Regulation of YAP by mTOR and autophagy reveals a therapeutic target of tuberous sclerosis complex.

Genetic studies have shown that the tuberous sclerosis complex (TSC) 1-TSC2-mammalian target of Rapamycin (mTOR) and the Hippo-Yes-associated protein 1 (YAP) pathways are master regulators of organ size, which are often involved in tumorigenesis. The crosstalk between these signal transduction pathways in coordinating environmental cues, such as nutritional status and mechanical constraints, is crucial for tissue growth. Whether and how mTOR regulates YAP remains elusive. Here we describe a novel mouse model of TSC which develops renal mesenchymal lesions recapitulating human perivascular epit

Trimethylamine N-oxide induces macrophage M1 polarization via the Piezo1/Yes-associated protein pathway.

Trimethylamine N-oxide (TMAO), a gut microbiota-derived metabolite, has been implicated in cardiometabolic inflammation; however, the intracellular signaling mechanisms linking TMAO to macrophage polarization remain incompletely defined. This study aimed to investigate whether TMAO promotes macrophage M1 polarization through a Piezo1-Yes-associated protein (YAP) signaling axis. Gain- and loss-of-function approaches were performed in RAW264.7 cells and primary bone marrow-derived macrophages, including Piezo1 overexpression, small interfering RNA-mediated knockdown of Piezo1 or YAP, and pharmac

Unraveling the Hippo pathway: YAP/TAZ as central players in cancer metastasis and drug resistance.

In regulating cellular plasticity, epithelial to mesenchymal transition (EMT), and tumor progression across a broad range of cancer types, the Hippo signaling pathway depends on YAP (Yes-associated protein) and TAZ (transcriptional coactivator with PDZ binding motif) as core effectors. This pathway can become dysregulated, disrupting tissue homeostasis and promoting oncogenic processes such as metastasis, immune evasion, and therapy resistance. This review explores the multifaceted roles of YAP/TAZ in lung, breast, ovarian, liver, and renal cancers, detailing their interactions with key signal

Mechanistic insights into Hippo-YAP pathway activation for enhanced DFU healing.

With the increasing prevalence of diabetes, diabetic foot ulcers (DFUs) have become a global health challenge, significantly impacting patients' quality of life and placing a substantial burden on healthcare systems. Among various immune cell subsets, M2-polarized macrophages play a pivotal role in tissue repair and inflammation resolution. This study uses single-cell RNA sequencing (scRNA-seq) and bulk RNA sequencing to comprehensively investigate the role of the TFAP2A-LIFR-Hippo-YAP signaling axis in regulating macrophage M2 polarization and its critical function in DFU wound healing. Amelioration of NAFLD by sleeve gastrectomy-triggered hepatocyte regeneration in mice - experimental research.

BACKGROUND: Sleeve gastrectomy (SG) is known to alleviate non-alcoholic fatty liver disease (NAFLD) and restore liver function; however, its underlying mechanism remains unclear. MATERIALS AND METHODS: We investigated the effect of SG on the metabolic phenotype of diet-induced obese (DIO) mice. Postoperative stained liver images were analyzed to determine the hepatocyte proliferation phenotype. Single-cell RNA sequencing was used to characterize the regeneration signals of the liver after SG in DIO mice, and real-time quantitative reverse transcription PCR was performed to verify the above re

Wnt/β-catenin-YAP axis in the pathogenesis of primary intraosseous carcinoma NOS, deriving from odontogenic keratocyst.

Odontogenic tumors (OGTs), which originate from cells of odontogenic apparatus and their remnants, are rare entities. Primary intraosseous carcinoma NOS (PIOC), is one of the OGTs, but it is even rarer and has a worse prognosis. The precise characteristics of PIOC, especially in immunohistochemical features and its pathogenesis, remain unclear. We characterized a case of PIOC arising from the left mandible, in which histopathological findings showed a transition from the odontogenic keratocyst to the carcinoma. Remarkably, the tumor lesion of this PIOC prominently exhibits malignant attributes

p21-activated kinase 4 drives hepatic stellate cell activation and liver fibrosis through the phosphorylation of YAP at T428.

