DNA vaccination is known to elicit robust cellular and humoral responses to encoded antigen. The co-administration of costimulatory molecules CD80 (B7-1), CD86 (B7-2) and CD154 (CD40L) has been shown to enhance immune responses in several murine models. The role of specific costimulatory molecules in non-rodent species remains incompletely characterized. In these studies, we demonstrate that the co-administration of CD80 and CD86, but not CD154, to an existing candidate subunit DNA vaccine (ESAT-6) against bovine tuberculosis, enhances protection after aerosol challenge with virulent Mycobacte

Vaccination has been identified as a promising control strategy for tuberculosis in both humans and cattle. Recent heterologous prime-boost approaches combining BCG vaccination with subunit boosts have shown considerable promise in both fields. However, the identification of further protective antigens is still a research priority. In this paper we have established the response hierarchy in Mycobacterium bovis infected or BCG vaccinated cattle of 6 Mycobacterium tuberculosis-derived proteins that were protective against M. tuberculosis in the guinea pig aerosol challenge model. Two of these pr

Bovine tuberculosis (BTB) is an infectious disease of livestock and wildlife species that is caused by pathogenic members of the Mycobacterium tuberculosis complex such as Mycobacterium bovis. Due to the introduction of M. bovis-infected bison in the 1920s, BTB is now endemic in wood bison (Bison bison athabascae) population within the Wood Buffalo National Park (WBNP) in northern Canada. This disease poses a grave threat to the long-term survival of this ecologically and culturally important species and has the potential to cause zoonotic TB and spill over to BTB-free livestock and other biso

Identifying the causes of tuberculosis (TB) chronicity in cattle herds in Spain is a complex endeavour, mainly due to the multiple factors involved in persistence and the clonal population structure of the Mycobacterium tuberculosis complex. This study assessed the genomic diversity among M. bovis isolates belonging to SB0121, the most prevalent genotype in Spain, in chronically-infected herds. A total of 70 M. bovis isolates from 22 herds, located in six Spanish provinces, in which M. bovis SB0121 was isolated in at least three different sampling events were sequenced. Forty-three isolates fr

Mycobacterium bovis is the causative pathogen of bovine tuberculosis (bTB), a disease that affects cattle and other mammals, including humans. Currently, there is no efficient vaccine against bTB, underscoring the need for novel immunization strategies. The M72 fusion protein, composed of three polypeptides derived from Mycobacterium tuberculosis and M. bovis, has demonstrated protective efficacy against M. tuberculosis in clinical trials when combined with the AS01E adjuvant. Given the established efficacy of nanocapsule formulations as vaccine delivery systems, this study evaluated a novel i

Bovine tuberculosis (bTB) is a zoonotic bacterial disease of animals that causes substantial economic losses in underdeveloped countries such as Ethiopia. A cross-sectional study was conducted from May 2021 to August 2021 in the Wolaita zone, southern Ethiopia using a designed questionnaire to assess risk, perception, and protective behavior against bTB. For this purpose, a total of 250 randomly chosen meat handlers working at six main, purposively selected abattoirs were interviewed, and the data were analyzed using STATA v. 13 software. Of the total participants interviewed, 37.0% (57/152) o

The Bacillus Calmette-Guérin (BCG) vaccination provides partial protection against, and reduces severity of pathological lesions associated with bovine tuberculosis (bTB) in cattle. Accumulating evidence also suggests that revaccination with BCG may be needed to enhance the duration of immune protection. Since BCG vaccine cross-reacts with traditional tuberculin-based diagnostic tests, a peptide-based defined antigen skin test (DST) comprising of ESAT-6, CFP-10, and Rv3615c to detect the infected among the BCG-vaccinated animals (DIVA) was recently described. The DST reliably identifies bTB-in

Bovine tuberculosis (bTB) is a zoonosis whose main reservoir and source of infection is cattle. Continuous efforts for disease effective control and eradication are made worldwide, including the identification of bTB-suggestive lesions in slaughtered animals. Since granulomatous lesions could have different causes, laboratory diagnosis is important to confirm the disease etiology. The aim of this study was to evaluate the accuracy of the macroscopic bTB diagnosis during abattoir inspection compared to laboratory confirmation, and to identify macroscopic lesions characteristics associated with

Bovine tuberculosis (bTB), a neglected zoonotic disease, is endemic in cattle in many Sub-saharan African countries, yet its contribution to tuberculosis (TB) burden is understudied. Rapid urbanisation and increase in demand for animal proteins, including dairy products, increases the risk of spill over. This study compared the latent tuberculosis infection (LTBI) risk in children, a proxy-measure for recent TB infection, in children living in high cattle density areas to children from the general population in Cameroon. Cross-sectional study in the Centre Region of Cameroon in 2021, recruitin

BACKGROUND: The fusion protein H65, composed of Mycobacterium tuberculosis (TB) ESX-secreted antigens, has improved the bacillus Calmette-Guerin-induced immune protection in a mouse model of bovine TB when formulated in the liposomal adjuvant CAF01. In this study, we aimed to evaluate the protective efficacy of an attenuated Mycobacterium bovis strain - a mutant in mce2 and phoP genes - combined with H65+CAF01 immunization. We evaluated the protection of MbΔmce2-phoP alone or combined with H65+CAF01 against M. bovis challenge in mice.
METHODS: Groups of BALBc mice were inoculated with the vac

