Contrastive Disentanglement for Quantitative Ultrasound Muscle Atrophy Evaluation.

OBJECTIVE: Muscleatrophy reduces the quality of life and increases morbidity and mortality from other diseases. The development of non-invasive muscle atrophy evaluation method is of great practical value. The lack of gold standard for pathological grading usually allows only the duration of weightlessness as a criterion for the degree of atrophy. However, the adaptive reductive remodeling of muscle physiology and structure shows a trend of nonlinear changes in time. Consequently, using weightlessness time as a benchmark for the degree of atrophy is inaccurate. METHODS: This paper proposes a

Regulation of body fluid volume and electrolyte concentrations in spaceflight.

Despite a number of difficulties in performing experiments during weightlessness, a great deal of information has been obtained concerning the effects of spaceflight on the regulation of body fluid and electrolytes. Many paradoxes and questions remain, however. Although body mass, extracellular fluid volume, and plasma volume are reduced during spaceflight and remain so at landing, the changes in total body water are comparatively small. Serum or plasma sodium and osmolality have generally been unchanged or reduced during the spaceflight, and fluid intake is substantially reduced, especially d

Neurobiological responses of fish to altered gravity conditions: a review.

In vertebrates (including man), altered gravitational environments such as weightlessness can induce malfunctions of the inner ears, based on an irregular dislocation of the inner ear otoliths from the corresponding sensory epithelia. This dislocation leads to an illusionary tilt, since the otolithic inputs are not confirmed by the other sensory organs, which results in an intersensory conflict. Vertebrates in the orbit therefore face severe orientation problems. In humans, the intersensory conflict may additionally lead to a malaise, commonly referred to as space motion sickness (SMS). During

Histomorphometric analysis of rat skeleton following spaceflight.

Male Sprague-Dawley rats were placed in orbit for 7 days aboard the space shuttle. Bone histomorphometry was performed in the long bones and lumbar vertebrae of flight rats and compared with data derived from ground-based control rats. Trabecular bone mass was not altered during the 1st wk of weightlessness. Strong trends were observed in flight rats for decreased periosteal bone formation in the tibial diaphysis, reduced osteoblast size in the proximal tibia, and decreased osteoblast surface and number in the lumbar vertebra. For the most part, histological indexes of bone resorption were nor

Do we have the guts to go? The abdominal compartment, intra-abdominal hypertension, the human microbiome and exploration class space missions.

Humans are destined to explore space, yet critical illness and injury may be catastrophically limiting for extraterrestrial travel. Humans are superorganisms living in symbiosis with their microbiomes, whose genetic diversity dwarfs that of humans. Symbiosis is critical and imbalances are associated with disease, occurring within hours of serious illness and injury. There are many characteristics of space flight that negatively influence the microbiome, especially deep space itself, with its increased radiation and absence of gravity. Prolonged weightlessness causes many physiologic changes th

Abdominal LIPUS Stimulation Prevents Cognitive Decline in Hind Limb Unloaded Mice by Regulating Gut Microbiota.

Weightlessness usually causes disruption of the gut microbiota and impairs cognitive function. There is a close connection between gut microbiota and neurological diseases. Low-intensity pulsed ultrasound (LIPUS) has a beneficial effect on reducing intestinal inflammation. So we wondered if abdominal LIPUS stimulation can have a positive impact on weightlessness induced cognitive decline by reducing intestinal dysfunction. The findings revealed that the hind limb unloaded mice exhibited evident disruption in intestinal structure and gut microbial homeostasis, along with impairment in their lea

Medical imaging in microgravity.

Microgravity environment induces significant pathophysiological changes in humans. As long-term space habitation and interplanetary missions become commonplace in the next century, researchers and imaging specialists must embrace the unique challenges of microgravity research in developing innovative uses of imaging technology. Microgravity results in cephalad fluid shift, loss of electrolytes, loss of muscle and bone mass, anemia, reduced immune response, variability in gastric emptying and hepatic metabolism, increased bowel transit, and development of space motion sickness. Medical imaging

Ultrasound-based Assessment of Jugular Vein Alterations in Response to Simulated Microgravity and Countermeasures: A Systematic Review.

During spaceflight, assessment of jugular vein (JV) alterations is critical as it reflects systemic responses to fluid redistribution and carries a risk of thrombosis, a potentially fatal condition if undetected. Imaging options during spaceflight are restricted, making ultrasound the only feasible modality. However, available data on JV alterations are from small astronaut cohorts (typically n <12), limiting statistical power and robust validation. As a result, ground-based microgravity simulation studies are required to characterise microgravity-induced physiological changes, including JV al

Vestibular function and sensory interaction in space flight.

