Systemic lupus erythematosus (SLE) is a complex autoimmune disease with diverse clinical presentations characterized by the presence of autoantibodies to nuclear components. Toll-like receptor (TLR)7, TLR8, and TLR9 sense microbial or endogenous nucleic acids and are implicated in the development of SLE. In mice TLR7-deficiency ameliorates SLE, but TLR8- or TLR9-deficiency exacerbates the disease because of increased TLR7 response. Thus, both TLR8 and TLR9 control TLR7 function, but whether TLR8 and TLR9 act in parallel or in series in the same or different cell types in controlling TLR7-media

BACKGROUND: Toll-like receptor (TLR) ligands have been explored as vaccine adjuvants for tumor and virus immunotherapy, but few TLR ligands affecting schistosoma vaccines have been characterized. Previously, we developed a partially protective DNA vaccine encoding the 26-kDa glutathione S-transferase of Schistosoma japonicum (pVAX1-Sj26GST).
METHODOLOGY/PRINCIPAL FINDINGS: In this study, we evaluated a TLR7/8 ligand (R848) and a TLR9 ligand (CpG oligodeoxynucleotides, or CpG) as adjuvants for pVAX1-Sj26GST and assessed their effects on the immune system and protection against S. japonicum. We

Dengue manifestations range from a mild form, dengue fever (DF), to more severe forms such as dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS). The ability of the host to present one of these clinical forms could be related to polymorphisms located in genes of the Toll-like receptors (TLRs) which activate the pro-inflammatory response. Therefore, the genotyping of single nucleotide genetic polymorphisms (SNPs) in TLR3 (rs3775291 and rs6552950), TLR4 (rs2737190, rs10759932, rs4986790, rs4986791, rs11536865, and rs10983755), TLR7 (rs179008 and rs3853839), and TLR8 (rs3764880, rs574

Highly active antiretroviral therapy (HAART) for HIV-1 infection suppresses but does not cure the disease due to persistent latent viral reservoirs. The "shock and kill" strategy aims to eradicate these reservoirs completely. In exploring the role of TLR8 activation in this context, extensive efforts have been devoted to identify compound 23a, a potent and selective TLR8 agonist with strong agonistic activity (EC50 = 19 nM) and high selectivity (∼1300-fold) over TLR7. Notably, compound 23a exhibits potent reactivation of latent HIV-1 reservoirs in both infected cell lines and primary PBMCs fro

Endosomal toll-like receptors (TLRs) TLR7, TLR8, and TLR9 play an important role in systemic lupus erythematosus (SLE) pathogenesis. The proteolytic processing of these receptors in the endolysosome is required for signaling in response to DNA and single-stranded RNA, respectively. Targeting this proteolytic processing may represent a novel strategy to inhibit TLR-mediated pathogenesis. Human alpha 1 antitrypsin (hAAT) is a protease inhibitor with anti-inflammatory and immunoregulatory properties. However, the effect of hAAT on endosomal TLRs remains elusive. In this study, we first tested the

In colorectal cancer (CRC), systemic inflammation is associated with poor prognosis, but the underlying mechanisms are not fully characterized. Tumor necrosis may contribute to systemic inflammation by inducing interleukin (IL)-6 signaling, and proinflammatory cytokines such as IL-6 and IL-8, and matrix metalloproteinase (MMP)-8 also are linked to adverse CRC outcomes. Because Toll-like receptors (TLRs) are important mediators of inflammatory responses, we investigated the roles of TLR2 and TLR4 in CRC-associated systemic inflammatory responses, especially tumor necrosis. In 118 patients with

Recent subunit vaccines based on purified protein antigens have shown reduced immunogenicity and efficacy, highlighting the need for adjuvants to improve the level and quality of the induced immune responses. Toll-like receptors (TLR) are innate immunity receptors that represent promising targets for the development of agonist-based vaccine adjuvants. Combinations of TLR ligands represent an attractive strategy for developing better adjuvants, but remain underexplored due to technical difficulties related to the co-delivery of molecules with distinct physicochemical properties. In this study,

