Molecular characterization of the T cell costimulatory receptors CD28 and CTLA4 in the European sea bass.

For the activation of T cells, it is necessary the specific recognition of the peptide by the T cell receptors (TCR) in the surface of antigen-presenting cells (APCs) and additional signals delivered by costimulatory receptors. In fish, knowledge about the presence of these costimulatory signals is limited and functional evidence almost absent. Thus, in this study, we have identified the stimulatory CD28 and the inhibitory cytotoxic T-lymphocyte-associated protein 4 (CTLA4) coreceptors in the European sea bass (Dicentrarchus labrax), and evaluated their transcription. In parallel, the transcri

Resistance gene identification from Larimichthys crocea with machine learning techniques.

The research on resistance genes (R-gene) plays a vital role in bioinformatics as it has the capability of coping with adverse changes in the external environment, which can form the corresponding resistance protein by transcription and translation. It is meaningful to identify and predict R-gene of Larimichthys crocea (L.Crocea). It is friendly for breeding and the marine environment as well. Large amounts of L.Crocea's immune mechanisms have been explored by biological methods. However, much about them is still unclear. In order to break the limited understanding of the L.Crocea's immune mec

Two HSP90 genes in mandarin fish Siniperca chuatsi: identification, characterization and their specific expression profiles during embryogenesis and under stresses.

HSP90 plays important roles in multiple cellular stress responses. Here, two cytoplasmic HSP90 isoforms, ScHSP90α and ScHSP90β, were identified from Siniperca chuatsi. Their cDNA and gDNA structures, amino acid sequence features, and sequence identities and phylogenetic analysis with other species were described. Their expression profiles during embryonic development in different tissues and under stressful conditions were analyzed using real-time quantitative PCR. During embryogenesis, transcripts of both genes were detected at low levels during the early developmental stages and were up-regu

Identification and characterization of three TLR1 subfamily members from the orange-spotted grouper, Epinephelus coioides.

Toll-like receptors (TLRs), which play important roles in host defense against pathogen infection, are the most intensively studied pattern recognition receptors (PRRs). In this study, we identified three novel TLR1 subfamily members, including TLR1 (EcTLR1b), TLR2 (EcTLR2b) and TLR14 (EcTLR14), from the orange-spotted grouper (Epinephelus coioides). EcTLR1b and EcTLR2b displayed low sequence identity with the previously reported grouper TLR1 (EcTLR1a) and TLR2 (EcTLR2a), respectively. The open reading frames (ORFs) of EcTLR1b, EcTLR2b and EcTLR14 contain 2484 bp, 2394 bp and 2640 bp, which en

A novel approach to AVT and IT studies in fish brain and pituitary: in vitro perfusion technique.

The study was designed to develop a new procedure for perfusion of brain and pituitary explants collected from three-spined stickleback (Gasterosteus aculeatus) and round goby (Neogobius melanostomus). The procedure was elaborated for studies of arginine vasotocin (AVT) and isotocin (IT) release from explants of both species. AVT and IT, analogs of mammalian vasopressin and oxytocin, are neurohormones produced in hypothalamus and released in neurohypophysis of Teleostei. Both nonapeptides are used as biomarkers of fish well being. Three perfusion sets were applied to test the method of medium

Excitatory action of GABA in the terminal nerve gonadotropin-releasing hormone neurons.

The terminal nerve (TN)-gonadotropin-releasing hormone (GnRH) neurons have been suggested to function as a neuromodulatory system that regulates the motivational and arousal state of the animal and have served as a model system for the study of GnRH neuron physiology. To investigate the synaptic control of the TN-GnRH neurons, we analyzed electrophysiologically the effect of GABA on the TN-GnRH neurons. GABA generally hyperpolarizes most of the neurons in the adult brain by activating GABA(A) receptors while the activation of GABA(A) receptors depolarizes some specific neurons in the mature br

A novel antimicrobial peptide Larimicin78-102 from large yellow croaker (Larimichthys crocea) shows potent antibacterial activity in vitro and enhances resistance to vibrio fluvialis infection in vivo.