Animals with bovine tuberculosis (bTB) develop granulomatous lesions as part of a complex immune response to chronic antigenic stimulus involving innate and adaptive mechanisms; however, the association of the microbiota with the development of and susceptibility to bTB remains poorly understood. This study characterizes the bacterial communities at different body sites involved in the pathophysiology of bTB, highlighting the gut-lung axis and their association with disease. Lungs, gut, thoracic, and mesenteric lymph nodes samples were collected from 31 bovines from a single herd (n = 19 bTB;

In areas with low bovine tuberculosis (bTB) prevalence, such as Santa Catarina state, Brazil, effective surveillance is essential for disease eradication. Current strategies may miss high-risk farms by inadequately considering spatial risk factors. This study used Kernel Density Estimation (KDE) to analyze spatial risk patterns in Santa Catarina, Brazil, leveraging the official veterinary service's (CIDASC's Sigen+ database) farm data, testing history, and animal movement records. Results revealed that while existing surveillance targets many high-risk areas, some remain unmonitored. Practices

INTRODUCTION: Bovine tuberculosis (TB) in water buffalo (Bubalus bubalis) is primarily diagnosed using intra vitam tests, as the intradermal tuberculin test (IDT) and the interferon-gamma release assay (IGRA), both of which detect cell-mediated immunity (CMI). However, a subset of infected animals fails to mount a detectable CMI response, posing a significant risk of undetected transmission. Serological tests assessing the humoral immune response could provide a valuable complementary tool for identifying infected animals that escape detection through traditional CMI-based assays.

PURPOSE: Tumor necrosis factor inhibitors (TNFi) are known to increase the risk of tuberculosis (TB) reactivation, though cases involving Mycobacterium bovis are rarely reported.
CASE PRESENTATION/RESULTS: We describe a case of disseminated TB with M. bovis in a 78-year-old woman with a negative Interferon-Gamma-Release Assay (IGRA), taking adalimumab due to rheumatoid polyarthritis, which resulted in a fatal outcome. The atypical clinical and histopathological features were initially interpreted as sarcoidosis. The case occurred in Switzerland, an officially bovine tuberculosis-free country.

BACKGROUND: Bovine tuberculosis (bTB) is a chronic disease caused by members of the Mycobacterium tuberculosis complex (MTBC) that ultimately leads to the development of progressive granulomatous lesions. Although the disease is widespread, especially in crossbred cattle in Ethiopia, routine investigations and surveillance are lacking. Thus, the aim of this study was to determine the prevalence, associated risk factors, and species of mycobacteria causing bTB in slaughtered cattle at four slaughterhouses in Central Ethiopia.
METHODS: Postmortem examination of 7,640 cattle was conducted using

BACKGROUND: Tuberculosis in cattle is caused by Mycobacterium tuberculosis complex (MTBC) species. Apart from MTBC, different Nontuberculous Mycobacteria (NTM) species have also been isolated from cattle. The presence of NTM infection in bovines makes the diagnosis of bovine tuberculosis (bTB) a cumbersome task. Therefore, a cross sectional study was conducted to isolate and characterize different Mycobacterium spp. from a slaughterhouse situated in Kolkata, a city in the eastern part of India.
RESULTS: Out of 258 morbid samples, 98 isolates were found to be positive for bacterial growth, and

BACKGROUND: Bovine Tuberculosis is a respiratory disease caused by the pathogen Mycobacterium bovis (M. bovis) that infects cattle. Though rare, this disease can also affect humans, as well as domestic and wild animals, making it a serious concern. Therefore, searching for alternative and new vaccines with high efficiency and safety is the main goal in bovine tuberculosis prophylaxis. New vaccines, known as vector vaccines, have the potential to become safe and effective alternatives to the traditional BCG vaccine. In this study, two major immunodominant proteins of M. bovis Esat-6 and TB10.4

Bovine tuberculosis (TB) is an important zoonotic disease that is caused by Mycobacterium bovis. Eradication efforts in developed countries have reduced the prevalence of this disease significantly. TB can be difficult to diagnose based only on the clinical signs; therefore, it is usually diagnosed in the field with the tuberculin skin test and diagnostic blood tests, including the lymphocyte proliferation assay, the interferon (IFN)-γ assay, and enzyme-linked immunosorbent assay. The aim of this study was to compare the tuberculin and IFN-γ tests. A total of 110 animals were evaluated by tube

Bovine tuberculosis (bTB) remains a problem on many dairy farms in Mexico, as well as a public health risk. We previously found a high frequency of Mycobacterium bovis DNA in colostrum from dairy cows using a nested PCR to detect mpb70. Since there are no reliable in vivo tests to determine the effectiveness of booster Mycobacterium bovis BCG vaccination against bTB, in this work we monitored M. bovis DNA in colostrum by using this nested PCR. In order to decrease the risk of adverse reactions in animals likely containing viable M. bovis, a single application of BCG and a subunit vaccine (EEP-

Vaccination of cattle against bovine tuberculosis could be an important strategy for the control of disease either where there is a wildlife reservoir of Mycobacterium bovis infection or in developing countries where it is not economically feasible to implement a 'test and slaughter' control program. Advances in the understanding of protective immune responses to M. bovis infection in cattle and the advent of new molecular biological techniques, coupled with the sequencing of the M. bovis genome have provided opportunities for the rational development of improved tuberculosis vaccines.
