NADPH-diaphorase in the developing rat: lower brainstem and cervical spinal cord, with special reference to the trigemino-solitary complex.

A previous study indicated that in adult rat, a distinctive neuronal group in the dorsomedial division of the subnucleus oralis of the spinal trigeminal nucleus (SpVo) and the rostrolateral part of the nucleus of the solitary tract (Sn) is stained for nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d), and suggested that the labeled structures are involved with sensorimotor reflexive functions. This study aimed to characterize the developmental expression of NADPH-d in SpVo and Sn, including other areas of the lower brainstem and cervical spinal cord, by means of the enzyme histo

Prevalence of effusion in the tympanic cavity in dogs with dysfunction of the trigeminal nerve: 18 cases (2004-2013).

BACKGROUND: Animals with disorders involving the trigeminal nerve or its nuclei in the brainstem can have effusion in the tympanic cavity ipsilateral to the side of the neurological deficits. The tensor veli palatini muscle (TVP), innervated by the mandibular branch of the trigeminal nerve, opens the pharyngeal orifice of the auditory tube. With denervation of the TVP, dysfunction of the auditory tube may occur, which could lead to effusion. HYPOTHESIS/OBJECTIVES: To determine the prevalence of effusion in the tympanic cavity in dogs with disorders involving the trigeminal nerve. ANIMALS: Ei

Imaging the trigeminal nerve.

Of all cranial nerves, the trigeminal nerve is the largest and the most widely distributed in the supra-hyoid neck. It provides sensory input from the face and motor innervation to the muscles of mastication. In order to adequately image the full course of the trigeminal nerve and its main branches a detailed knowledge of neuroanatomy and imaging technique is required. Although the main trunk of the trigeminal nerve is consistently seen on conventional brain studies, high-resolution tailored imaging is mandatory to depict smaller nerve branches and subtle pathologic processes. Increasing devel

Localization of proprioceptive neurons innervating the muscle spindles of pig extraocular muscles studied by horseradish peroxidase labelling.

Each muscle of the extraocular muscles, containing abundant muscle spindles, was exposed to horseradish peroxidase (HRP) on 8 young pigs (2-month-old, 20-30 kg in body weight, both sexes). The results obtained are follows: The HRP-labelled neurons innervating the superior rectus muscle were always found in a crescent ventro-medio-dorsal fashion in the most medial position of the contralateral oculomotor nucleus. The HRP-labelled cells for the medial rectus muscle appeared close to the superior rectus group in the ipsilateral nucleus. The labelled cells for the inferior rectus muscle appeared i

Trigeminal Neuropathy Ascribed to a Probable Intrinsic Brainstem Schwannoma of the Pons: A Case Report.

Brainstem schwannomas are very rare, only 11 cases have been reported in the literature so far. We report a small intraparenchymal brainstem schwannoma of the pons, in a 37-year-old female patient who presented with a four-day history of numbness at the mandibular division of the trigeminal nerve on the left side of her face. Trigeminal neuralgia was diagnosed, and magnetic resonance imaging (MRI) showed a small intraparenchymal lesion at the level of the nuclei of the left trigeminal nerve present at the junction between the pons and left brachium pontis. A biopsy wasn't feasible in this smal

Spinocerebellar ataxia type 4 (SCA4): Initial pathoanatomical study reveals widespread cerebellar and brainstem degeneration.

Spinocerebellar ataxia type 4 (SCA4), also known as 'hereditary ataxia with sensory neuropathy', represents a very rare, progressive and untreatable form of an autosomal dominant inherited cerebellar ataxia (ADCA). Due to a lack of autopsy cases, no neuropathological or clinicopathological studies had yet been performed in SCA4. In the present study, the first available cerebellar and brainstem tissue of a clinically diagnosed and genetically-confirmed German SCA4 patient was pathoanatomically studied using serial thick sections. During this systematic postmortem investigation, along with an o

Tissue distribution of Ret, GFRalpha-1, GFRalpha-2 and GFRalpha-3 receptors in the human brainstem at fetal, neonatal and adult age.