Carfentanil, an ultrapotent synthetic opioid, poses a severe public health threat because of its rapid brain penetration and potent activation of μ-opioid receptors. Conventional anti-drug vaccines are limited by the poor hapten immunogenicity and dominant anti-carrier immunity. Here, we report a molecularly encoded synthetic carfentanil vaccine (MECVPLA-PEI) integrating a carfentanil hapten, TLR7/8 agonist, and T-helper epitope within a polymeric scaffold to coordinate B and T cell activation. MECVPLA-PEI exhibited markedly enhanced lymph node targeting, with 21.2-fold higher accumulation at

The transcription factor interferon regulatory factor 5 (IRF5) functions as an important mediator of the inflammatory response downstream of MyD88-dependent TLRs. Whereas dysregulation of IRF5 activity has been implicated in the development of numerous autoimmune diseases including systemic lupus erythematosus, the factors that modulate TLR-induced IRF5 post-translational modifications are poorly understood. The focus of this study was to identify novel kinases in TLR-MyD88-IRF5 signaling. We performed a kinome-wide siRNA screen in human THP-1 monocytic cells and identified serine/threonine pr

Common variable immunodeficiency (CVID) is the most common symptomatic primary antibody deficiency and is associated with recurrent infections and chronic inflammatory diseases. We evaluated the ability of Toll-like receptor (TLR) ligands to induce secretion of chemokines, cytokines and type I interferons by peripheral blood mononuclear cells (PBMCs) from CVID patients. High levels of CXCL10, CCL2, CXCL9, CCL5, CXCL8, and IL-6 were detected in sera of CVID patients compared with healthy controls. Increased chemokine levels were observed in unstimulated PBMCs, but after stimulation with TLR2 an

Recognition of RNA fragments by Toll-like receptor 7 (TLR7) and TLR8 helps to initiate the innate immune response against pathogens. An outstanding question is why RNA fragments generated during clearance of apoptotic cells fail to activate TLR7 and TLR8 signaling. Here we show that select 2'-O-methyl (2'-OMe) guanosine RNA fragments, including those derived from host RNAs, function as potent TLR7 and TLR8 antagonists and reduce TLR7 sensing in vivo. Mechanistically, these fragments bind to an antagonistic site on these proteins via their 5'-end 2'-OMe guanosine. These findings indicate that h

Copper plays an important role in tumor growth and metastasis. Copper chelation has been confirmed to be an effective strategy for breast cancer therapy through antiangiogenesis. In this work, a copper chelating coil-comb block copolymer RGD-PEG-b-PGA-g-(TETA-DTC-PHis) (RPTDH) was synthesized and used to prepare nanoparticles for loading resiquimod (R848), a TLR7 and TLR8 agonist, thus to combine antiangiogenesis and immune activation to treat breast cancer. RPTDH has strong copper-chelating ability and could self-assemble to form spherical nanoparticles with significant pH-sensitivity. R848 w

Immunomodulators that can amplify the in situ vaccine effect by regulating radiation-induced immunosuppression have been proven to play a pivotal role in improving the radioimmunotherapeutic efficacy of tumors. Nevertheless, they still face challenges to be addressed for effective tumor radioimmunotherapy, such as poor tumor specificity, rapid clearance from lesions, and potential side effects. Herein, a folate receptors (FRs)-targeting and protein sulfenic acid (PSA)-reactive gold nanosystem FA-dAu-CDR is rationally designed and fabricated in which the toll-like receptor 7/8 immunoagonists ar

Toll-like receptors 7 and 8 (TLR7/8) play pivotal roles in modulating the body's immune response, and their antagonists provide promising treatment options for autoimmune diseases. Here, by modifying the structure of TLR7 agonist SMU-L11, we developed a series of TLR7/8-specific antagonists with therapeutic potential for psoriasis. Experimental validation revealed that structural restriction of the N1 substituent was critical for the pharmacological efficacy of the inverted parent scaffold. Among derivatives, SMU-R39 was the optimal compound, demonstrating potent TLR7/8 inhibitory activity. In

The Toll-like receptor (TLR) family comprises a class of proteins that play a critical role in the innate immune system. Toll-like receptors TLR7 and TLR8 are transmembrane receptors that recognize single-stranded RNA. Aberrant activation of TLR7/8 is associated with the progression of certain autoimmune diseases, such as lupus. Therefore, highly selective inhibitors of TLR7/8 would be an effective therapeutic approach. Here, we have discovered a small molecule inhibitor, KBD4466 (19), which demonstrates excellent cellular activity, favorable pharmacokinetic properties, potent inhibition of cy
