Surgical removal of a fish hook from the oesophagus of a turtle.

An adult male Macquarie turtle was presented for examination after swallowing a fish hook. Radiographs taken to determine the position of this foreign body within the gastrointestinal tract revealed the fish hook lodged in the turtle's oesophagus. Surgical removal of the fish hook involved an approach through the soft tissues of the lateral neck. The turtle recovered well and was returned to the wild.

The distribution of cholinergic neurons in the central nervous system of turtles.

The distribution of cholinergic neurons was studied in painted and red-eared turtles using antisera against choline acetyltransferase (ChAT). The results showed that the organization of cholinergic systems in the central nervous system of turtles was remarkably similar to that previously described in diverse other species. For example, the present studies revealed the presence of cholinergic neurons in the striatum, the basal telencephalon, the isthmo-tectal system, and a region of cells near the cerebellar peduncle. The motoneurons of the III, IV, V, VI, VII, X and XII cranial nerves and the

Oxidative cost of breathing in the turtle Chrysemys picta bellii.

We estimated the cost of breathing of turtles by measuring ventilation and oxygen consumption during air breathing and CO2 breathing. We assumed that any increment in oxygen consumption due to hypercapnic hyperpnea was due to the metabolic cost of the increased breathing. Six turtles were studied while breathing air and then 5% CO2 in air after at least 12 h breathing each gas. For the measurements, the turtles were submerged unrestrained in water at 20 degrees C and were free to raise their heads into a ventilated chamber. Tidal volumes were measured by the pressure changes in the chamber, an

Membrane recycling in the cone cell endings of the turtle retina.

The ultrastructural effects of dark, light, and low temperature were investigated in the cone cell endings of the red-eared turtle (Pseudemys scripta elegans). Thin sections revealed that in dark-adapted retinas maintained at 22 degrees C, the neural processes which contact the cone cells at the invaginating synapses penetrated deeply into the photoreceptor endings. When dark-adapted retinas were illuminated for 1 h at 22 degrees C, the invaginating processes were apparently extruded from the synaptic endings. On the other hand, 1-h exposure to a temperature of 4 degrees C in the dark caused t

Ventilation, gas exchange and metabolic scaling of a sea turtle.

Ventilation of green turtles (Chelonia mydas) was affected by the position in which the animal was placed: supine animals breathed slowly 0.07 breaths/min) and deeply (8.0 L/breath); prone animals breathed more rapidly (0.43 breaths/min) and more shallowly (3.5 L/breath). From the respiratory exchange ratio and other indicators it appears that green turtles hyperventilate during exercise and hypoventilate during recovery. O2 consumption of the resting sea turtle (0.024 L-kg-1-h-1) is similar to that of other large turtles. Maximal O2 consumption (0.25L-kg-1-h-1) is greater than that of other l

ANALGESIC EFFICACY OF SUBCUTANEOUS MELOXICAM IN RED-EARED SLIDER TURTLES (TRACHEMYS SCRIPTA).

Meloxicam is a commonly used analgesic for reptiles. There are few published studies regarding the efficacy of meloxicam and dosing is typically extrapolated from mammals. We evaluated the analgesic efficacy of two meloxicam doses in red-eared slider turtles (Trachemys scripta). We hypothesized that subcutaneously administered 1 mg/kg meloxicam would significantly increase thermal hindlimb withdrawal latency for a greater time period than 0.4 mg/kg meloxicam. Twenty red-eared sliders were randomly assigned to one of three groups: 1) 0.9% saline (NaCl) (0.5 ml/kg), 2) 0.4 mg/kg meloxicam (Melox

The nasal cavity in sea turtles: adaptation to olfaction and seawater flow.

The nasal cavity of tetrapods has become phylogenetically adapted to the environment in terms of function, respiration, and olfaction. In addition, the nasal cavity of sea turtles plays an important role in seawater flow and water olfaction, unlike that of terrestrial species. Here, we describe the functional, morphological, and histological characteristics of the nasal cavity, and the odorant receptors encoded in the genome of sea turtles. The nasal cavity of sea turtles is well-suited to its complicated functions, and it significantly differs from those of other animals, including terrestria

Species and population specific gene expression in blood transcriptomes of marine turtles.

BACKGROUND: Transcriptomic data has demonstrated utility to advance the study of physiological diversity and organisms' responses to environmental stressors. However, a lack of genomic resources and challenges associated with collecting high-quality RNA can limit its application for many wild populations. Minimally invasive blood sampling combined with de novo transcriptomic approaches has great potential to alleviate these barriers. Here, we advance these goals for marine turtles by generating high quality de novo blood transcriptome assemblies to characterize functional diversity and compare

Thermosensitive sex chromosome dosage compensation in ZZ/ZW softshell turtles, Apalone spinifera.

Sex chromosome dosage compensation (SCDC) overcomes gene-dose imbalances that disturb transcriptional networks, as when ZW females or XY males are hemizygous for Z/X genes. Mounting data from non-model organisms reveal diverse SCDC mechanisms, yet their evolution remains obscure, because most informative lineages with variable sex chromosomes are unstudied. Here, we discovered SCDC in turtles and an unprecedented thermosensitive SCDC in eukaryotes. We contrasted RNA-seq expression of Z-genes, their autosomal orthologues, and control autosomal genes in Apalone spinifera (ZZ/ZW) and Chrysemys pi

SAFETY OF MULTIPLE-DOSE INTRAMUSCULAR KETOPROFEN TREATMENT IN LOGGERHEAD TURTLES (CARETTA CARETTA).

