Activation of transducin by a Xenopus short wavelength visual pigment.

Phototransduction in cones differs significantly from that in rods in sensitivity, kinetics, and recovery following exposure to light. The contribution that the visual pigment makes in determining the cone response was investigated biochemically by expressing a Xenopus violet cone opsin (VCOP) cDNA in COS1 cells and assaying the light-dependent activation of transducin. Light-exposed VCOP stimulated [35S]guanosine 5'-(gamma-thio)triphosphate nucleotide exchange on bovine rod transducin in a time-dependent manner with a half-time for activation of 0.75 min, similar to that of bovine rhodopsin.

Establishment and characterization of Xenopus oviduct cells in primary culture.

Based on a previously established procedure for Xenopus hepatocytes, we describe tubular oviduct cells in primary culture which continue to secrete substantial quantities of egg jelly for several days, as can be visualized microscopically. Freshly isolated cells exhibited a culture shock response [A. P. Wolffe, J. F. Glover, and J. R. Tata (1984) Exp. Cell Res. 154, 581], from which they recovered by the third day in culture. This recovery was characterized by (a) the diminished synthesis of heat shock proteins hsp 70 and hsp 85, (b) the cessation of the drop in number of estrogen receptor, an

The ontogeny of androgen receptors in the CNS of Xenopus laevis frogs.

Androgenic steroids have been implicated in the development of sex differences in Xenopus laevis frogs. In order to determine when neurons first acquire the ability to concentrate androgen, we prepared autoradiograms of CNS in developing frogs following injection of tritiated dihydrotestosterone (DHT). X. laevis tadpoles and juveniles from stage 60 to 2 months post-metamorphosis (PM) were injected with [3H]DHT. Brain and spinal cord autoradiograms from these animals were examined for the presence of labelled cells. The pattern of [3H]DHT labelling in stage-64 tadpoles and in PM juveniles was s

Selective binding of soybean agglutinin to the olfactory system of Xenopus.

The binding patterns of four different lectins conjugated to horseradish peroxidase were investigated in the nervous system of juvenile Xenopus borealis. Only the lectin soybean agglutinin revealed a very selective binding pattern, which was restricted to the olfactory system. The olfactory and vomeronasal epithelia, the olfactory and accessory olfactory nerves and the olfactory and accessory olfactory bulbs were all labelled. The ventral portions of the olfactory nerve and bulb were however more intensely labelled than their dorsal portions. The rest of the brain and spinal cord did not bind

Xenopus as a model system for studying pigmentation and pigmentary disorders.

Human pigmentary disorders encompass a broad spectrum of phenotypic changes arising from disruptions in various stages of melanocyte formation, the melanogenesis process, or the transfer of pigment from melanocytes to keratinocytes. A large number of pigmentation genes associated with pigmentary disorders have been identified, many of them awaiting in vivo confirmation. A more comprehensive understanding of the molecular basis of pigmentary disorders requires a vertebrate animal model where changes in pigmentation are easily observable in vivo and can be combined to genomic modifications and g

Xenopus as a platform for discovery of genes relevant to human disease.

Congenital birth defects result from an abnormal development of an embryo and have detrimental effects on children's health. Specifically, congenital heart malformations are a leading cause of death among pediatric patients and often require surgical interventions within the first year of life. Increased efforts to navigate the human genome provide an opportunity to discover multiple candidate genes in patients suffering from birth defects. These efforts, however, fail to provide an explanation regarding the mechanisms of disease pathogenesis and emphasize the need for an efficient platform to

Developmental neurogenesis in mouse and Xenopus is impaired in the absence of Nosip.

BACKGROUND: Genetic deletion of Nosip in mice causes holoprosencephaly, however, the function of Nosip in neurogenesis is currently unknown. RESULTS: We combined two vertebrate model organisms, the mouse and the South African clawed frog, Xenopus laevis, to study the function of Nosip in neurogenesis. We found, that size and cortical thickness of the developing brain of Nosip knockout mice were reduced. Accordingly, the formation of postmitotic neurons was greatly diminished, concomitant with a reduced number of apical and basal neural progenitor cells in vivo. Neurospheres derived from Nosip

Xenopus tropicalis as a test system for developmental and reproductive toxicity.

The usefulness of Xenopus tropicalis as a model species to investigate endocrine disruption and developmental reproductive toxicity was assessed. In our test system tadpoles were exposed to test substances from shortly after hatching until metamorphosis, including the period of gonadal differentiation. Effects on the sex hormone and thyroid hormone axes were evidenced as skewed sex ratios, malformations of reproductive organs, altered cytochrome (CYP19) (aromatase) activity, and gene expression in gonads and brain, as well as changed thyroid histology and time to metamorphosis. Reproductive to

FoxN3 is required for craniofacial and eye development of Xenopus laevis.

A functional knockdown of FoxN3, a member of subclass N of fork head/winged helix transcription factors in Xenopus laevis, leads to an abnormal formation of the jaw cartilage, absence or malformation of distinct cranial nerves, and reduced size of the eye. While the eye phenotype is due to an increased rate of apoptosis, the cellular basis of the jaw phenotype is more complex. The upper and lower jaw cartilages are derivatives of a subset of cranial neural crest cells, which migrate into the first pharyngeal arch. Histological analysis of FoxN3-depleted embryos reveals severe deformation and f

Breathing and calling: neuronal networks in the Xenopus laevis hindbrain.

