Elevated Expression of EphA10 in Gastric Cancer Tissues: A Potential Biomarker for Tumor Metastasis and Poor Survival.

Ephrin type-A receptor 10 (EphA10) is a receptor tyrosine kinase involved in tumor cell proliferation and metastasis in certain cancers. The expression profile and function of EphA10 in gastric cancer (GC) have not been well studied. The present study aimed to evaluate EphA10 expression and its clinicopathological significance in gastric cancer. Data from TCGA/GEPIA database were used to assess EphA10 mRNA expression in GC. EphA10 expression in GC tissues was analyzed via IHC to validate these findings. CCK8, flow cytometry, and Transwell migration assays were used to evaluate EphA10 functions

Sex-specific survival but not tissue wasting in the KPP mouse model of pancreatic cancer-induced cachexia.

UNLABELLED: Cancer cachexia, a multifactorial condition resulting in muscle and adipose tissue wasting, reduces the quality of life of many people with cancer. Despite decades of research, therapeutic options for cancer cachexia remain limited. Cachexia is highly prevalent in people with pancreatic ductal adenocarcinoma (PDAC), and many animal models of pancreatic cancer are used to understand mechanisms underlying cachexia. One such model is the Kras LSL-G12D , Ptf1a Cre-ER/+ , Pten flox/flox (KPP) model, which utilizes an inducible Cre recombinase to allow tumor development to be initiated a

Treatment strategies and survival of patients with connective tissue disease and pulmonary arterial hypertension: a COMPERA analysis.

OBJECTIVES: Pulmonary arterial hypertension (PAH) occurs in various connective tissue diseases (CTDs). We sought to assess contemporary treatment patterns and survival of patients with various forms of CTD-PAH. METHODS: We analysed data from COMPERA, a European pulmonary hypertension registry, to describe treatment strategies and survival in patients with newly diagnosed PAH associated with SSc, SLE, MCTD, UCTD and other types of CTD. All-cause mortality was analysed according to the underlying CTD. For patients with SSc-PAH, we also assessed survival according to initial therapy with endothe

Amputations for extremity soft tissue sarcoma in an era of limb salvage treatment: Local control and survival.

BACKGROUND: Despite multimodality limb salvage treatment (LST) for locally advanced extremity soft tissue sarcoma (ESTS), some patients still need an amputation. Indications for amputation and oncological outcome for these patients are described. METHODS: Between 1996 and 2016, all patients who underwent an amputation for ESTS were included. Patients who underwent an amputation as primary or as non-primary treatment formed Group I and II, respectively. RESULTS: Thirty-nine patients were included, 16 in Group I (41%) and 23 in Group II (59%). Tumor size or local recurrence which could not be

[Influence of the donor age on graft survival : Is the demographic change also important for corneal tissue donation?].

The success of corneal transplantation highly depends on the quality of the used graft. Various factors play a crucial role such as a perfect stroma without optically relevant, centrally located cloudiness or changes (e. g. scars) or an adherent Descemet membrane. One of the most important parameters is the quality of the endothelial cell layer with a sufficiently large endothelial cell count. An open question is so far whether the donor age affects corneal quality and therefore has an impact on the success of transplantation. A comprehensive review of the available literature revealed that a

Soft Tissue Dehiscence and Survival of Customized CAD/CAM Titanium Subperiosteal Implants: A Systematic Review and Metaanalysis.

PURPOSE: This systematic review and meta-analysis aimed to assess the prevalence of soft tissue dehiscence and survival rates of modern customized titanium subperiosteal implants (CSIs). A secondary objective was to determine whether the anatomical site (maxilla vs. mandible) is a significant predictor of these outcomes. MATERIALS AND METHODS: An electronic search was conducted across PubMed, Scopus, and Google Scholar for clinical studies published between January 2010 and April 2026 involving 3D-printed (DMLS) or milled titanium CSIs. Primary outcomes focused on soft tissue dehiscence, whil

MOTS-c, a mitochondrial-derived peptide, ameliorates lysosomal membrane permeability and improves survival of soft tissue transplantation.