Occurrence and localization of receptor components of the glial cell line-derived neurotrophic factor (GDNF) family ligands, the Ret receptor tyrosine kinase and the GDNF family receptor (GFR) alpha-1 to -3, were examined by immunohistochemistry in the normal human brainstem at fetal, neonatal, and adult age. Immunoreactive elements were detectable at all examined ages with uneven distribution and consistent pattern for each receptor. As a rule, the GFRalpha-1 and GFRalpha-2 antisera produced the most abundant and diffuse tissue labelling. Immunoreactive perikarya were observed within sensory

Electron microscopic analysis of lesion-induced changes in synaptic structure and immunogold labeling of neurotransmitters within the feline trigeminal nucleus.

This report uses lesion and postembedding immunogold protocols to examine the ultrastructural details of lesion-induced synaptic and neurotransmitter changes in the feline trigeminal nucleus. Electron microscopic (EM) analysis concentrated on lamina II (substantia gelatinosa) of the subnucleus pars caudalis (PC) which is one relay site or trigeminal fibers involved in nociception. Special attention was directed to analysis of reoccupation of synaptic sites vacated by primary afferent degeneration. Primary afferents are caused to degenerate by performing unilateral retrogasserian rhizotomy. Aft

Distribution of glycogen phosphorylase and cytochrome oxidase in the central nervous system of the turtle Trachemys dorbigni.

Glycogen phosphorylase (GP) and cytochrome oxidase (CO) activities were mapped histochemically in the brain of the turtle Trachemys dorbigni. In the telencephalon, both activities occurred in the olfactory bulb, in all cortical areas, in the dorsal ventricular ridge, striatum, primordium hippocampi and olfactory tubercle. In the diencephalon, they were identified in some areas of the hypothalamus, and in rotundus and geniculate nuclei. Both reactions were detected in the oculomotor, trochlear, mesencephalic trigeminal nuclei, the nucleus of the posterior commissure, torus semicircularis, subst

Nature of neck motor unit activities evoked by different trigeminal inputs in rats.

Recent reports have suggested functional interactions between the masticatory and head motor systems. However, the neurophysiological mechanisms behind these phenomena have not been thoroughly investigated. In this study, the motor unit activity was recorded bilaterally from the dorsal neck splenius muscles (SPL) while the cell bodies of trigeminal afferent fibers were stimulated at either the trigeminal mesencephalic nucleus (Me5) or the Gasserian ganglion (5Gn) in 13 barbiturate-anesthetized Wistar rats. After Me5 electrical stimulation, SPL motor unit responses were recorded ipsilaterally (

Segmental pattern and nuclei in the human embryonic brain at stage 13.

Studies of serial sections and reconstructions of embryos at stage 13 (32) postovulatory days) revealed that the cranial neural tube in the investigated embryos possesses 15 neuromers: 1 telencephalic, 3 diencephalic, 3 mesencephalic, and 8 rhombencephalic. Within the primary efferent column all nuclei of the cranial nerves may be distinguished. Also fibers entering brain from cranial nerve ganglia form the common afferent tract. The relation of neuromers to the motor nuclei of the cranial nerves is following: M1-oculomotor, M2-trochlear, Rh2-trigeminal, Rh4-facial, Rh6-glossopharyngeal, Rh7-v

The trigeminal trophic syndrome: an unusual cause of face pain, dysaesthesias, anaesthesia and skin/soft tissue lesions.

The trigeminal trophic syndrome is an unusual consequence of trigeminal nerve injury that results in facial anaesthesia, dysaesthesia and skin ulceration. Limited knowledge is available. The aim of this study was to increase the knowledge of this syndrome by performing a retrospective medical record review and case series report. Fourteen cases were identified. The female:male ratio was 6:1. Mean age of onset was 45 years (range 6-82). The cause was iatrogenic in most. Latent period to onset ranged from days to almost one decade. The majority (n = 12) had bothersome dysaesthesias. Most (n = 9)

The digastric reflex evoked by tooth-pulp stimulation in the cat and its modulation by stimuli applied to the limbs.

