Resistance breaking Root-knot nematodes carry a fitness cost associated with defective feeding site development.

Root-knot nematodes (RKNs) cause an estimated 157 billion dollars in annual yield losses worldwide. In tomato, resistance to RKNs is conferred by the single dominant resistance gene Mi-1, which has been widely integrated into commercial cultivars. Prolonged and widespread use of Mi-1 has led to the emergence of resistance-breaking populations in tomato fields worldwide; however, the consequences of resistance breaking for nematode performance on susceptible hosts remain poorly understood. Here, we compared infection outcomes of two closely related strains of Meloidogyne javanica, VW4 (Mi-1-avi

Synergistic Enhancement of Soybean Resistance to Cyst Nematodes via Chemical Modulation of Salicylic Acid-Gibberellic Acid Crosstalk.

Soybean cyst nematode (SCN, Heterodera glycines) causes major yield losses, and Rhg1 locus-mediated genetic resistance is becoming less effective. Hormonal signaling pathways, particularly salicylic acid (SA) and gibberellic acid (GA), are increasingly implicated in nematode resistance, but their use in plant protection remains underexplored. Here, we tested exogenous application of chemical modulators of these pathways for effects on SCN resistance and dependence on the presence or absence of Rhg1-b. We found that foliar application of the SA mimic acibenzolar-S-methyl (ASM) or the systemic a

Siderophore-producing Bacillus and free-living nematodes are associated with soil suppressiveness to banana root-knot nematodes.

The control of soil-borne diseases is crucial for ensuring global food security. Here, we investigate the impact of the root-knot nematode (Meloidogyne) on banana continuous cropping over a period of 11 years. The results show significant root infestation initially, but disease incidence declined markedly from the 7th cropping year onwards. Soil community profiling revealed that this intriguing onset of nematode suppressiveness was associated with changes in free-living nematode populations and rhizosphere microbiome composition. Rhizosphere microbiome analyses and strain isolation pinpointed

Morphological and Molecular Characterization of a New Root-Knot Nematode, Meloidogyne limonae n. sp. (Nematoda: Meloidogynidae), Parasitizing Lemon in China.

Root-knot nematodes of the genus Meloidogyne parasitize the roots of thousands of plants and can cause severe damage and yield losses. Here, we report a new root-knot nematode, Meloidogyne limonae n. sp., parasitizing "lemon" (Citrus limon) in Hainan Province, South China. Lemon trees infected by the root-knot nematode showed poor-quality lemons, chlorosis of foliage, weak growth, and numerous root galls with white females and egg masses protruding outside. Phylogenetic trees of sequences within the ribosomal and mitochondrial DNA demonstrated that this species differs clearly from other previ

The Arabidopsis transcription factor TCP9 modulates root architectural plasticity, reactive oxygen species-mediated processes, and tolerance to cyst nematode infections.

Infections by root-feeding nematodes have profound effects on root system architecture and consequently shoot growth of host plants. Plants harbor intraspecific variation in their growth responses to belowground biotic stresses by nematodes, but the underlying mechanisms are not well understood. Here, we show that the transcription factor TEOSINTE BRANCHED/CYCLOIDEA/PROLIFERATING CELL FACTOR-9 (TCP9) modulates root system architectural plasticity in Arabidopsis thaliana in response to infections by the endoparasitic cyst nematode Heterodera schachtii. Young seedlings of tcp9 knock-out mutants

Papain-like cysteine proteases: key players at molecular battlefields employed by both plants and their invaders.

Papain-like cysteine proteases (PLCPs) play crucial roles in plant-pathogen/pest interactions. During these parasitic interactions, PLCPs act on non-self substrates, provoking the selection of counteracting inhibitors and other means to evade proteolysis. We review examples of PLCPs acting on molecular battlefields in the extracellular space, plant cytoplasm and herbivore gut. Examples are maize Mir1 (Maize inbred resistance 1), tomato Rcr3 (Required for Cladosporium resistance-3), Pseudomonas AvrRpt2 and AurPphB, insect DvCAL1 (Diabrotica virgifera cathepsin L-like protease-1) and nematode Mi

A deep learning framework for species-level identification of root-knot nematodes using perineal pattern images.

The use of machine learning is an emerging approach in precision agriculture, including pest identification, disease diagnostics, and soil health monitoring. Root-knot nematodes (Meloidogyne spp.) are a key group of soil-dwelling metazoans that cause plant diseases and lead to significant yield losses in major crops. Identification of these nematodes is essential for pest management. Manual identification of large samples is time-consuming, labour-intensive, and subject to inter- and intra-rater variations, which may affect the consistency and reliability of results. Moreover, the number of sk

Genomic and Transcriptomic Insights into the Evolution and Parasitic Strategy of the Woody-Plant Nematode Pratylenchus vulnus.

The root-lesion nematode Pratylenchus vulnus parasitizes a wide range of hosts, including woody perennials such as walnut (Juglans regia) and grapevine (Vitis vinifera), significantly damaging roots and reducing yields. Here, we present a high-quality, chromosome-level genome assembly of P. vulnus (61.7 Mb across six chromosomes). Comparative genomic analysis revealed high collinearity in protein-coding genes between P. vulnus and the root-knot nematode included in this analysis, Meloidogyne graminicola, indicating a close evolutionary relationship between these genera of plant parasites. Mechanistic elucidation of a terpenoid nano-bionematicide for the management of root-knot nematodes, Meloidogyne incognita infecting tomato.

