The bacterial protein RelE severely restricts prokaryotic cell growth, probably by acting as a global inhibitor of translation. It is ubiquitous in prokaryotes as part of the RelE-RelB toxin-antitoxin system, and may be activated by nutritional stress. When the relE gene from Escherichia coli was expressed inducibly in a human osteosarcoma cell line, it was shown to retard growth and to lead to cell death by apoptosis. RelE is therefore unusual among bacterial toxins in possessing broad activity against both prokaryotes and eukaryotes, perhaps by acting on evolutionarily conserved components o

Here, we have used a comprehensive, system-wide approach to delineate the function of the virulence-associated proteins B and C 36 (VapBC36) toxin-antitoxin (TA) system in Mycobacterium tuberculosis pathophysiology. We show that VapC36 interacts with cognate and noncognate antitoxins. We demonstrate that serX is degraded in vitro by VapC36. We show that VapC36 binds copper, which enhances its ribonuclease activity. Furthermore, we report that deletion of vapBC36 or vapC36 enhances the susceptibility of M. tuberculosis upon exposure to copper in vitro. Compared to the wild-type strain, ΔBC36 wa

Streptococcus suis serotype 2 is a major zoonotic pathogen causing meningitis, septicemia, streptococcal toxic shock-like syndrome (STSLS) in swine and humans. This review synthesizes discoveries over the past decade, organized around the infection progression, colonization, bloodstream survival, immune evasion, blood-brain barrier disruption, excessive inflammation culminating in STSLS. We dissect regulatory architecture including two-component systems, Ser/Thr kinase STK, small RNAs, toxin-antitoxin modules, and stringent response. We analyze functional effectors enabling niche adaptation, i

The Mycobacterium tuberculosis genome harbors an unusually high number of toxin-antitoxin (TA) systems. These TA systems have been implicated in establishing the nonreplicating persistent state of this pathogen during latent tuberculosis infection. More than half of the M. tuberculosis TA systems belong to the VapBC (virulence associated protein) family. In this work, we first identified the RNA targets for the M. tuberculosis VapC-mt11 (VapC11, Rv1561) toxin in vitro to learn more about the general function of this family of toxins. Recombinant VapC-mt11 cleaved 15 of the 45 M. tuberculosis t

Tuberculosis is a worldwide plague caused by the pathogen Mycobacterium tuberculosis (M. tb). Toxin-antitoxin (TA) systems are genetic elements abundantly present in prokaryotic organisms and regulate important cellular processes. MazEF is a TA system implicated in the formation of "persisters cells" of M. tb, which contain more than 10 such members. However, the exact function and inhibition mode of each MazF are not fully understood. Here we report crystal structures of MazF-mt3 in its apo form and in complex with the C-terminal half of MazE-mt3. Structural analysis suggested that two long b

AIMS: Klebsiella michiganensis is a medically important bacterium that has been subject to relatively little attention in the literature. Interrogation of sequence data from K. michiganensis strains in our collection has revealed the presence of multiple large plasmids encoding type II toxin-antitoxin (TA) systems. Such TA systems are responsible for mediating a range of phenotypes, including plasmid stability ('addiction') and antibiotic persistence. In this work, we characterize the hipBA TA locus found within the Klebsiella oxytoca species complex (KoSC).
METHODS AND RESULTS: The HipBA TA

Retrons are bacterial immune systems that prevent phage propagation by initiating a toxic response within infected hosts. Previously characterized retrons produce abundant multicopy single-stranded DNA (msDNA) by reverse transcription, which acts as an antitoxin in the absence of phage. However, we describe here a non-canonical mechanism for type VI retrons, which do not produce detectable msDNA in the absence of phage yet still provide defense. Focusing on Retron-Vpa2 from V. parahaemolyticus, we show broad defense against phages and identify triggers of the system when expressed in E. coli.

