Prostaglandin synthesis and its inhibition by cyclic AMP and forskolin in postpartum follicles of the guppy (Poecilia reticulata).

Prostaglandin synthesis in vitro and its inhibition by dibutyryl cyclic AMP and forskolin were examined in postpartum follicles of the guppy. The follicles were cultured for up to 72h under chemically-defined conditions, and the synthesis of prostaglandins was determined by radioimmunoassay. The results show unequivocally that the ovarian follicles devoid of embryos can synthesize PGE and PGF from both endogenous precursors as well as added arachidonic acid. That this could be inhibited by indomethacin is indicative of active synthesis rather than release of preformed prostaglandins. Dibutyryl

Transcriptomic analysis of post-mating neural reprogramming and its coordinated control of ovarian development in the mud crab Scylla paramamosain.

The cerebral ganglion and eyestalk are pivotal regulators of reproduction in decapod crustaceans. To understand how mating drives ovarian development, we performed a comprehensive transcriptomic analysis of these neural tissues in the mud crab, Scylla paramamosain. Our findings reveal that mating induces significant molecular reprogramming, coordinating post-mating neuroendocrine functions. Functional specialization was also evident: the cerebral ganglion showed enrichment in pathways for tight junctions and cytoskeletal reorganization, while the eyestalk primarily activated second messenger s

Warm and cold temperatures have distinct germline stem cell lineage effects during Drosophila oogenesis.

Despite their medical and economic relevance, it remains largely unknown how suboptimal temperatures affect adult insect reproduction. Here, we report an in-depth analysis of how chronic adult exposure to suboptimal temperatures affects oogenesis using the model insect Drosophila melanogaster. In adult females maintained at 18°C (cold) or 29°C (warm), relative to females at the 25°C control temperature, egg production was reduced through distinct cellular mechanisms. Chronic 18°C exposure improved germline stem cell maintenance, survival of early germline cysts and oocyte quality, but reduced

Cold-Sensing TRP Channels and Temperature Preference Modulate Ovarian Development in the Model Organism Drosophila melanogaster.

Temperature is perceived primarily via transient receptor potential (TRP) channels, which are integral to the molecular machinery sensing environmental and cellular signals. Functional evidence of TRP channels' involvement in regulating cold-induced developmental/reproductive responses remains scarce. Here, we show that mutations affecting cold-sensing TRP channels antagonize the reduction in ovarian development induced by low temperatures (reproductive dormancy) in Drosophila melanogaster. More specifically, mutants for brv1, trp, and trpl significantly lowered dormancy levels at 12 °C and ex

Activin B participates in ovarian development in Japanese eel (Anguilla japonica) via coordinated regulation of reproductive signaling and metabolic pathways.

The Japanese eel (Anguilla japonica) is a commercially valuable catadromous species widely cultured in East Asia. However, large-scale artificial propagation remains challenging. Activin B, a dimeric protein belonging to the transforming growth factor-β (TGF-β) superfamily, plays key roles in vertebrate gonadal development, yet its function in eel ovarian development has not been characterized. In this study, we cloned the inhbb gene from Japanese eel, expressed its recombinant Activin B protein, and evaluated its effects on ovarian development under artificial induction conditions. Adult fema

Lipoprotein biology during induced oogenesis in the short-finned eel, Anguilla australis-hepatic target gene expression and accumulation of nutrient sources into developing oocytes.

Production of good quality eggs still plagues mass propagation efforts of anguillid eels. Understanding the lipid dynamics of artificially maturing eels will inevitably benefit captive rearing practices due to the importance of lipids for egg quality. Fortnightly sampling of shortfinned eels (Anguilla australis) subjected to hypophysation for 10 weeks allowed us to gather important information relating to multiple aspects of lipid physiology. Accordingly, expected increases in gonadosomatic (from 2.20 ± 0.14 to 23.20 ± 6.01%) and hepatic somatic indices (from 0.89 ± 0.04 to 1.93 ± 0.01%), as w

Morphology of the female reproductive system and spermatheca in Notonecta maculata Fabricius, 1794 (Heteroptera: Notonectidae).