Understanding the molecular mechanisms that govern hepatic stellate cell (HSC) fate is essential for developing effective antifibrotic therapies. Yes-associated protein (YAP) plays a pivotal role in organ fibrogenesis and HSC activation, but its upstream regulatory signals in liver fibrosis remain largely unknown. Here, we demonstrate the role of p21-activated kinase 4 (PAK4), an emerging therapeutic target, in the activating phosphorylation of YAP and subsequent HSC activation. PAK4 expression was markedly upregulated in activated HSCs from patients and mice with liver fibrosis and correlated

Blockade of YAP alleviates hepatic fibrosis through accelerating apoptosis and reversion of activated hepatic stellate cells.

Yes-associated protein (YAP) is a significant downstream protein in the Hippo signaling pathway with important functions in cell proliferation, apoptosis, invasion and migration. YAP also plays a role in the progression and development of various liver diseases. In hepatic fibrosis development and reversion, the proliferation and apoptosis of activated hepatic stellate cells (HSCs) play a critical role. However, the contribution of YAP to hepatic fibrosis progression and reversion and the underlying mechanism have not been investigated. Here we investigated the expression and function of YAP i

The epidermal growth factor receptor regulates YAP signalling and promotes diabetic retinal fibrosis by modulating the degradation of SAV1.

PURPOSE: The epidermal growth factor receptor (EGFR) and Yes-associated protein (YAP) signalling pathways are two intrinsic mechanisms that typically control fibrosis. The intracellular molecules that regulate these two pathways remain unclear. Here, we investigated how EGFR and YAP signalling control retinal Müller cell-related diabetic retinal fibrosis in vivo and in vitro. METHODS: Human retinal Müller cells (MIO-M1) were transfected in vitro with a Flag-tagged EGFR expression plasmid or an EGFR-targeting siRNA. Western blotting and coimmunoprecipitation (Co-IP) were used to assess the rol

YAP/TAZ as a mechanochemical signal-resolution circuit.

Mechanical forces regulate cell fate, tissue homeostasis, regeneration, fibrosis and cancer through interconnected mechanotransduction networks in which Yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ) serve as major transcriptional effectors. This review develops a mechanochemical signal-resolution circuit framework that integrates evidence from mechanobiology, Hippo signaling, nuclear transport, chromatin regulation, extracellular matrix remodeling and mechanical memory. The framework defines YAP/TAZ regulation as a distributed process in which mechan

Hepatocellular carcinoma is one of the deadliest cancers occurring worldwide. Substantial evidence suggests that the Hippo-YAP pathway as a promising therapeutic target. Phyllanthus niruri is known for pharmacological activities, including hepatoprotective, anti-viral, and anti-inflammatory effects. However, there is a lacuna of molecular studies on how the bioactives in Phyllanthus niruri interacts with hippo signaling to enhance its potential for liver cancer management. This study aimed to delineate the effects of Phyllanthus niruri extract on cell proliferation and apoptosis, and its mecha

L1CAM-ILK-YAP Mechanotransduction Drives Proliferative Activity of Epithelial Cells in Middle Ear Cholesteatoma.

Middle-ear cholesteatoma (cholesteatoma) is a chronic otitis media with an enhanced proliferation of epithelial cells. Negative pressure in the middle ear is thought to be important for the etiology of cholesteatoma. However, the mechanism of cholesteatoma formation remains unclear. Integrin-linked protein kinase (ILK), an important modulator of actin cytoskeletal dynamics, interacts with extracellular matrix and results in the up-regulation of mechanotransduction effector Yes-associated protein (YAP). The L1 cell adhesion molecule (L1CAM) has recently been reported as an activator of the mech

aYAP modRNA reduces cardiac inflammation and hypertrophy in a murine ischemia-reperfusion model.

