Characterization of widespread canine leishmaniasis among wild carnivores from Spain.

Visceral Leishmaniasis (VL) is an emerging zoonotic parasitic disease caused by Leishmania infantum in Mediterranean countries, with sand flies (Phlebotomus spp.) as vectors and dogs as the main domestic reservoir. The role of wild carnivores in the epidemiology of leishmaniasis is still controversial. In order to determine the prevalence of natural infection with L. infantum in wild carnivores from Spain, we analyzed 217 samples by PCR and western blotting and used restriction fragment length polymorphism (RFLP) to compare the patterns present in wild carnivores with those of domestic dogs fr

Spatiotemporal dynamics and ecological determinants of cutaneous Leishmaniasis in Errachidia Province, southeastern Morocco: A 16-year municipality-level analysis with SARIMA forecasting.

Leishmaniasis is a vector-borne parasitic disease transmitted by female sandflies of the genus Phlebotomus and affects approximately 1.2 million people annually in more than 90 countries worldwide. Morocco remains one of the most affected countries in North Africa, where the disease continues to represent a major public health concern. This study aimed to analyze the spatial and temporal trends of leishmaniasis incidence across all municipalities of Errachidia Province between 2010 and 2025 and to assess the influence of ecological, demographic, and socio-economic factors on its distribution.

Development and application of species ID and insecticide resistance assays, for monitoring sand fly Leishmania vectors in the Mediterranean basin and in the Middle East.

BACKGROUND: Development of insecticide resistance (IR) in sand fly populations is an issue of public health concern, threatening leishmaniasis mitigation efforts by insecticide-based vector control. There is a major knowledge gap in the IR status of wild populations worldwide, possibly attributed to the unavailability of specialized tools, such as bioassay protocols, species baseline susceptibility to insecticides and molecular markers, to monitor such phenomena in sand flies. METHODOLOGY/PRINCIPAL FINDINGS: Sand fly populations from (semi-)rural regions of Greece, Turkey and Iran were sample

Canine leishmaniosis in Tunisia: Growing prevalence, larger zones of infection.

BACKGROUND: Discovered by Nicolle and Comte in 1908 in Tunisia, Leishmania infantum is an intracellular protozoan responsible for zoonotic canine leishmaniosis (CanL) and zoonotic human visceral leishmaniasis (HVL). It is endemic in several regions of the world, including Tunisia, with dogs considered as the main domestic reservoir. The geographic expansion of canine leishmaniosis (CanL) has been linked to global environmental changes that have affected the density and the distribution of its sand fly vectors. METHODOLOGY/PRINCIPAL FINDINGS: In this study, a cross-sectional epidemiological su

Quality by Design-Driven Process Development of Severe Fever With Thrombocytopenia Syndrome Vaccine.

Owing to the biological activity of the vaccine, the complicated production process, sterility, and uniformity of the product, the producing process of the vaccine is complicated and the product quality hard to control. In recent years, with the development of basic science such as cell biology, molecular biology, and metabolic engineering, bioprocess engineering research has developed rapidly. Therefore, U.S. Food and Drug Administration and European Medicines Agency conduct stringent control over the development of biomedical process engineering and product quality. This case study describes

Therapeutic effects of favipiravir against severe fever with thrombocytopenia syndrome virus infection in a lethal mouse model: Dose-efficacy studies upon oral administration.

Severe fever with thrombocytopenia syndrome (SFTS), caused by SFTS virus (SFTSV), is a viral hemorrhagic fever with a high case fatality rate. Favipiravir was reported to be effective in the treatment of SFTSV infection in vivo in type I interferon receptor knockout (IFNAR-/-) mice at treatment dosages of both 60 mg/kg/day and 300 mg/kg/day for a duration of 5 days. In this study, the efficacy of favipiravir at dosages of 120 mg/kg/day and 200 mg/kg/day against SFTSV infection in an IFNAR-/- mouse infection model was investigated. IFNAR-/- mice were subcutaneously infected with SFTSV at a 1.0

First evidence of intraclonal genetic exchange in trypanosomatids using two Leishmania infantum fluorescent transgenic clones.

BACKGROUND: The mode of reproduction in Leishmania spp has been argued to be essentially clonal. However, recent data (genetic analysis of populations and co-infections in sand flies) have proposed the existence of a non-obligate sexual cycle in the extracellular stage of the parasite within the sand fly vector. In this article we propose the existence of intraclonal genetic exchange in the natural vector of Leishmania infantum. METHODOLOGY/PRINCIPAL FINDINGS: We have developed transgenic L. infantum lines expressing drug resistance markers linked to green and red fluorescent reporters. We hy

Seroprevalence of antibodies to sandfly fever Sicilian virus in a sample population in Tuscany, Italy.

Toscana virus is the main phlebovirus circulating in Tuscany during the warm season, thus, a seroprevalence study was performed in the same area to estimate the antibody prevalence rates for sandfly fever Sicilian virus (SFSV) that is endemic in the Mediterranean countries. The low seroprevalence observed in this study shows that this virus does not play an important role in the etiology of febrile illness in central Italy.

Serological survey of Toscana virus infections in a high-risk population in Italy.

Toscana virus is the most important agent responsible for meningitis in central Italy. We report a serosurveillance study, using an immunoenzymatic assay, of 360 serum samples harvested from a high-risk population occupationally exposed to Toscana virus in two regions of Italy, Tuscany and Piedmont. The results indicates a seroprevalence of Toscana virus of 77.2% in the forestry workers, particularly in the Tuscany region. This fact is strictly correlated with the ecological niches specific for the survival of Toscana virus arthropod vector.

Insights into animal reservoir hosts for the circulation of Leishmania parasite in the sustainable phase of National Kala-azar Elimination Programme in India.

BACKGROUND: India, currently in the sustained phase of visceral leishmaniasis elimination, continues to face challenges due to reports of indigenous cases from emerging foci, detection of parasitic DNA in multiple sand fly species, and the potential involvement of animal reservoirs in disease transmission. Although Leishmania donovani has traditionally been regarded as anthroponotic, domestic animals may play an under-recognized role in maintaining transmission. METHODS: A multicentric study was conducted across seven districts in endemic (Bihar, Jharkhand, West Bengal) and emerging foci (Ker

Species Composition of Phlebotominae Sand Flies (Diptera: Psychodidae) in Mashhad City, an Endemic Focus of Anthroponotic Cutaneous Leishmaniasis, Northeastern Iran.

BACKGROUND: Phlebotomine sand flies are the primary vectors of cutaneous leishmaniasis. We conducted a field survey and comprehensive literature review to investigate the species composition and bionomics of sand flies in Mashhad, a major pilgrimage and tourist city in northeastern Iran. This location is critical due to its high population mobility, which can influence disease dynamics. METHODS: Sand flies were collected monthly from spring to winter 2022 using sticky paper traps at 108 sites across Mashhad's diverse ecological zones. Species were identified, and population densities were cal

Quantifying the Infectiousness of Post-Kala-Azar Dermal Leishmaniasis Toward Sand Flies.

BACKGROUND: On the Indian subcontinent, visceral leishmaniasis (VL) incidence is on track to reach elimination goals by 2020 in nearly all endemic districts. Although not included in official targets, previous data suggest post-kala-azar dermal leishmaniasis (PKDL) patients can act as an infection reservoir. METHODS: We conducted xenodiagnosis on 47 PKDL patients and 15 VL patients using laboratory-reared Phlebotomus argentipes. In direct xenodiagnosis, flies were allowed to feed on the patient's skin for 15 minutes. For indirect xenodiagnosis, flies were fed through a membrane on the patient
