the rostro-ventral tip of the telencephalon blocked the induction of the olfactory bulbs from the telenceph

Environmentally-relevant concentrations of the antipsychotic drugs sulpiride and clozapine induce abnormal dopamine and serotonin signaling in zebrafish brain.

The presence of drugs in surface and groundwaters adversely affects the physiological function of non-target organisms due special activities that can pose a serious threats to various forms of aquatic life. Psychotropic drugs are one of the most commonly used drugs in the world. Hence, the aim of this study was to investigate the effect of environmentally-relevant concentrations of the antipsychotic drugs, sulpiride and clozapine, on dopaminergic (DAergic) and serotonergic (5-HTergic) neurotransmitter systems in the brain of zebrafish. Adult zebrafish (AB strain) were exposed to the environme

Dorsal thalamic nuclei in Caiman crocodilus.

In Caiman crocodilus, identification of nuclei that comprise the dorsal thalamus was determined by: injections of retrograde tracers into cortex/pallium; injections of retrograde tracers into the noncortical telencephalon; and injections of anterograde tracers into thalamic nuclei. With the exception of nucleus dorsolateralis anterior, which has bilateral projections, all other dorsal thalamic nuclei send axons to terminate in the ipsilateral telencephalon. Nuclei that only projected to cortex/pallium were: dorsolateralis anterior; diagonalis; and dorsal geniculate. Neuronal aggregates that se
