Molecular epidemiology and genetic characterization of Theileria orientalis in cattle.

Theileria orientalis (T. orientalis) is a benign species of Theileria that is found all throughout the world, including Egypt. The purpose of this work was to determine the current status of T. orientalis infection in cattle, as well as to define T. orientalis genetic diversity using the major piroplasm surface protein (MPSP) genes. In 2020, blood samples from 500 asymptomatic cattle were collected, which live in four different governorates at northern Egypt and examined using PCR assay based on MPSP gene. Overall, 44 blood samples (8.8%, 44/500) were positive for T. orientalis, and the highes

Molecular assessment of Theileria equi and Babesia caballi prevalence in horses and ticks on horses in southeastern France.

Equine piroplasmosis (EP) is a tick-borne disease caused by Babesia caballi and Theileria equi that is potentially emerging in non-endemic countries. We conducted a descriptive study to investigate EP prevalence and spatial distribution in an endemic region: the Camargue and the Plain of La Crau in France. In spring 2015 and 2016, we carried out sampling at stables (total n = 46) with a history of horses presenting chronic fever or weight loss. Overall, we collected blood from 632 horses, which were also inspected for ticks; these horses had been housed in the target stables for at least 1 yea

Hematology and biochemical values in equines naturally infected with Theileria equi in Nigeria.

Equine piroplasmosis (EP) is a tick-borne disease of equines with resultant economic consequences. To date, little information is available regarding the effects of EP on the health of equids in Nigeria. Therefore, this study was carried out to ascertain the effects of natural sub-clinical infection with T. equi on the hematology and biochemical profiles of horses and donkeys. Blood and serum samples were subjected to hematology and serum biochemistry analyses, respectively. Data corresponding to PCR-positive (infected) and PCR-negative (uninfected) was computed to ascertain changes in any of

Vector ecology of equine piroplasmosis.

Equine piroplasmosis is a disease of Equidae, including horses, donkeys, mules, and zebras, caused by either of two protozoan parasites, Theileria equi or Babesia caballi. These parasites are biologically transmitted between hosts via tick vectors, and although they have inherent differences they are categorized together because they cause similar pathology and have similar morphologies, life cycles, and vector relationships. To complete their life cycle, these parasites must undergo a complex series of developmental events, including sexual-stage development in their tick vectors. Consequentl

The epidemiology of tick-borne haemoparasites as determined by the reverse line blot hybridization assay in an intensively studied cohort of calves in western Kenya.

The development of sensitive surveillance technologies using PCR-based detection of microbial DNA, such as the reverse line blot assay, can facilitate the gathering of epidemiological information on tick-borne diseases, which continue to hamper the productivity of livestock in many parts of Africa and elsewhere. We have employed a reverse line blot assay to detect the prevalence of tick-borne parasites in an intensively studied cohort of indigenous calves in western Kenya. The calves were recruited close to birth and monitored for the presence of infectious disease for up to 51 weeks. The fina

Review of equine piroplasmosis.

Equine piroplasmosis is caused by one of 2 erythrocytic parasites Babesia caballi or Theileria equi. Although the genus of the latter remains controversial, the most recent designation, Theileria, is utilized in this review. Shared pathogenesis includes tick-borne transmission and erythrolysis leading to anemia as the primary clinical outcome. Although both parasites are able to persist indefinitely in their equid hosts, thus far, only B. caballi transmits across tick generations. Pathogenesis further diverges after transmission to equids in that B. caballi immediately infects erythrocytes, wh

Infection rates of Theileria sergenti in Haemaphysalis longicornis ticks collected from the field in Japan.

For a period of one year Haemaphysalis longicornis ticks were collected in a pasture from vegetation by dragging flannel cloth and from soil samples at monthly intervals. Nymphal ticks were assessed for Theileria infection. Salivary glands were stained with methylgreen-pyronin and examined for parasite masses. Nymphal H. longicornis infected with Theileria sergenti were found in all samples during 12 months including the winter. After shortening the prefed period on rabbits to 24 hours, the parasite masses could be detected in the salivary glands of nymphs collected in the grazing season, from

Theileria parva: the nature of the immune response and its significance for immunoprophylaxis.

Theileria parva is a tick-borne haemoprotozoan parasite of cattle and buffalo which is responsible for considerable economic losses to cattle farming in Eastern, Central and Southern Africa. Infection with the parasite results in an acute lymphoproliferative disorder with high mortality, but animals which survive infection are solidly immune to homologous challenge. Such immunity can be reproduced by infecting cattle with the parasite and treating them with tetracyclines or theilericidal drugs, but the widespread use of this technique as a method of control is hindered by its dependence on liv

Serodiagnosis of Babesia motasi (Wales), Theileria recondita (Wales) and Cytoecetes phagocytophila infection in sheep.