Myocardial recovery from ischemia-reperfusion (IR) is shaped by the interaction of many signaling pathways and tissue repair processes, including the innate immune response. We and others previously showed that sustained expression of the transcriptional co-activator yes-associated protein (YAP) improves survival and myocardial outcome after myocardial infarction. Here, we asked whether transient YAP expression would improve myocardial outcome after IR injury. After IR, we transiently activated YAP in the myocardium with modified mRNA encoding a constitutively active form of YAP (aYAP modRNA).

RNF207 promotes colorectal cancer growth by regulating the Hippo-YAP pathway via enhanced MST1 ubiquitination and degradation.

BACKGROUND: Colorectal cancer ranks second in global mortality rates, yet its molecular pathogenesis remains incompletely understood. Accumulating evidence indicates that E3 ubiquitin ligases modulate the activity and stability of critical oncoproteins via ubiquitination, positioning these enzymes as promising therapeutic targets for cancer intervention. The E3 ligase Ring finger protein 207 (RNF207), characterized as a regulator involved in heart disease, has not been investigated in colorectal cancer. METHODS: We constructed RNF207 overexpression and knockdown colorectal cancer cell lines,

YAP and long non-coding RNAs in immune cells: context-dependent regulation and a hypothesis-generating synthesis of their potential crosstalk.

Yes-associated protein (YAP) is a transcriptional co-activator classically associated with tissue growth, mechanotransduction, and oncogenesis. Emerging evidence now identifies YAP as a critical regulator of immune cell function, influencing inflammatory signaling, antiviral responses, immune cell differentiation, and effector activity. In immune cells, YAP integrates biochemical and biomechanical cues to shape context-dependent outcomes, acting either as a promoter or suppressor of immune activation depending on cell type and microenvironment. Long non-coding RNAs (lncRNAs) have also emerged

Pannexin 1 crosstalk with the Hippo pathway in malignant melanoma.

In this study, we explored the intricate relationship between Pannexin 1 (PANX1) and the Hippo signaling pathway effector, Yes-associated protein (YAP). Analysis of The Cancer Genome Atlas (TCGA) data revealed a significant positive correlation between PANX1 mRNA and core Hippo components, YAP, TAZ, and Hippo scaffold, IQGAP1, in invasive cutaneous melanoma and breast carcinoma. Furthermore, we demonstrated that PANX1 expression is upregulated in invasive melanoma cell lines and is associated with increased YAP protein levels. Notably, our investigations uncovered a previously unrecognized int

Hippo effector, Yorkie, is a tumor suppressor in select Drosophila squamous epithelia.

Mammalian Yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ) and Drosophila Yorkie (Yki) are transcription cofactors of the highly conserved Hippo signaling pathway. It has been long assumed that the YAP/TAZ/Yki signaling drives cell proliferation during organ growth. However, its instructive role in regulating developmentally programmed organ growth, if any, remains elusive. Out-of-context gain of YAP/TAZ/Yki signaling often turns oncogenic. Paradoxically, mechanically strained, and differentiated squamous epithelia display developmentally programmed con

Shh-Yap signaling controls hepatic ductular reactions in CCl4 -induced liver injury.

Carbon tetrachloride (CCl4 ) exposure can induce hepatic ductular reactions. To date, however, the related mechanism remains largely unknown. Sonic hedgehog (Shh) and Yes-associated protein (Yap) signaling are correlated with liver injury and regeneration. Herein, we investigated the role of Shh and Yap signaling in the fate of ductular reaction cells in CCl4 -treated livers and the possible mechanisms. Wild-type and Shh-EGFP-Cre male mice were exposed to CCl4 (2 mL/kg), and then treated with or without the Shh signaling inhibitor Gant61. The level of liver injury, proliferation of ductular re
