The gorgonian coral Leptogorgia virgulata contains thyroxine, or a thyroxine-like substance, referred to here as G-T(4). Thyroxine and insulin inhibit the aggregation capacity of platelets. To study the mechanisms of the antiaggregation action of thyroxine and insulin, platelets obtained from patients with diffuse toxic goiter and from rabbits with experimental hyperinsulinemia were incubated with acetylsalicylic acid and indomethacin. Acetylsalicylic acid or indomethacin were found to suppress the aggregation activity of thyrotoxicosis patients' platelets. We calculated a free triiodothyronine (FT3) index on 124 patients who exhibited symptoms and signs of hyperthyroidism and elevations of the free thyroxine (FT4) index on initial screening. A thyrotropin-releasing hormone (TRH) test served as the final arbiter of thyroid function if the clinical presentation was not characteristic of hyperthyroidism or if the FT3 index was not elevated. Thyroxine (T4) transport in the serum of rats deficient in vitamin A was studied by electrophoresis after incubation of serum samples with radioactive T4. In serum from vitamin A-deficient rats, the amount of radioactivity bound to the prealbumin zone of the electrophoretograms decreased, whereas there was an increase in the amount of radioactivity bound to the albumin zone. The rehabilitative effects of exogenous thyroxine on infant development of undernourished animals were examined. Thyroxine injection accelerated the rate of body growth, age at eye opening, tooth eruption, and several emitted behaviors in undernourished infant animals. The data indirectly support previous findings that undernutrition induces hypothyroidism and that exogenous thyroxine might be an effective rehabilitative agent.

A patient with psychomotor retardation secondary to delayed treatment of cretinism developed abnormal dystonic movements in the absence of other signs of toxicity during levothyroxine replacement therapy at a serum thyroxine level of 16 mug./100 ml. The dystonic movements disappeared when the serum thyroxine level fell. The abnormal movements were considered to be related to high thyroxine levels in this patient with pre-existing central nervous system dysfunction.

Compartmental analysis of the peripheral distribution of labeled thyroxine was applied to various groups of subjects with thyrotoxicosis and hypothyroidism.
