Characterization of trihalomethane (THM)-induced renal dysfunction in the rat. II: Relative potency of THMs in promoting renal dysfunction.

Single non-lethal doses (3 mmol/kg) of chloroform (CHCl3), dichlorobromomethane (CHCl2Br), dibromochloromethane (CHClBr2), and bromoform (CHBr3) were administered by intraperitoneal injection to male Sprague-Dawley rats and proximal tubular secretion and reabsorption was assessed at varied times following treatment. Each of the trihalomethanes (THMs) at this dose inhibited proximal tubular secretion, as indicated by decreased in vitro renal cortical slice accumulation of organic anion p-aminohippuric acid (14C PAH). The time of maximal THM interference with 14C PAH uptake occurred at 8 h, with

Removal assessment of disinfection by-products (DBPs) from drinking water supplies by solar heterogeneous photocatalysis: A case study of trihalomethanes (THMs).

Solar heterogeneous photocatalysis was used to remove trihalomethanes (THMs) from drinking water. THMs, mainly trichloromethane (TCM), tribromomethane (TBM), bromodichloromethane (BDCM) and dibromochloromethane (DBCM) are one of the main class of disinfection by-products (DBPs). THMs were determined by HSGC-MS with detection limits (LODs) ranging from 0.5 μg L-1 to 0.9 μg L-1 for TCM and BDCM, respectively. Results show that a great proportion of THMs present in water are finally transferred to air as a result of their high volatility in the order TCM > BDCM > DBCM > TBM. The use of band-gap s

Microbial degradation of typical amino acids and its impact on the formation of trihalomethanes, haloacetonitriles and haloacetamides during chlor(am)ination.

Nitrogenous disinfection by-products (N-DBPs) in chlorinated drinking water are receiving increasing attention due to their elevated toxicities. An effective strategy to control the formation of N-DBPs is to reduce their nitrogenous precursors (e.g., amino acids [AAs], believed to be the important N-DBP precursors) before disinfection. So far, little information is available about the effectiveness of conventional microbial degradation at controlling the formation of N-DBPs. In this study, the biodegradability of 20 AAs was investigated, and the impacts of microbial degradation for the selecte

Investigating chlorine consumption through semi-mechanistic modeling and its implications for trihalomethane formation: A comparison of diverse drinking water.

In many regions, drinking water disinfection is crucial for the inactivation of microorganisms. However, chemical reactions between chlorine and natural organic matter and inorganic compounds can result in unwanted formation of disinfection by-products (DBPs). 25 non-chlorinated tap water samples from different regions of Germany were chlorinated to evaluate the chlorine consumption and formation of DBPs. Most of the water samples quickly consumed the initially dosed chlorine (NaOCl as 1 mg/L Cl2), leaving less than 10% residual chlorine after 2 h. A strong correlation between ultra-violet abs

Induction of genetic damage in human lymphocytes and mutations in Salmonella by trihalomethanes: role of red blood cells and GSTT1-1 polymorphism.

The brominated trihalomethanes (THMs) are mutagenic and carcinogenic disinfection by-products frequently found in chlorinated drinking water. They can be activated to mutagens by the product of the glutathione S-transferase-Theta (GSTT1++-1) gene in Salmonella RSJ100, which has been transfected with this gene. To evaluate this phenomenon in humans, we have examined the genotoxicity of a brominated THM, bromoform (BF), using the Comet assay in human whole blood cultures exposed in vitro. No differences were found in the comet tail length between cultures from GSTT1-1(+) versus GSTT1-1(-) indivi

Multi-exposure cancer and non-cancer risk assessment of trihalomethanes in drinking water supplies - A case study of Eastern region of India.

The lifetime cancer risk and the hazard index of trihalomethanes (THMs) through oral ingestion, dermal absorption, and inhalation exposure from supply water of five WTPs were analysed. THMs concentration varied from plant to plant and was found to be in the range of 274-511µg/l, which is much higher than the prescribed USEPA standards of 80µg/l. Chloroform was the most dominant THM followed by bromodichloromethane (BDCM), and dibromochloromethane (DBCM). Cancer risk analysis through multi-pathways exposure reveals that residents had a higher cancer risk through oral ingestion than other two ro

Machine learning-driven proteomic profiling reveals seminal plasma biomarkers linking trihalomethane exposure to impaired sperm quality.

The decline in sperm quality has emerged as a major public health concern with significant implications for male fertility and population sustainability. However, high-accuracy biomarkers for sperm quality are lacking, and the environmental drivers remain obscure. Trihalomethanes (THMs), ubiquitous disinfection by-products in drinking water, have been linked to declining semen quality worldwide, yet the molecular mechanisms underlying THM-induced reproductive toxicity remain underexplored. In this propensity score-matched case-control study designed to maximize internal validity, we identified

Impact of Climate and Hydrological Variability on Drinking Water Production and Trihalomethane Levels: A Case Study in Barcelona, Spain (2010-2024).

Surface water-based utilities increasingly face challenges in drinking water production during prolonged droughts and heavy rainfall events. We assessed the impact of climate and hydrological variability on trihalomethane (THM) levels in two drinking water treatment plants in Barcelona: one river-based (Llobregat plant) and one reservoir-based (Ter plant). We examined data from 15 years (2010-2024) using generalized additive models (GAMs) to evaluate the change (β) in chloroform, bromodichloromethane, dibromochloromethane, bromoform, and total THMs (THM4), by extreme (≤percentile 10, ≥percenti

Association of Blood Trihalomethane Concentrations with Risk of All-Cause and Cause-Specific Mortality in U.S. Adults: A Prospective Cohort Study.

Water chlorination can lead to the formation of disinfection byproducts, including trihalomethanes (THMs). However, few epidemiologic studies have explored associations between THM exposure and mortality. This study included 6720 adults aged ≥40 years from the National Health and Nutrition Examination Survey 1999-2012 who had blood THM concentrations quantified. A higher risk of all-cause mortality was found across increasing quartile concentrations of blood chloroform (TCM) and total THMs (TTHMs; sum of all four THMs) (both p for trend = 0.02). Adults in the highest quartile of TCM and TTHM c

Associations of public water system trihalomethane exposure during pregnancy with spontaneous preterm birth and the cervicovaginal microbial-immune state.

BACKGROUND: Water total trihalomethanes (TTHMs) are disinfectant byproducts found in municipal water supplies. TTHM exposure has been linked to cancer and may be associated with adverse reproductive outcomes. A non-optimal cervicovaginal microbiota and low cervicovaginal beta-defensin-2 levels are associated with increased risk of spontaneous preterm birth. Whether TTHM exposure increases the risk of spontaneous preterm birth or alters the cervicovaginal microbial or immune state is unknown. OBJECTIVE: Investigate associations of water TTHM levels with spontaneous preterm birth, a non-optimal

Can CO2 sensors in the ventilation system of a pool facility help reduce the variability in the trihalomethane concentration observed in indoor air?

Volatile and hazardous compounds are formed during the chlorination of pool water. Monitoring components in the air, such as the four trihalomethanes; chloroform, dichlorobromomethane, dibromochloromethane and bromoform (tTHM), is challenging. Carbon dioxide (CO2) sensors are used for controlling air quality in different buildings and can be installed in ventilation systems for continuous surveillance and monitoring purposes. However, such sensors are not used in indoor swimming facilities. In this study, samples of tTHM and CO2 were collected and analysed, along with other air and water quali
