Polystyrene microparticles can affect the health status of freshwater fish - Threat of oral microplastics intake.

Plastic waste pollution is considered one of the biggest problems facing our planet. The production and use of these materials has led to huge amounts of plastic waste entering the aquatic environment and affecting aquatic life. In our experiment, the effect of polystyrene microparticles (PS-MPs; 52.5 ± 11.5 μm) on individual juvenile rainbow trout (Oncorhynchus mykiss) was tested at three different dietary concentrations of 0.5, 2 and 5 % for six weeks. At the end of the experiment, various health parameters of exposed organisms were compared with the control group. The haematological profile

New insights on in vitro biotransformation of anticoagulant rodenticides in fish.

The assessment of the bioaccumulation potential of chemicals is an essential and mandatory part of their regulatory environmental risk and hazard assessment. So far, in vitro data on fish metabolism is rarely available for biocidal active substances such as anticoagulant rodenticides. In this case study we present in vitro biotransformation rates of eight biocidal and one pharmaceutical anticoagulants in rainbow trout (Oncorhynchus mykiss) liver subcellular S9 fraction (RT-S9) determined following the Organisation for Economic Co-operation and Development test guideline 319B method at two diff

Frustrative reward omission increases aggressive behaviour of inferior fighters.

Animals use aggressive behaviour to gain access to resources, and individuals adjust their behaviour relative to resource value and own resource holding potential (RHP). Normally, smaller individuals have inferior fighting abilities compared with larger conspecifics. Affective and cognitive processes can alter contest dynamics, but the interaction between such effects and that of differing RHPs has not been adjudged. We investigated effects of omission of expected reward (OER) on competing individuals with contrasting RHPs. Small and large rainbow trout (Oncorhynchus mykiss) were conditioned t

Experimentally induced marine flexibacteriosis in Atlantic salmon smolts Salmo salar. II. Pathology.

The fish disease marine flexibacteriosis is characterised by necrotic lesions on the body, head, fins, and occasionally gills, with erosive lesions on the external surface as the prominent clinical sign. In Australia, the main species affected are Atlantic salmon Salmo salar and rainbow trout Oncorhynchus mykiss in sea-cage culture in Tasmania. Using a dose-dependent trial to determine pathology, 2 forms of the disease were noted in Atlantic salmon. The acute form occurs within 2 to 3 d after inoculation at high doses (1 x 10(8) cells ml(-1)) and is characterised by the disintegration of the e

A matter of potential concern: natural organic matter alters the electrical properties of fish gills.

Natural organic matter (NOM) is an important constituent of aquatic environments; however, its influence on aquatic biota remains poorly studied. In the current study, NOM was isolated from nine different sites in southern Ontario, Canada, by the on-site treatment of water by reverse osmosis, followed by cation exchange. NOM from each site was reconstituted to 10 mg of C/L and pH 7.0 and exposed to either adult rainbow trout implanted with indwelling catheters or to in vitro primary cultures of the gill epithelium grown on semipermeable membranes. In both the in vivo and in vitro preparations,

Toxicity of Clostridium botulinum type E neurotoxin to Great Lakes fish: implications for avian botulism.

Since 1999, large-scale mortalities of fish-eating birds have been observed on the Great Lakes, and more specifically on Lake Erie. Type E botulism has been established as the primary cause of death. The mechanism of type E botulism exposure in fish-eating birds is unclear. Given that these birds are thought to eat live fish exclusively, it seems likely that their prey play a key role in the process, but the role of fish as potential transport vectors of botulinum neurotoxin type E (BoNT/E) to birds has not been adequately investigated. Between June 2003 and April 2004 a methodological model f

Increasing Risk Factors for Imported and Domestic Gnathostomiasis in the United States.

Gnathostomiasis is a foodborne zoonotic helminthic infection, commonly described in Asia and Latin America, which may follow the consumption of raw fish, eels, amphibians, and reptiles infected with muscle-encysted larvae of Gnathostoma species nematodes. After an inoculum of as little as one infective larva and an incubation period of months to years, most infections are characterized by intermittent migratory swellings due to subdermal larval migration. Less commonly, larval migration to the central nervous system may result in radiculomyelopathy or eosinophilic meningoencephalitis with high

The contribution of polymer additives to microplastic toxicity: A long-term study with Oncorhynchus mykiss exposed to polystyrene microparticles with different hexabromocyclododecane content.