Distal ischemic necrosis remains a major challenge in reconstructive surgery. Mitochondria and lysosomes interact via signaling and membrane contacts to maintain cellular homeostasis. Mitochondrial-derived peptide MOTS-c, encoded by the MT-RNR1/12S rRNA open reading frame, enhances mitochondrial function by reducing reactive oxygen species (ROS) and stabilizing the membrane potential, potentially preserving lysosomal integrity and reducing lysosomal membrane permeabilization (LMP). This study investigated the protective effects and underlying mechanisms of MOTS-c in ischemic flaps. RNA sequenc

Impact of liver-specific survival motor neuron (SMN) depletion on central nervous system and peripheral tissue pathology.

Spinal muscular atrophy (SMA) is caused by mutations in the Survival Motor Neuron 1 (SMN1) gene. While traditionally viewed as a motor neuron disorder, there is involvement of various peripheral organs in SMA. Notably, fatty liver has been observed in SMA mouse models and SMA patients. Nevertheless, it remains unclear whether intrinsic depletion of SMN protein in the liver contributes to pathology in the peripheral or central nervous systems. To address this, we developed a mouse model with a liver-specific depletion of SMN by utilizing an Alb-Cre transgene together with one Smn2B allele and o

Effects of Microstructured and Anti-Inflammatory-Coated Cochlear Implant Electrodes on Fibrous Tissue Growth and Neuronal Survival.

Cochlear implants are well established devices for treating severe hearing loss. However, due to the trauma caused by the insertion of the electrode and the subsequent formation of connective tissue, their clinical effectiveness varies. The aim of the current study was to achieve a long-term reduction in connective tissue growth and impedance by combining surface patterns on the electrode array with a poly-L-lactide coating containing 20% diclofenac. Three groups of six guinea pigs each (control, structure, structure with diclofenac in the coating) were implanted for four weeks. The hearing th

Initial Treatment Strategies Show No Survival Difference in Early-Stage Salivary Gland Mucosa-Associated Lymphoid Tissue Lymphoma.

PURPOSE: Given the indolent nature of mucosa-associated lymphoid tissue (MALT) lymphoma, immediate intervention is not always necessary, and a consensus on the optimal treatment modality remains elusive. This study aimed to evaluate survival outcomes of different initial treatments for early-stage (stage I-II) salivary gland MALT lymphoma. METHODS: Using data from the Surveillance, Epidemiology, and End Results program, we included patients diagnosed between 2000 to 2021. Initial treatments included surgery alone, radiotherapy alone, surgery combined with radiotherapy, chemotherapy alone, or

The role of endogenous tissue-type plasminogen activator in neuronal survival after ischemic stroke: friend or foe?

Endogenous protease tissue-type plasminogen activator (tPA) has highly efficient fibrinolytic activity and its recombinant variants alteplase and tenecteplase are established as highly effective thrombolytic drugs for ischemic stroke. Endogenous tPA is constituted of five functional domains through which it interacts with a variety of substrates, binding proteins and receptors, thus having enzymatic and cytokine-like effects to act on all cell types of the brain. In the past 2 decades, numerous studies have explored the clinical relevance of endogenous tPA in neurological diseases, especially

A short-term Borrelia burgdorferi infection model identifies tissue tropisms and bloodstream survival conferred by adhesion proteins.

Borrelia burgdorferi, the causative agent of Lyme disease in the United States, is able to persist in the joint, heart, skin, and central nervous system for the lifetime of its mammalian host. Borrelia species achieve dissemination to distal sites in part by entry into and travel within the bloodstream. Much work has been performed in vitro describing the roles of many B. burgdorferi outer surface proteins in adhesion to host cell surface proteins and extracellular matrix components, although the biological relevance of these interactions is only beginning to be explored in vivo. A need exists

Predictors of diagnosis and survival in idiopathic pulmonary fibrosis and connective tissue disease-related usual interstitial pneumonia.

BACKGROUND: Although usual interstitial pneumonia (UIP) appears to portend better survival when associated with connective tissue disease (CTD-UIP), little is known about the presenting clinical, radiologic, and pathologic features that differentiate pathologically confirmed UIP with CTD from idiopathic pulmonary fibrosis (IPF). In patients with atypical radiologic and clinical features, what specific findings predict underlying IPF vs. CTD-UIP diagnosis and their respective long term survival? METHODS: A large retrospective cohort analysis was done of consecutive patients seen from 1995 thro

Spermatogonial survival after cryopreservation and short-term orthotopic immature human cryptorchid testicular tissue grafting to immunodeficient mice.

