Striking mycotoxin tolerance and zearalenone elimination capacity of the decaying wood associated yeast Sugiyamaella novakii (Trichomonascaceae).

BACKGROUND: Mycotoxin-producing fungal species and their mycotoxins pose a global threat for crop production and for human and animal health. Given the increasing demand for healthier food and feed, alternative non-pesticide approaches for reducing fungal infections in crops and eliminating mycotoxin contamination in feedstock are becoming more prevalent. For such purposes, yeast species can be good candidates. Thus, the present study examined the mycotoxin tolerance and mycotoxin elimination ability of several yeast strains belonging to the Trichomonascaceae family. RESULTS: While none of th

Deoxynivalenol exposure-related male reproductive toxicity in mammals: Molecular mechanisms, detoxification and future directions.

An increasing body of evidence indicates that exposure to widespread, environmental and food contaminants such as mycotoxins may cause endocrine disorders and infertility. Deoxynivalenol (DON), which is a toxic secondary metabolite produced by Fusarium fungi, can lead to multiple harmful effects in humans and animals, such as hepatotoxicity, nephrotoxicity, immunotoxicity, gastrointestinal toxicity, neurotoxicity, genetic toxicity and carcinogenicity. Recently, there has been growing concern about DON-induced male infertility. Exposure to DON and its metabolites can damage the structure and fu

Quercetin Ameliorates Deoxynivalenol-Induced Intestinal Injury and Barrier Dysfunction Associated with Inhibiting Necroptosis Signaling Pathway in Weaned Pigs.

Quercetin (Que) is a flavonol compound found in plants, which has a variety of biological activities. Necroptosis, a special form of programmed cell death, plays a vital role in the development of many gastrointestinal diseases. This study aimed to explore whether Que could attenuate the intestinal injury and barrier dysfunction of piglets after deoxynivalenol (DON) exposure through modulating the necroptosis signaling pathway. Firstly, twenty-four weaned piglets were used in a 2 × 2 factorial design and the main factors, including Que (basal diet or diet supplemented with 100 mg/kg Que) and D

A human cell-free translation screen identifies the NT-2 mycotoxin as a ribosomal inhibitor that binds the peptidyl transferase center.

Translation inhibitors are invaluable for probing ribosome function and therapeutic applications, but systematic discovery in human systems is limited by the lack of scalable, screening-compatible cell-free platforms. Here, we establish a robust high-throughput screening using human lysates that bypasses cellular cytotoxic effects. After screening ~28,000 small molecules, we identified known and a novel translation inhibitor, including NT-2, a trichothecene mycotoxin produced by the pathogenic Fusarium sporotrichioides. NT-2 suppressed protein synthesis in human cells and yeast lysates, while

The Role of AFB1, OTA, TCNs, and Patulin in Forensic Sciences: Applications in Autopsy, Criminal Investigations, and Public Health Prevention.

Mycotoxins, specifically aflatoxin B1 (AFB1), ochratoxin A (OTA), trichothecenes (TCNs), and patulin, are a group of secondary metabolites that can contaminate food, leading to severe health implications for humans. Their detection and analysis within forensic toxicology are crucial, particularly as they can be implicated in cases of poisoning, foodborne illnesses, or lethal chronic exposure. However, little is known about the application that mycotoxins could have in forensic investigations and especially about the possibility of extracting and quantifying these molecules on tissues or post-m

Nitric oxide (NO)-mediated mitochondrial damage plays a critical role in T-2 toxin-induced apoptosis and growth hormone deficiency in rat anterior pituitary GH3 cells.

T-2 toxin, a major compound of trichothecenes, induces cell apoptosis and growth hormone (GH) deficiency and causes considerable growth retardation in animals and human cells. However, the mechanism underlying its growth suppression still remains unclear. Recent studies have suggested that ROS induced cell apoptosis and animal feed intake reduction, but there are limited reports on the role of RNS in T-2 toxin-mediated mitochondrial damage, cell apoptosis and growth retardation. Herein, T-2 toxin-induced GH3 cell damage and apoptosis were tested by MTT assay, LDH leakage and flow cytometry, re

Strategies to prevent mycotoxin contamination of food and animal feed: a review.

Mycotoxins are fungal secondary metabolites that have been associated with severe toxic effects to vertebrates produced by many important phytopathogenic and food spoilage fungi including Aspergillus, Penicillium, Fusarium, and Alternaria species. The contamination of foods and animal feeds with mycotoxins is a worldwide problem. We reviewed various control strategies to prevent the growth of mycotoxigenic fungi as well as to inhibit mycotoxin biosynthesis including pre-harvest (resistance varieties, field management and the use of biological and chemical agents), harvest management, and post-

Short-term dietary deoxynivalenol exposure negatively affects performance, intestinal and reproductive functions in laying hens.

