Identification and characterization of Theileria annulata heat-shock protein 90 (HSP90) isoforms.

Heat-shock proteins (HSPs) refer to a group of proteins whose synthesis is enhanced upon sudden increase in temperature or exposure to a variety of other stressors. In this study, Theileria annulata (T. annulata) HSP90 was identified and characterized as a first step to understand the function of this molecule in T. annulata-infected cells. Our results indicated the existence in the genome of T. annulata of two HSP90 genes: one located in chromosome one (TaHSP90-Chr1) and the other in chromosome four (TaHSP90-Chr4). The amino acid alignment between the two isoforms has shown identity and simil

Delayed type hypersensitivity theilerin test in cattle vaccinated against Theileria annulata infection.

Theilerin was prepared and its total nitrogen was determined at level of 1 mg/ml. Seventy-two Holstein Friesian cattle recovered from acute theileriosis or vaccinated received 0.1 ml of theilerin intradermally at normal concentration of total nitrogen or other dilutions (1 / 10 and 1 / 100). Some 60(% of calves and 66.67% of milking cows receiving normal theilerin showed more than 1 mm increase in skin thickness at the site of inoculation (mean value of 1.86 mm for calves and 3.46 mm for milking cows). The pathological examination of the inoculation site showed infiltration of lymphocytes. No

Oxidative Stress and Inflammatory Responses in Horses Naturally Infected with Theileria equi.

Equine piroplasmosis, caused by the intraerythrocytic protozoan Theileria equi, is a globally distributed tick-borne disease characterized by persistent infection and variable clinical manifestations. Although oxidative stress has been implicated in the pathogenesis of several hemoprotozoan diseases, information regarding redox imbalance and its relationship with inflammatory responses in horses naturally infected with T. equi remains limited. This study aimed to evaluate oxidative stress, antioxidant status, inflammatory cytokines, and clinicopathological alterations in horses naturally infec

First molecular detection of Theileria haneyi infection in horses in Southern Spain.

BACKGROUND: Equine piroplasmosis (EP) is a tick-borne disease of equids caused by the hemoparasites Theileria equi, Babesia caballi, and Theileria haneyi. T. haneyi, a close relative of T. equi that causes mild clinical signs under experimental conditions, has been reported in several countries, adding complexity to EP ecology, diagnosis, and treatment. However, its presence in Spain remains unknown, which is concerning given the country's importance in equine genetics and international trade. Hereby, we investigated the presence of T. haneyi in blood samples from equids in Spain. METHODS: Bl

Epidemiological and molecular study of Theileria spp. in sheep and goats in Erbil, Iraq.

Theileria spp. are a group of hemoprotozoal parasites pose a significant threat to small ruminants in tropical and subtropical regions of the world. Because of that and due to the limited data on the genetic diversity of Theileria species in Erbil Governorate, northern Iraq, this study was conducted to assess the prevalence of Theileria spp. in sheep and goats using both Giemsa-stained blood smears and PCR methods as a diagnostic criterion. The overall infection rate detected by blood smears was 26.2% in sheep and 18.3% in goats, while PCR showed higher infection rates, 47.7% in sheep and 38.3

Haemaphysalis longicornis ticks are unable to transstadially transmit Theileria haneyi to horses.

INTRODUCTION: The recent discovery of Theileria haneyi, a tick-borne hemoparasite that causes mild clinical signs of equine piroplasmosis, has added complexity to the diagnosis of this reportable disease, which is prevalent among equids globally. Knowledge gaps regarding competent tick vectors that can transmit T. haneyi and the recent outbreak of Haemaphysalis longicornis in the US has prompted us to conduct this study. Our objective was to investigate whether H. longicornis can transstadially transmit T. haneyi to horses. MATERIALS AND METHODS: Haemaphysalis longicornis larvae (0.5 g) and n

First molecular evidence of Theileria lestoquardi in small ruminants in northern Iran.

