Synthetic microbial community to enhance the fermentation and feed valorization of pineapple waste.

Global livestock industry is confronted with severe feed-supply pressure, and valorizing agro-industrial by-products has become a promising way to alleviate feed resource shortages. Large quantities of pineapple waste are produced annually during pineapple processing, but antinutritional factors greatly limit its direct application in pig diets. Pineapple waste (PW) is a promising agro-industrial by-product for feed production; however, its high levels of antinutritional factors and structural carbohydrates limit its utilization in monogastric animals. This study aimed to develop a synthetic m

Chrysin Ameliorates Valproic Acid Induced Autism Spectrum Behavioral Phenotypes Through Corticohippocampal Histone Deacetylase and Translational Modulation of SHANK3 Expression in Juvenile Rats.

Autism spectrum disorder (ASD) is a neurodevelopmental disorder marked by repetitive behaviors, social deficits, and comorbid phenotypes, with rising prevalence. Its unclear pathogenesis and symptom heterogeneity hinder therapy development. Chrysin, a flavone from bee products and plants, shows diverse biological effects but limited ASD studies. Therefore, this study examines chrysin's impact on ASD behaviors and comorbidities. Pregnant Wistar rats received 600 mg/kg valproic acid (VPA) on Embryonic day (ED) 12.5 intraperitoneally to induce ASD phenotypes. Neurodevelopmental milestones were ev

Apigenin accelerates fracture healing by enhancing endochondral differentiation and inflammation resolution via targeting NF-κB signaling.

BACKGROUND: Fractures are common yet lack effective pharmacological treatments. Apigenin, a natural flavone, has anti-inflammatory and pro-osteogenic effects. Here, we examined its impact on fracture healing-a multistep process involving hematoma formation, soft callus formation (endochondral differentiation), hard callus formation (osteogenic differentiation), and bone remodeling-and investigated the underlying mechanisms. METHODS: Femoral fracture and calvarial injury models were established to evaluate the effects of apigenin. Local apigenin treatment was initiated at postoperative time po

Integrated network pharmacology and UHPLC-Q-TOF LC-MS/MS phytochemical profiling demonstrate tilianin-mediated restoration of PDE10A/cAMP/PKA/CREB signaling in an MK-801-induced schizophrenia-like model.

BACKGROUND: Schizophrenia is a complex neuropsychiatric disorder characterized by impairments in cognition, perception, and social behavior. Current antipsychotic medications primarily target dopaminergic signaling but often exhibit limited efficacy in treating negative and cognitive symptoms. Moreover, long-term antipsychotic therapy is frequently associated with clinically relevant adverse effects, including metabolic syndrome and extrapyramidal symptoms (EPS). Phosphodiesterase 10A (PDE10A) has emerged as a promising non-dopaminergic therapeutic target involved in regulating cyclic nucleoti

Apigenin enhances Nrf2-induced chaperone-mediated autophagy and mitigates α-synuclein pathology: Implications for Parkinson's disease therapy.

BACKGROUND: The aggregation of α-synuclein (SNCA) in dopaminergic neurons of the substantia nigra is a key factor in the pathogenesis of Parkinson's disease (PD). Despite years of drug discovery efforts targeting SNCA aggregation, no disease-modifying drugs have been approved to date. The failure of numerous clinical trials can be attributed, at least in part, to the difficulty in identifying potent compounds during preclinical investigations. OBJECTIVE: Establish a screening approach based on molecular docking and autophagic flux detection to identify natural compounds from new perspectives

Synergistic effect of morin and paclitaxel impedes cell proliferation through PI3K/AKT/STAT3 signaling axis inhibition in gastric cancer.

Gastric cancer (GC) is one of the most prevalent malignant cancers, with currently unsuccessful treatment strategies for patients. Increased PI3K/AKT/STAT-3 pathway activity has been observed in patients with GC. Morin (MRN), which is a flavonoid, exhibits significant anticancer activity by inhibiting the PI3K/AKT signaling pathway. However, monotherapy with MRN has faced challenges due to poor bioavailability and rapid elimination. This study investigated the combined effects of MRN and paclitaxel (PTX) on apoptosis induction and their molecular mechanisms in GC cells (HGT-1). After 24 h of M

Nobiletin Alleviates Npy1r-Mediated Insulin Secretion Deficiency of Islet β-Cells via the Clock-Modulatory Signaling.

Current research indicates that insulin secretion deficiency in β-cells contributes to Type 2 diabetes mellitus (T2DM), which is associated with neuropeptide Y1 receptor (Npy1r) overexpression from neuropeptide Y (NPY) system dysregulation. To date, limited literature has explored nobiletin (NOB) as a circadian modulator for restoring β-cell function through Npy1r regulation. This study investigates NOB's stimulatory effects on insulin secretion via Npy1r and clock-modulatory signaling to elucidate its underlying mechanism. The findings demonstrated that NOB ameliorated hyperglycemia and promo

Nobiletin Reduces LPS-Induced Neuroinflammation through TLR4/MyD88/NF-κB and Oxidative Stress via Nrf2/HO-1 Signaling in Human Microglial HMC3 Cells.

Neuroinflammation and oxidative stress (OS) are the major contributors to the onset and progression of neurodegenerative diseases (NDs), where microglial activation and dysregulated inflammatory signaling exacerbate neuronal injury. Nobiletin (NOB), a polymethoxylated flavonoid abundant in citrus fruits, has been reported to possess excellent bioactivities; however, its effects in combating inflammation and OS in human microglial cells (HMC3) have not been comprehensively examined. In this study, we investigated the effects of NOB on lipopolysaccharide (LPS)-induced inflammatory and oxidative

Negletein as a multifaceted therapeutic approach in overcoming MRSA-induced pneumonia and septic arthritis amidst antimicrobial resistance.

Antimicrobial resistance in Staphylococcus aureus, especially methicillin resistant strains, contributes to severe pneumonia and septic arthritis and undermines the efficacy of standard antibiotics. We explore an antivirulence approach that targets MgrA, a global transcriptional regulator of pathogenic programs and resistance traits, and identify negletein as a small molecule that engages this target. At efficacious exposures negletein is nonbactericidal, separating virulence control from growth and with the potential to reduce selection pressure compared with bactericidal strategies. Thermal

Acacetin targets STING to alleviate the destabilization of the medial meniscus-induced osteoarthritis in mice.

Osteoarthritis (OA) is a common joint disorder affecting about 7% of the global population, primarily characterized by the gradual loss of articular cartilage. This degeneration results from local inflammation, matrix depletion, and direct cartilage damage. A critical element in this process is the activation of the stimulator of the interferon genes (STING) pathway. Emerging evidence highlights its potential as a therapeutic target, with natural products showing promise as inhibitors. Our study centers on Acacetin, a basic unit of polyketides known for its anti-inflammatory properties. Prior

7,8-Dihydroxyflavone leads to survival of cultured embryonic motoneurons by activating intracellular signaling pathways.

Brain-derived neurotrophic factor (BDNF), a member of the neurotrophin family and a ligand for the tropomyosin-receptor kinase B (TrkB), mediates neuronal survival, differentiation, and synaptic plasticity. However, BDNF is not used to treat neurodegenerative diseases because of its poor pharmacokinetic profile, side effects, and absence of survival properties in clinical trials. Consequently, alternative approaches such as TrkB receptor agonist application are gaining importance. 7,8-Dihydroxyflavone (7,8-DHF), a member of the flavonoid family, has been described as a robust TrkB receptor ago
