Harnessing glycerol for secondary metabolite biosynthesis in microorganisms.

The increase in glycerol availability as a byproduct of biodiesel production has attracted attention in its utilization as a carbon source for microbial fermentation. Compared with traditional carbon sources such as glucose, glycerol enhances the production of various secondary metabolites such as aromatic compounds, polyols, and lipids in microorganisms such as Komagataella phaffii and Yarrowia lipolytica. For instance, in K. phaffii, glycerol increased the titers of p-coumarate, naringenin, and resveratrol compared with glucose-based media, particularly when combined with specific amino acid

The digestive and defensive basis of carcass utilization by the burying beetle and its microbiota.

Insects that use ephemeral resources must rapidly digest nutrients and simultaneously protect them from competitors. Here we use burying beetles (Nicrophorus vespilloides), which feed their offspring on vertebrate carrion, to investigate the digestive and defensive basis of carrion utilization. We characterize gene expression and microbiota composition in the gut, anal secretions, and on carcasses used by the beetles. We find a strict functional compartmentalization of the gut involving differential expression of immune effectors (antimicrobial peptides and lysozymes), as well as digestive and

Functional characterization of a novel aspartic acid rich lipase, TALipC, from Trichosporon asahii MSR54: solvent-dependent enantio inversion during esterification of 1-phenylethanol.

A novel lipase gene TAlipC was isolated from Trichosporon asahii MSR54 and functionally expressed in Pichia pastoris. The protein was His-tagged and purified to homogeneity by affinity chromatography. Sequence comparison revealed a high homology with lipases from Cryptococcus sp. It had a GX type oxyanion hole and a GHSLG-type conserved penta-peptide similar to those in the lipases from Yarrowia lipolytica. The enzyme had optimal activity at pH 8 and 50 °C. It was specific for long chain fatty acid groups of p-nitrophenol esters and triacylglycerols, showing regio- and enantio-selectivity. It

Synthetic microbial communities of heterotrophs and phototrophs facilitate sustainable growth.

Microbial communities comprised of phototrophs and heterotrophs hold great promise for sustainable biotechnology. Successful application of these communities relies on the selection of appropriate partners. Here we construct four community metabolic models to guide strain selection, pairing phototrophic, sucrose-secreting Synechococcus elongatus with heterotrophic Escherichia coli K-12, Escherichia coli W, Yarrowia lipolytica, or Bacillus subtilis. Model simulations reveae metabolic exchanges that sustain the heterotrophs in minimal media devoid of any organic carbon source, pointing to S. elo

Cell death in Ustilago maydis: comparison with other fungi and the effect of metformin and curcumin on its chronological lifespan.

Ustilago maydis is a Basidiomycota fungus, in which very little is known about its mechanisms of cell survival and death. To date, only the role of metacaspase1, acetate and hydrogen peroxide as inducers of cell death has been investigated. In the present work, we analyzed the lifespan of U. maydis compared with other species like Sporisorium reilianum, Saccharomyces cerevisiae and Yarrowia lipolytica, and we observed that U. maydis has a minor lifespan. We probe the addition of low concentrations metformin and curcumin to the culture media, and we observed that both prolonged the lifespan of

Microbial succession and interaction in vacuum-packed beef: a longitudinal study of bacterial and fungal dynamics.

The microbial dynamics of vacuum-packed (VP) beef are shaped by interactions between bacterial and fungal communities, influencing spoilage and meat quality during storage. While bacterial succession is well studied, fungal roles remain underexplored. We examined microbial communities in VP beef over 85 days using spike-in, qPCR, 16S/18S rRNA gene amplicon sequencing, culture-based methods, whole genome sequencing, and co-culture experiments. Initially dominated by Pseudomonas and Brochothrix, the bacterial community shifted toward lactic acid bacteria (LAB) by day 15. Fungal communities remai

Hydrolysis of Gluten-Derived Celiac Disease-Triggering Peptides across a Broad pH Range by RmuAP1: A Novel Aspartic Peptidase Isolated from Rhodotorula mucilaginosa.

An aspartate peptidase with proteolytic activity toward gluten was identified from an isolated red yeast Rhodotorula mucilaginosa strain. This peptidase consists of 425 amino acids, comprising an N-terminal signal peptide, a propeptide, and a C-terminal catalytic domain. The catalytic domain, termed RmuAP1CD, could be secreted by the recombinant oleaginous yeast Yarrowia lipolytica, whose genome contains the expression cassette for RmuAP1CD. RmuAP1CD exhibited optimum activity at pH 2.5 when acting on bovine serum albumin. Moreover, it facilitated the hydrolysis of gluten-derived immunogenic p

From hive to laboratory - biotechnological potential of microorganisms from honey.

Honey is a valuable source of microorganisms that may have potential applications in various fields of science and industry. The high sugar content and low pH inhibit the growth of most microorganisms; however, some are capable of adapting to these conditions. Moreover, the bioactive components of honey, such as hydrogen peroxide and flavonoids, exhibit antimicrobial activity, which limits the proliferation of pathogens while simultaneously promoting the selection of microorganisms with beneficial properties. These include bacteria from the genera Lactobacillus, Fructilactobacillus, Leuconosto

Biosynthesis and Microbial Production of Carminic Acid: From Pathway Elucidation to Synthetic Biology.

Carminic acid (CA) is a high-value natural anthraquinone pigment used in foods, cosmetics, textiles, and pharmaceuticals, but its current industrial supply depends largely on extraction from the scale insect Dactylopius coccus, creating constraints in yield, cost, sustainability, and allergen control. This review summarizes recent progress from pathway elucidation to microbial production. We first outline the structure, occurrence, applications, and biosynthetic logic of CA, emphasizing the convergence of type III polyketide assembly with insect-associated tailoring reactions, especially C-gly

Host analysis-guided selection and targeted engineering (HASTE) of Lipomyces tetrasporus for the conversion of CO2-derived feedstocks.

Cost-competitive bioproduction calls for utilizing cellulosic sugars or acetate from electro-reduction of CO2. Here, we identified a novel Lipomyces tetrasporus strain (i.e., NRRL Y-11562 from 24 Lipomyces species) capable of effectively assimilating diverse substrates (acetate, glucose, and xylose). We characterized this new chassis by integrative 13C-metabolic flux analysis, metabolomic analysis, and RNA sequencing with large language model-based data processing. During acetate metabolism, L. tetrasporus demonstrated a highly active TCA cycle, rapid citrate and malate accumulation, strong NA
