Identification of Wnt-5a Receptors Important in Diabetic and Non-Diabetic Corneal Epithelial Wound Healing.

PURPOSE: Persistent epithelial alterations such as delayed wound healing are a key feature of diabetic corneal disease. Previously, we reported that epigenetic changes in the diabetic cornea led to the suppression of Wnt-5a, and that addition of Wnt-5a accelerated wound healing. In this study, we set to determine which Wnt receptor(s) mediated Wnt-5a induced stimulation of diabetic corneal epithelial wound healing. METHODS: Human limbal epithelial cells (LECs) were isolated from postmortem diabetic and non-diabetic donor eyes for single-cell RNA sequencing (scRNA-seq) and DNA methylation anal

Expression of pluripotency and multipotency factors in human ocular surface tissues.

AIM: Mechanisms that control ocular surface stem cells (SCs) are unclear. Recent studies have shown that several adult SCs express pluripotency markers. Our objective was to analyze the expression of key molecules of pluripotency in human ocular surface tissues as well as in cultivated limbal epithelium. METHODS: Four samples of human corneal, limbal and on amniotic membrane cultivated limbal epithelium (HLEC-AM), as well as bulbar and fornical conjunctiva were analyzed. Human embryonic stem (ES) cells and human umbilical vein endothelial cells served as controls. Expression of corneal epithe

Cultivated human conjunctival epithelial transplantation for total limbal stem cell deficiency.

PURPOSE: To determine the feasibility of cultivated conjunctiva as a viable epithelial sheet for transplantation and corneal resurfacing in eyes with limbal stem cell deficiency (LSCD). METHODS: Human corneal epithelial (HCE) and human conjunctival epithelial (HCjE) cells were cultivated on human amniotic membrane (AM) to confluence and then air lifted to allow further stratification and differentiation. Denuded AM and cultivated HCE and cultivated HCjE cells were then transplanted into 18 eyes of rabbits with induced LSCD. The cultivated and engrafted epithelia were examined by transmission

Topical Rho kinase inhibitor rescues TRPV4 deficiency-induced an impairment of corneal epithelial wound healing by enhancing limbal stem cell proliferation.

PURPOSE: Transient receptor potential vanilloid 4 (TRPV4), a mechanosensitive cation channel, is essential for cell proliferation in peripheral cornea during epithelial repair. We examined whether topical administration of a Rho kinase inhibitor, Y-27632, rescues impaired epithelial healing in TRPV4 knockout (KO) mice. METHODS: TRPV4 KO mice underwent 2 mm corneal epithelial debridement and were topically treated every 6 h with Y-27632 or PBS. Epithelial closure was monitored by fluorescein staining. Cell proliferation was evaluated by BrdU incorporation. Corneal sensitivity was assessed with