Although microplastic toxicity in fish has been extensively studied, the role of polymer additives remains insufficiently understood. This study investigated the combined effects of polystyrene (PS) microplastics and the brominated flame retardant hexabromocyclododecane (HBCD) on the health of rainbow trout (Oncorhynchus mykiss) following six weeks of dietary exposure. Fish were exposed to PS particles with varying HBCD content (0.0 mg/g - PS-HBCDfree, 0.2 mg/g - PS-HBCDlow, 1.0 mg/g - PS-HBCDhigh) or to HBCD alone (HBCDonly). The combination of PS and HBCD induced the most pronounced biologic

The Mucosal Immune System of Teleost Fish.

Teleost fish possess an adaptive immune system associated with each of their mucosal body surfaces. Evidence obtained from mucosal vaccination and mucosal infection studies reveal that adaptive immune responses take place at the different mucosal surfaces of teleost. The main mucosa-associated lymphoid tissues (MALT) of teleosts are the gut-associated lymphoid tissue (GALT), skin-associated lymphoid tissue (SALT), the gill-associated lymphoid tissue (GIALT) and the recently discovered nasopharynx-associated lymphoid tissue (NALT). Teleost MALT includes diffuse B cells and T cells with specific

NASA/American Cancer Society High-Resolution Flow Cytometry Project-I.

BACKGROUND: The NASA/American Cancer Society (ACS) flow cytometer can simultaneously analyze the electronic nuclear volume (ENV) and DNA content of cells. This study describes the schematics, resolution, reproducibility, and sensitivity of biological standards analyzed on this unit. METHODS: Calibrated beads and biological standards (lymphocytes, trout erythrocytes [TRBC], calf thymocytes, and tumor cells) were analyzed for ENV versus DNA content. Parallel data (forward scatter versus DNA) from a conventional flow cytometer were obtained. RESULTS: ENV linearity studies yielded an R value of

cDNA sequence, ligand biding, and regulation of galanin/GMAP in mouse brain.

The cDNA encoding the 29 amino acid-long neuropeptide galanin and its flanking peptide galanin message associated peptide (GMAP), has been cloned and sequenced from mouse hypothalamic cDNA. The primary sequence of mouse galanin is GWTLNSAGYLLGPHAIDNHRSFSDKHGLT, followed by an amidation signal GKR. There are now 12 galanin sequences known: human, porcine, dog, rat, bovine, chicken, sheep, alligator, bowfin, dogfish, trout and mouse. The N-terminal 14 amino acids are identical in all of these species and the whole primary sequence of mouse galanin is identical to that of rate galanin. The mouse

Development of a Novel Virus-Like Particle-Based Vaccine Against PRV-1 Suitable for DIVA Strategies.

Piscine orthoreovirus genotype 1 (PRV-1) is an emerging viral pathogen in salmon aquaculture that causes Heart and Skeletal Muscle Inflammation (HSMI), with high prevalence in salmon-producing countries such as Chile. A significant obstacle in PRV-1 vaccine development is the inability to culture the virus in vitro, which limits the scalability and production of traditional inactivated or DNA-based vaccine strategies. This study describes the development of a novel virus-like particle (VLP)-based vaccine against PRV-1. Recombinant VLP were produced by co-expressing the six structural proteins

Nephrocalcinosis in farmed salmonids: diagnostic challenges associated with low performance and sporadic mortality.

Disease conditions that involve multiple predisposing or contributing factors, or manifest as low performance and/or low-level mortality, can pose a diagnostic challenge that requires an interdisciplinary approach. Reaching a diagnosis may also be limited by a lack of available clinical profile parameter reference ranges to discriminate healthy fish from those affected by specific disease conditions. Here, we describe our experience investigating poorly performing rainbow trout (Oncorhynchus mykiss) in an intensive recirculation aquaculture, where reaching a final diagnosis of nephrocalcinosis

Isolation, identification, virulence potential and genomic features of Tenacibaculum piscium isolates recovered from Chilean salmonids.

