Hypoxia response in glioblastoma cells: effect of trehalose on macropinocytosis, autophagy and cell survival.

In glioblastoma multiforme, the most malignant brain tumor in adults, the hypoxic milieu is believed to contribute to tumor aggressiveness and resistance to therapy. Here, human glioblastoma U373-MG and T98G cells were exposed to hypoxia (1 % oxygen) or normoxia (18 % oxygen), and treated with trehalose, a natural disaccharide increasingly studied for its therapeutic potential in cancer. In all samples under hypoxia, HIF-1α stabilization was accompanied by a decrease in Nrf2 and p62/SQSTM1 proteins; redox imbalance also occurred, as documented by increased levels of ROS and parallel lowering o

Isolation and antioxidant evaluation of trehalose from Phylloporia tamilnadensis: a structural and functional study.

Fungal secondary metabolites exhibit diverse bioactivities with significant therapeutic relevance. In this study, we report the first-ever isolation and antioxidant evaluation of trehalose, a disaccharide purified from Phylloporia tamilnadensis. The compound was isolated through silica gel column chromatography and subsequently crystallised via slow methanol evaporation. Structural elucidation was performed using UV-Vis, FTIR, NMR, and single-crystal X-ray diffraction (SCXRD) analyses. SCXRD confirmed the compound as α-D-glucopyranosyl-α-D-glucopyranoside (trehalose), crystallising in the orth

Functional analysis of the CRP and TreR-treBC regulon in trehalose utilization of Vibrio parahaemolyticus.

Vibrio parahaemolyticus is an important foodborne pathogen recognized for its adaptability to various environmental conditions. Understanding its metabolic pathways is crucial for elucidating its survival and pathogenicity. This study investigates the metabolism of trehalose as a carbon source in V. parahaemolyticus and explores its effects on bacterial growth and virulence. We found that V. parahaemolyticus can utilize trehalose as a sole carbon source for growth and identified the presence of treBC operon, which is crucial for trehalose degradation. Notably, the ΔtreB and ΔtreC strains exhib

[Effects of freeze-drying bovine pericardium using a combination of polyethylene glycol and trehalose].

The freeze-drying is a technology that preserves biological samples in a dry state, which is beneficial for storage, transportation, and cost saving. In this study, the bovine pericardium was treated with a freeze-drying protectant composed of polyethylene glycol (PEG) and trehalose (Tre), and then freeze-dried. The results demonstrated that the mechanical properties of the pericardium treated with PEG + 10% w/v Tre were superior to those of the pericardium fixed with glutaraldehyde (GA). The wet state water content of the rehydrated pericardium, determined using the Karl Fischer method, was (

Shedding Light on the Trehalose-Enabled Mucopermeation of Nanoparticles with Label-Free Raman Spectroscopy.

Nanoparticle-based drug delivery systems have attracted significant interest owing to their promise as tunable platforms that offer improved intracellular release of cargo therapeutics. However, significant challenges remain in maintaining the physiological stability of the mucosal matrix due to the nanoparticle-induced reduction in the matrix diffusivity and promotion of mucin aggregation. Such aggregation also adversely impacts the permeability of the nanoparticles, and thus, diminishes the efficacy of nanoparticle-based formulations. Here, an entirely complementary approach is proposed to t

Effect of Trehalose Supplementation on Autophagy and Cystogenesis in a Mouse Model of Polycystic Kidney Disease.

Autophagy impairment has been demonstrated in the pathogenesis of autosomal dominant polycystic kidney disease (ADPKD) and could be a new target of treatment. Trehalose is a natural, nonreducing disaccharide that has been shown to enhance autophagy. Therefore, we investigated whether trehalose treatment reduces renal cyst formation in a Pkd1-hypomorphic mouse model. Pkd1 miRNA transgenic (Pkd1 miR Tg) mice and wild-type littermates were given drinking water supplemented with 2% trehalose from postnatal day 35 to postnatal day 91. The control groups received pure water or 2% sucrose for the con

Use of the 2,3-diacyl-trehalose and the purified protein derivative in the serodiagnosis of tuberculosis in AIDS.

The effect of the human immunodeficiency virus (HIV) infection on IgG production against purified protein derivative (PPD) and 2,3-diacil-trehalose (SL-IV) was investigated by an enzyme-linked immunosorbent assay (ELISA) test. Comparison between the antigens showed that immunocompetent patients produce preferentially antibodies to SL-IV than to PPD (73.3% versus 63.3%). Combination of these results showed an increase of the sensitivity to 80%, which decreased over the spectrum of immunodepression caused by HIV. In the tuberculous HIV seropositive group the sensitivities of SL-IV and PPD were 3

Disruption of paramyosin compromises muscle integrity and alters systemic trehalose metabolism in Helicoverpa armigera.

Trehalose is a principal circulating sugar in insects, maintaining a critical balance between energy supply and storage through systemic insulin signalling. Among different tissues, insect muscle functions as a metabolic hub, coordinating trehalose utilization and glucose uptake to sustain energy homeostasis. In Helicoverpa armigera, we examined whether paramyosin (key muscle gene) silencing influence carbohydrate metabolism. HaPrm silencing led to muscle defects and delayed pupation, indicating a crucial role of HaPrm in development. Transcriptomics revealed dysregulation of genes involved in

Adaptive Laboratory Evolution of Baker's Yeast for Improved Trehalose Accumulation and Freeze-Thaw Tolerance.

