Nonhormonal therapies for hot flashes in menopause.

Numerous reports in the medical literature and popular media have discussed the effectiveness of various nonhormonal agents in reducing menopausal hot flash symptoms. Data for these therapies are limited, and most of the studies have been conducted in women with a history of breast cancer. Selective serotonin reuptake inhibitors and venlafaxine have been shown to reduce hot flashes by 19 to 60 percent and were well tolerated by study participants. Soy isoflavones reduced hot flashes by 9 to 40 percent in some trials, but most trials showed no difference compared with placebo. Black cohosh and

Carbon dynamics in mycorrhizal symbioses is linked to carbon costs and phosphorus benefits.

The nutrient and carbon (C) allocation dynamics in mycorrhizal hyphal networks cause variation in costs and benefits for individual plants and fungi and influence the productivity, diversity and C cycling in ecosystems. We manipulated light and phosphorus (P) availability in a pot experiment with Trifolium subterraneum colonised by the arbuscular mycorrhizal (AM) fungus Glomus intraradices. Stable (13)C-labelling was used to trace assimilated CO(2) to the mycorrhizal fungus in roots and soil using compound-specific isotope ratio mass spectrometry. We used the neutral lipid fatty acid 16:1omega

Global urban environmental change drives adaptation in white clover.

Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities

Herbal preparations for the treatment of hair loss.

Though hair does not serve any crucial physiological function in modern humans, it plays an important role in our self-esteem. Androgenic baldness (androgenic alopecia) and circular/spot baldness (alopecia areata) are the most common forms of hair loss. Many active ingredients of synthetic origin are available for treatment; however, they have a number of limitations. Their effectiveness and safety are questionable and the amount of time needed to achieve the effect is both long and unclear. This has increased interest in finding an alternative approach against hair loss using preparations con

Development of three copper metalloenzymes in clover leaves.

Subterranean clover (Trifolium subterraneum L. cv Seaton Park) was grown in solution cultures containing adequate nitrogen both with and without Cu. After Cu deficiency had developed, Cu(2+) was added to some deficient plants and Cu content, protein content, and activities of three Cu metalloenzymes (diamine oxidase [EC1.4.3.6], ascorbate oxidase [EC1.10.3.3] and o-diphenol oxidase [EC1.10.3.1]) were assayed in young and recently matured leaf blades over 11 days during the development of the next three leaves.Copper deficiency had little effect on protein concentrations, but markedly depressed

Determination of coumarin via a two-step chromogenic reaction coupled with capillary electrophoresis in legume seeds.

Coumarin is a naturally occurring secondary metabolite found in many plants and exhibits various biological activities. However, robust and reliable techniques for the simultaneous qualitative and quantitative analysis of coumarin are still lacking. This study focused on optimizing the chromogenic reaction of coumarin with the Gibbs reagent by utilizing borate as a chelating agent to enable sensitive detection through capillary electrophoresis (CE). It was observed that alkaline hydrolysis of coumarin at pH 12.1 produced the strongest color response in the presence of borate, which was further

Impact of Alligator Weed (Alternanthera philoxeroides) Invasion on Floral Composition and Soil Microbiota.

Plant invasion modifies the aboveground and belowground biota directly or indirectly via allelopathic effect. This study aimed to ascertain if the invasive species Alternanthera philoxeroides impacts plant diversity, plant composition, and soil microbes or not. The soil microbial activity (CO2 release) and soil pH were also recorded. The plant communities invaded and uninvaded with Alternanthera philoxeroides were examined using the quadrat method. Soil samples were collected from 0 to 10 cm depth, and the culture method was used for soil microbial analysis. The plant species richness and soil

Relative amounts of soluble and insoluble forms of phosphorus and other elements in intraradical hyphae and arbuscules of arbuscular mycorrhizas.

Transport of phosphorus (P) into host plants and its release to root cells is an important function of arbuscular mycorrhizal fungi (AMF). However, relatively little is known about the forms and water solubilities of P compounds in specific locations in the intraradical fungal structures. We determined concentrations and solubility of P components in these structures in white clover (Trifolium repens L.). Plants were grown in the field (colonised by indigenous AMF) or in the glasshouse (inoculated with Glomus intraradices). Mycorrhizas were cryo-fixed in liquid nitrogen immediately (control) o

Optical parametric oscillator-based photoacoustic detection of hydrogen cyanide for biomedical applications.

A versatile, continuous wave, optical parametric oscillator is used in combination with photoacoustic spectroscopy for long-term trace gas experiments of volatile compounds emitted by biological samples. The optical parametric oscillator-based spectrometer (wavelength near 3 μm, 8-MHz linewidth, output power ∼1 W) is successfully tested for the detection of hydrogen cyanide (HCN) emission from clover leaves, and Pseudomonas bacteria; in addition, the presence of HCN in exhaled human breath is measured. For specific experiments, the spectrometer is operated continuously up to 10 days and has a

Red clover RNA virus 1: functional analysis of ORF2 protein, associations with co-infecting viruses, and seed transmission.

