Insulin-like growth factor 1 modulates bioengineered tooth morphogenesis.

Regenerative therapy to replace missing teeth is a critical area of research. Functional bioengineered teeth have been produced by the organ germ method using mouse tooth germ cells. However, these bioengineered teeth are significantly smaller in size and exhibit an abnormal crown shape when compared with natural teeth. The proper sizes and shapes of teeth contribute to their normal function. Therefore, a method is needed to control the morphology of bioengineered teeth. Here, we investigated whether insulin-like growth factor 1 (IGF1) can regulate the sizes and shapes of bioengineered teeth,

Expression of aquaporin isoforms during human and mouse tooth development.

Previously, we described the development of hyaluronan (HA) deposition in human tooth germ tissues that are consistent with water transport in different stages of tooth development. The aquaporins (AQP) constitute a family of membrane water channels that are expressed in many organs. However, there are no data available about the expression pattern of aquaporin water channels in dental structures. In the present study we have characterised the expression of six different aquaporin isoforms (AQP1-5, AQP-9) in developing human and mouse tooth germs by immunohistochemistry using isoform specific

Histogenesis and disappearance of the teeth of the Mekong giant catfish, Pangasianodon gigas (Teleostei).

Juveniles of the Mekong giant catfish, Pangasianodon gigas (Teleostei), have 3 sorts of tooth-upper and lower jaw teeth, palatal teeth, and pharyngeal teeth--but adults are toothless. To investigate the histogenesis and disappearance of the teeth, we made serial sections of the mouth and teeth of juvenile fish at 10 developmental stages (from ca. 8.5 to ca. 30 cm in total length) and examined them under scanning electron microscope and light microscope. Observations of teeth and surrounding tissues in the serial sections revealed the process of tooth resorption by active odontoclast-like cells

Eruption of Bioengineered Teeth: A New Approach Based on a Polycaprolactone Biomembrane.

Obtaining a functional tooth is the ultimate goal of tooth engineering. However, the implantation of bioengineered teeth in the jawbone of adult animals never allows for spontaneous eruption due mainly to ankylosis within the bone crypt. The objective of this study was to develop an innovative approach allowing eruption of implanted bioengineered teeth through the isolation of the germ from the bone crypt using a polycaprolactone membrane (PCL). The germs of the first lower molars were harvested on the 14th day of embryonic development, cultured in vitro, and then implanted in the recipient si

Delayed calcification and congenitally missing teeth. Case report.

A case of delayed differentiation and/or calcification, that is, development, followed by accelerated growth of the maxillary second premolars is presented. This condition was accompanied by the congenital absence of mandibular second premolars even though maxillary and mandibular third molar germs were present. The case was followed over a five-year period. It seems that agenesis of any tooth might be found in conjunction with delay in development of any other tooth. Moreover, full development of a tooth germ (followed by an accelerated growth) as well as pathological changes may occur, despi

A histochemical study of the development of premaxilla and maxilla during secondary palate formation in the mouse embryo.

The development of premaxilla and maxilla in the mouse fetus during secondary palate formation from the 12th to the 16th days of gestation was histochemically assessed. To determine the developmental stages, a classification based on the morphogenesis of the limbs, or the "limb score" (LS) was employed. The stage of LS coincided with the gestational age from the 13th to the 15th days. Early on the 12th day, alkaline phosphatase (ALPase) activity was intense in the mesenchyme lateral to the incisor tooth bud and latero-inferior to the inferior orbital nerve. Subsequently, osteoblasts differenti

Bone and cartilage resorption in relation to tooth development in the anterior part of the mandible in cichlid fish: a light and TEM study.

This paper presents ultrastructural features of the contact region between particular tooth germs and Meckel's cartilage prior to, during, and after initial resorption of the perichondral bone and of the cartilage in the cichlids Hemichromis bimaculatus and Astatotilapia burtoni. Imminent resorption opposite such teeth is announced by the presence, in this region, of a particular cell type, considered to be a stage in the cytodifferentiation of osteoclasts. Slightly later, an osteoclast with typical ruffled border is seen to open a fenestra in the perichondral bone which surrounds Meckel's car

Analysis of dental anomalies in the Siberian mole, Talpa altaica (Insectivora, Talpidae).

We re-examined tooth variation in specimens of the Siberian mole, Talpa altaica, from the collection of the Siberian Zoological Museum and discuss the mechanisms of dental evolution. The number of teeth counted in 1789 specimens ranged from 34 to 47, and supernumerary, absent, and connate teeth were observed. The most frequent tooth anomaly was an absent tooth in the premolar region (200 maxillary first premolars and 190 mandibular third premolars), which does not support Fujita and Kirino's terminal reduction hypothesis in the mandible [Fujita T, Kirino T. Ectopic Activation of Fgf8 in Dental Mesenchyme Causes Incisor Agenesis and Molar Microdontia.

Putatively, tooth agenesis was attributed to the initiation failure of tooth germs, though little is known about the histological and molecular alterations. To address if constitutively active FGF signaling is associated with tooth agenesis, we activated Fgf8 in dental mesenchyme with Osr-cre knock-in allele in mice (Osr2-creKI; Rosa26R-Fgf8) and found incisor agenesis and molar microdontia. The cell survival assay showed tremendous apoptosis in both the Osr2-creKI; Rosa26R-Fgf8 incisor epithelium and mesenchyme, which initiated incisor regression from cap stage. Effects of alendronate on tooth eruption and molar root formation in young growing rats.