Notonecta maculata Fabricius, 1794 is an ecologically important freshwater predator of mosquito larvae and other aquatic invertebrates and has potential as a biological indicator of water quality. This study presents the first detailed microscopic anatomical and histological description of the female reproductive system of N. maculata, based on stereomicroscopy, light microscopy, and scanning electron microscopy. The female reproductive system consists of a pair of ovaries, each with seven telotrophic meroistic ovarioles, a spermatheca, and a genital chamber. Each ovariole is differentiated in

Functional characterisation of Fe (II) and 2OG-dependent dioxygenase TcALKBH4 in the red flour beetle, Tribolium castaneum.

Alpha-ketoglutarate-dependent dioxygenase ALKB homologue 4 (ALKBH4) is a member of the Fe (II) and 2-oxoglutarate-dependent ALKB homologue family that plays important roles in epigenetic regulation by alkyl lesions removal in mammals. However, the roles of ALKBH4 in insects are not clear. Here, TcALKBH4 was cloned and functionally characterised in Tribolium castaneum. Temporal expression revealed that TcALKBH4 was highly expressed in early embryos and early pupae. Spatial expression showed that TcALKBH4 was highly expressed in the adult testis, and followed by the ovary. RNA interference targe

Molecular and transcriptional characterization of GTHs and mPRα during ovarian maturation in rock bream Oplegnathus fasciatus.

The membrane progestin receptor α (mPRα) plays an important role in the regulation of oocyte meiotic maturation in fish. However, the endocrine regulation of multiple spawning in fish with asynchronous ovarian development remains to be determined. We identified four genes (FSHβ, LHβ, CGα and mPRα) that are widely expressed in both the reproductive and non-reproductive tissues of rock bream Oplegnathus fasciatus and examined their expression during ovarian maturation. The level of FSHβ transcripts in the pituitary remained high during vitellogenic stages, when the plasma levels of estradiol-17β

Characterizing the symbiotic relationship between Wolbachia (wSpic) and Spodoptera picta (Lepidoptera: Noctuidae): From genome to phenotype.

Wolbachia is a genus of symbiotic bacteria prevalent in arthropods, with diverse effects on host reproduction and fecundity; however, it is unclear how Wolbachia modulates the host reproductive system. In this study, a novel Wolbachia strain, wSpic, was identified in the Noctuid moth Spodoptera picta and its effect on the reproduction of this host was investigated. We sequenced and annotated the 1,339,720 bp genome of wSpic. We identified a total of five WO phage regions in the genome and found no evidence of any plasmids associated with wSpic. Evolutionary analysis revealed that wSpic belongs

Identification of two sterol carrier protein-2 like genes in the yellow fever mosquito, Aedes aegypti.

Two genes encoding sterol carrier protein-2 like proteins are identified from fourth instar cDNAs of the yellow fever mosquito, Aedes aegypti. The predicted AeSCP-2like1 (AeSCP-2L1) and AeSCP-2like2 (AeSCP-2L2) proteins are small, acidic and lacking the peroxisomal targeting sequence at the C-termini. Purified recombinant AeSCP-2L1 and -2L2 bind to cholesterol with a Kd of 5.4 x 10(-6) M and 2.6 x 10(-6) M, respectively. The Kd values of AeSCP-2L1 and -2L2 to palmitic acid are 3.7 x 10(-7) M and 2.6 x 10(-7) M, respectively. Both genes are expressed predominantly in gut tissues. The transcript

Oogenesis and plasma levels of sex steroids in cultured females of brown trout (Salmo trutta linnaeus, 1758) in Chile.

Naturalized brown trout populations in Chile are a valuable genetic resource with aquaculture potential. The oogenesis of a three-year-old brown trout cultured population was studied in southern Chile. Gonadosomatic index (GSI), oocyte growth, gonadal microscopic characteristics, and plasma levels of estradiol-17beta (E2), testosterone (T), and 17alpha-hydroxyprogesterone (17alpha-HP) were measured bimonthly for a nine-month period before spawning. The maximum GSI level (22%) was similar to that described for other salmonids, although it was reached in May, more than one month before the popul

An ovary-enriched piRNA controls ovarian development via Gadd45a-JNK-mediated nurse-cell apoptosis in the arbovirus vector Aedes albopictus.

