The heterogeneous sulfoethylation of cellulose, xylan, α-1,3-glucan, glucomannan, pullulan, curdlan, galactoglucomannan, and agarose was studied using sodium vinylsulfonate (NaVS) as reagent in presence of sodium hydroxide and iso-propanol (i-PrOH) as slurry medium. The influence of the concentration of polymer, water, and NaOH (solid or aqueous solution) on the degree of substitution (DS) was investigated. The sulfoethylation rendered the polysaccharides studied water-soluble. Sulfoethylation of heteropolysaccharides yielded products with higher DS compared to the conversion of homopolysaccha

Solventogenic Clostridium species can efficiently produce n-butanol and other valuable chemicals via acetone-butanol-ethanol (ABE) fermentation from plant-based feedstocks. For economic and ecological sustainability, cheap and abundant substrates such as lignocellulosic and hemicellulosic residues from agricultural or forestry side streams are preferable. Cereal brans, rich in hemicellulose, represent a promising substrate. However, for direct fermentation of this material, only low product titers are reported. In this study, we characterized the utilization of arabinoxylan, the main polysacch

Bacterial cellulose/arabinoxylan composite (BA) is widely used as plant cell wall analog for binding polyphenols to form antioxidant dietary fiber (ADF), which is a potential functional ingredient. This study aimed to utilize ferulic acid (FA) to prepare ADF (FA-BA complex) and investigate its functional properties and digestive behavior. Results showed that the FA-BA complex was successfully prepared, with FA content of 20.44 μg/mg and the crystallinity index of 87.56 %. The complex exhibited stronger DPPH and ABTS antioxidant activities than the FA-BA mixture. The in vitro digestion assay in

Hetero-trans-β-glucanase (HTG) is an Equisetum plant enzyme that covalently grafts cellulose (donor substrate) to xyloglucan heptasaccharide (XXXG; Xyl3.Glc4 as acceptor substrate). Commercial exploitation of this technology requires bulk XXXG production. Novel acceptor substrates bearing two or more XXXG moieties could potentially cross-link cellulose chains. We explored bulk XXXG production, the synthesis of novel (including multimeric) XXXG conjugates and novel coloured derivatives, and explored new activators of HTG action. Commercial tamarind gum (crude xyloglucan) was digested with xylog

In this study, arabinoxylans (AX) with various molecular weights (Mw) and bound ferulic acid (FA) contents were prepared to compare their effects on the gelatinization, short-term retrogradation and digestive properties of pea starch (PeS). The results indicated that all AX samples could obviously impede the pasting process of PeS and inhibit the short-term retrogradation of PeS-based gels during refrigeration by hindering the rearrangement and double helical associations of amylose. More precisely, AXs with low Mw and the highest FA content (H-FAX) exhibited the strongest intervention ability

Xylanases are widely used in baking, seafood processing, and paper production, but their performance is often compromised under high-salt, acidic, or alkaline conditions, limiting broader industrial deployment. Identifying robust xylanases from saline-alkali environments is therefore of practical importance. Here, we report a GH10 xylanase gene, XynE102, mined from a saline-alkali soil metagenome from Karamay, Xinjiang. The deduced amino acid sequence shares 69.17% identity with a xylanase from Cellvibrionaceae bacterium (GenBank accession HEY7885703.1). XynE102 was cloned and heterologously e

Ammonium persulfate (APS)-assisted hydrothermal pretreatment (HP) is an emerging method for xylan depolymerization, yet its potential for integrated valorization of all biomass fractions remains underexplored. This study aimed to selectively produce xylo-oligosaccharides (XOS) and xylose from corncob via controlled APS-HP, evaluate XOS bioactivity, and ferment xylose hydrolysate to xylonic acid without detoxification, while assessing enzymatic digestibility of solid residues. Under 0.5 % APS at 150 °C for 90 min, an optimal XOS yield reached 40.90 % (based on xylan in corncob), whereas 1 % APS

The gut microbiota in the human body is an important component that plays a pivotal role in the ability of the host to prevent diseases and recover from these diseases. If the human microbiome changes for any reason, it affects the overall functioning of the host. Healthy and vigorous gut microbiota require dietary fiber supplementation. Recently, oligosaccharides have been found to play a significant role in the modulation of microbiota. Several such oligosaccharides, i.e., xylooligosaccharides (XOS), mannooligosaccharides (MOS), and arabino-xylooligosaccharides (AXOS), are derived from hemic

