Metabolic and photosynthetic consequences of blocking starch biosynthesis in the green alga Chlamydomonas reinhardtii sta6 mutant.

Upon nutrient deprivation, microalgae partition photosynthate into starch and lipids at the expense of protein synthesis and growth. We investigated the role of starch biosynthesis with respect to photosynthetic growth and carbon partitioning in the Chlamydomonas reinhardtii starchless mutant, sta6, which lacks ADP-glucose pyrophosphorylase. This mutant is unable to convert glucose-1-phosphate to ADP-glucose, the precursor of starch biosynthesis. During nutrient-replete culturing, sta6 does not re-direct metabolism to make more proteins or lipids, and accumulates 20% less biomass. The underlyi

Docosahexaenoic acid supplementation, vascular function and risk factors for cardiovascular disease: a randomized controlled trial in young adults.

BACKGROUND: A high consumption of omega-3 long-chain polyunsaturated fatty acids, and particularly docosahexaenoic acid (DHA), has been suggested to reduce the risk of cardiovascular disease (CVD). However, while DHA supplementation may have benefits for secondary prevention, few studies have investigated the role of DHA in the primary prevention of CVD. Here, we tested the hypothesis that DHA supplementation improves endothelial function and risk factors for CVD. METHODS AND RESULTS: Healthy volunteers (n=328), aged 18 to 37 years, were randomly assigned to 1.6 g DHA/day (from a microalgae s

Protective effect of Chlorella vulgaris on sub-chronic hepatic and testicular toxicity induced by acetamiprid in rats.

Acetamiprid (ACMP) is a widely used neonicotinoid insecticide that has been linked to oxidative stress-induced toxicity and physiological changes in mammals. This study investigates ACMP-induced hepatic and testicular toxicity in rats and the potential protective role of Chlorella vulgaris (C. vulgaris), a green microalga used as an antioxidant against ACMP-induced toxicity. Thirty-two male rats were allocated into four groups of eight each, as follows: (1) The CTRL group received only water; (2) The ACMP group received 21.7 mg/kg body weight (BW) of ACMP; (3) The C. vulgaris group received 15

Fenton and photo-Fenton strategies for sustainable pesticide decontamination. A review.

Herbicides, insecticides, and fungicides are synthetic pesticides widely used to control or destroy pests in agriculture. Their toxic persistence as residues in waters and soils is hazardous for plants and animals, necessitating their removal. This article presents a critical and comprehensive review of recent trends of Fenton and photo-Fenton strategies to destroy such pollutants from 2021 to May 2025. In synthetic solutions, homogeneous Fenton with H2O2/Fe2+ yielding oxidant •OH was limited to pH = 3.0 and by Fe(OH)3 sludge precipitation upon neutralization. Homogeneous Fenton-like processes

Nabumetone and flufenamic acid pose a serious risk to aquatic plants: A study with Chlamydomonas reinhardtii as a model organism.

The aquatic environment is constantly under threat due to the release of numerous pollutants. Among them, pharmaceuticals constitute a huge and diverse group. Non-steroidal anti-inflammatory drugs (NSAIDs) are increasingly found in water bodies, but knowledge about their potential toxicity is still low. In particular, there is a lack of information about their influences on aquatic plants and algae. We estimated the susceptibility of the microalgae Chlamydomonas reinhardtii to nabumetone (NBT) and flufenamic acid (FFA), focusing on photosynthesis. Due to the differences in the structures of th

Biosorption of rare earth elements from luminophores by G. sulphuraria (Cyanidiophytina, Rhodophyta).

Lanthanides are indispensable constituents of modern technologies and are often challenging to acquire from natural resources. The demand for REEs is so high that there is a clear need to develop efficient and eco-friendly recycling methods. In the present study, freeze-dried biomass of the polyextremophile Galdieria sulphuraria was employed to recover REEs from spent fluorescent lamps (FL) luminophores by pretreating the freeze-dried biomass with an acid solution to favour ion exchange and enhance the binding sites on the cell surface available for the metal ions. Lanthanides were extracted f

A 15-Year Retrospective Review of Ciguatera in the Madeira Islands (North-East Atlantic, Portugal).

