An artificial intelligence model (euploid prediction algorithm) can predict embryo ploidy status based on time-lapse data.

BACKGROUND: For the association between time-lapse technology (TLT) and embryo ploidy status, there has not yet been fully understood. TLT has the characteristics of large amount of data and non-invasiveness. If we want to accurately predict embryo ploidy status from TLT, artificial intelligence (AI) technology is a good choice. However, the current work of AI in this field needs to be strengthened. METHODS: A total of 469 preimplantation genetic testing (PGT) cycles and 1803 blastocysts from April 2018 to November 2019 were included in the study. All embryo images are captured during 5 or 6

In Vivo Study of Dynamics and Stability of Dendritic Spines on Olfactory Bulb Interneurons in Xenopus laevis Tadpoles.

Dendritic spines undergo continuous remodeling during development of the nervous system. Their stability is essential for maintaining a functional neuronal circuit. Spine dynamics and stability of cortical excitatory pyramidal neurons have been explored extensively in mammalian animal models. However, little is known about spiny interneurons in non-mammalian vertebrate models. In the present study, neuronal morphology was visualized by single-cell electroporation. Spiny neurons were surveyed in the Xenopus tadpole brain and observed to be widely distributed in the olfactory bulb and telencepha

Loss of co-chaperone TopJ impacts adhesin P1 presentation and terminal organelle maturation in Mycoplasma pneumoniae.

Mycoplasma pneumoniae is a wall-less human respiratory tract pathogen that colonizes mucosal epithelium via a polar terminal organelle having a central electron-dense core and adhesin-related proteins clustered at a terminal button. A mutant lacking J-domain co-chaperone TopJ is non-cytadherent and non-motile, despite having a core and normal levels of the major cytadherence-associated proteins. J-domain co-chaperones work with DnaK to catalyse polypeptide binding and subsequent protein folding. Here we compared features of the topJ mutant with other cytadherence mutants to elucidate the contr

4D Imaging of Brown Algal Cells.

In vivo imaging of brown algal cells in 3D is extremely challenging because of the presence of pigments, such as fucoxanthin and chlorophyll, that diffract light. Moreover, brown algae live in seawater, a high ionic environment that can change the fluorochrome behavior or cause aggregates. Despite the importance of in vivo monitoring the developmental process of brown algal tissues, 4D imaging (x, y, z, t) on a conventional fluorescence microscope is limited. Here, we propose a detailed protocol using a new orange-emitting fluorochrome, styryl benzoindoleninium sulfonate (SBIS), suitable for l

In vivo visualisation of different modes of action of biological DMARDs inhibiting osteoclastic bone resorption.

OBJECTIVES: Osteoclasts play critical roles in inflammatory bone destruction. Precursor cell migration, cell differentiation, and functional cell activation are all in play. Biological disease-modifying antirheumatic drugs (DMARDs) have been shown to significantly inhibit both bone erosion as well as synovitis, although how such agents reduce osteoclastic bone destructionin vivo has not been fully explained. Here, we used an intravital time-lapse imaging technique to directly visualise mature osteoclasts and their precursors, and explored how different biological DMARDs acted in vivo. METHODS

In vivo corneal microscopy: from image to insight - where are we going?

In vivo confocal microscopy (IVCM) enables noninvasive imaging of the human cornea at cellular resolution, making it an indispensable diagnostic tool in clinical ophthalmology and biomedical research. The combination of the Heidelberg Retina Tomograph (HRT) and the Rostock Cornea Module (RCM) has established itself as the gold standard for corneal cellular imaging. This review summarizes the imaging principles, technological advancements, and clinical applications of IVCM. It highlights recent advancements, including automated large-area mosaicking of the subbasal nerve plexus, three-dimension

Least-detectable and age-related local in vivo bone remodelling assessed by time-lapse HR-pQCT.

We previously developed an image analysis approach for the determination of local sites of bone remodelling using time-lapse in vivo HR-pQCT. The involved image filtering for removing noise was chosen rather aggressively, and also removed some effects of the bone remodelling. In this paper, we quantify these filtering settings using ex vivo reproducibility HR-pQCT images, and determine the least-detectable bone remodelling using in vivo reproducibility HR-pQCT images, as well as testing whether the approach is capable of capturing age-related bone remodelling by use of in vivo long-term HR-pQC

Comparison of euploid blastocyst expansion with subgroups of single chromosome, multiple chromosome, and segmental aneuploids using an AI platform from donor egg embryos.

PURPOSE: This retrospective observational study compares how different classes of blastocyst genotypes from egg donor cycles differentially blastulate and expand using a standard assay. METHODS: Quantitative measurements of expansion utilized a customized neural network that segments all sequential time-lapse images during the first 10 h of expansion. RESULTS: Analyses were performed using two developmental time perspectives using time-lapse imaging. The first was the time to blastocyst formation (tB), which broadly reflects variations in developmental rate. Euploidy peaked at 100-115 h from

Self-organizing actin networks drive sequential endocytic protein recruitment and vesicle release on synthetic lipid bilayers.

Forces generated by actin assembly assist membrane invagination during clathrin-mediated endocytosis (CME). The sequential recruitment of core endocytic proteins and regulatory proteins, and assembly of the actin network, are well documented in live cells and are highly conserved from yeasts to humans. However, understanding of CME protein self-organization, as well as the biochemical and mechanical principles that underlie actin's role in CME, is lacking. Here, we show that supported lipid bilayers coated with purified yeast Wiskott Aldrich Syndrome Protein (WASP), an endocytic actin assembly

Non-invasive molecular assessment of human embryo development and implantation potential.