Sea turtles are frequently presented for rehabilitation with injuries for which analgesic treatment is warranted. Ketoprofen is a nonsteroidal anti-inflammatory drug (NSAID) widely used in clinical veterinary medicine for musculoskeletal pain relief. Pharmacokinetics of 2 mg/kg IM have been studied in loggerhead sea turtles (Caretta caretta) as a single and a repeated dose q24hr for 3 days. Safety of longer term administration has not been performed, however, and NSAID use carries a risk of potential complications, including gastrointestinal ulceration, kidney damage, and bleeding. Sensory-evoked turning locomotion in red-eared turtles: kinematic analysis and electromyography.

We examined the limb kinematics and motor patterns that underlie sensory-evoked turning locomotion in red-eared turtles. Intact animals were held by a band-clamp in a water-filled tank. Turn-swimming was evoked by slowly rotating turtles to the right or left via a motor connected to the shaft of the band-clamp. Animals executed sustained forward turn-swimming against the direction of the imposed rotation. We recorded video of turn-swimming and computer-analyzed the limb and head movements. In a subset of turtles, we also recorded electromyograms from identified limb muscles. Turning exhibited

Relating fibropapilloma tumor severity to blood parameters in green turtles Chelonia mydas.

Fibropapillomatosis is a neoplastic disease that is commonly found in the green turtles Chelonia mydas in tropical and subtropical regions of the world. In the current project, juvenile green turtles were captured with large-mesh tangle nets in the Indian River Lagoon and on nearshore reefs of Indian River County, Florida, USA, in 1998 and 1999. The purpose of the study was to determine the relationship between the severity of the disease and the general health of green turtles as indicated by blood parameters. All turtles were measured and examined, and the overall severity of the disease was

Physiological responses to freezing in hatchlings of freeze-tolerant and -intolerant turtles.

Freeze tolerance is a complex cold-hardiness adaptation that has independently evolved in a diverse group of organisms, including several ectothermic vertebrates. Because little is known about the mechanistic basis for freeze tolerance in reptiles, we compared responses to experimental freezing in winter-acclimatized hatchlings representing nine taxa of temperate North American turtles, including ones that tolerated freezing and others that did not. Viability rates of hatchlings frozen to -3 degrees C for 72 h ranged from 0 to 100%. Tolerance to freezing was poor in Sternotherus odoratus, Grap

Effects of anoxia on intracellular free Ca2+ in isolated cardiomyocytes from turtles.

One of the most important negative consequences of hypoxic stress in the mammalian myocardium is a breakdown in intracellular calcium homeostasis. This study examines the effects of anoxic stress on intracellular calcium regulation in isolated ventricular myocytes from a hypoxia tolerant vertebrate, the western painted turtle (Chrysemys picta bellii). Isolated calcium tolerant cardiomyocytes from turtle hearts were mounted on a glass cover slip that formed the bottom of a sealed, Plexiglas perfusion chamber. Free [Ca2+]i (determined by FURA2 fluorescence) in isolated turtle cardiomyocytes aver

Organization of geniculocortical projections in turtles: isoazimuth lamellae in the visual cortex.

The projection from the dorsal lateral geniculate complex to the visual cortex in Pseudemys and Chrysemys turtles was examined by using the anterograde transport of horseradish peroxidase (HRP) in vitro and the retrograde transport of HRP in vivo. In vitro HRP injections into the lateral forebrain bundle were used to fill geniculocortical axons anterogradely, which were then analyzed in cortical wholemount preparations. Geniculocortical axons gain access to the visual cortex along its entire rostral-caudal extent. They course in slightly curved trajectories for up to 2 mm from the lateral edge

Effect of induced hypercapnia on anaerobic metabolic rate of anoxic musk turtles.

To evaluate the possible effect of induced hypercapnia on anaerobic metabolic rate during anoxia, musk turtles (Sternotherus odoratus) were submerged in N2-equilibrated water at 10 degrees C for 3 days either with (anoxic hypercapnic) or without (anoxic normocapnic) elevated aquatic PCO2 (30-40 Torr). Control animals had access to air at 10 degrees C. Plasma [lactate] was significantly higher (P less than 0.01) in the normocapnic [59.4 +/- 7.4 (SD) mM; n = 22] than in the hypercapnic (47.4 +/- 8.5 mM; n = 19) anoxic turtles, although the hypercapnic turtles had lower blood pH (P less than 0.05

Histopathologic Evaluation, Ultrastructure, and Molecular Characterization of Acanthocephalan Infections in Freshwater Turtles.

Turtle acanthocephalans are a depauperate group of helminths, yet they parasitize a quarter of all turtle species in North America. Despite acanthocephalans being common in turtles, their potential as disease-causing agents remains unknown. In other vertebrate hosts, acanthocephalans are known to cause pathologic changes upon attachment (via with their armed proboscis) to the intestinal wall of the host. Herein, we report new turtle host associations, ultrastructural characterization, and molecular data for turtle acanthocephalans. However, the primary goal of this study was to evaluate whethe

Gonadal development in scorpion mud-turtles, Kinosternon scorpioides, in a controlled environment.

Research on gonadal development including sexual differentiation of gonads in chelonians is essential for understanding the mechanisms that determine offspring sex and, consequently, the sex ratio of egg clutches. This directly impacts population structure, as sex ratios influence reproductive dynamics and species' long-term viability. The aim of this study was to characterize the gonadal development of scorpion mud-turtle (Kinosternon scorpioides). A total of 85 embryos from stages 12-26 were analyzed, kept under digital temperature control (28.5 ± 0.5°C). The reproductive system was dissecte
