Relationship between mobile phone use and multidimensional frailty: evidence from UK Biobank.

OBJECTIVE: To investigate the longitudinal temporal relationships between weekly mobile phone call time and four dimensions of frailty. METHODS: Data were derived from three instances (T1, T2, T3) of the UK Biobank. Frailty encompassed four dimensions: psychological, physical, social, and cognitive. Separate Random Intercept Cross-Lagged Panel Models (RI-CLPM) were constructed for each frailty dimension with weekly mobile phone call time, measured by self-reported weekly mobile phone call time standardized to baseline. Chi-square difference tests and the BIC were adopted for model comparisons

Modelling cognitive outcomes in the UK Biobank: Education, noradrenaline and frontoparietal networks.

Education is often used as a surrogate measure of so called cognitive reserve (CR) benefiting cognitive functioning in later years. In line with Robertson's theory we tested here a hypothesis that education acting on the noradrenergic system strengthen the right fronto-parietal networks to facilitate CR and maintain cognition throughout the lifetime. We used machine learning and mediation analysis to model interactions between neurobiological features (genetic variants in noradrenergic signalling, structural and functional fronto-parietal connectivity) and education (proxy of CR) on cognitive

Association between cannabis use and risk of metabolic disease in UK biobank.

BACKGROUND AND AIM: The rising global burden of metabolic diseases-including hypertension, type 2 diabetes, hyperlipidemia, obesity, and non-alcoholic fatty liver disease-underscores the urgency of understanding their modifiable risk factors, including cannabis use. We aimed to investigate the association between cannabis use and risk of metabolic diseases across different body mass index levels. METHODS AND RESULTS: We analyzed 91,002 UK Biobank participants without prevalent metabolic diseases at baseline. Multivariable Cox models evaluated associations between cannabis use and incident met

The epidemiology of cubital tunnel syndrome: a UK Biobank case-control study.

INTRODUCTION: Cubital tunnel syndrome is the second most prevalent upper limb compression neuropathy. Limitations in non-operative and surgical treatment warrant an adjunctive preventative approach targeting modifiable risk factors. However, risk factors are poorly characterized given methodologic shortcomings in previous studies. METHODS: In this case-control study in the UK Biobank (1971 cases, 398,639 controls), multivariable logistic regression with extensive covariate correction was conducted to assess the association between cubital tunnel syndrome and sex, age, ethnicity, socioeconomic

Menopause and arterial stiffness index: insights from the women's UK Biobank cohort.

BACKGROUND: Menopause is a significant physiological transition characterized by hormonal changes that can influence cardiovascular health. One key concern is increased arterial stiffness, a predictor of cardiovascular disease and adverse cardiovascular events. However, the independent association between menopause and arterial stiffness, beyond traditional cardiovascular risk factors, remains unclear. This study investigates the relationship between menopause and arterial stiffness index in the women's UK Biobank cohort. METHODS: This cross-sectional study included 52,891 women from the UK B

Relationships between heart shape, function, and disease in 38,858 UK biobank participants.

BACKGROUND: Cardiac functional metrics such as ejection fraction, strain, and valve excursion are important diagnostic and prognostic measures of cardiac disease. However, they ignore a large amount of systolic shape change information available from modern cardiovascular magnetic resonance (CMR) examinations. We aimed to automatically quantify multidimensional shape and motion scores from CMR, investigate covariates, and test their discrimination of disease in the UK Biobank compared against standard functional metrics. METHODS: An automated analysis pipeline was used to obtain quality-contr

Genetic architecture of telomere length in 462,666 UK Biobank whole-genome sequences.

Telomeres protect chromosome ends from damage and their length is linked with human disease and aging. We developed a joint telomere length metric, combining quantitative PCR and whole-genome sequencing measurements from 462,666 UK Biobank participants. This metric increased SNP heritability, suggesting that it better captures genetic regulation of telomere length. Exome-wide rare-variant and gene-level collapsing association studies identified 64 variants and 30 genes significantly associated with telomere length, including allelic series in ACD and RTEL1. Notably, 16% of these genes are know

Identifying healthy individuals with Alzheimer's disease neuroimaging phenotypes in the UK Biobank.

BACKGROUND: Identifying prediagnostic neurodegenerative disease is a critical issue in neurodegenerative disease research, and Alzheimer's disease (AD) in particular, to identify populations suitable for preventive and early disease-modifying trials. Evidence from genetic and other studies suggests the neurodegeneration of Alzheimer's disease measured by brain atrophy starts many years before diagnosis, but it is unclear whether these changes can be used to reliably detect prediagnostic sporadic disease. METHODS: We trained a Bayesian machine learning neural network model to generate a neuroi

Neuroticism and sudden cardiac death: a prospective cohort study from UK biobank.

OBJECTIVE: There is a paucity of evidence on the risk of sudden cardiac death (SCD) according to the degree of neuroticism. We sought to evaluate the association between neuroticism and the long-term risk of SCD. METHODS: From the UK Biobank nationwide prospective cohort, participants free from previous SCD, ventricular arrhythmias, implantable cardioverter-defibrillator (ICD) insertion, depression, schizophrenia, and bipolar disorder were selected. The 12-item scale of neuroticism measurement (neuroticism score) was categorized into high (≥ 3) and low (< 3) groups. The primary outcome was SC

Association Between Regular Laxative Use and Incident Dementia in UK Biobank Participants.

BACKGROUND AND OBJECTIVES: The use of over-the-counter laxatives is common in the general population. The microbiome-gut-brain axis hypothesis suggests that the use of laxatives could be associated with dementia. We aimed to examine the association between the regular use of laxatives and the incidence of dementia in UK Biobank participants. METHODS: This prospective cohort study was based on UK Biobank participants aged 40-69 years without a history of dementia. Regular use of laxatives was defined as self-reported use in most days of the week for the last 4 weeks at baseline (2006-2010). Th

Serum biomarkers are altered in UK Biobank participants with mosaic chromosomal alterations.

Age-related clonal expansion of cells harbouring mosaic chromosomal alterations (mCAs) is one manifestation of clonal haematopoiesis. Identifying factors that influence the generation and promotion of clonal expansion of mCAs are key to investigate the role of mCAs in health and disease. Herein, we report on widely measured serum biomarkers and their possible association with mCAs, which could provide new insights into molecular alterations that promote acquisition and clonal expansion. We performed a cross-sectional investigation of the association of 32 widely measured serum biomarkers with

Cystic fibrosis F508del carriers and cancer risk: Results from the UK Biobank.

Cystic fibrosis (CF) carriers carrying one defective copy of a CFTR germline mutation are common in the general population. A recent study reported associations of CF carriers with risk for cancers of digestive organs and pancreatic cancer. In the current study, we assessed associations of CFTR F508del carriers with the risk for 54 types of cancers in the UK Biobank, a large population-based study. In Caucasians, compared to the carrier rate of 3.15% (12 357/392274) in noncancer subjects, the rate was significantly higher in cancer patients overall (2621/79619 = 3.29%), especially in patients