Tooth eruption consists of the movement of teeth from the bony crypt in which they initiate their development to the occlusal plane in the oral cavity. Interactions between the tooth germ and its surrounding alveolar bone occur in order to offer spatial conditions for its development and eruption. This involves bone remodeling during which resorption is a key event. Bisphosphonates are a group of drugs that interfere with the resorption of mineralized tissues. With the purpose of investigating the effects of sodium alendronate (a potent bisphosphonate inhibitor of osteoclast activity) on alveo

Interactions of the tooth and bone during development.

The tooth works as a functional unit with its surrounding bony socket, the alveolar bone. The growth of the tooth and alveolar bone is co-ordinated so that a studied distance always separates the 2, known as the tooth-bone interface (TBI). Lack of mineralization, a crucial feature of the TBI, creates the space for the developing tooth to grow and the soft tissues of the periodontium to develop. We have investigated the interactions between the tooth and its surrounding bone during development, focusing on the impact of the developing alveolar bone on the development of the mouse first molar (M

Differential expression of cxcl-14 during eruptive movement of rat molar germs.

Tooth eruption at the early postnatal period is strictly controlled by the molecules secreted mainly from follicular tissues, which recruit monocytes for osteoclast formation. In this study, it was hypothesized that different molecules can be expressed according to the stages of tooth eruption. Rat molar germs together with follicles were extracted and DD-PCR was performed from the root formation stage 2nd molars germs (after eruptive movement) and cap stage 3rd molar germs (before movement) at postnatal day 9. Cxcl-14, a potent chemoattractant, was detected as one of the differentially expres

Three-dimensional volume tomographic study of the imaging accuracy of impacted teeth: MSCT and CBCT comparison--an in vitro study.

The aim of this study was to analyze the imaging accuracy of cone beam computed tomography (CBCT) data sets compared with multislice spiral computed tomography (MSCT) data sets in determining the exact mesiodistal width of unerupted porcine tooth germs and to compare the radiologically obtained results of width measurements with the actual mesiodistal dimension of the tooth germs. In MSCT and CBCT data sets, the largest diameter of 24 tooth germs was determined with the aid of the mesial and distal contact points. The reference method used was mesiodistal width measurement using sliding callip

The Shh signalling pathway in tooth development: defects in Gli2 and Gli3 mutants.

The expression of genes involved in the Sonic Hedgehog signalling pathway, including Shh, Ptc, Smo, Gli1, Gli2 and Gli3, were found to be expressed in temporal and spatial patterns during early murine tooth development, suggestive of a role in early tooth germ initiation and subsequent epithelial-mesenchymal interactions. Of these Ptc, Smo, Gli1, Gli2 and Gli3 were expressed in epithelium and mesenchyme whereas Shh was only detected in epithelium. This suggests that Shh is involved in both lateral (epithelial-mesenchymal) and planar (epithelial-epithelial) signalling in early tooth development

In vivo and in vitro study of the effects of chlorpromazine on tooth mineralization in rats and mice.

The effects of chlorpromazine (CPZ) on tooth mineralization were examined using incisor dentine in adult rats and cultured tooth germs of mandibular first molars dissected from mouse embryos. CPZ (10, 50 and 250 mg/kg, s.c.) substantially inhibited dentine mineralization as evaluated by contact microradiographs. Plasma calcium and phosphorus concentrations were not decreased by CPZ (10 and 50 mg/kg). Physicochemical effects were not involved in the action of CPZ on the mineralization. In vitro experiments showed that CPZ (1 and 10 microM) inhibited mineralization and alkaline phosphatase (ALP)

Gestational protein restriction alters the RANKL/OPG system in the dental germ of offsprings.

UNLABELLED: Evaluate the dentinogenesis in the offspring of rats submitted to gestational protein restriction (GPR). DESIGN: The offspring were evaluated at the 21st day of gestation (21 dG). Assessments were made of morphological parameters and the RANKL/OPG system - bone tissue maturation markers - in the upper incisor tooth germ. Pregnant 10-week-old female Wistar rats were divided into normal protein (NP, 17% casein, n = 5) and low protein (LP, 6% casein, n = 5) diet groups. At 21 dG, the offspring maxillae were collected for histomorphometric and immunohistochemical analyses. RESULTS: T

Histology of tooth attachment tissues and plicidentine in Varanus (Reptilia: Squamata), and a discussion of the evolution of amniote tooth attachment.

Few recent studies have examined the histological basis for tooth attachment in squamates. In the past few years, a surge of interest in this topic has led to the intriguing suggestion that the major tissues derived from the tooth germ (enamel, dentine, cementum and alveolar bone), are conservative and are present in all amniotes. In this study, we describe the histology and development of the tooth attachment complex in Varanus rudicollis, the rough-neck monitor. We provide the first published evidence for the role of cementum and alveolar bone in tooth attachment in varanoid lizards. In Vara