The indirect fluorescent antibody test (IFAT) was used to diagnose some tick-borne infections of sheep, Babesia motasi (Wales), Theileria recondita (Wales) and Cytoecetes phagocytophila. Antigen was prepared from blood derived from splenectomised sheep except for C phagocytophila which was derived from a normal animal. A field survey was made to assess the prevalence of B motasi and T recondita in North Wales and a comparison made between the titres using the B motasi (Wales) antigen with those previously reported. IFA titres reported in the homologous system were consistently lower than those

Effect of chronic trypanosomiasis on immunization against East Coast fever.

Two experiments were carried out in which uninfected cattle, or cattle chronically infected with Trypanosoma congolense, were immunized by the infection and treatment method against East Coast fever (ECF; Theileria parva infection). Chronic trypanosomiasis did not prevent cattle mounting an effective immunological response to ECF immunization and resisting subsequent lethal challenge. There appeared to be no difference in the level or quality of immunity between uninfected cattle and trypanosome-infected cattle. Thus, T. congolense infection on its own does not appear to provide a constraint t

Tafenoquine succinate inhibits the growth of the equine piroplasmosis hemoparasites Theileria equi and Babesia caballi.

BACKGROUND: Equine piroplasmosis (EP) is a tick-borne disease of equids caused by the intraerythrocytic apicomplexan parasites Theileria equi, Babesia caballi and the recently identified Theileria haneyi. Acute cases can be severe, with anemia, jaundice, abortion or sudden death. Survivors remain lifelong carriers, serving as reservoirs for tick-borne and iatrogenic transmission. No vaccines are currently available, and control strategies rely heavily on accurate diagnostics and chemotherapeutic intervention. Imidocarb dipropionate (ID) is the current standard of care for both acute treatment

In silico comparative proteomics and structural analysis of Theileria species reveals conserved druggable targets for anti-theilerial therapy.

Theileriosis, a tick-borne disease caused by apicomplexan parasites of the genus Theileria, poses a major constraint to livestock productivity worldwide. Existing chemotherapeutics are increasingly limited by host toxicity and emerging resistance, underscoring the need for parasite-specific interventions. In this study, we integrated comparative proteomics, subtractive genomics, and structure-based modeling to systematically identify conserved and novel druggable targets across four clinically important Theileria species (T. parva, T. annulata, T. orientalis Fish Creek, and T. orientalis Goon

Distribution and Abundance of the Tick-Borne Piroplasm Theileria cervi Parasitizing White-Tailed Deer (Odocoileus virginianus) in Massachusetts.

Pathogen surveillance contributes to the prediction of outbreaks and implementation of strategies to minimize disease transmission. White-tailed deer (Odocoileus virginianus) host important disease vectors and often support the proliferation of associated pathogens. Such vectors include ticks: hematophagous acarids that facilitate pathogen transmission between animals through blood feeding. Piroplasmida, an order of blood-borne protozoa including Babesia and Theileria, exploits this feeding to infect definitive hosts. Accordingly, we investigated the prevalence of Theileria cervi infections in

Prevalence of Babesia bigemina and Theileria orientalis in Domestic Cattle (Bos taurus) Across the Three Geo-Climatic Zones of Sri Lanka.

Theileria orientalis and Babesia bigemina are hemoprotozoan parasites that adversely affect domestic cattle (Bos taurus) productivity and health worldwide. Rapid climatic changes in different geo-climatic zones observed over the past few years may have altered the distribution and transmission of tick-borne pathogens and vectors. Though these parasites are reported in Sri Lanka in surveillance records, there is a limited understanding of their current distribution pattern. The present study aimed to determine the prevalence and genetic characterization of B. bigemina and T. orientalis in cattl

In vivo assessment of buparvaquone resistant Theileria annulata populations: genetic structure, transmission dynamics, drug susceptibility and pharmacokinetics.

Tropical theileriosis, caused by the protozoan parasite Theileria annulata and transmitted by several species of ixodid ticks of the genus Hyalomma, is an economically important disease of bovines. Concerningly, studies conducted in recent years have shown an increase in the rate of failure when using the primary drug of treatment, buparvaquone (BPQ), particularly in infection caused by T. annulata populations bearing V135A and P253S mutations on the Cytochrome b (Cyto b) gene of the parasite mitochondrial genome. The aim of this study was to demonstrate the relationship between BPQ-resistance

Genetic characterization and identification of a recent discovered genotype of Theileria orientalis (Piroplasmida: Theileriidae) in water buffaloes (Bubalus bubalis) from the Amazon region, Brazil.

Theileria orientalis is typically considered a benign protozoan parasite of cattle, although certain genotypes can cause significant economic losses in the livestock industry. Despite Brazil harboring the largest buffalo (Bubalus bubalis) herd in the Western Hemisphere, information regarding T. orientalis infections in this species remains scarce. This study aimed to identify and characterize T. orientalis genotypes circulating in water buffaloes from the State of Pará, located in the Amazon region of Brazil. The major piroplasm surface protein (mpsp) gene of T. orientalis was detected by PCR