In vitro fertilization (IVF) is the most common assisted reproductive technology used to treat infertility. Embryo selection for transfer in IVF cycles relies on the morphological evaluation by embryologists, either by conventional microscopic assessment or more recently by time-lapse imaging systems. Despite the introduction of time-lapse imaging improvements in IVF success rates have failed to materialize, therefore alternative approaches are needed. Recent studies have shown that embryos resulting in successful pregnancy differ in their secretome and metabolism compared to embryos that fail

Three-dimensional imaging of human cutaneous forearm bite marks in human volunteers over a 4 day period.

INTRODUCTION: Human bite marks are often sustained during sexual, domestic or child abuse. Currently, analysis of these marks involves digital photography techniques along with an expert forensic odontologist opinion. Photographs often focus closely on the bite mark and give little context to the anatomical location of the injury. Due to variation in camera models and expertise of the photographer, photograph quality can affect its interpretation. Additionally, it can sometimes be days between injury and examination, allowing the injury pattern and colour to alter, making it harder to analyse.

Visualization of Nuclease- and Serum-Mediated Chromatin Degradation with DNA-Histone Mesostructures.

This study analyzed the nuclease- and serum-driven degradation of millimeter-scale, circular DNA-histone mesostructures (DHMs). DHMs are bioengineered chromatin meshes of defined DNA and histone compositions designed as minimal mimetics of physiological extracellular chromatin structures, such as neutrophil extracellular traps (NETs). Taking advantage of the defined circular shape of the DHMs, an automated time-lapse imaging and image analysis method was developed and used to track DHM degradation and shape changes over time. DHMs were degraded well by 10 U/mL concentrations of deoxyribonuclea

Infection of zebrafish embryos with live fluorescent Streptococcus pneumoniae as a real-time pneumococcal meningitis model.

BACKGROUND: Streptococcus pneumoniae is one of the most important causes of bacterial meningitis, an infection where unfavourable outcome is driven by bacterial and host-derived toxins. In this study, we developed and characterized a pneumococcal meningitis model in zebrafish embryos that allows for real-time investigation of early host-microbe interaction. METHODS: Zebrafish embryos were infected in the caudal vein or hindbrain ventricle with green fluorescent wild-type S. pneumoniae D39 or a pneumolysin-deficient mutant. The kdrl:mCherry transgenic zebrafish line was used to visualize the b

Time-lapse RESOLFT nanoscopy empowered by deep learning.

Capturing the dynamic interplay of proteins and organelles within cells requires microscopy methods with high spatial and temporal resolution. Reversible Saturable OpticaL Fluorescent Transition (RESOLFT) demonstrated great potential for live cell nanoscopy by using patterned light and reversibly switching fluorescent probes. Despite the latest improvement in both switching probes and optical schemes, the duration and frame rate of the time-lapse are still limited by photobleaching and spatial sampling steps. Here, we present two distinct strategies based on deep learning to restore low signal

High content 3D imaging by dual-view oblique plane microscopy.

Oblique plane microscopy (OPM) is a form of light-sheet fluorescence microscopy (LSFM) employing a single microscope objective at the sample for both fluorescence excitation and detection. Dual-view OPM (dOPM) is an optically folded form of OPM. We present an improved dOPM system employing a 60×/1.2NA water immersion primary objective and measure the spatial resolution and fluorescence collection efficiency for illumination angles of 35° and 45° with respect to the coverslip. Illumination at 35° provides slightly better lateral resolution and collection efficiency. Collection efficiency measur

The Laminin-Derived Peptide C16 Interacts With β1 Integrin, and Is Internalized by Vesicles Through Endocytic Pathway.

Breast cancer is one of the leading causes of mortality worldwide. The tumor microenvironment plays a critical role in cancer progression. This microenvironment is composed of various cells embedded in the extracellular matrix (ECM). Laminin-111, a major ECM glycoprotein, produces bioactive peptides that influence tumor biology. We have shown that the laminin-derived peptide C16 (KAFDITYVRLKF), located in the short arm of the γ1 chain, regulates migration, invasion, and invadopodia formation in different cancer cells. Our findings suggest that the regulatory mechanisms underlying the effects o

Large-scale time series microscopy of neovessel growth during angiogenesis.

During angiogenesis, growing neovessels must effectively navigate through the tissue space as they elongate and subsequently integrate into a microvascular network. While time series microscopy has provided insight into the cell activities within single growing neovessel sprouts, less is known concerning neovascular dynamics within a large angiogenic tissue bed. Here, we developed a time-lapse imaging technique that allowed visualization and quantification of sprouting neovessels as they form and grow away from adult parent microvessels in three dimensions over cubic millimeters of matrix volu

Time-lapse in vivo dynamics of human corneal immune cells reveals a density-diffusivity relationship.

PURPOSE: We used time-lapse in vivo confocal microscopy (IVCM) combined with random walk modelling to investigate the homeostatic migratory dynamics of naïve dendritiform cells in human cornea. Specifically, we asked whether diffusion coefficient (D), which quantifies the tissue area a cell surveys over time, varies with local cell density and morphological parameters. METHODS: Two-dimensional time-lapse IVCM imaging was obtained from the subbasal nerve plexus at the corneal whorl of 55 healthy young adults. Dendritiform cells without visible dendrites were tracked at approximately 5-min inte