Antimicrobial peptides (AMPs) are considered a key component of innate immunity, playing a vital role in host defense. In the study, a novel functional gene, named Larimicin, was identified from large yellow croaker Larimichthys crocea. The Larimicin gene was widely distributed in multiple tissues of healthy L. crocea and was significantly induced in the liver after Vibrio alginolyticus or Vibrio parahaemolyticus infection. Larimicin78-102, a truncated peptide derived from Larimicin, showed broad-spectrum antimicrobial activity and a binding affinity with LPS. It exhibited effective bactericid

Characterization and functional analysis of integrin αV in Sebastes schlegelii: Implications for apoptosis, adhesion, and migration in intestinal cells.

Integrins, as essential cell adhesion molecules, consist of an α subunit and a β subunit that interact with extracellular matrix proteins and cell surface ligands to mediate cellular adhesion and signaling. The integrin αV subfamily is widely expressed on the cell surface and plays a critical role in regulating cell growth, apoptosis, and various cellular processes. To explore the function of integrin αV in teleosts, we retrieved the integrin αV (SsITGαV) sequences from the Sebastes schlegelii genome and assessed the tissue expression and response to Edwardsiella tarda stimuli of SsITGαV. Accacoelium contortum (Trematoda: Accacoeliidae) a trematode living as a monogenean: morphological and pathological implications.

BACKGROUND: Accacoelium contortum (Rudolphi, 1819) Monticelli, 1893 is a frequent but poorly known trematode found on gills, pharynx and digestive tract of the ocean sunfish Mola mola (L.). Although the morphology of A. contortum agrees with that of a typical endoparasitic trematode, with two relatively small suckers and no large holdfasts, this parasite is normally ectoparasitic. The main objective of this paper is to explore this peculiar host-parasite relationship. METHODS: A total of 106 ocean sunfish were examined for the presence of A. contortum. The oropharyngeal chamber (gills and pha

Transcriptomic Analysis Revealed the Regulatory Mechanisms of Oocyte Maturation and Hydration in Orange-Spotted Grouper (Epinephelus coioides).

Oocyte maturation and hydration are regulated by a complex interplay of various hormones and local factors. We have investigated the morphological changes of follicles and serum steroid levels during the HCG (human choionic gonadotophin)-induced oocyte maturation in the orange-spotted grouper. For the first time, a large-scale transcriptomic analysis of follicles during the maturation has been conducted in a fish species which produce pelagic oocytes. Eight cDNA libraries of follicle samples, from full-grown immature follicles to mature follicles, were constructed. A total of 402,530,284 high-

Molecular identification and expression analysis of TAB1 from orange-spotted grouper (Epinephelus coioides).

Transforming growth factor-β activated kinase 1 (TAK1) is a crucial signal transducer in multiple signaling pathways. TAK1 binds TAB1, TAB2, and TAB3, which act as activators and adaptors that specifically regulate the activation of TAK1. To date, the role of TABs is largely unknown in fish. In the present study, a TAB1 cDNA sequence was identified in grouper (Epinephelus coioides), and designated EcTAB1. The full-length open reading frame of EcTAB1 is 1, 521 bp; it encodes 506 amino acids that contains an N-terminal PP2C domain. Many important functional sites in mammalian TAB1 were conserved

Cloning, characterisation, and expression analysis of the cathepsin D gene from rock bream (Oplegnathus fasciatus).

Cathepsins are lysosomal cysteine proteases belonging to the papain family, members of which play important roles in normal metabolism for the maintenance of cellular homeostasis. Rock bream (Oplegnathus fasciatus) cathepsin D (RbCTSD) cDNAs were identified by expressed sequence tag analysis of a lipopolysaccharide-stimulated rock bream liver cDNA library. The full-length RbCTSD cDNA (1644 bp) contained an open reading frame of 1191 bp encoding 396 amino acids. Alignment analysis revealed that the active sites and N-glycosylation sites of the deduced protein were well conserved. Phylogenetic a

Molecular cloning of leucocyte cell-derived chemotaxin-2 gene in croceine croaker (Pseudosciaena crocea).

Leucocyte cell-derived chemotaxin-2 (LECT2) was originally demonstrated to have a chemotactic activity against human neutrophils in vitro. Current evidence suggests that LECT2 may be a multifunctional protein involved in cell growth, differentiation and autoimmune. A full-length cDNA clone of the LECT2 gene, 595bp in size, was isolated from the fish croceine croaker (Pseudosciaena crocea). It's 3'-UTR was much shorter (112nts) than that of trout LECT2 gene (210nts). Its deduced amino acid sequence of 151 residues had 39.7-75.5% identity to that of other animals.