Tenacibaculum piscium, a gram-negative bacterium isolated from the skin ulcers of sea-farmed fish, has only been described in Norway. In the present study, we examined 16 Chilean Tenacibaculum isolates recovered from different organs in moribund and dead Atlantic salmon (Salmo salar), Rainbow trout (Oncorhynchus mykiss) and Coho salmon (Oncorhynchus kisutch) cultured at different fish farms between 2014 and 2018. The present study applied biochemical, phenotypic, fatty acid and whole-genome sequence-based analyses to confirm the taxonomic status of the Chilean isolates. The obtained results ar

Processing plant and machinery sanitation and hygiene practices associate with Listeria monocytogenes occurrence in ready-to-eat fish products.

Listeria monocytogenes causes the foodborne illness listeriosis, which exhibits high fatality among people in risk groups. The incidence of listeriosis has increased in Europe, which raises concerns about L. monocytogenes occurrence in foodstuffs. Ready-to-eat seafood products are considered particularly risky vehicles. Poor hygiene at processing facilities predisposes them to L. monocytogenes contamination, which can be controlled by stringent self-checking system measures. We examined the association of fish-processing plant operational and hygiene practices with the occurrence of L. monocyt

Viral haemorrhagic septicaemia virus (VHSV Id) infections are detected more consistently using syndromic vs. active surveillance.

The eradication of viral haemorrhagic septicaemia virus (VHSV Id) from Finnish brackish-water rainbow trout Oncorhynchus mykiss farms located in the restriction zone in the Province of Åland, Baltic Sea, failed several times in the 2000s. The official surveillance programme was often unable to find VHSV-positive populations, leading to the misbelief in the fish farming industry that virus eradication could be achieved. The ability of 3 other surveillance programmes to detect infected fish populations was compared with the official programme. One programme involved syndromic surveillance based

Chronic toxicity of heavy fuel oils to fish embryos using multiple exposure scenarios.

The chronic toxicity to rainbow trout (Oncorhynchus mykiss) embryos of heavy fuel oil (HFO) 6303, weathered HFO 6303, HFO 7102, and medium South American (MESA) crude oil was assessed by different exposure regimes. These included water accommodated fractions (WAF; water in contact with floating oil), chemically enhanced WAF (CEWAF; oil dispersed with Corexit 9500), and effluent from columns of gravel coated with stranded oil. Heavy fuel oil WAF was nontoxic and did not contain detectable concentrations of hydrocarbons, likely because the high density and viscosity of HFO prevented droplet form

Salt in the wound: consequences of changing salinities on marine disease.

Salinity plays an important role in the physiology of marine organisms and their pathogens. As the climate continues to change, marine and estuarine salinities may become more extreme and irregular, potentially altering host-pathogen interactions. In this review, we explore and highlight the role of salinity on a range of pathosystems, outlining impacts to the host, pathogen and their interactions. We present fisheries and aquaculture-focused case studies, including behavioural preferences for freshwater in sea louse-infested trout and the use of freshwater as a delouser in salmon aquaculture,

A Nutritional Bioenergetic Model for Farmed Fish: Effects of Food Composition on Growth, Oxygen Consumption and Waste Production.

The study of flow and transformation of energy and nutrients via mathematical modelling provides an in silico tool approach for designing scientific experiments, improving precision in aquaculture production and reducing the need for experimental animals. The proposed nutritional bioenergetics model is based on the dynamic energy budget (DEB) theory, a mechanistic framework to study individual metabolism. The model is an extension of the typical DEB models in that it includes a digestion module where the protein and non-protein food components contribute to assimilation via the concept of a sy

The application of Global Burden of Animal Diseases methodology to aquatic animal production.

The Global Burden of Animal Diseases (GBADs) programme's key objective â€" to provide a systematic approach to determine the burden of animal disease â€" is as relevant to aquatic as to terrestrial animal production systems. However, to date GBADs methods have mainly been applied to terrestrial animal production systems. The challenges in applying GBADs methods, notably the Animal Health Loss Envelope (AHLE), vary considerably by production system. The authors demonstrate how the AHLE can be calculated for rainbow trout production in England and Wales and acknowledge that its application to ot