Ovine theileriosis as a critical agent in small ruminant production, can cause lethal infections. Different species of Theileria have been reported in various parts of the world, and each species causes different diseases in the host. This is the first molecular study to investigate the prevalence of ovine theileriosis and identify the dominant Theileria species in northern Iran. A number of 220 small ruminants, including sheep and goats, were randomly sampled from 22 flocks. Peripheral blood smears were stained by the Giemsa staining method. As well as for species identification, all samples

Hematology and biochemical values in equines naturally infected with Theileria equi in Nigeria.

Equine piroplasmosis (EP) is a tick-borne disease of equines with resultant economic consequences. To date, little information is available regarding the effects of EP on the health of equids in Nigeria. Therefore, this study was carried out to ascertain the effects of natural sub-clinical infection with T. equi on the hematology and biochemical profiles of horses and donkeys. Blood and serum samples were subjected to hematology and serum biochemistry analyses, respectively. Data corresponding to PCR-positive (infected) and PCR-negative (uninfected) was computed to ascertain changes in any of

Molecular detection of Theileria species and Babesia caballi from horses in Nigeria.

Equine piroplasmosis (EP) is an infectious, tick-borne disease caused by the hemoprotozoan parasites, Theileria equi, Babesia caballi, and a recently reported new species, T. haneyi. Infections by these apicomplexan parasites limit performance and cause economic losses for the horse industry. Equine piroplasmosis is widespread in the northern regions of Nigeria, where an increasing portion of the animal population is composed of horses. This disease has remained epidemiologically challenging, especially as the movement of horses increases across Nigeria. In this study, blood samples from 300 h

The effects of trans-chalcone and chalcone 4 hydrate on the growth of Babesia and Theileria.

BACKGROUND: Chemotherapy is a principle tool for the control and prevention of piroplasmosis. The search for a new chemotherapy against Babesia and Theileria parasites has become increasingly urgent due to the toxic side effects of and developed resistance to the current drugs. Chalcones have attracted much attention due to their diverse biological activities. With the aim to discover new drugs and drug targets, in vitro and in vivo antibabesial activity of trans-chalcone (TC) and chalcone 4 hydrate (CH) alone and combined with diminazene aceturate (DA), clofazimine (CF) and atovaquone (AQ) we

Genetic characterization of Babesia and Theileria parasites in water buffaloes in Sri Lanka.

Water buffaloes are thought to be the reservoir hosts for several hemoprotozoan parasites that infect cattle. In the present study, we surveyed Sri Lankan bred water buffaloes for infections with Babesia bovis, Babesia bigemina, Theileria annulata, and Theileria orientalis using parasite-specific PCR assays. When 320 blood-derived DNA samples from water buffaloes reared in three different districts (Polonnaruwa, Mannar, and Mullaitivu) of Sri Lanka were PCR screened, B. bovis, B. bigemina, and T. orientalis were detected. While T. orientalis was the predominant parasite (82.5%), low PCR-positi

Detection and differentiation of ovine Theileria and Babesia by reverse line blotting in China.

A reverse line blot (RLB) assay was developed for detection and specific identification of the different ovine Theileria and Babesia parasites. In a polymerase chain reaction (PCR), the hypervariable region 4 (V4 region) of the 18S ribosomal DNA gene was amplified with a set of general primers specific for members of the genera Theileria and Babesia. Meanwhile, specific oligonucleotide probes were designed and bound on membrane. After one single-PCR amplification, the amplified fragment was hybridized against different generic and species-specific probes. It was able to detect four species, i.

Prevalence and diagnosis of Babesia and Theileria infections in horses in Italy: a preliminary study.

Babesia caballi and Theileria equi are the causative agents of equine piroplasmosis. In this preliminary epidemiological study, 412 horses reared in central and northern Italy were sampled and three diagnostic methods compared, namely, the microscopy, the indirect fluorescent antibody test (IFAT) and a PCR. Possible risk factors (such as area, season, breed, activity, sex, age, and grazing) associated with serological positivity were evaluated. A seroprevalence of 68.4% was found: 12.4% of the animals had anti-T. equi antibodies, 17.9% anti-B. caballi antibodies and 38.1% had antibodies agains

Theileria parva: sporozoite entry into bovine lymphocytes is not dependent on the parasite cytoskeleton.

