Neural correlates of blink suppression and the buildup of a natural bodily urge.

Neuroimaging studies have elucidated some of the underlying physiology of spontaneous and voluntary eye blinking; however, the neural networks involved in eye blink suppression remain poorly understood. Here we investigated blink suppression by analyzing fMRI data in a block design and event-related manner, and employed a novel hypothetical time-varying neural response model to detect brain activations associated with the buildup of urge. Blinks were found to activate visual cortices while our block design analysis revealed activations limited to the middle occipital gyri and deactivations in

Role of corticotropin-releasing hormone in the amygdala and bed nucleus of the stria terminalis in the behavioral, pain modulatory, and endocrine consequences of opiate withdrawal.

The extra-hypothalamic actions of corticotropin-releasing hormone (CRH) have been accorded an important role in coordinating responses to stressors and contributing to the consequences of drug abuse. Recent proposals suggest that CRH actions in the bed nucleus of the stria terminalis coordinate responses to tonic/unpredictable stressors whereas these actions in the central nucleus of the amygdala coordinate responses to phasic/predictable stressors. We used in situ hybridization histochemistry and site-specific microinjections of a CRH receptor antagonist to study the role of CRH in opiate wit

State-dependent stimulus control: cueing attributes of ethanol "hangover" in rats.

In Experiment 1, twelve Sprague-Dawley rats were trained in a two-choice food-reinforced drug discrimination task (10-min sessions) using the state-dependent interoceptive stimulus attributes of ethanol's (ETOH) delayed or rebound effects (EDE) versus "normal" basal homeostasis. Rats were injected with either 4 g/kg ETOH or equivalent volumes of saline (SAL) 18 hr before the sessions. Each rat was injected with an additional 1 ml/kg injection of SAL 15 min before the sessions. EDE training sessions were always followed by a "day off." SAL sessions were conducted between 36-96 hr after an EDE t

Non-invasive neuromodulation of sub-regions of the human insula differentially affect pain processing and heart-rate variability.

The insula is a portion of the cerebral cortex folded deep within the lateral sulcus covered by the overlying opercula of the inferior frontal lobe and superior portion of the temporal lobe. The insula has been parsed into sub-regions based upon cytoarchitectonics and structural and functional connectivity with multiple lines of evidence supporting specific roles for each of these sub-regions in pain processing and interoception. In the past, causal interrogation of the insula was only possible in patients with surgically implanted electrodes. Here, we leverage the high spatial resolution comb

Bridging dream and wake states: a new tool for consciousness research.

In this study, the Consciousness States Comparison Scale: REM Version (CSCS-REM) was developed as a self-report instrument to assess dream consciousness during REM sleep in comparison with waking consciousness. Existing instruments capture only limited aspects of phenomenology and provide few opportunities for systematic cross-state evaluation. By anchoring waking consciousness as a reference point, the CSCS-REM quantifies relative differences in phenomenological experience and serves as a tool for empirically testing the predictions of the continuity and discontinuity hypotheses in consciousn

A systematic review of associations between functional connectivity, mood and cognition in patients with irritable bowel syndrome.

Irritable bowel syndrome (IBS) affects 9.3%–35.5% of the population, with women at greater risk. It is often comorbid with anxiety, depression, and cognitive dysfunction. Understanding the neural mechanisms underlying these comorbidities is crucial for identifying IBS pathology and developing targeted treatments. Functional magnetic resonance imaging (fMRI) offers valuable insights into brain connectivity and psychopathology. This review evaluates research on the link between functional connectivity, mood, and cognition in IBS. We systematically searched Embase, PubMed, and PsycINFO for studie

Reinforcing and discriminative stimulus effects of 4-fluorobutyrfentanyl, 4-fluoroisobutyrfentanyl, and isobutyrfentanyl in male rats.

RATIONALE: 4-fluorobutyrfentanyl (4F-BF), 4-fluoroisobutyrfentanyl (4F-iBF), and isobutyrfentanyl (iBF) are isomers of butyrfentanyl. Despite these fentanyl analogs being directly involved in numerous fatal overdoses, their abuse liability profiles have yet to be fully elucidated. OBJECTIVES: This study evaluated the reinforcing effects of 4F-BF, 4F-iBF, and iBF on drug self-administration and their relationship to the interoceptive effects of heroin via drug discrimination. METHODS: Rats were trained to self-administer heroin (50 μg/kg/inf) under a fixed ratio (FR) 1 schedule. After 10 days

Identifying diagnostic biomarkers in functional motor disorders through multimodal behavioral, neurophysiological, and imaging assessment using explainable machine learning.

BACKGROUND: Functional motor disorders (FMDs) represent a frequent and disabling neurological condition. The lack of reliable diagnostic biomarkers and their heterogeneity might affect diagnosis. We identified multimodal biomarkers distinguishing FMDs from healthy controls (HCs) using machine-learning approaches. METHODS: In this multicenter cross-sectional study, consecutive adults with a clinically established FMDs diagnosis (n = 75, 74.7% female; mean age 44.20 ± 12.92) and age- and sex-matched HCs (n = 75; 58.6% female; 48.42 ± 11.67) were recruited. All participants underwent standardize

EEG microstate dynamics reveal progressive sensorimotor network dysfunction across levels of anxiety severity in drug-naïve major depressive disorder.

