The potential anesthetic effect of ketamine in combination with xylazine was examined in the common carp (Cyprinus carpio). Anesthesia (indicated by cessation of voluntary swimming and loss of responses to pinching the skin with forceps and pricking the tail with a needle) was induced by mixing 14.4 ml of ketamine (5% solution), or 11.4 ml of xylazine (2% solution), or both, in 10 l of water containing 10 fish/group. Xylazine alone induced sedation and anesthesia in 5-12.3 min with a mean duration of 7.3 min. Ketamine alone induced similar effects in 7.2-9.7 min for an average of 24.2 min. Com

Following hind leg intramuscular injections of ketamine, xylazine, and acepromazine, 4 of 6 rabbits exhibited self-mutilation of the digits. At necropsy, the affected sciatic nerve appeared enlarged. Lymphohistiocytic perineural inflammation and fibrosis were observed, together with nerve degeneration. Neuronal regeneration as the reason for self-mutilation is discussed.

The depressant effects of midazolam and xylazine on the central nervous system (CNS) were evaluated in 12 dogs. Xylazine was administered to six dogs (1.1 mg/kg intravenously [IV]) followed in 5 minutes by midazolam (1.0 mg/kg intramuscularly [IM]). In a second group of six dogs, xylazine (2.2 mg/kg IM) was followed in 5 minutes by midazolam (1.0 mg/kg IV). Both drug regimens induced rapid and profound sedation or anesthesia. Duration of action varied with the doses and routes of administration. Dogs given the high dose of xylazine IM had an arousal time of 95.4 +/- 8.9 minutes and a walking t

INTRODUCTION: Xylazine, an α2-adrenergic agonist used as a veterinary sedative, has emerged as an adulterant of illicit opioids, contributing to fatal overdoses in North America and has recently been detected in Europe. In France, despite alarmist media reports, data on xylazine exposures are scarce.
METHODS: We retrospectively reviewed intentional xylazine exposures reported to the eight French poison control centers from January 2010 to December 2024. Cases were extracted from the French National Database of Poisonings. Inclusion required intentional use; severity was assessed using the Poi

Xylazine adulterated Fentanyl is considered one of the deadliest drug threats ever faced in the United States. However, due to the complexity of the illegal drug supply, xylazine detection in the frame of drug-checking (harm reduction) remains an unsolved challenge, one which has cost numerous lives across North America. In this work, we present two sensor platforms for detecting xylazine in drug-checking applications. Both platforms are based on molecularly imprinted polymers (MIPs) using vinyl phosphonic acid as the main monomer. By combining the MIPs with thermometric and colorimetric reado

There are only a few cases of drug-facilitated sexual assaults on children reported in the literature so far. Here, a case of a four-year-old boy is presented. He was unconscious, and the accompanying adults reported that the child had been at a playground on his own. Returning home, he complained of having been stung and collapsed immediately. Urine and serum samples of the child were investigated. In the toxicological analysis, xylazine, a sedative and muscle relaxant used in animals, was detected. Subsequent quantification by GC/MS after solid-phase extraction revealed 0.053 mg/L xylazine i

Hypoxemia is a commonly observed complication during the chemical immobilization of wild ruminants. If severe and left untreated, it can predispose animals to arrhythmias, organ failure, and capture myopathy. The following prospective study was designed to measure the degree of hypoxemia in wapiti that were immobilized with a combination of xylazine and tiletamine-zolazepam and to assess the response to nasal oxygen therapy. Pulse oximetry and arterial blood gas analysis were used to assess the degree of hypoxemia prior to nasal insufflation of oxygen and to demonstrate any beneficial effects

BACKGROUND: Testing street drugs for the presence of active adulterants such as fentanyl and xylazine can provide the user some confidence as to the safety of their drugs.
METHODS: We obtained 3 different commercially available xylazine and 3 different commercially available fentanyl test strips and evaluated them for analytical sensitivity using drug standards. The specificity of the fentanyl strips against fentanyl analogues was also assessed. Powdered fentanyl, xylazine, and fentanyl analogue standards were dissolved in water and serially diluted to bracket the manufacturer's stated test s