The main conclusion from the present study is that T. parva sporozoite entry is dependent on a functional host cell actin cytoskeleton and is not driven by the parasite. Treating lymphocytes with cytochalasin D resulted in a dose-dependent reduction in the levels of host cell infection. However, the primary effect was to block sporozoite binding and only at the highest concentration (20 microM) was sporozoite internalization significantly reduced. In fact at lower concentrations (1-10 microM) cytochalasin treatment lead to a relative increase in sporozoite internalization. The results are cons

Preparation of antibodies directed to the Babesia ovata- or Theileria sergenti-parasitized erythrocytes.

To investigate the surface antigens of the bovine red blood cells (RBCs) parasitized by Babesia ovata or Theileria sergenti, attempts were made to produce monoclonal antibodies (mAbs) with BALB/c mice. Comparable numbers of hybridomas producing anti-piroplasm mAbs, as well as anti-bovine RBC mAbs, were obtained from the mice immunized with B. ovata- or T. sergenti-PRBCs. However, mAbs directed to the surface of parasitized RBCs (PRBCs) were obtained only from the mice immunized with B. ovata-PRBCs, but not from those immunized with T. sergenti-PRBCs. When serum samples from the immunized mice

Isolation and preliminary characterisation of a previously unidentified Theileria parasite of cattle in Kenya.

A Theileria parasite was isolated from cattle on a ranch in Kenya where it caused mild theileriosis in approximately one third of the cattle exposed to natural tick challenge. The parasite was isolated by inoculation of blood into two experimental cattle. Blood from one of these cattle was used to infect two splenectomised cattle which developed high piroplasm parasitaemias and severe anaemia. A blood stabilate was prepared from one of the splenectomised cattle and produced high parasitaemias in splenectomised cattle. Immunofluorescence tests showed that the unidentified Theileria species was

Isolation of a Theileria species from Eland (Taurotragus oryx) infective for cattle.

Theileria infections were induced in cattle by feeding ticks on them from 3 sources: (a) adult rhipicephalid ticks obtained from the vegetation in a paddock containing an eland EAO at the Animal Orphanage, Nairobi National Park, Kenya, (b) Rhipicephalus appendiculatus adults fed as nymphs on the same eland, (c) R. pulchellus adults fed as nymphs on an eland W 68 captured in the Machakos district of Kenya. Both eland were harbouring Theileria parasites at the time nymphal ticks were fed. Mild infections were produced when adult ticks from these 3 batches were applied to cattle associated with l

Activity of octyl gallate against drug-sensitive and buparvaquone-resistant Theileria annulata.

Theileria annulata, the causative agent of tropical theileriosis, poses a significant threat to cattle industries, a challenge further intensified by increasing drug resistance. In this study, the food-grade phenolic compound octyl gallate (OG) was evaluated for its antitheilerial activity and identified as a potent and selective inhibitor of T. annulata. Comparative screening of gallic acid (GA) derivatives revealed that OG exhibited the strongest activity against T. annulata-transformed cell lines, with IC50 values of 374.0 nM in TaNM cells and 448.7 nM in the buparvaquone-resistant TaXJS ce

mRNA vaccine design using the proteome of Theileria annulata through immunoinformatics approaches.

Theileriosis exerts a substantial impact on ruminants, resulting in significant economic losses within the animal husbandry sector. The current vaccine, a live attenuated parasite, has several limitations that hinder effective disease control. This study utilized immunoinformatics to prioritize potential vaccine candidates and pointed to the design of a novel mRNA vaccine against Theileria annulata using in silico methods. Nine antigenic proteins were selected using various software, and their epitopes were identified through immunoinformatics tools. These epitopes were assessed for their biol