The Asian tiger mosquito (Aedes albopictus) is a rapidly expanding invasive species and a global vector of arboviruses. Its transmission depends on the female's blood meal-driven reproductive cycle. Although mosquito PIWI-interacting RNAs (piRNAs) research has predominantly focused on antiviral immunity, their roles in reproduction remain poorly understood. Here we profiled sex-biased piRNA expression in Ae. albopictus and identified a female-enriched piRNA, AalpiRNA-18529. This piRNA accumulates in pre-vitellogenic ovaries and is rapidly downregulated after a blood meal. Gain-of-function expe

Ras/MAPK signaling mediates adipose tissue control of ovarian germline survival and ovulation in Drosophila melanogaster.

From insects to humans, oogenesis is tightly linked to nutritional input, yet little is known about how whole organism physiology matches dietary changes with oocyte development. Considering that diet-induced adipose tissue dysfunction is associated with an increased risk for fertility problems, and other obesity-associated pathophysiologies, it is critical to decipher the cellular and molecular mechanisms linking adipose nutrient sensing to remote control of the ovary and other tissues. Our previous studies in Drosophila melanogaster have shown that amino acid sensing, via the amino acid resp

Exposure effects of levonorgestrel on oogenesis in the fathead minnow (Pimephales promelas).

The synthetic progestin levonorgestrel is commonly utilized in human oral contraceptives. It enters the environment as a component of wastewater treatment plant effluent, and has been measured at low ng/L concentrations in surface waters. It has been shown to activate fish androgen receptors, causing the physical masculinization of females, changes in reproductive behavior, and decreases in fecundity. In the present study, the effects of levonorgestrel exposure on early-stage oogenesis in the fathead minnow (Pimephales promelas) was examined. Adult females were exposed to 0, 10, or 100 ng/L le

Broodstock management and hormonal manipulations of fish reproduction.

Control of reproductive function in captivity is essential for the sustainability of commercial aquaculture production, and in many fishes it can be achieved by manipulating photoperiod, water temperature or spawning substrate. The fish reproductive cycle is separated in the growth (gametogenesis) and maturation phase (oocyte maturation and spermiation), both controlled by the reproductive hormones of the brain, pituitary and gonad. Although the growth phase of reproductive development is concluded in captivity in most fishes-the major exemption being the freshwater eel (Anguilla spp.), oocyte

Reproduction and love: strategies of the organism's cellular defense system?

A novel view is presented which states that primordial germ cells and their descendants can be regarded as 'cancerous cells' which emit signals that activate a whole array of cellular defensive mechanisms by the somatoplasm. These cells have become unrestrained in response to the lack of typical cell adhesion properties of epithelial cells. From this point of view: (1) the encapsulation of oocytes by follicle cells, vitelline membrane and egg shell; (2) the suppression of gonadal development in larval life; (3) the production of sex steroid hormones and of vitellogenin; and (4) the expulsion o

In vivo direct cell-penetrating peptide mediated protein transduction system in Acyrthosiphon pisum.

OBJECTIVE: The principal delivery method for CRISPR-based genome editing in insects is now based on microinjection into single cells or embryos. The direct protein transduction systems cannot be employed in aphids because oogenesis occurs without apparent vitellogenesis. Given the limited timing of injection into the embryonic stage in oviparous aphids, a protein delivery system from the hemolymph to the germline and embryos would be a useful tool for genome editing. This study reports a newly developed direct protein delivery system for aphids using cell-penetrating peptides (CPPs). CPPs are

Vitellogenin receptor mediates heat adaptability of oocyte development in mud crabs and zebrafish.

Climate-driven warming affects the reproduction of oviparous ectotherms. However, whether oviparous ectotherms possess a protection mechanism against heat stress for oocyte development, which is essential for maintaining the continuity of animal populations, is largely unknown. Under high temperatures, female mud crabs (Scylla paramamosain) typically have well-formed ovaries, while a few crabs were found to experience oocyte development failure. To investigate the heat stress protection mechanism of oocyte development in mud crabs, we construct a chromosome-level genome of this species and ide