Cas9-based genome engineering is a powerful tool for yeast strain development, but its use in the food industry is limited due to GMO concerns. As a non-GMO alternative, adaptive laboratory evolution (ALE) was applied to enhance trehalose accumulation, a stress protectant, in Saccharomyces cerevisiae. To avoid ethanol-centric metabolism and promote storage carbohydrate production, ALE was conducted under nitrogen limitation with ethanol as the sole carbon source. The evolved strain 65EV showed a 2.3-fold increase in trehalose (11.51% vs 5.25%), resulting in enhanced cell viability (77.7% vs 5.

Colloidal protein stability and surface coverage by trehalose: bridging molecular dynamics and experimental work.

The aim of this study was to assess to what extent a simulated trehalose monolayer can predict the minimum molar trehalose to protein ratios required to preserve the colloidal stability of lyophilized lactate dehydrogenase, catalase, and β-galactosidase. We employed an iterative molecular dynamics (MD) approach to quantify the number of trehalose molecules required to saturate the protein's accessible surface area. These in-silico values were compared to experimentally determined colloidal stability thresholds (tetramer recovery) in lyophilized formulations, defined as the minimum molar ratio

Stabilizing lysosome to facilitate verteporfin-based anticancer photodynamic therapy via trehalose co-assembled nanogel.

Trehalose (Tr) is a naturally occurring disaccharide extensively used as a stabilizing and cryoprotective agent for protein and cell preservation. Owing to its unique bioactivities, Tr-based biomaterials have recently emerged as a promising class of functional materials for biomedical applications. In this work, we reveal that Tr can protect lysosomes from the damage caused by verteporfin (VP), a clinically used photosensitizer. Inspired by this finding, a Tr nanogel (NG) ionically crosslinked with ferric ion was designed for the enhanced photodynamic therapy (PDT) of VP using a murine 4T1 bre

H2S-dependent trehalose-induced salt tolerance in tomato involves phenylpropanoid and cyanoamino acid pathways.

Soil salinity limits tomato growth. Trehalose (Tre) and hydrogen sulfide (H₂S) individually enhance salt tolerance, but their combined effect is unknown. Here, we investigated seedling physiological and transcriptomic responses to exogenous Tre and the H2S donor sodium hydrosulfide (NaHS), alone or in combination, under salt stress. Our results demonstrated that both Tre and NaHS significantly alleviated salt-induced growth inhibition, with the combined treatment showing the most pronounced effects. Pharmacological experiments using the Tre inhibitor validamycin A (VA) and the H2S scavenger hy

Inhibitors of trehalose-6-phosphate synthase activity in fungal pathogens compromise thermal tolerance pathways.

UNLABELLED: Infections caused by fungal pathogens such as Candida and Cryptococcus are associated with high mortality rates, partly due to limitations in the current antifungal arsenal. This highlights the need for antifungal drug targets with novel mechanisms of action. The trehalose biosynthesis pathway is a promising antifungal drug target because trehalose biosynthesis is essential for virulence in Cryptococcus neoformans and Candida albicans and is also a mediator of fungal stress responses, such as thermotolerance. To exploit its untapped antifungal potentials, we screened the St. Jude 3

Trehalose, a natural disaccharide, reduces stroke occurrence in the stroke-prone spontaneously hypertensive rat.

Cerebrovascular disease, a frequent complication of hypertension, is a major public health issue for which novel therapeutic and preventive approaches are needed. Autophagy activation is emerging as a potential therapeutic and preventive strategy toward stroke. Among usual activators of autophagy, the natural disaccharide trehalose (TRE) has been reported to be beneficial in preclinical models of neurodegenerative diseases, atherosclerosis and myocardial infarction. In this study, we tested for the first time the effects of TRE in the stroke-prone spontaneously hypertensive rat (SHRSP) fed wit

Trehalose reduces retinal degeneration, neuroinflammation and storage burden caused by a lysosomal hydrolase deficiency.

UNLABELLED: The accumulation of undegraded molecular material leads to progressive neurodegeneration in a number of lysosomal storage disorders (LSDs) that are caused by functional deficiencies of lysosomal hydrolases. To determine whether inducing macroautophagy/autophagy via small-molecule therapy would be effective for neuropathic LSDs due to enzyme deficiency, we treated a mouse model of mucopolysaccharidosis IIIB (MPS IIIB), a storage disorder caused by deficiency of the enzyme NAGLU (alpha-N-acetylglucosaminidase [Sanfilippo disease IIIB]), with the autophagy-inducing compound trehalose.

Strategies to reduce osmotic stress during cryopreservation of red blood cells when using trehalose.

The aim of this study was to determine optimal conditions for cryopreserving red blood cells (RBCs) using trehalose. We assessed the extent of trehalose uptake by RBCs during incubation at 37 °C and following freezing-and-thawing. Additionally, we examined whether betaine could alleviate osmotic stress at various stages of the cryopreservation process, including trehalose loading, freezing, and return to isotonic conditions after thawing. Trehalose uptake during incubation at 37 °C was limited, whereas freezing-and-thawing RBCs in trehalose-containing solutions led to substantially higher intr

Discovery of 2-Cinnamoylbenzimidazoles as Novel Insect and Fungal Trehalose-6-Phosphate Phosphatase Inhibitors.

Trehalose-6-phosphate phosphatase (TPP), a key enzyme in trehalose biosynthesis, represents a promising target for pest and fungal control due to the absence of TPP in higher animals. However, few TPP inhibitors have been reported, and broad-spectrum agents targeting both insects and fungi remain scarce. In this study, 2-cinnamoylbenzimidazole was identified as a dual-targeting inhibitor scaffold against insect and fungal TPPs. Structure-guided optimization by pocket size yielded 5-1 (Ki = 1.3 μM) for insect TPP and 5-3 (Ki = 17.3 μM) for fungal TPP. Molecular docking revealed that 2-cinnamoyl
