Limbal relaxing incisions using a reference point and corneal topography for intraoperative identification of the steepest meridian.

PURPOSE: To examine the efficacy and safety of topography-based limbal relaxing incision (LRI) surgery. METHODS: Forty-four eyes of 36 consecutive patients who underwent cataract surgery more than 1 month previously, had refractive against-the-rule astigmatism of ≥2.00 diopters (D), and were scheduled to undergo LRI surgery were enrolled in the study. Patients were randomized into two groups-a topography-based LRI group (19 eyes of 14 patients) and a conventional LRI group (25 eyes of 22 patients). The topography-based LRI procedure comprised 3 steps: placing a mark on the cornea and conjunct

Expression of herpes simplex associated antigens in ocular tissues following induced herpetic anterior uveitis.

Timing and distribution of the expression of herpes simplex virus, type 1, associated antigen (HSV-1 AA) positive cells were studied immunohistochemically, following unilateral inoculation of HSV-1 into the anterior chamber of BALB/c mice. Inflammatory cells appeared in the corneal limbus, the anterior chamber angle, and Schlemm's canal of the inoculated eyes within 24 hours. HSV-AA positive cells developed in this period in the non-pigmented epithelium (which corresponds to the human pigmented epithelium) of the iris and the corneal endothelium; they also appeared in the ciliary body by the d

Inhibitor of DNA-Binding 3 Is a Novel Regulator of Limbal Epithelial Cell Migration Via the EphA2/Akt Signaling Pathway.

PURPOSE: Upon corneal injury, early and late transit amplifying (TA) cells, the progeny of epithelial stem cells, migrate to the site of the wound, which facilitates its closure. Understanding the targetable signals that guide such cell migration is essential for the development of novel wound-healing strategies. METHODS: Single-cell RNA sequencing (scRNA-seq) was conducted in wild-type cornea. To investigate the role of inhibitor of DNA binding 3 (ID3) in the limbal epithelium, bulk RNA-seq was conducted in limbal epithelial cells with ID3 knockdown. hTCEpi cells were transfected with small

Three-year outcomes of cultured limbal epithelial allografts in aniridia and Stevens-Johnson syndrome evaluated using the Clinical Outcome Assessment in Surgical Trials assessment tool.

Limbal stem cell deficiency (LSCD) is an eye disorder in which the stem cells responsible for forming the surface skin of the cornea are destroyed by disease. This results in pain, loss of vision, and a cosmetically unpleasant appearance. Many new treatments, including stem cell therapies, are emerging for the treatment of this condition, but assessment of these new technologies is severely hampered by the lack of biomarkers for this disease or validated tools for assessing its severity. The aims of this study were to design and test the reliability of a tool for grading LSCD, to define a set

Foxp3+ regulatory T cells reside within the corneal epithelium and co-localize with limbal stem cells.

In this study we investigated the presence of resident Foxp3+ regulatory T cells (Tregs) within the cornea and assessed the role of resident Tregs in corneal epithelial wound healing. Using a mouse model, we showed that in the steady state Foxp3+Tregs are either in close proximity or co-localize with ABCG2+ limbal stem cells. We also showed that these Tregs reside within the epithelial layer and not the corneal stroma. In addition, using a mouse model of mechanical injury, we demonstrated that depletion of Tregs from the cornea prior to corneal mechanical injury, using subconjunctival injectio