The vestibular system and vestibulo-visual interaction were examined in 11 astronauts by the electrooculographic (EOG) method during short- and long-term flights on days 2, 3, 5, 9, 22, 50, 164, and 169 (experiments OPTOKINES and LABYRINTH). In space (flight days 2 and 3), they showed enhanced spontaneous vertical nystagmus, and disorders of tracking of vertical and diagonal movements of the stimulus which improved after active head movements. Early increasing of the reactivity of the cupulo-endolymphatic system (flight days 2-3) was replaced after 5 days of flight with a reduction of the vest

Retinal blood vessel diameter changes with 60-day head-down bedrest are unaffected by antioxidant nutritional cocktail.

Long-term human spaceflight can lead to cardiovascular deconditioning, but little is known about how weightlessness affects microcirculation. In this study, we examined how the retinal microvessels and cerebrovascular regulation of 19 healthy male participants responded to long-term head-down bedrest (HDBR), an earth-based analog for weightlessness. In addition, we examined whether an anti-inflammatory/antioxidant cocktail could prevent the vascular changes caused by HDBR. In all study participants, we found a decrease in retinal arteriolar diameter by HDBR day 8 and an increase in retinal ven

Cardiovascular autonomic nervous system responses and orthostatic intolerance in astronauts and their relevance in daily medicine.

BACKGROUND: The harsh environmental conditions during space travel, particularly weightlessness, impose a major burden on the human body including the cardiovascular system. Given its importance in adjusting the cardiovascular system to environmental challenges, the autonomic nervous system has been in the focus of scientists and clinicians involved in human space flight. This review provides an overview on human autonomic research under real and simulated space conditions with a focus on orthostatic intolerance. METHODS: The authors conducted a targeted literature search using Pubmed. RESUL

Effect of altered gravity on the neurobiology of fish.

In vertebrates (including humans) altered gravitational environments such as weightlessness can induce malfunction of the inner ears due to a mismatch between canal and statolith afferents. Coherence between brain cortical function and neurocognitive performance during changed gravity conditions.

Previous studies of cognitive, mental and/or motor processes during short-, medium- and long-term weightlessness have only been descriptive in nature, and focused on psychological aspects. Until now, objective observation of neurophysiological parameters has not been carried out--undoubtedly because the technical and methodological means have not been available--, investigations into the neurophysiological effects of weightlessness are in their infancy (Schneider et al. 2008). While imaging techniques such as positron emission tomography (PET) and magnetic resonance imaging (MRI) would be hard

Numerical Model of the Eye for Understanding Acute Microgravity-Induced Ocular Changes.

INTRODUCTION: Intraocular pressure (IOP) changes with acute microgravity exposure defy easy explanation. IOP increases above supine levels acutely in weightlessness, but studies show both central venous pressure and intracranial pressure (ICP) fall below supine values. These reductions would be expected to reduce IOP in weightlessness. We developed and validated a physiologically relevant numerical model of the eye to investigate how these ocular changes could occur. METHODS: A finite element numerical model of the 50th percentile adult human globe, optic nerve, and surrounding tissues was co

Arterial and venous cerebral blood flow responses to long-term head-down bed rest in male volunteers.

NEW FINDINGS: What is the central question of this study? A heterogeneous cerebral blood flow (CBF) response in the cerebral arteries has been demonstrated in several physiological conditions, and it might be attributable to different physiological properties. However, the whole cerebral haemodynamic response to weightlessness remains unknown. What is the main finding and its importance? Long-term head-down bed rest caused a heterogeneous CBF response between the anterior and posterior cerebral arteries and between the cerebral arteries and veins. Especially, in contrast to the anterior cerebr

Organs of rats flown for 19.5 days on board the Cosmos-782 biosatellite were investigated morphologically and cytochemically. After flight, as a result of drastic decrease or loss of static load on the musculeskeletal system in zero-g, the rats developed mineral metabolism changes that morphologically appeared as osteoporosis of spongy bones, their periosteocytic osteolysis, as well as inhibition of red blood cell precursors in bone marrow, atrophy and dystrophy of skeletal muscles. Metabolic Profiles of Encapsulated Chondrocytes Exposed to Short-Term Simulated Microgravity.

The mechanism by which chondrocytes respond to reduced mechanical loading environments and the subsequent risk of developing osteoarthritis remains unclear. This is of particular concern for astronauts. In space the reduced joint loading forces during prolonged microgravity (10-6 g) exposure could lead to osteoarthritis (OA), compromising quality of life post-spaceflight. In this study, we encapsulated human chondrocytes in an agarose gel of similar stiffness to the pericellular matrix to mimic the cartilage microenvironment. We then exposed agarose-chondrocyte constructs to simulated microgra