Xenopus laevis is an aquatic anuran with a complex vocal repertoire. Unlike terrestrial frogs, vocalizations are independent of respiration, and a single muscle group--the laryngeal dilators--produces underwater calls. We sought to identify the premotor neural network that underlies vocal behaviors. Vocal patterns generated by premotor networks control laryngeal motor neurons in cranial nucleus (n.) IX-X. Glottal motor neurons, active during respiration, are also present in n.IX-X. We used horseradish peroxidase (HRP), Lucifer yellow, and fluorescently conjugated dextrans to characterize the o

Xenopus Bicaudal-C is required for the differentiation of the amphibian pronephros.

The RNA-binding molecule Bicaudal-C regulates embryonic development in Drosophila and Xenopus. Interestingly, mouse mutants of Bicaudal-C do not show early patterning defects, but instead develop polycystic kidney disease (PKD). To further investigate the molecular mechanism of Bicaudal-C in kidney development, we analyzed its function in the developing amphibian pronephros. Bicaudal-C mRNA was present in the epithelial structures of the Xenopus pronephros, the tubules and the duct, but not the glomus. Inhibition of the translation of endogenous Bicaudal-C with antisense morpholino oligomers (

Effects of BmK AS on Nav1.2 expressed in Xenopus laevis oocytes.

In the present study, the pharmacological effects of BmK AS, a beta-like scorpion toxin on rNav1.2 alpha-subunit expressed in Xenopus laevis oocytes were investigated using a two-electrode voltage-clamp recording. It was found that the voltage dependence of rNav1.2 inactivation was significantly shifted towards positive membrane potential by 500 nM BmK AS, whereas the activation curves of rNav1.2 were unruffled at the same dosage. The inactivation curves of both slow and fast inactivation currents were positively moved about 12.8 and 9.7 mV, respectively. In addition, the persistent currents o

Regional gene expression in the epithelia of the Xenopus tadpole gut.

In recent years much progress has been made in the understanding of the genes and mechanisms involved in specification of the cells of the endoderm, which give rise to the epithelium of the gut and respiratory system. However, little is known about the way in which the gut becomes patterned along its anterior-posterior axis, that is, how boundaries are established between the different epithelia of the gut tube. Here we show that the expression patterns of five genes divide the Xenopus tadpole gut epithelium into at least four regions along this axis in the undifferentiated, 3-day-old gut (sta

A screen for targets of the Xenopus T-box gene Xbra.

Brachyury (T), a member of the T-box gene family, is essential for the formation of posterior mesoderm and notochord in vertebrate development. Expression of the Xenopus homologue of Brachyury, Xbra, causes ectopic ventral and lateral mesoderm formation in animal cap explants and co-expression of Xbra with Pintallavis, a forkhead/HNF3beta-related transcription factor, induces notochord. Although eFGF and the Bix genes are thought to be direct targets of Xbra, no other target genes have been identified. Here, we describe the use of hormone-inducible versions of Xbra and Pintallavis to construct

The influence of the olfactory placode on the development of the telencephalon in Xenopus laevis.

Removal of the sensory plate in Xenopus laevis embryos was performed to study the influence of the olfactory anlage on the development of the forebrain. Embryos, which at stage 22-23 underwent removal of the olfactory anlagen, were killed from stage 47 to 60. In 79% of the animals, two olfactory organs reformed and gave origin to two olfactory nerves which contacted the forebrain. In this instance, the telencephalic hemispheres developed normally. In 14% of the animals, one olfactory organ reformed which contacted the brain by means of one olfactory nerve. This resulted in the development of a

The control of mealanoblast differentiation in the periodic albino mutant of Xenopus.

Studies on the incidence of melanophores in older ventral trunk tissues and in isolated regions of periodic albino embryos of Xenopus suggest that melanin granule formation in mutant melanoblasts depends on an environmental contribution which arises at stage 43 in the endodermal tissues.

The deleterious effects of fungicides and herbicides on Xenopus laevis embryos.

Groups of 30 Xenopus laevis embryos, at "tail-bud" stage (Nieukoop-Faber stages 22-24) were exposed to 0.1-2 ppm concentrations of various pesticides for 1 to 10 days. The pesticides used were chloranil and dichlone (both are fungicidal and herbicidal); diquat (herbicide); and nabam (fungicide). The parameters examined were mortality, gross morphology, histology, and behavior. Chloranil (1.25 to 1.75 ppm) treated embryos showed abnormalities of the otolith, optic cup, and general pigmentation. Their movement was sporadically convulsive and they were unable to maintain proper balance. Dichlone

Divergent redox states define regenerative outcomes between salamanders and Xenopus.

Salamanders possess the extraordinary ability to regenerate limbs throughout life, whereas Xenopus lose this capacity after metamorphosis, producing only rudimentary "spikes" after limb amputation. Here, we investigate the underlying mechanisms governing these distinct regenerative outcomes by focusing on the role of endogenous oxygen consumption and reactive oxygen species (ROS) dynamics. The gene expression, oxygen consumption and redox state analysis indicated that Xenopus undergo intense oxidative metabolism and DNA damage during metamorphosis, which was conspicuously absent in salamanders

Overlapping action of T3 and T4 during Xenopus laevis development.

Thyroid hormones are involved in many biological processes such as neurogenesis, metabolism, and development. However, compounds called endocrine disruptors can alter thyroid hormone signaling and induce unwanted effects on human and ecosystems health. Regulatory tests have been developed to detect these compounds but need to be significantly improved by proposing novel endpoints and key events. The Xenopus Eleutheroembryonic Thyroid Assay (XETA, OECD test guideline no. 248) is one such test. It is based on Xenopus laevis tadpoles, a particularly sensitive model system for studying the physiol