In recent years, xylazine has emerged as a cutting agent combined with illicit drugs to extend their effects. The present study aimed to discover drug use patterns associated with xylazine-positive and -negative urine toxicology drug screens and to assess whether xylazine can be used as a marker for exposure to designer drugs/new psychoactive substances in our study population. This is a retrospective analysis of urine toxicology results from two different analytical platforms: a targeted, structurally confirmatory, high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) ass

A recent twist to the ongoing tragedy of the opioid epidemic is the adulteration of fentanyl with the veterinary tranquilizer xylazine, an α2-adrenoreceptor agonist. Unfortunately, our knowledge of how fentanyl and xylazine interact pharmacologically in different behavioral assays is limited. We examined fentanyl and xylazine alone, with 4 antagonists, and in multiple dose ratio combinations using a 52.5 °C hot-plate antinociception assay in Swiss-Webster male and female mice. Both fentanyl and xylazine produced full, dose-dependent increases in antinociception. In antagonism studies, naltrexo

RATIONALE: Xylazine, a sedative typically used in veterinary medicine, has been increasingly detected as an adulterant in the unregulated opioid supply and present in opioid overdose deaths. Therefore, xylazine-adulterated fentanyl is a growing public health concern. People who use drugs have reported that xylazine changes and prolongs the effects of fentanyl.
OBJECTIVES: We used standard operant drug discrimination procedures to better understand how xylazine impacts the discriminative stimulus/interoceptive effects of fentanyl.
METHODS: Male and female Long-Evans rats (n = 23) were trained

Xylazine, an α2-adrenergic receptor agonist commonly used as a veterinary sedative, has gained notoriety for its increasing misuse in combination with opioids, such as fentanyl, to potentiate its effects. This trend has led to a rise in cases of xylazine poisoning. Although timely diagnosis and intervention have reduced fatalities, the postmortem pathological manifestations and lethal blood concentration of xylazine alone remain poorly characterized due to its common co-occurrence in toxicology with fentanyl. This study presented a case of homicide involving fatal intoxication solely from xyla

Medetomidine has been increasingly found in the illegal opioid supply across the country over the last few years. In 2024, it replaced xylazine as the most common adulterant of the illegal opioid supply in the Philadelphia area. Medetomidine is estimated to be over 100 times as potent and selective for the alpha-2 receptor than xylazine. This leads to increased sedative effects compared with other alpha-2 agonists and in some patients, a severe autonomic withdrawal syndrome when medetomidine is abruptly stopped. Severe hypertension, tachycardia, vomiting, and tremors have been seen, often requ

BACKGROUND: Xylazine, a veterinary tranquilizer, has emerged as a widespread adulterant in the U.S. illicit drug supply, detected in over 90 % of street fentanyl samples in some regions and identified in a growing number of overdose deaths. While xylazine's health risks are well-documented, little is known about how its presence influences substance use behaviors. We aimed to explore behavioral changes among people encountering xylazine in illicit drug markets.
METHODS: We conducted semi-structured in-depth interviews with people with recent overdose reversal experiences in two Midwestern cit

Xylazine is a veterinary sedative that is frequently detected in the illicit drug supply, often found mixed with illicitly manufactured fentanyl (IMF). It has been detected in the blood of overdose victims and patients who use illicit drugs. Xylazine is not approved for use in humans. It is an alpha-2-adrenergic receptor agonist that causes deep sedation that is non-responsive to naloxone, the antidote for opioid overdose. Chronic exposure to xylazine has been linked to severe wounds that can progress to amputation. Medetomidine is another related veterinary sedative that has more recently eme

Xylazine exposure is common in some US cities, but a commercial assay for routine laboratory testing for xylazine is not currently available. We evaluated a pre-release version of the ARK Diagnostics immunoassay for qualitative detection of xylazine/4-hydroxyxylazine in urine. Studies were conducted using either the semi-quantitative assay application (A. Roche Cobas 503 analyzer) or the qualitative assay application (B. Beckman Coulter AU480 analyzer). Study specimens consisted of deidentified patient urine samples submitted for routine drugs-of-abuse testing. Measurements of xylazine (X) wer