The detrimental effects of deoxynivalenol (DON) on broiler chickens are well documented; however, studies on laying hens fed diets naturally contaminated with this mycotoxin remain limited. To evaluate the impact of short-term DON exposure, 600 56-week-old Dekalb White laying hens were fed either a marginally contaminated control diet (CON) or a diet containing 2.45 mg/kg DON for 16 weeks. Production performance was impaired by DON as early as 8 weeks of exposure. Although no changes were observed in the physical characteristics of the eggshells, mRNA expression analyses revealed disruptions i

Concentration and prevalence of mycotoxins in poultry feed and poultry products: a global systematic review and meta-analysis.

Mycotoxins are secondary metabolites produced by fungi that commonly contaminate animal feed, especially poultry feed, impacting poultry productivity and causing adverse effects on animal health. Ochratoxin A, deoxynivalenol, fumonisins, aflatoxins, zearalenone, and trichothecene are of particular concern due to their high toxicity, prevalence, and potential to accumulate as residues in animal-derived foods, such as offal, muscle tissues, and eggs. This study aims to conduct a systematic review and meta-analysis of the occurrence, prevalence, and co-occurrence of mycotoxins in poultry feed and

Antifungal activity and biochemical characteristics of natamycin against Fusarium graminearum.

Fusarium head blight (FHB) caused by Fusarium graminearum is a destructive fungal disease of wheat. Natamycin is a natural polyene macrolide antifungal compound exhibiting broad-spectrum antifungal activity, but its efficacy and mechanism against F. graminearum remain unclear. In this study, the sensitivity of 47 F. graminearum strains from different regions of China to natamycin was determined. The results showed that natamycin had a good inhibitory effect with the EC50 value ranging from 0.93 to 4.04 μg/mL and a mean EC50 value of 2.20 ± 0.59 μg/mL. No resistance association was found betwee

Corneal transcriptomic signatures of mycotoxin exposure reveal distinct immune and metabolic responses.

Fungal keratitis (FK) is a vision-threatening infection in which Fusarium species not only invade the cornea but may also release mycotoxins that exacerbate tissue injury. Here, we established a corneal exposure model to delineate the direct effects of Fusarium-derived mycotoxins independent of live fungal infection, using deoxynivalenol (DON), fumonisin B1 (FB1), and T-2 toxin as representative compounds. Hstological analyses revealed that all three mycotoxins markedly impaired corneal epithelial repair, induced stromal disorganization, and promoted inflammatory cell infiltration. At the mole

Deoxynivalenol and Fumonisin B1 in Gilthead Seabream Diets: Impact on Growth Performance, Hematology, Immunology, and Histopathology.

As aquafeed formulations include more plant-based materials, mycotoxin contamination is becoming relevant for fish farming. Deoxynivalenol (DON) and fumonisin B1 (FB1) frequently occur in fish feeds and may impair fish performance and health. This study evaluated the impact of DON and FB1 on growth and health parameters in gilthead seabream (Sparus aurata). Fish were fed ad libitum for seven weeks with six diets assessed in triplicate: DON A (300 ppb), DON B (2000 ppb), DON C (5000 ppb), FB1 A (5 ppb), FB1 B (10 ppb), FB1 C (40 ppb). The highest contamination levels significantly reduced feed

Low dose dietary contamination with deoxynivalenol mycotoxin exacerbates enteritis and colorectal cancer in mice.

BACKGROUND: The mycotoxin deoxynivalenol (DON) is a frequent contaminant of grain and cereal products worldwide. Exposure to DON can cause gastrointestinal inflammation, disturb gut barrier function, and induce gut dysbiosis in vivo under basal conditions, but little is known about the effects of DON ingestion in individuals with pre-existing gastrointestinal disease. OBJECTIVES: Mice were orally exposed to 10 and 100 μg/kg bw/day of DON, corresponding to 10 to 100-fold human tolerable daily intake concentrations, and to the translation in mice of current human daily intake. The effects of DO

Does Deoxynivalenol Affect Amoxicillin and Doxycycline Absorption in the Gastrointestinal Tract? Ex Vivo Study on Swine Jejunum Mucosa Explants.

The presence of deoxynivalenol (DON) in feed may increase intestinal barrier permeability. Disturbance of the intestinal barrier integrity may affect the absorption of antibiotics used in animals. Since the bioavailability of orally administered antibiotics significantly affects their efficacy and safety, it was decided to evaluate how DON influences the absorption of the most commonly used antibiotics in pigs, i.e., amoxicillin (AMX) and doxycycline (DOX). The studies were conducted using jejunal explants from adult pigs. Explants were incubated in Ussing chambers, in which a buffer containin

Deepoxy-deoxynivalenol retains some immune-modulatory properties of the parent molecule deoxynivalenol in piglets.

Deoxynivalenol (DON) is the most abundant trichothecene in food and feed. It causes both acute and chronic disorders of the human and animal intestine, liver and the immune system. The structural basis for the toxicity of DON has not been fully elucidated. Using the pig as a target and a model species for human, the toxicity of DON and its deepoxy-metabolite (DOM-1) was compared. Animals were exposed by gavage to 1 and 0.5 nmol toxin/kg b.w./day for 2 and 3 weeks respectively. Whatever the dose/duration, DOM-1 was less toxic than DON in terms of weight gain and emesis. In the 3-week experiment