Anxiety symptoms are highly prevalent in major depressive disorder (MDD) and significantly influence clinical outcomes, yet neurophysiological mechanisms underlying anxiety severity remain poorly understood. To characterize neurophysiological alterations associated with different levels of anxiety severity in drug-naïve MDD, we used resting-state electroencephalogram (EEG) microstate analysis. Resting-state EEG was recorded from 113 MDD patients with varying anxiety levels (DDA) and 60 healthy controls (HC). Patients were stratified into low (DDLA), moderate (DDMA), and severe (DDSA) groups us

Resting-state functional connectivity related to empathy and sensory processing sensitivity.

INTRODUCTION: Sensory processing sensitivity (SPS) is a personality trait that has both positive features (SPS+) such as sensitivity to subtle cues, and negative features (SPS-) such as sensitivity to unpleasant stimuli or situations. Both SPS feature clusters are related to empathy. This study examined the neural correlates of SPS as it relates to empathy in the brain at rest. METHODS: In a sample of university students (N = 172), we identified links between SPS facets and multiple aspects of empathy measured via self-report. Subsequently, resting-state functional magnetic resonance imaging

The role of exercise in post-traumatic stress disorder: clinical evidence, sleep, cognition, and psychotherapy augmentation.

Post-traumatic stress disorder (PTSD) constitutes a profoundly incapacitating psychiatric disorder distinguished by the presence of intrusive recollections, heightened arousal, disruptions in sleep patterns, cognitive impairments, and diminished quality of life (QoL). Notwithstanding the advancements achieved in pharmacological treatments and trauma-oriented psychotherapeutic interventions, a considerable number of patients encounter persistent residual symptoms, underscoring the necessity for supplementary therapeutic modalities. This review scrutinizes the nascent role of physical exercise a

Dynamic Brain Activation and Connectivity in Elite Golfers During Distinct Golf Swing Phases: An fMRI Study.

Background/Purpose: Skilled motor performance depends on the action-observation networks (AONs), which supports the internal simulation of perceived movements. While expertise effects are well-documented in sports, neuroimaging evidence in golf is scarce, particularly on temporal dynamics across swing phases. This study examines how golf expertise modulates AON activation and functional connectivity during temporally distinct swing phases (pre-hitting vs. hitting) and assesses implications for predictive-coding models of motor skill. Methods: Fifty-seven participants (elite golfers: n = 28; co

Virtual Reality-Based Running Exposure to Target the Acute Urge to Be Physically Active in Anorexia Nervosa: A Case Series.

INTRODUCTION: Anorexia nervosa (AN) is a severe and complex eating disorder (ED), whose incidence has increased following the COVID-19 pandemic, straining specialized healthcare systems worldwide. Although patients often resort to compulsive exercise as a means of weight control, prolonged fasting intensifies the urge to move, leading to increased motor restlessness. Despite progress in understanding AN, interventions often overlook the crucial role that the urge to be physically active plays in reinforcing the ED itself. This case series tested the feasibility of a Virtual Reality-based Runni

Pre-exposure to eutylone attenuates its own aversive effects but has no impact on cocaine or MDMA: A possible role of eutylone's hybrid pharmacology.

Previous research has reported that pre-exposure to a variety of drugs of abuse can impact (reduce) the aversive effects of themselves and other abused compounds, often as a function of their shared pharmacological activity. In this context, the present series of studies investigated the effects of history with the synthetic cathinone eutylone on the aversive effects of cocaine, MDMA, and itself in adult, male Sprague-Dawley rats. Given eutylone's structural similarities with its parent compound methylone, it was predicted that its ability to attenuate cocaine- and MDMA-induced taste avoidance

Meditation Linked to Enhanced MRI Signal Intensity in the Pineal Gland and Reduced Predicted Brain Age.

Growing evidence demonstrates that meditation practice supports cognitive functions, including attention and interoceptive processing, and is associated with structural changes across cortical networks, including prefrontal regions and the insula. However, the extent of subcortical morphometric changes linked to meditation practice is less appreciated. A noteworthy candidate is the pineal gland, a key producer of melatonin, which regulates circadian rhythms that augment sleep-wake patterns and may also provide neuroprotective benefits to offset cognitive decline. Increased melatonin levels, as

Neural reactivity to a racial trauma cue in an African American woman with alcohol use disorder and PTSD: Case report.

Alcohol use disorder (AUD) and posttraumatic stress disorder (PTSD) are common co-occurring diagnoses that disproportionately affect racial minorities and women. Black/African American women are more likely to experience racial trauma, which compounds AUD+PTSD comorbidity. Limited research has investigated the effect of racial trauma on AUD+PTSD using functional magnetic resonance imaging (fMRI). We used a novel script-driven imagery procedure during MRI for racial trauma in a 26-year-old, self-identifying Black woman enrolled in a 12-week randomized clinical trial for individuals with AUD+PTS