Burkholderia pseudomallei is the causative agent of melioidosis, a disease with a mortality rate of up to 40% even with treatment. Despite the ability of certain antibiotics to control initial infection, relapse occurs in treated patients. The inability of antibiotics to clear this bacterial infection is in part due to persistence, an evasion mechanism against antibiotics and the effect of host defenses. Evaluation of antibiotic efficacy against B. pseudomallei revealed that up to 48% of in vitro grown populations can survive in a persister state. Toxin-antitoxin (TA) systems have been previou

Tetanus is a vaccine-preventable disease that still commonly occurs in many low-income and middle-income countries, although it is rare in high-income countries. The disease is caused by the toxin of the bacterium Clostridium tetani and is characterised by muscle spasms and autonomic nervous system dysfunction. Global vaccination initiatives have had considerable success but they continue to face many challenges. Treatment for tetanus aims to control spasms and reduce cardiovascular instability, and consists of wound debridement, antitoxin, antibiotics, and supportive care. Recent research has

Many human Gram-negative bacterial pathogens express a Type Three Secretion Apparatus (T3SA), including among the most notorious Shigella spp., Salmonella enterica, Yersinia enterocolitica and enteropathogenic Escherichia coli (EPEC). These bacteria express on their surface multiple copies of the T3SA that mediate the delivery into host cells of specific protein substrates critical to pathogenesis. Shigella spp. are Gram-negative bacterial pathogens responsible for human bacillary dysentery. The effector function of several Shigella T3SA substrates has largely been studied but their potential

Persisters represent a transient, antibiotic-tolerant subpopulation within isogenic bacterial populations, contributing to infection relapses. However, the mechanisms driving persister formation and resuscitation remain elusive. Here, we developed nano-flow cytometry (nFCM)-based methods for single-cell quantification of toxin (T) RelE and antitoxin (A) RelB levels, as well as for monitoring persister states through cell wall growth. We demonstrate that bacteria elevate the T/A ratio through two distinct TA expression modalities to withstand bacteriostatic antibiotic challenge, with T/A = 1.0

Bacteria and phages are engaged in a persistent evolutionary struggle. To survive constant phage predation, bacteria have evolved a highly diverse and multi-layered immune arsenal that determines the success of therapeutic phage infection. Bacterial defenses include receptor blockade, DNA restriction systems, CRISPR-Cas adaptive immunity, and secondary messenger signaling systems that induce effector-mediated cell death. Recent mechanistic advances have elucidated systems such as Thoeris (gcADPR-activated SIR2 effectors depleting NAD⁺), CBASS (cyclic nucleotide-activated effectors disrupting c

Mycobacterium tuberculosis (Mtb) survives host-imposed stress through dynamic metabolic adaptation and growth modulation. Toxin-antitoxin (TA) systems and cell cycle regulators, including RelBE1 and MadR1, are increasingly recognized as contributors to these processes, yet their physiological roles remain unclear. In this perspective, we propose that these systems function as genomically encoded metabolic control modules that couple carbon source availability to persistence. The relBE1 loci, encoded adjacent to the α-ketoglutarate decarboxylase gene (kgd), and the cell division regulator, madR

BACKGROUND: Clostridium botulinum (C. botulinum) is known for producing one of the strongest neurotoxins, namely botulinum neurotoxin (BoNT). This toxin is responsible for causing botulism, which is considered a severe neurological disease.
OBJECTIVES: This comprehensive narrative review highlights the dual nature of C. botulinum as both a serious health threat and a groundbreaking therapeutic agent. Methods: A comprehensive literature search was conducted in Google Scholar, PubMed/Medline, and Scopus (1971-2025, 2020-2025 literary focus) using keywords related to botulism pathogenesis, clini

Subunit intranasal vaccines offer the prospect of inducing combined systemic-mucosal immunity against mucosally transmitted infections such as human immunodeficiency virus. However, although human studies have demonstrated the induction of active immunity, secretory immunoglobulin A (sIgA) responses are variable, and no study has demonstrated protection by accepted vaccine-licensing criteria as measured by direct toxin-neutralizing activity. Using the genetically inactivated mutant diphtheria toxoid CRM(197) in a bioadhesive polycationic polysaccharide chitosan delivery system, we found that a

Staphylococcus aureus is one of the leading agents of nosocomial and community-acquired infections. In this study, we explored the genomic characterization of eight methicillin-resistant clinical isolates of S. aureus from Dhaka, Bangladesh. Notably, all strains were resistant to penicillin, cephalosporins, and monobactams, with partial susceptibility to meropenem and complete susceptibility to amikacin, vancomycin, and tigecycline antibiotics. The strains were found to have an average genome size of 2.73 Mbp and an average of 32.64% GC content. Multi-locus sequence typing analysis characteriz