The first ciguatera fish poisoning (CFP) in Portugal dates from 2008 when 11 people reported CFP symptoms after consuming a 30 kg amberjack caught around the Selvagens Islands (Madeira Archipelago). Since then, 49 human poisonings have been reported. The emergence of a new threat challenged scientists and regulators, as methods for toxic microalgae analyses and ciguatoxin (CTX) detection were not implemented. To minimise the risk of ciguatera, the Madeira Archipelago authorities interdicted fisheries in Selvagens Islands and banned the capture of amberjacks weighing more than 10 kg in the enti

Phosphate-regulated expression of the SARS-CoV-2 receptor-binding domain in the diatom Phaeodactylum tricornutum for pandemic diagnostics.

The worldwide COVID-19 pandemic caused by the SARS-CoV-2 betacoronavirus has highlighted the need for a synthetic biology approach to create reliable and scalable sources of viral antigen for uses in diagnostics, therapeutics and basic biomedical research. Here, we adapt plasmid-based systems in the eukaryotic microalgae Phaeodactylum tricornutum to develop an inducible overexpression system for SARS-CoV-2 proteins. Limiting phosphate and iron in growth media induced expression of the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein from the P. tricornutum HASP1 promoter in the wi

Dissolution rate and growth performance reveal struvite as a sustainable nutrient source to produce a diverse set of microbial protein.

To provide for the globally increasing demand for proteinaceous food, microbial protein (MP) has the potential to become an alternative food or feed source. Phosphorus (P), on the other hand, is a critical raw material whose global reserves are declining. Growing MP on recovered phosphorus, for instance, struvite obtained from wastewater treatment, is a promising MP production route that could supply protein-rich products while handling P scarcity. The aim of this study was to explore struvite dissolution kinetics in different MP media and characterize MP production with struvite as sole P-sou

Spirulina inhibits hepatocellular carcinoma through activating p53 and apoptosis and suppressing oxidative stress and angiogenesis.

BACKGROUND: Most hepatocellular carcinoma cases are diagnosed at late stages of the disease, which makes it the second cause of cancer mortality worldwide. For advanced-stage patients, chemotherapeutic drugs are the best treatment option; however, their adverse effects and high cost are still major obstacles for effective treatment. Spirulina microalga is a rich source of nutritional and bioactive elements and potential pharmaceuticals, which has an -proliferative effect against several cancer cell lines. It also has a prophylactic effect against the early stages of some cancer models, includi

Cu2+-assisted laccase from Trametes versicolor enhanced self-polyreaction of triclosan.

Laccase-mediated humification processes (L-MHPs) can be used to polymerize and transform phenolic pollutants in water. However, the mechanism on Cu2+ impacts the self-polymerization of multi-purpose antimicrobial agent triclosan during L-MHPs is less understood. Here, the influence of divalent metal ions (DMIs) on Trametes versicolor laccase activity was investigated. Particularly, the performance of Cu2+-assisted laccase in polymerizing and transforming triclosan was systematically characterized. Compared with DMI-free, the activity of laccase was obviously accelerated with Cu2+ present due t

Development of an LC-MS/MS method to simultaneously monitor maitotoxins and selected ciguatoxins in algal cultures and P-CTX-1B in fish.

Ciguatera fish poisoning is a serious human health issue that is highly localized to tropical and sub-tropical coastal areas, affecting many of the indigenous island communities intrinsically linked to reef systems for sustenance and trade. It is caused by the consumption of reef fish contaminated with ciguatoxins and is reported as the most common cause of non-bacterial food poisoning. The causative toxins bioaccumulate up the food web, from small herbivorous fish that graze on microalgae of the genus Gambierdiscus into the higher trophic level omnivorous and carnivorous fish predating on the

Diffusion-based process for carbon dioxide uptake and isoprene emission in gaseous/aqueous two-phase photobioreactors by photosynthetic microorganisms.