The digastric reflex evoked by electrical stimulation of tooth pulp in anaesthetized cats was studied together with the effects on this reflex of stimulating other parts of the body. The threshold for the digastric reflex generally lay in the range of stimulus intensities which would excite a large proportion of the pulpal afferent fibres which suggested that a large amount of central summation was required to evoke the reflex. During the course of 25/27 experiments, the threshold for the reflex increased. It was also found that repeated application of suprathreshold stimuli produced first an

Diaphragmatic cramp as a possible cause of noncardiac chest pain and referred mandibular pain.

The initial assumption that sudden acute chest pain may be of cardiac origin is justifiable, but when this proves not to be the case the patient is left with little explanation of the cause. It is suggested here that diaphragmatic cramp may be a cause of some undiagnosed noncardiac chest pains associated with mandibular referred pain. The phrenic nerve provides both motor and sensory innervation to the diaphragm, while the trigeminal nerve carries sensation from the mandibular teeth. Both nerves originate in separate nuclei close together in the lower medulla. Interconnections between these nu

Rabies RNA synthesis, detected with cDNA probes, as a marker for virus transport in the rat nervous system.

The kinetics of viral RNA synthesis in different parts of the rat brain, infected with fixed or street rabies virus strains, is correlated with their anatomical neuronal connections with the masseter muscles, using hybridization with rabies cDNA probes. Viral RNA synthesis is first detected in the brain-stem and in the pons where the direct anatomical projection of the masseter muscle nervous arborization into the sensory and motor nuclei is located, through the trigeminus nerve. Rabies RNA detection is delayed in the other regions of the rat brain depending on the time course of virus transpo

A Dynamic Circuit Hypothesis for the Pathogenesis of Blepharospasm.

Blepharospasm (sometimes called "benign essential blepharospasm," BEB) is one of the most common focal dystonias. It involves involuntary eyelid spasms, eye closure, and increased blinking. Despite the success of botulinum toxin injections and, in some cases, pharmacologic or surgical interventions, BEB treatments are not completely efficacious and only symptomatic. We could develop principled strategies for preventing and reversing the disease if we knew the pathogenesis of primary BEB. The objective of this study was to develop a conceptual framework and dynamic circuit hypothesis for the pa

Migraine: where and how does the pain originate?

Migraine is a complex neurological disease with a genetic background. Headache is the most prominent and clinically important symptom of migraine but its origin is still enigmatic. Numerous clinical, histochemical, electrophysiological, molecular and genetical approaches form a puzzle of findings that slowly takes shape. The generation of primary headaches like migraine pain seems to be the consequence of multiple pathophysiological changes in meningeal tissues, the trigeminal ganglion, trigeminal brainstem nuclei and descending inhibitory systems, based on specific characteristics of the trig

Neuropathology of Joubert syndrome.

Very little documentation of the neuropathologic changes in Joubert syndrome exists. This paper presents a detailed postmortem neuropathologic study of a clinically and radiographically well-documented case of Joubert syndrome. In addition to aplasia of the cerebellar vermis and fragmentation of the dentate nuclei, there was marked dysplasia of structures at the pontomesencephalic junction and caudal medulla. There was abnormal decussation of the superior cerebellar peduncles and an enlarged iter (rostral 4th ventricle) with elongated tegmental nuclei (including the locus coeruleus). Neurons o

Subcortical neuroimaging signature of painful disorders of the trigeminal nerve: a UK Biobank study.

BACKGROUND: Trigeminal neuralgia, classified as a painful disorder of the trigeminal nerve (PDTN), is characterized by brief, electric shock-like or stabbing pain localized mainly to the maxillary and mandibular regions. The severity of these paroxysmal attacks severely impairs quality of life and, in extreme cases, may lead to suicidal ideation. We hypothesized that PDTN shares a common neuroanatomical signature with other chronic pain disorders, while also exhibiting disorder-specific alterations. METHODS: High-resolution T1-weighted structural MRI scans from 85 individuals diagnosed with p
