Genetic Diversity of a Brazilian Strain Collection of Xanthomonas citri subsp. citri Based on the Type III Effector Protein Genes.

Exclusion and eradication or management based on an integrated approach with less susceptible varieties, copper-based bactericides, and windbreaks are the two main strategies used to prevent or control citrus canker. Field tolerance or resistance to citrus canker is not found in the most important commercial sweet orange cultivars, and pathogen-derived resistance has been developed and applied in different crops to obtain resistant genotypes to plant pathogens. We describe the development of DNA primers and probes based on the type III effector genes avrXacE1, avrXacE2, avrXacE3, avrBs2, pthA4

Genetic relationship among worldwide strains of Xanthomonas causing canker in citrus species and design of new primers for their identification by PCR.

Partial sequence analysis of the ribosomal operon in Xanthomonas axonopodis allowed discrimination among strains causing the A, B, and C types of citrus bacterial canker (CBC) and quantification of the relationship of these organisms with other species and pathovars in the same genus. Sets of primers based on sequence differences in the internally transcribed spacer and on a sequence from the plasmid gene pthA involved in virulence were designed for specific identification of xanthomonads causing CBC diseases. The two sets were validated with a collection of Xanthomonas strains associated with

GpaR Drives Type III Secretion and Virulence Through Direct Activation of the HrpG-HrpX Cascade in Xanthomonas oryzae pv. oryzicola.

Xanthomonas oryzae pv. oryzicola (Xoc), the causal agent of bacterial leaf streak of rice, relies on a functional type III secretion system (T3SS) for full virulence and for induction of the hypersensitive response (HR) in nonhost plants. Although the HrpG-HrpX regulatory cascade is central to T3SS gene expression in Xanthomonas, the upstream regulatory mechanisms controlling this pathway in Xoc remain insufficiently understood. In the present study, we identified GpaR as a previously unrecognised transcriptional activator required for T3SS-associated pathogenicity in Xoc. Deletion of gpaR sig

The RpfF Acyl-ACP thioesterase of major rice pathogen Xanthomonas oryzae pv. oryzae is essential for maintenance of membrane function and virulence.

In Xanthomonas, RpfF is a bifunctional enzyme having both enoyl-ACP dehydratase and acyl-ACP thioesterase activities catalyzed by a single active site. While the hydratase activity is essential for producing quorum-sensing signals of the diffusible signal factor (DSF) family, the physiological role of the thioesterase activity remained unclear. Singh and coworkers have reported that deletion of the gene encoding RpfF (a ∆rpfF strain) of the rice pathogen X. oryzae pv. oryzae (Xoo) resulted in loss of membrane integrity (Singh et al, 2022). We report that the loss in membrane integrity is due t

In vitro susceptibility of 33 clinical case isolates of Xanthomonas maltophilia. Inconsistent correlation of agar dilution and of disk diffusion test results.

Xanthomonas maltophilia is an emerging nosocomial pathogen, possibly selected by a changing antimicrobial usage and patient population. In the present study, we tested the susceptibility of 33 recent clinical case isolates to 12 commonly employed antimicrobials. Trimethoprim-sulfamethoxazole (1:19 ratio) and doxycycline were uniformly the most active agents; ciprofloxacin and fleroxacin were slightly less active and, along with tetracycline and ceftazidime, more variable in their potency. Interestingly, the disk diffusion method routinely overstated the activity of ciprofloxacin (12% very majo

GC-TOF/MS-based metabolomics analysis to investigate the changes driven by N-Acetylcysteine in the plant-pathogen Xanthomonas citri subsp. citri.

N-Acetylcysteine (NAC) is an antioxidant, anti-adhesive, and antimicrobial compound. Even though there is much information regarding the role of NAC as an antioxidant and anti-adhesive agent, little is known about its antimicrobial activity. In order to assess its mode of action in bacterial cells, we investigated the metabolic responses triggered by NAC at neutral pH. As a model organism, we chose the Gram-negative plant pathogen Xanthomonas citri subsp. citri (X. citri), the causal agent of citrus canker disease, due to the potential use of NAC as a sustainable molecule against phytopathogen