BACKGROUND: Fertility preservation has become an urgent clinical requisite for prepubertal male cancer patients undergoing gonadotoxic treatment. As these patients do not yet produce spermatozoa for freezing, only immature tissue is available for storage. We studied the survival and proliferative activity of spermatogonia and Sertoli cells after cryopreservation of cryptorchid testicular tissue pieces followed by xenografting for 21 days. METHODS AND RESULTS: Single pieces of tissue from cryptorchid testes (2-9 mm(3)) of young boys (2-12 years) were cryopreserved, thawed and transplanted into

Delayed treatment with recombinant human tissue factor pathway inhibitor improves survival in rabbits with gram-negative peritonitis.

To determine whether treatment with recombinant human tissue factor pathway inhibitor (TFPI), an inhibitor of the extrinsic coagulation pathway, can improve survival in a clinically relevant model of gram-negative sepsis, rabbits were given an intraperitoneal inoculation of a suspension containing hemoglobin (40 microg/mL), porcine mucin (150 microg/mL), and viable Escherichia coli O18:K1 (1.0 +/- 0.5 x 10(5) cfu/kg). Treatment with gentamicin (5 mg/kg every 12 h for five doses) was instituted 4 h after induction of peritonitis. At the same time point, rabbits were randomized to receive a 24-h

Prolongation of renal allograft survival in DLA-tissue typed beagles after third-party leucocyte and erythrocyte transfusion.

In a dog model, both transfusion of leucocytes and of erythrocytes induced lymphocytotoxic antibodies. The results, obtained in DLA identical littermate and DLA nonidentical nonrelated beagle recipient pairs, suggest a correlation between DLA-type and lymphocytotoxic immune response. Two weeks after the last of three transfusions from different blood donors, kidneys from unrelated DLA-mismatched donrs were transplanted to the beagle recipients. A standard postoperative immunosuppressive regimen was given. No correlation between erythrocyte or leucocyte induced immune response was observed, but

Machine Learning-Based Recurrence and Survival Prediction in a Large Institutional Upper Extremity Soft Tissue Sarcoma Cohort.

BACKGROUND AND OBJECTIVES: Upper extremity soft tissue sarcomas (UE STS) present unique management challenges; existing prognostic nomograms were developed from mixed anatomic cohorts. This study develops anatomically specific machine learning (ML) models to predict UE-STS outcomes and benchmarks them against established tools. METHODS: Time-to-event models for local recurrence (LR), distant metastasis (DM), and overall survival (OS) were developed in 188 patients treated at a single sarcoma center, treating death as a competing risk. Five survival algorithms were compared within a nested cro

SLC7A11 in liver tissue homeostasis and disease: Cellular mechanisms linking ferroptosis, redox regulation, and hepatic cell survival.

Liver diseases involve major disturbances in iron metabolism, redox homeostasis, and lipid peroxidation; therefore, ferroptosis is considered as an important link between cellular stress and liver damage/progression. This article describes solute carrier family 7 member 11 (SLC7A11), the functional subunit of the cystine/glutamate antiporter, System Xc-, as a major context-dependent regulator of hepatic ferroptosis. SLC7A11 supports cystine uptake, GSH synthesis, and detoxification of lipid peroxides through glutathione peroxidase 4 (GPX4), which in turn provides protection from ferroptosis ca

Computational Morphological Assessment of Bladder Cancer Tissue Is Prognostic of Recurrence and Overall Survival Following Transurethral Resection.

PURPOSE: Current risk assessment tools for bladder cancer following transurethral resection of the bladder tumor (TURBT) depend on pathological examination of resected tissue, with the consequent intra- and inter-reviewer variability. Improved prognostic tools could enable increased monitoring and aggressive interventions for high-risk patients while reducing the frequency of invasive testing for low-risk patients. METHODS: We present an automated tumor risk assessment method based on quantitative features of nuclear pleomorphism and polarity extracted from digitized hematoxylin and eosin sli