Sweet sorghum bagasse (SSB) was steam pretreated in the conditions of 190 °C for 5 min to assess its amenability to the pretreatment and enzymatic hydrolysis. Results showed that pretreatment conditions were robust enough to pretreat SSB with maximum of 87% glucan and 72% xylan recovery. Subsequent enzymatic hydrolysis showed that the pretreated SSB at 2% substrate consistency resulted in maximum of 70% glucan-glucose conversion. Increasing substrate consistency from 2% to 16% led to a significant reduction in glucan conversion. However, the decrease ratio of glucan-glucose conversion was the

YesT, a putative protein from Bacillus subtilis ATCC 6633 that has been provisionally classified as a rhamnogalacturonan acetyl esterase (RGAE) in CE-12 family, was cloned, expressed in Escherichiacoli Rosetta (DE3), and purified. The enzyme is monomeric with a molecular mass of 37 kDa and presents thermophilic properties similar to RGAE from Aspergillus aculeatus, although YesT is more alkaliphilic. The study of inhibitors confirmed the importance of the His and the nucleophilic Ser for the esterase activity, apart from the Asp from the catalytic triad. This enzyme also presents broad substra

Lint percentage (LP) is a core determinant of cotton yield. Here, we systematically characterized the evolutionary impacts and molecular mechanisms of GhFLA9_D12 on fiber development. Population genomics revealed a significant selective sweep at the locus, where the superior haplotype HapA (T/INS) correlates with higher LP and underwent continuous selection during modern breeding. This favorable variation was likely introgressed from G. barbadense into G. hirsutum. Mechanistically, GhFLA9_D12 regulates fiber development via divergent expression and protein functions. Transcriptionally, a 162-b

Arabinoxylan (AX)-cellulose interactions are central to plant cell wall architecture and biomass recalcitrance. Here, six AX variants differing in the extent and placement of arabinose (Ara) substitutions at O2, O3, and both O2/O3 are examined using molecular dynamics simulations. AX-native (5 Ara groups) reveals a marked surface-dependent variation in its interactions, displaying greater interfacial association with the (110) hydrophilic cellulose than with the (100) hydrophobic cellulose surface. Similarly, AXO3-all, characterized by O3 Ara substitution reveals comparatively reduced associat

From November 1981 to March 1982, an outbreak of scleritis and/or iritis occurred among patients treated with a special series of hemodialyzers. Several works demonstrated that acetylated carbohydrate derivatives derived from hemicellulose played a primary role in the outbreak and that uronic acid residues were present in the carbohydrate derivatives. Based on these previous findings, we tried to clarify the structure of the polysaccharide causing eye lesions. Application of anion-exchange chromatography enabled us to isolate an acidic polysaccharide from the dialyzer extracts. We found that x

The aim of the study was to determine the effects of environmental factors (harvest year and growing location), genotype, and their interactions on the content, molar mass, degree of arabinose substitution, and ferulic acid content of arabinoxylans (AXs) in organically grown rye grain and the corresponding low-extraction rye flours. The experimental material consisted of grain from five population cultivars and two hybrid rye cultivars. Genotype, harvest year, growing location, and their interactions affected most of the analyzed arabinoxylan characteristics, although the significance and magn

Composite hydrogels from whey protein isolate and polysaccharides are promising for encapsulating bioactive compounds like curcumin. This study extracted arabinoxylan from sesame industry waste to create a stable hydrogel system using minimal protein (13%w/v) and polysaccharides (0.65%w/v). The hydrogels displayed significant elasticity and stiffness, confirmed by TPA and dynamic oscillatory tests. The water holding capacity of >99 % and syneresis of <0.7 % demonstrated the efficiency of the composite hydrogel system. Zeta potential values demonstrated the hydrogel's stability and high conduct

The intake of dietary fibers is related with important benefits for human health. We produced two different arabinoxylan fibers with (FAX) and without ferulic acid linked (AX), 12.5 and 0.1 mg g-1 of ferulic acid respectively, by subcritical water extraction of wheat bran. Both FAX and AX fibers were used as supplement in bread production, while non-supplemented bread was used as control. Through an enzymatic deconstruction process we investigated the effect of bread making on the fibers, the preservation of their molecular structure (A/X ratio of 0.13 and Mw of 105 Da) and the interaction wit

Fusarium oxysporum f.sp. melonis, a pathogen of melon (Cucumis melo L.), was grown in shaken cultures at 26 degrees C in a mineral salts medium containing glucose, xylan and apple pectin as carbon sources. The extracellular enzymic complex obtained from these cultures showed lytic activity on plant tissues, causing maceration of melon fruits, potato tubers and carrot roots. Protoplasts were isolated from melon fruits when the maceration was carried out under appropriate osmotic conditions. This fact suggest a possible relationship between the enzymes produced by Fusarium oxysporum f.sp. meloni