A previously undescribed group of related plant viruses has recently been identified within the Benyviridae family. This group is called reclovirids, named after the type species, red clover RNA virus 1 (RCRV1). The single genomic RNA of most reclovirids contains two main open reading frames (ORFs): a large, well-conserved 5′-proximal ORF1 that encodes a viral replicase and a less conserved 5′-distal ORF2 that encodes a polypeptide of unknown function with a C4-type zinc finger motif. In this study, the ORF2 proteins of RCRV1 and a Gymnadenia rhellicani virus, another reclovirid, are found to

Use of Fermented Red Clover Isoflavones in the Treatment of Overactive Bladder in Postmenopausal Women: A Randomized, Double-Blinded, Placebo-Controlled Trial.

Postmenopausal women are at risk of developing an overactive bladder (OAB). Conventional vaginal estrogen has shown promise for symptom relief. Isoflavones have proven effective as an alternative to estrogen treatment against menopause-related symptoms. However, its effect on OAB symptoms has not been studied. This study investigates if fermented red clover isoflavones reduce OAB symptoms in postmenopausal women. In this randomized, double-blinded, placebo-controlled trial, women were administered red clover extract (RCE) or a placebo twice daily for three months. Women filled out the Internat

Continuous directed evolution of isoflavone synthase to mitigate feedback inhibition: combine use of a novel developed bacteria-based biosensor and high-throughput droplet sorting.

Genistein, a bioactive isoflavone with therapeutic potential in treating oxidative stress, cardiovascular diseases, and cancer, faces production limitations during microbial biosynthesis due to product feedback inhibition of isoflavone synthase (IFS). To overcome this bottleneck, a biosensor-assisted continuous directed evolution platform was developed. Specifically, a genistein-specific biosensor was engineered using the Bradyrhizobium japonicum transcription factor FrrA, capable of distinguishing genistein from its precursor, (2S)-naringenin. The biosensor was systematically optimized to eli

Production of Recombinant Hemoglobin in Tremella fuciformis Yeast-like Cells Through Selection of Hemoglobins and Secretion Elements.

Hemoglobins have attracted increasing interest for their potential applications in alternative protein foods and food biotechnology, but the use of fungal yeast-like cells for recombinant hemoglobin production remains underexplored. In this study, Tremella fuciformis yeast-like cells (YLCs) were evaluated as a potential host system by screening candidate hemoglobins and secretion elements. Clover hemoglobin from Trifolium subterraneum (CHb), Vitreoscilla hemoglobin (VHb), and Vigna unguiculata leghemoglobin 2 (Lb2) were heterologously expressed in T. fuciformis YLCs. The estimated yields of CH

Effects of selected condensed tannins on Cryptosporidium parvum growth and proliferation in HCT-8 cell cultures.

Infections with Cryptosporidium spp. constitute a substantial public health burden and are responsible for widespread production losses in cattle herds. Reducing disease and shedding of Cryptosporidium spp. oocysts is an important One Health goal. There are very few therapeutic options available to treat cryptosporidiosis. Interest in plant bioactive compounds to mitigate the spread of anthelmintic resistance in ruminants has led to investigation of these phytocompounds against other parasitic taxa. Condensed tannins (CTs) are plant secondary metabolites that have shown potential against nemat

Structures and kinetics for plant nucleoside triphosphate diphosphohydrolases support a domain motion catalytic mechanism.

Extracellular nucleoside triphosphate diphosphohydrolases (NTPDases) are enzymes that hydrolyze extracellular nucleotides to the respective monophosphate nucleotides. In the past 20 years, NTPDases belonging to mammalian, parasitic and prokaryotic domains of life have been discovered, cloned and characterized. We reveal the first structures of NTPDases from the legume plant species Trifolium repens (7WC) and Vigna unguiculata subsp. cylindrica (DbLNP). Four crystal structures of 7WC and DbLNP were determined at resolutions between 1.9 and 2.6 Å. For 7WC, structures were determined for an -apo

Biochemical and Physiological Responses of Weeds to the Application of a Botanical Herbicide Based on Cinnamon Essential Oil.

The use of chemical herbicides induces negative impacts on the environment, animals, and human health. It also leads to the development of herbicide-resistant weeds. In this context, natural and efficacious herbicides are highly sought after. Essential oils are natural compounds with antibacterial, fungicidal, and phytotoxic properties. For this reason, we studied the post-emergence phytotoxic effect of cinnamon essential oil (cinnamon EO) from Cinnamomum cassia under greenhouse conditions, testing it against Trifolium incarnatum (T. incarnatum) and Lolium perenne (L. perenne). The content of