Photosynthesis for the generation of fuels and chemicals from cyanobacteria and microalgae offers the promise of a single host organism acting both as photocatalyst and processor, performing sunlight absorption and utilization, as well as CO(2) assimilation and conversion into product. However, there is a need to develop methods for generating, sequestering, and trapping such bio-products in an efficient and cost-effective manner that is suitable for industrial scale-up and exploitation. A sealed gaseous/aqueous two-phase photobioreactor was designed and applied for the photosynthetic generati

A nuclear CobW/WW-domain factor represses the CO2-concentrating mechanism in the green alga Chlamydomonas reinhardtii.

Microalgae induce a CO2-concentrating mechanism (CCM) to maintain photosynthesis when CO2 is limited. Because this system consumes a substantial portion of photosynthetically generated ATP, its suppression when CO2 levels rise is critical for energy balance, yet the underlying mechanism remains unclear. Here, we identify a nuclear repressor of the CCM in the green alga Chlamydomonas reinhardtii. A pull-down screen for interacting partners of the master activator CCM1/CIA5 revealed an uncharacterized protein that tightly associates with CCM1. This protein, CCM1-binding protein 1 (CBP1), combine

High-productive recovery and purification of superoxide dismutase from Tetraselmis chuii using integrated ultrasonication-aqueous two-phase system.

Superoxide dismutase (SOD) is the first line of antioxidant defense against reactive oxygen species, and it has found wide application in cosmetic and supplementary products. Thus, supplementation of exogenous SOD can restore the antioxidant defense system. Tetraselmis chuii is rich source of SOD, has low toxicity, exhibits high resistance toward culture conditions and grows rapidly, gaining significant attention. However, whole-cell SOD has poor bioavailability due to degradation of the SOD in gastrointestinal tract. Therefore, SOD bioseparation is needed for the nutraceutical/biopharmaceutic

Optimizing the algae-bacteria biofilm reactor for imidacloprid wastewater treatment: An evaluation of hydraulic retention times for enhanced efficiency and energy savings.

Recent observations have highlighted the rapidly growing prevalence of emerging contaminants such as Imidacloprid (IMI) within our environment. These insecticidal pollutants, coexisting with more traditional contaminants, have become predominant in aquatic systems, posing risks to both human and ecological well-being. Among the various wastewater treatment approaches tested, biofilm reactors are currently gaining prominence. In this study, we employed an Algae-Bacteria Biofilm Reactor (ABBR) to concurrently address both conventional and emergent contaminants, specifically IMI, over an extended

Physiological, biochemical and transcription effects of roxithromycin before and after phototransformation in Chlorella pyrenoidosa.

Photodegradation is an important transformation pathway for macrolide antibiotics (MCLs) in aquatic environments, but the ecotoxicity of MCLs after phototransformation has not been reported in detail. This study investigated the effects of roxithromycin (ROX) before and after phototransformation on the growth and physio-biochemical characteristics of Chlorella pyrenoidosa, and its toxicity were explored using transcriptomics analysis. The results showed that 2 mg/L ROX before phototransformation (T0 group) inhibited algae growth with inhibition rates of 53.06%, 54.17%, 47.26%, 31.27%, and 28.3

Effects of light quality and nutrient availability on accumulation of mycosporine-like amino acids in Gymnodinium catenatum (Dinophycea).

A Portuguese Gymnodinium catenatum Graham strain was studied for its ultraviolet (UV) photoprotective pigments. This strain presented high absorption in the UVA region, in particular in the near UVA region around 370nm, followed by the far-UVA region around 340nm. Absorption in the near-UVA increased when grown under fluorescent when compared to halogen light. This was even more relevant when grown under nutrient-limiting conditions, which even surpassed absorption in the blue region, closely resembling absorption in natural plankton assemblages reported in the literature.