Plant-parasitic nematodes, particularly Meloidogyne incognita, represent a major constraint to global vegetable production and cause substantial yield losses. Although azadirachtin exhibits strong nematicidal potential, its practical application is limited by instability under field conditions. In this study, a terpenoid-based azadirachtin nano-formulation (Terpaz®, TNF) was developed and evaluated through an integrated approach encompassing physicochemical characterization, in vitro bioassays, molecular docking and molecular dynamics simulations, enzyme inhibition assays, root invasion studie

Hirschmanniella mucronata Effector HmENG1 Interacts with OsGAPDH to Promote Nematode Infection in Rice.

β-1,4-Endoglucanase (ENG) is a canonical effector protein secreted by the esophageal glands of plant-parasitic nematodes. Homology analysis identified a β-1,4-endoglucanase homolog in Hirschmanniella mucronata, designated as HmENG1. In situ hybridization demonstrated the specific localization of Hmeng1 transcripts within the esophageal glands of H. mucronata. Recombinant HmENG1 exhibited cellulase activity in vitro. In planta RNAi experiments confirmed that silencing Hmeng1 significantly reduced the infection capability of H. mucronata. In addition to its potential role in degrading plant cell

Root-knot nematodes (Meloidogyne spp.) a threat to agriculture in Mexico: biology, current control strategies, and perspectives.

Root-knot nematodes (RKN) are sedentary parasites of the roots of plants and are considered some of the most damaging pests in agriculture. Since RKN target the root vascular system, they provoke host nutrient deprivation and defective water transport, causing above-ground symptoms of growth stunting, wilting, chlorosis, and reduced crop yields. In Mexico RKN infestations are primarily dealt with by treating with synthetic chemically based nematicides that are preferred by farmers over available bioproducts. However, due to environmental and human health concerns chemical control is increasing

One pot synthesis of N-7 alkylated xanthine derivatives and their exploration as antioxidant, antimicrobial and nematicidal agents.

The widespread presence of economically dominant pathogens has significantly hindered agricultural growth and productivity by affecting the quality and yield of crops. The recent deregistration of essential bactericides and nematicides have underscored the urgency for the development of novel agrochemical candidates. In response, N-7 alkylated xanthine derivatives were synthesized using various alkylbromides and characterized with the help of various spectroscopic techniques. The synthesized derivatives were explored for antioxidant, antibacterial, antifungal and antinemic properties. A compre

Hijacking the Host Clock: A Nematode Effector Antagonizes Soybean Circadian Defense and Translation Control.

The majority of commercially cultivated soybean cyst nematode (SCN)-resistant soybean cultivars rely on the Rhg1 locus to defend against SCN, the most economically destructive soybean root pathogen. The decline of Rhg1's effectiveness due to its continuous use creates an urgent need for alternative genetic sources to sustain SCN resistance. Although pathogen effectors are known to perturb host circadian clocks in other plant-pathogen systems, whether and how plant-parasitic nematodes interfere with the host clock to promote virulence remains unexplored. Here, we report that the soybean circadi

Cross-Talk Between a Nematode Effector-Targeted Metabolic Enzyme and a Lectin Receptor Kinase Regulates Wheat Immunity.

The cereal cyst nematode (Heterodera avenae) is an obligate plant parasite that severely threatens wheat production. Previous studies have shown that its secreted effector HaGLAND5, suppresses host immunity and promotes nematode parasitism by enhancing acetylation at lysine 39 of TaEMB3003, the E2 subunit of the wheat pyruvate dehydrogenase complex. However, how this molecular interaction precisely regulates the immune response remains unclear. In this study, we identified a wheat L-type lectin receptor kinase, TaLecRK-S.4, and demonstrated its interaction with TaEMB3003. Genetic analyses show

miRNA-Mediated Regulation of Meloidogyne arenaria Responses in Wild Arachis.

MicroRNAs (miRNAs) are key post-transcriptional regulators of plant development and stress responses, with many being conserved across diverse plant lineages. In this study, we investigated the expression profiles of miRNAs and their corresponding target genes in Arachis stenosperma, a wild peanut relative that exhibits robust resistance to root-knot nematodes (RKN). Small RNA sequencing of nematode-infected roots identified 107 miRNA loci, of which 93 corresponded to conserved miRNA families and 14 represented novel candidates, designated as miRNOVO. Among these, 18 miRNAs belonging to 11 con

Effects of MAPK Homologous Genes on Chemotaxis and Egg Hatching in Meloidogyne incognita.

Root-knot nematodes, known for their broad host range and the challenges associated with their control, cause significant losses in global agricultural productivity. The mitogen-activated protein kinase (MAPK) pathway amplifies signals through a phosphorylation cascade to regulate downstream transcription factors or effector proteins, which is essential for maintaining normal physiological functions in nematodes. This study presents a comprehensive characterization of the MAPK signaling cascade regulatory network in Meloidogyne incognita, elucidating the regulatory roles of Mi-let-60, Mi-lin-4

Flavonoid synthesis is crucial for Trichoderma asperellum-induced systemic resistance to root-knot nematodes in tomato plants.

Trichoderma spp. can enhance plant resistance against a wide range of biotic stressors. However, the fundamental mechanisms by which Trichoderma enhances plant resistance against Meloidogyne incognita, known as root-knot nematodes (RKNs), are still unclear. Here, we identified a strain of Trichoderma asperellum (T141) that could effectively suppress RKN infestation in tomato (Solanum lycopersicum L.). Nematode infestation led to an increase in the concentrations of reactive oxygen species (ROS) and malondialdehyde (MDA) in roots but pre-inoculation with T141 significantly decreased oxidative s
